    Notes: Abstract The effects of the novel antiasthmatic/antiallergic compound flezelastine on LPS-induced actions were investigatedin vitro andin vivo. In monocytes, IL-1β generation stimulated by LPS was inhibited dose dependently.In vivo, LPS-induced fever in rats, which is at least partly driven by the release of IL-1β, was also inhibited by flezelastine. These findings suggest that flezelastine inhibits IL-1 synthesis and/or releasein vitro andin vivo.

    Description / Table of Contents: Summary The role of cyclic AMP in the regulation of renin release (RR) was studied in isolated rat kidneys, which were perfused at constant pressure in a single-pass system with a modified Krebs-Henseleit solution. Isoproterenol (IP) (2×10−9 to 2×10−6 M) and 3-isobutyl-1-methyl-xanthine (IBMX) (4×10−7 to 7×10−5 M) induced a dose-dependent increase of renal perfusate flow, glomerular filtration rate and urinary sodium excretion. RR was stimulated up to 10-fold above control values within 5 min. At the lowest concentrations IP stimulated RR, but did not affect renal haemodynamics. When IP and IBMX were administered concomitantly, a supraadditive stimulation of RR was observed. Dibutyryl-cAMP (db-cAMP) and 8-Br-cGMP (10−6 to 10−4 M) produced a similar dose-dependent vasodilation and natriuresis, but differed in their action on RR. Within 15 min after the start of the infusion, db-cAMP increased RR up to 4-fold, whereas 8-Br-cGMP was without an effect. These results suggest that IP, IBMX and db-cAMP stimulated RR by increasing the concentrations of cAMP in the epitheloid cells and independently of changes in renal haemodynamics and sodium excretion.
    Notes: Zusammenfassung Die Beteiligung von cyclischem AMP an der Regulation der Reninfreisetzung wurde an isolierten Rattennieren untersucht, die in einem nicht rezirkulierenden System mit einer modifizierten Krebs-Henseleit-Lösung druckkonstant perfundiert wurden. Isoprenalin (IP) (2×10−9 bis 2×10−6 M) und 30-Isobutyl-1-Methyl-Xanthin (IBMX) (4×10−7 bis 7×10−5 M) erhöhten dosisabhängig die Nierendurchströmung, die glomeruläre Filtrationsrate und die Natriumausscheidung. Die Reninfreisetzung stieg bis auf das 10fache der Ausgangswerte an. Während der Infusion der niedrigsten Dosen steigerte IP die Reninfreisetzung ohne die renale Hämodynamik zu beeinflussen. Die gemeinsame Verabreichung von IP und IBMX fürte zu einer supraadditiven Stimulation der Reninfreisetzung. Dibutyryl-cAMP und 8-Br-cGMP (10−6 bis 10−4 M) bewirkten eine gleichartige dosisabhängige Vadodilatation und Natriurese aber unterschieden sich in ihrer Wirkung auf die Reninfreisetzung. Dibutyryl-cAMP stimulierte innerhalb von 15 min die Reninabgabe aus der Niere bis zum 4fa-chen der Kontrollwerte, während 8-Br-cGMP die Reninfreisetzung nicht beeinflußte. Diese Befunde lassen annehmen, daß IP, IBMX und Dibutyryl-cAMP die Reninfreisetzung unabhängig von Änderungen der renalen Hämodynamik und der Natriumausscheidung durch eine Erhöhung der Konzentration von cyclischem AMP in den epitheloiden Zellen beeinflussen.

    Description / Table of Contents: Summary Description of 23 patients (21 women, 2 men) with an average age of 36.6 (19–68) years, who were hypokalemic during 6.5 years on the average (range 1/2–16 years). The cause of the potassium depletion was malnutrition (anorexia nervosa, vomiting) and/or abuse of laxatives and/or diuretics. With increasing duration of potassium depletion renal function deteriorated; in two cases terminal renal failure developed. Histology of the kidneys (9 cases) showed the picture of chronic abacterial interstitial nephritis. Urinalysis was negative or non-specific. The blood pressure levels were normal or low, hypertensive values being exceptional. Aside of hypokalemia a tendency to hyponatriemia, hypochloremia and metabolic alcalosis was observed, the latter turning into hypokalemic normochloremic acidosis with advancing renal insufficiency. Plasma renin activity and aldosterone concentration or excretion frequently were elevated, but no close correlation was found between these parameters or with the blood pressure. Bacterial infection of the urinary tract occurred, if at all, in the late phase and seems to be complication rather than the cause of the kidney disease. The discussion of other possible pathogenetic factors leads to the conclusion that the term “chronic kaliopenic nephropathy” is justified. Some diagnostic and therapeutic consequences are mentioned.
    Notes: Zusammenfassung Es wurden 23 Patienten (21 Frauen, 2 Männer) im Alter von 36,6 (19–68) Jahren untersucht, bei denen infolge Mangelernährung (Anorexia nervosa, Erbrechen) und/oder Abusus von Laxantien und/oder Diuretika während 6,5 (1/2–16) Jahren eine teilweise schwere Hypokaliämie nachweisbar war. Mit zunehmender Dauer der Hypokaliämie entwickelte sich eine progrediente Einschränkung der Nierenfunktion, in 2 Fällen eine terminale Niereninsuffizienz. Histologisch (9 Fälle) fand sich das Bild einer chronischen abakteriellen interstitiellen Nephritis. Der Urinbefund war negativ oder uncharakteristisch, der Blutdruck normal oder niedrig, hypertone Werte waren die Ausnahme. Neben der Hypokaliämie bestand eine Tendenz zur Hyponatriämie und Hypochlorämie sowie eine metabolische Alkalose, die mit zunehmender Niereninsuffizienz in eine hypokaliämische normochlorämische Acidose überging. Plasma-Reninaktivität und Aldosteron-Konzentration bzw. -exkretion waren häufig erhöht, ohne daß eindeutige Korrelationen untereinander und zum Blutdruck bestanden. Eine bakterielle Harnwegsinfektion ist ein Spätsymptom und allenfalls Folge, nicht Ursache der Nierenerkrankung. Die Diskussion sonstiger möglicher pathogenetischer Mechanismen führt zu der Schlußfolgerung, daß die Bezeichnung „chronische kaliopenische Nephropathie“ gerechtfertigt ist. Einige diagnostische und therapeutische Konsequenzen werden erörtert.

    Notes: In the geminate recombination reaction of radical ion pairs with overall triplet multiplicity, locally excited triplet molecules are generated. The magnetic field effect (of the hyperfine type) on the triplet yield is usually determined by measuring the triplet absorbance both at zero and at higher applied magnetic fields. It is shown that the magnitude of this magnetic field effect decreases with time if the decay of the triplet concentration measured in the microsecond range is not of first order. This has been demonstrated experimentally using the linked acceptor–donor compound pyrene-(CH2)16-DMA as the test molecule in acetonitrile.
