    Notes: Garnet films of composition (Gd, Bi)3(Fe, Ga)5O12 are grown by liquid–phase epitaxy on (111) oriented substrates of gadolinium gallium garnet. The films are close to compensation. Using laser annealing straight vertical compensation walls (CW) are generated. Monomode rib waveguides are fabricated by ion beam etching so that a CW is located close to a rib center. The CW induces a nonreciprocal phase shift of the TE0 mode. The dependence of this phase shift on the position of the CW within the rib is calculated. Experimentally a phase shift of 0.7 cm−1 at 1.3 μm wavelength is observed. The deviation from the theoretical value is discussed. Such waveguides are essential to realize polarization–independent optical isolators. © 2001 American Institute of Physics.

    Notes: Magnetic garnet films of composition (Nd,Lu,Bi)3(Fe,Al,Ga)5O12 are grown by liquid-phase epitaxy on [111] oriented substrates of gadolinium gallium garnet. They have positive uniaxial anisoptropy and support lattices of parallel stripe domains. A monomode optical rib waveguide is prepared by photolithography and ion-beam etching parallel to the (112¯) direction, which is the preferred direction of the stripe domains. A magnetic domain pattern can be created such that a domain wall is located at the rib center while the adjacent walls stick to the flanks of the rib. At a wavelength of 1.3 μm a nonreciprocal phase shift of the guided fundamental transverse electric mode is observed. © 1999 American Institute of Physics.

    Notes: The Philadelphia translocation was demonstrated by two-colour fluorescence in situ hybridization (FISH) in decalcified paraffin sections of bone marrow from patients with chronic myelogenous leukaemia. FISH was combined with immunocytochemical detection of different membrane-bound or cytoplasmic antigens. With this new technique, the cells bearing the 9;22 translocation can be identified morphologically, as well as immunocyto-chemically, in tissue sections.

    Notes: Abstract Ninety-one Hodgkin's lymphomas (HD), 52 non-Hodgkin lymphomas (NHL) and 33 specimens of non-neoplastic lymphatic tissues were investigated by polymerase chain reaction (PCR) for the presence of the bcl-2/JH gene rearrangement. The majority of the HD cases were drawn from the files of the German Hodgkin trial where diagnoses are established by a panel of four independent histopathologists. Using the very sensitive PCR method which detected 1 positive among 10000 negative cells, the bcl-2/JH gene rearrangement was found in 7/52 NHL and 3/16 tonsils with follicular hyperplasia, but in none of the 91 HD. The bcl-2 protein, however, was expressed by malignant cells of B and T cell lymphomas and by the giant tumour cells in 2/13 HD lymphocyte predominant, 11/28 HD nodular sclerosing I, 14/17 HD nodular sclerosing II, 10/27 HD mixed cellularity and 3/3 HD lymphocyte depleted. The bcl-2/JH rearrangement is thus independent of protein over-expression, the latter being found in all types of lymphomas. Our results do not confirm the findings of others who have detected the bcl-2/JH rearrangement in HD. These discrepancies may be explained by differences in choice of material, the gene rearrangement actually occuring in bystander cells but not in Reed-Sternberg or Hodgkin cells, or by contamination.

    Notes: Summary Command fibers located in the circumesophageal connectives which modify scaphognathite and heart rhythms have been mapped and characterized in the crab,Cancer magister. Behavior: Crabs show a variety of responses to external stimuli often including simultaneous cessation of cardiac and scaphognathite “pumping”. Habituation and a return to prestimulus rhythms results from continued stimulation. The response to short stimulus durations, on the other hand, generally outlasts the stimulus indicating the playing-out of a motor program. Neurophysiology: Small bundles of fibers have been isolated from desheathed connectives. Activity in these fibers resulting from stimulation of various anterior sensory receptors was recordeden passant with suction electrodes. When sensory stimulation produced both electrical activity in the nerves under examination and a cardiac and/or scaphognathite response it was assumed such units were involved in inducing this response. This was tested by electrical stimulation delivered through the same electrode. Those units which produced similar responses to natural and artificial stimulation were deemed “command fibers”. It was invariably found that the minimum stimulating frequency needed to mimic naturally induced responses was much greater than the frequency at which the units discharged in response to those stimuli. During mapping experiments, command fibers were characterized with respect to their positions in the connectives and by the responses they produced at different frequencies of stimulation. 68% of the fibers identified affected both cardiac and scaphognathite systems, 29% the scaphognathites alone and 3% the heart alone. The frequency-response profiles of single bivalent command fibers were often different from the heart and scaphognathites. These findings help explain the responses of both systems to natural stimuli and also indicate that the circulatory and respiratory systems not only perform in concert, but are often under common control.

    Notes: Abstract Directional selectivities of mechanoreceptors that innervate filiform hairs on the crayfish tailfan were investigated with unidirectional, sinusoidal, water-motion stimuli. These recordings provide the first representative sample from filiform hair sensilla on the entire tailfan. The filiform hair receptors exhibit unimodal directional selectivity patterns that were well fitted by a cardioid function with a half-width of 122°. The preferred directions correspond to the major axis of hair motion, and are perpendicular to the orientation of lateral branches on the main hair shaft. Pooled plots of preferred directions demonstrate quadrimodal patterns on the telson and endopods which are associated with hair location, and a bimodal pattern on the exopods. For each appendage, the combination of the overall pattern of preferred directions with “coarse coding” of direction by individual receptors provides sensitivity to a full 0–360° range of water motion and the potential to discriminate the direction of water motion throughout this range. The results demonstrate several similarities to the wind-sensitive cercal receptor system in orthopteroid insects, and suggest that crustacean filiform hair receptors provide a sufficient sensory basis for behavioral orientation to water currents and shorelines.

    Notes: Abstract Formaldehyde fixation of biopsy specimens for routine purposes has often been held responsible for the poor reproducibility of immunohistochemical studies. Recently, antigen retrieval (AGR) using microwave irradiation was described as a potential tool to enhance immunostaining. A comparison of conventional staining and staining after microwave heating was performed for 52 markers, using tissues fixed in formaldehyde for 24 h, 1 to 6 weeks and 3 years respectively, as well as consultant case material. After adequate duration of fixation (24 h), only a few markers (17%) showed better results after AGR, but this percentage was increased to 50% when tissues were fixed for longer periods. Maximal enhancement was obtained in the group of consultant cases (58% of tested markers demonstrated better staining results), in which the period of fixation and tissue processing was unknown. To achieve reliable enhancement with AGR, continuous heating (100° C) should not be shorter than 20 min. In conclusion, AGR may become the most important tool to simplify and equalize immunohistochemical techniques, if critically evaluated.
