    Notes: We have measured the vibrational energy relaxation of azulene in the S2 state in ethane, carbon dioxide, and xenon over a wide density region by the time-resolved fluorescence spectroscopy. The reduced density of the solvent (the density divided by the critical density) has been changed from the gaslike one (lower than 0.05) to the liquidlike one (2.8 for ethane, 2.7 for carbon dioxide, and 2.0 for xenon). The density dependence of the relaxation rate is compared with that of azulene in the S0 state [D. Schwarzer, J. Troe, M. Votsmeier, and M. Zerezke, J. Chem. Phys. 105, 3121 (1996)]. Although the absolute rate largely depends on the electronic state, the density dependence of the relaxation rates is roughly the same for both electronic states, which can be considered as a side-proof of the isolated binary collision model. Possible origins for the validity of the isolated binary collision model are discussed. © 2000 American Institute of Physics.

    Notes: We have calculated nonlinear time correlation functions of a dilute hard-sphere gas numerically by using the Monte Carlo method, to find that the single particle dynamics in a dilute hard-sphere gas does not follow the Gaussian process. The deviation of the self-part of the dynamic structure factor from a Gaussian function is observed. This non-Gaussian character corresponds to those of Lennard-Jones liquids reported by Itagaki et al. [K. Itagaki, M. Goda, and H. Yamada, Physica A 265, 97 (1999)], if we scale the time unit by the collision frequency. Further, we trapped a particle in a harmonic well and calculated the time development of its distribution, in order to clarify the effect of collisions to the solvation dynamics. Both the Gaussian and the linear response assumptions are broken, and the deviation becomes larger as the curvature of the harmonic well gets larger. © 2001 American Institute of Physics.

    Notes: We have measured the time-resolved fluorescence spectra of azulene in the S2 state in compressed gases and in liquids. We have found that the band shape of the fluorescence changes significantly in the earlier time scale after the photoexcitation when large excess energy (about 6500 cm−1) is given. The change of the band shape is similar both in the compressed gases and in the liquids, although the time scales of the change are quite different. We have measured the excitation energy dependence of the fluorescence band shape of the isolated molecule separately, and shown that the time dependence of the fluorescence band shape in gases and liquids corresponds to the vibrational energy relaxation in the S2 state. Comparing with the excitation energy dependence of the fluorescence band shape of the isolated molecule, we have succeeded in determining the transient vibrational excess energy. The vibrational energy relaxation rates in the S2 state are 1–2 times faster than those in the ground state both in compressed gases and in liquids. © 2000 American Institute of Physics.

    Notes: Background We have demonstrated that carbohydrates in Cry j 1, the major allergen of Cryptomeria japonica pollen, play a major role in promoting Cry j 1-specific Th2 response. However, little is known as to whether the carbohydrates directly participate in allergic responses.Objective We sought to determine whether Cry j 1-related oligosaccharides function as IgE and/or T cell epitopes. In addition, the regulatory effect of Cry j 1-related oligosaccharide on Cry j 1-specific T cell responses was investigated.Methods Two monovalent oligosaccharides largely found on Cry j 1, Manα1-6(Manα1-3)(Xylβ1-2)Manβ1-4GlcNAcβ1-4(Fucα1-3)GlcNAc (M3FX), and GlcNAcβ1-2Manα1-6(GlcNAcβ1-2Manα1-3)(Xylβ1-2)Manβ1-4GlcNAcβ1-4(Fucα1-3)GlcNAc (GN2M3FX) were prepared. Manα1-2Manα1-6(Manα1-2Manα1-3)Manα1-6(Manα1-2Manα1-2Manα1-3)Manβ1-4GlcNAcβ1-4GlcNAc (M9A) was used as control. Competitive inhibition ELISA for Cry j 1-specific IgE was performed using these oligosaccharides as inhibitors. In addition, T cell lines specific for Cry j 1 or purified protein derivative of Mycobacterium tubecurosis (PPD) were established, and cellular responses against these oligosaccharides were investigated in the presence or absence of the respective antigens.Results Overall, neither M3FX nor GN2M3FX displayed inhibitory effect on the binding between IgE and Cry j 1. In addition, M3FX did not by itself stimulate Cry j 1 or PPD-specific T cells. However, M3FX significantly inhibited Cry j 1-induced proliferation and IL-4 production in Cry j 1-specific T cells. Such an inhibitory effect was not seen in PPD-specific T cell responses.Conclusion These results suggest that Cry j 1-related oligosaccharides are not major epitopes for IgE or T cells. However, these oligosaccharides have a novel potential to inhibit Cry j 1-specific T cell responses selectively.

    Notes: Background Kimura's disease (KD) is a chronic inflammatory disorder characterized by tumours in the head and neck region, enlarged lymph nodes, increased eosinophil counts and high serum IgE. Mast cells are known to play a central role in IgE-mediated allergic diseases through the release of inflammatory mediators like IL-4, IL-5 and chemokines. We hypothesized that mast cells may play a role in the pathogenesis of KD by regulating eosinophilic infiltration and IgE synthesis.Objective In order to investigate the role of mast cells in the pathogenesis of KD, we examined the expression of cytokines/chemokines in the lesions of KD.Methods We examined the number of tryptase+ cells, EG2+ cells, CD3+ cells, IL-4+ cells, IL-5+ cells, eotaxin+ cells, RANTES+ cells and CCR3+ cells in five specimens of KD versus normal tissues by immunohistochemistry. The sources of IL-4, IL-5, eotaxin and RANTES and the expression of CCR3 were examined by immunostaining of serial sections with antibodies to IL-4, IL-5, eotaxin, RANTES and CCR3, and antibodies to tryptase, ECP (EG2) and CD3.Results Mast cells, activated eosinophils, T cells, IL-4+ cells, IL-5+ cells, eotaxin+ cells, RANTES+ cells and CCR3+ cells were all increased in the lesions of KD as compared with those in normal tissue. Mast cells and T cells were the major source of IL-4, whereas mast cells, T cells and activated eosinophils were the main source of IL-5. Mast cells, T cells and activated eosinophils were the main source of eotaxin and RANTES.Conclusions The number of IL-4, IL-5, eotaxin and RANTES-expressing mast cells and T cells were increased in the lesions of KD. As mast cells are lesional resident cells, these cells may play an important role in the pathogenesis of KD by regulating IgE synthesis and orchestrating eosinophilic infiltration.

    Notes: Abstract We report here a case of granulocyte-colony stimulating factor (G-CSF)-producing esophageal carcinoma in a 66-year-old man. The International Union Against Cancer (UICC) staging was IV A, and a diagnosis of moderately differentiated squamous cell carcinoma was made, based on histopathology. The diagnosis was based on marked leukocytosis (41 500 leukocytes/mm3) and an elevated serum level of G-CSF (154 pg/ml). Immunofluorescent histochemistry and northern blot analysis confirmed the expression of G-CSF protein in cancer cells and its mRNA in cancer tissue. We paid special attention to any change in serum G-CSF levels during aggressive cancer treatment. Subtotal esophagectomy induced a significant decrease in G-CSF level. Adjuvant chemo-radiotherapy, targeting celiac lymph node metastasis, and radiotherapy, targeting solitary lung metastasis, together effectively maintained a low serum G-CSF level, despite a recurrence of the tumor in the lungs, in the form of multiple metastases, with an increase in serum G-CSF levels. The patient's clinical course suggested that serum G-CSF would be a useful marker for monitoring the effects of treatment on G-CSF-producing carcinoma.

    Notes: A computer program of the fractal analysis by the box-counting method was developed for the estimation of the fractal dimension of the three-dimensional fracture surface reconstructed by the stereo matching method. The image reconstruction and fractal analysis were then made on the fracture surfaces ofmaterials created by different mechanisms. There was a correlation between the fractal dimension of the three-dimensional fracture surface and the fractal dimensions evaluated by other methods on ceramics and metals. The effects of microstructures on the fractal dimension were also experimentally discussed

    Notes: Abstract  A thermosetting epoxy resin was found to form polymer particles spontaneously in a stationary reaction system using polyethers as solvents. Poly(oxypropylene) and its copolymer with poly(oxyethylene) were effectively used as reaction media. The diameter of the resultant particles depended on the structure of the amines used as hardeners as well as on the molecular weight and composition of the polyether media. The glass-transition temperatures of the particles revealed that a small amount of polyether used as the medium was incorporated in the epoxy resin particles during the polymerization.

    Notes: Abstract A new exercise protocol has been proposed, with respect to cardiopulmonary exercise testing, which starts at a low work rate (WR) and increases exponentially by a standard percentage of the previous work rate every minute: the test is termed STEEP (standardised exponential exercise protocol). The potential advantage of this protocol is that it can accommodate a wide range of subjects, since it allows a maximum to be attained with a relatively narrow variation of tolerance time, regardless of subjects exercise capacity. To date, only the VO 2max has been compared with that from the current standard ramp protocol. The ramp, however, also allows other important parameters of aerobic function to be estimated: the anaerobic threshold (AT); the response time constant; and ΔVO2/ΔWR. The aim of this study was, therefore, to clarify whether these aerobic parameters can be readily discerned from the responses to the STEEP protocol both from a theoretical and practical viewpoint. As a result of theoretical considerations, we demonstrated that the VO2 time constant and ΔVO2/ΔWR may not both be estimated uniquely. As a practical expedient, a procedure was proposed for estimating the parameter analogues. The preliminary results for six subjects between the STEEP and ramp protocols showed consistent positive correlation for VO 2max (r=0.997) and AT-VO2 (r=0.980), whereas the correlation for the VO2 time constant and ΔVO2/ΔWR were not significant. Further study is needed to clarify the reason(s) for the discrepancies both from a theoretical and practical viewpoint.
