Notes:
Summary After intraduodenal administration of3H-digitoxin,3H-digoxin and3H-ouabain in the anesthetized cat the radioactivity was determined in serum, urine and bile up to 3 h after the injection. 1. Both for3H-digitoxin and for3H-digoxin serum radioactivity reached its maximum value after approximately 2 h. For3H-digitoxin this maximum was considerably higher than for3H-digoxin. After the injection of3H-ouabain the serum radioactivity showed strongly varying levels in the individual animals. The differences may be due to varying absorption of the drug. 2. In the serum of human subjects no radioactivity could be detected after the oral administration of3H-ouabain. However, about 0.5–2% of the drug dosis given was excreted with the urine. 3. By means of51Cr labelled erythrocytes we determined the blood volume of the anesthetized cat. This parameter proved 49.0 ± 5.3 ml/kg. The amount of radioactive glycosides and their metabolites circulating with the blood could thus be calculated. 4. Both for3H-digitoxin and for3H-digoxin the excretion with the bile proved of greater relative importance than that with the urine.3H-ouabain, however, was excreted mainly via the kidneys. 5. Part of the3H-digitoxin was metabolized in the liver, although neither in the urine nor in the serum metabolites of3H-digitoxin could be demonstrated. Both for3H-digoxin and for3H-ouabain no metabolites could be found in bile and urine.
