    Notes: Summary Patients with AML in complete remission (CR) are immunosuppressed; remission lymphocytes of at least a part of the patients are able to recognize autologous leukemic blasts; the CR represents a minimal residual disease. Thus, important conditions for a potentially successful active immunotherapy are given. Results of most earlier randomized trials on immunotherapy revealed some, but limited benefit with respect to survival. Encouraging effects, however, have been achieved by immunizing CR patients with high-dose neuraminidase-treated allogeneic blasts. Because these data have not been confirmed until now we recently initiated a randomized study utilizing identical treatment protocols. The preliminary results of the ongoing study still do not allow to draw any firm conclusions. Specific monoclonal antibodies or autologous cytotoxic T-cells may be useful tools for future immunotherapy of AML.

    Notes: Summary Lysostaphin, a staphylococcus-derived staphylocidal substance, has widely been used in assays of granulocyte phagocytic and bactericidal capability. It rapidly kills extracellular bacteria. Thus, a separate determination of intracellular surviving bacteria can be performed. One prerequisite for this approach is the safe inactivation of lysostaphin (usually brought about by trypsin) before the intracellular bacteria are externalized for plating. This inactivation has been found by others to be incomplete. Data are presented demonstrating a safe inactivation of lysostaphin by trypsin, if the pH value is maintained within the alkaline range. A low variation of results is obtained by plotting the total number of bacteria killed per incubate vs the logarithm of initial bacterial inoculum or of the intracellular surviving bacteria, leading to linear regression lines. The variation of the results increases greatly for initial bacteria/granulocyte proportions of 〉 5/1. The results obtained for two differentSt. aureus strains are significantly different. Dexamethasone pretreatment (12 mg p.o. within 8 h) had no detectable influence, when fresh blood was assayed, while blood storage at room temperature for 12 h (without dexamethasone pretreatment) led to a significant functional impairment, mainly of bactericidal capability when analyzed in a pairwise fashion. A major limitation of this kind of assays is that killed bacteria cannot be determined directly.

    Notes: Summary In a randomized controlled clinical trial, the efficacy and safety of two low molecular weight heparin (LMWH) fractions in the prophylaxis of deep vein thrombosis (DVT) were assessed. One hundred twenty-six patients undergoing major abdominal surgery received alternatively 2,500 APTT units b.i.d. of two LMWH fractions or 5,000 APTT units b.i.d. of an unfractionated sodium mucosal heparin (UFH). LMWH 2 differed from LMWH 1 by presenting a lower mean molecular weight and a higher anti-Xa/APTT ratio in vitro. Patients were randomly allocated to the three groups, and the development of DVT was studied with the125I-fibrinogen uptake test (RFUT). The study was interrupted and the code broken prematurely because of otherwise unexplainable bleeding events. While no thrombosis and no severe bleeding were detected in the UFH group, three (7%) RFUT-positive DVT and two (5%) hemorrhagic complications occurred in the LMWH 1 group. No thrombosis and nine (22%) cases of severe bleeding were observed in the LMWH 2 group. Thus, the latter group differed significantly from the control group with regard to subjective and objective criteria for postoperative bleeding. Although these results do not allow general conclusions as to the value of LMWH fractions in the prevention of DVT, they indicate that these preparations just as ordinary heparin have a limited therapeutic range.

    Description / Table of Contents: Zusammenfassung Die Proteine aus Ductus thoracicus-Lymphe von 7 Patienten mit Lebercirrhose wurden mit Blutseren derselben Patienten sowie mit Lymph- und Blutseren von Normalpersonen nach Konzentration, elektrophoretischem Verhalten im Polyacrylamidgel, immunologischen Eigenschaften und Enzymgehalt verglichen. Auffallend war in den Lymphseren ein relativ geringer IgM-Gehalt, ein teilweise erhöhter IgA-Gehalt, eine hohe Albuminkonzentration, ein lebertypisches LDH-Isoenzymmuster, in 2 Lymphseren das Vorkommen eines lebereigenen Proteins und in einigen pathologischen Lymphseren das Auftreten eines Glykopeptids in hoher Konzentration.
    Notes: Summary The proteins of the thoracic duct lymph and blood serum from seven patients with liver cirrhosis and from normal persons were characterized by protein determination, electrophoresis on polyacrylamid gel, as well as by immunological and enzymatic methods. Differences between lymph and blood sera were observed. The lymph sera showed a low IgM content, a partially elevated IgA content, high albumin concentration, and altered LDH-isoenzyme pattern. In some lymph sera high concentrations of soluble protein from the liver and a glycopeptide were found.

    Notes: Abstract The enzyme glucose oxidase (GO) was covalently immobilized onto a poly(vinyl alcohol) hydrogel, cross-linked with glutardialdehyde and a polyazonium salt. To compare the kinetic parameters of immobilized GO with the known kinetic parameters of soluble GO, the diffusion cell method was used. Between two compartments, containing solutions with different glucose concentrations, a GO-containing hydrogel membrane was placed. Simultaneous diffusion through and enzymatic reaction in the membrane occurred. In this way diffusional effects of the membrane could be eliminated from the effective kinetic parameters to yield the inherent kinetic parameters. It appeared that the enzymatic reaction is independent of the oxygen concentration at oxygen concentrations ⩾0.22 mol m−3 (Michaelis constant for oxygen 〈 0.22 mol m−3). Further, the Michaelis constant for glucose does not change dramatically after immobilizing the enzyme. The maximal reaction rate is depending on the enzyme concentration. As the enzyme concentration in the membrane is not exactly known (mainly due to leakage of enzyme out of the membrane during membrane preparation), only an estimation of the turnover number can be made. The diffusion cell method is easy to carry out. Still, some recommendations can be made on the performance.

    Notes: Abstract. A novel disease affecting the brown-spotted grouper, Epinephelus tauvina Forskal, described here as Sleepy Grouper Disease1 (SGD), resulted in significant economic losses in some Singaporean marine net-cage farms from April to August 1992. Investigations suggested that the aetiological agent was a virus, probably introduced with imported groupers. The virus was provisionally identified as an iridovirus on morphological evidence. The disease caused extreme lethargy in affected fish with few visible external signs. Mortalities either occurred gradually over the week from onset of clinical signs, or over a shorter period and in large numbers if fish were stressed. Consistent tissue changes were seen by light microscopy in the spleen, heart and kidney of affected fish. Electron microscopy showed viral particles associated with damage to cell organelles. In an experimental infection, apparently healthy fish cohabiting with an infected fish developed similar lesions and died. The significance of SGD in grouper culture is discussed.
