    Notes: Abstract  We used a combination of 4 monoclonal antibodies (BM7, BM8 against MUC1, 5D3 aganist CK8,18,19 and HEA125 against human epithelial antigen) and a sensitive immunocytochemical staining using cytospin preparation to identify breast tumor cells in leukapheresis products (LP). This assay allowed detection of one tumor cell in 1×106 mononuclear cells (MC). In clinical specimens, tumor cells were detected in LP from 6 of 42 (14.3%) patients in the adjuvant treatment group, from 2 of 11 (18.2%) patients in the neoadjuvant treatment group and from 9 of 43 (20.1%) in the group of patients with metastatic disease. Tumor cell counts ranged from 0.25–5 cells in 1×106 normal cells per LP. The median tumor cell concentration was higher in specimens from patients with metastatic disease (median=0.96) than in specimens from patients in the adjuvant and neoadjuvant treament groups (median=0.5 and 0.75). No significant differences between the epithelial cell positive group and the epithelial cell negative group with respect to tumor size, lymph nodes involvement, tumor grade, histological type and receptor were found. We conclude that immunocytochemical staining of cytospin preparation is a sensitive and simple method to detect and quantitate breast cancer cells in LP.

    Notes: A nodavirus was isolated from diseased yellow grouper, Epinephelus awoara, larvae cultured in southern Taiwan. The histopathology and RT–PCR results confirmed that it was a fish nodavirus; its coat protein gene sequence was similar to that of red spotted grouper nervous necrosis virus (RGNNV) and it is named yellow grouper nervous necrosis virus (YGNNV). A new nodavirus-susceptible cell line, grouper brain (GB) was established and characterized from the brain tissue of yellow grouper. The GB cells multiplied well in Leibovitz’s L-15 medium supplemented with 10% foetal bovine serum at temperatures between 24 and 32 °C, and have been subcultured more than 80 times, becoming a continuous cell line. The GB cell line consists of fibroblast-like cells and some epithelioid cells. The cell line yielded titres of YGNNV up to 108.5 TCID50 mL–1. The GB cells effectively replicated the virus at 28 °C, which could be purified to homogeneity by caesium chloride gradient centrifugation. Electron microscopy studies showed that purified virus particles were 25–30 nm in diameter. The cytoplasm of infected cells was filled with aggregates of virus particles. These results indicate that the GB cell line is a significant tool for the study of fish nodaviruses.

    Notes: Stoichiometric, uniform, amorphous ZrO2 films with an equivalent oxide thickness of ∼1.5 nm and a dielectric constant of ∼18 were deposited by an atomic layer controlled deposition process on silicon for potential applications in metal–oxide–semiconductor (MOS) devices. The conduction mechanism is identified as Schottky emission at low electric fields and as Poole–Frenkel emission at high electric fields. The MOS devices showed low leakage current, small hysteresis (〈50 mV), and low interface state density (∼2×1011 cm−2 eV−1). Microdiffraction and high-resolution transmission electron microscopy showed a localized monoclinic phase of α-ZrO2 and an amorphous interfacial ZrSixOy layer which has a corresponding dielectric constant of 11. © 2001 American Institute of Physics.

    Notes: A lattice-matched δ-doped In0.34Al0.66As0.85Sb0.15/InP heterostructure field-effect transistor (HFET) which provides large band gap (∼1.8 eV), high Schottky barrier height (φB〉0.73 eV), and large conduction-band discontinuity (ΔEc〉0.7 eV) has been proposed. In0.34Al0.66As0.85Sb0.15/InP heterostructures are shown to be type II heterojunctions with the staggered band lineup. This HFET demonstrates a output conductance of less than 1 mS/mm. Two-terminal gate-source breakdown voltage is more than 20 V with a leakage current as low as 170 μA at room temperature. High three-terminal off-state breakdown voltage as high as 36 V, and three-terminal on-state breakdown voltage as high as 18.6 V are achieved. The gate voltage swing is also significantly improved. © 2000 American Institute of Physics.

    Notes: The thermal stability of ZrO2/ZrSixOy and ZrO2/ZrSixOy/SiNx thin films on silicon was examined by synchrotron radiation ultraviolet photoemission spectroscopy. The ZrO2/ZrSixOy layer deposited by atomic-layer-controlled deposition is stoichiometric, uniform, amorphous, and has an equivalent oxide thickness of ∼1 nm and a dielectric constant of ∼18 with low leakage current. These ZrO2/ZrSixOy samples are thermally stable in vacuum up to 880 °C at which the film decomposed to form ZrSi2, the most thermodynamically stable metal silicide at a per zirconium atom basis, and the desorption of SiO(g) and ZrO(g) accounted for the greatly reduced oxygen and zirconium photoemission intensities. The thermal stability of ZrO2/ZrSixOy is improved to 950 °C when deposited on a 0.5–0.7 nm SiNx film. © 2001 American Institute of Physics.

    Notes: Aim : To test whether supplements of Lactobacillus- and Bifidobacterium-containing yogurt (AB-Yogurt) affect the success of Helicobacter pylori eradication.Methods : One hundred and sixty H. pylori-infected patients were randomized into a triple-plus-yogurt group or a triple-only group, receiving 1 week of triple therapy with and without supplements of AB-Yogurt, respectively. In the triple-plus-yogurt group, AB-Yogurt was continued for 4 weeks after triple therapy. Eight weeks later, patients were assessed for the success of H. pylori eradication. The stool samples of 22 randomly selected patients, 11 from each group, were provided on enrolment, at the first week and at the fifth week for evaluation of the percentage of Bifidobacterium in anaerobes.Results : By intention-to-treat analysis, the triple-plus-yogurt group had a higher H. pylori eradication rate than the triple-only group (91% vs. 78%, P 〈 0.05). The per protocol H. pylori eradication rates were similar for both groups (93.5% vs. 89%, P = N.S.). Only patients supplemented with AB-Yogurt showed restoration of the percentage of Bifidobacterium in the anaerobes of stools at the fifth week to the level in the stools on enrolment.Conclusions : Supplement with AB-Yogurt can improve the intention-to-treat eradication rates of H. pylori, and can restore the depletion of Bifidobacterium in stools after triple therapy.

    Notes: Summary Ninety female patients were enrolled in this randomised, double-blind, placebo-controlled study to compare the anti-emetic effect of intravenous dexamethasone 8 mg with saline control in preventing patient-controlled analgesia-related nausea and vomiting following major orthopaedic surgery. The prophylactic administration of dexamethasone 8 mg significantly reduced the overall incidence of patient-controlled analgesia-related nausea and vomiting (p 〈 0.001) and the need for rescue anti-emetics (p 〈 0.01). Furthermore, patients who received dexamethasone showed a higher incidence of complete responses (no vomiting or need for rescue anti-emetic for a 24-h postoperative period) than those who received saline (p 〈 0.05). We conclude that dexamethasone 8 mg may be valuable for preventing patient-controlled analgesia-related nausea and vomiting in women undergoing major orthopaedic surgery.

    Notes: Objectives:  Amplification of the proto-oncogene c-erbB-2 (HER-2/neu) has been shown to be a prognostic marker in many kinds of cancer including oral squamous cell carcinoma (OSCC). In order to obtain further information on the c-erbB-2 gene product p185, it is necessary to quantify expression levels. In this study we used an enzyme-linked immunosorbent assay (ELISA) for the extracellular domain of p185 to determine whether a soluble oncoprotein fragment can be detected in the serum of OSCC patients.Method:  Sera from 84 OSCC patients, 51 breast cancer patients (as positive controls), and 15 healthy controls were assayed in an ELISA. To study c-erbB-2 overexpression in OSCC, and breast cancer tissue samples we used an immunohistochemical technique.Results:  The mean serum value (ng/ml, mean/SD) for the normal controls was 8.46/1.29. We chose the 95% level of normal controls as a cut-off to distinguish individuals with elevated levels. The breast cancer patients’ and OSCC patients’ serum values were 13.83/6.82 and 13.1/4.56, respectively. Significant differences (P 〈 0.0001) were observed between normal control and OSCC, normal control and the breast cancer group. Immunohistochemically detectable p185 (intermediate to high) was noted in 30 of 61 OSCC, and 24 of 51 breast cancer patients. There was a trend of association of serum oncoprotein fragment levels with tumor stages, but not with tumor sizes, nodal stages, metastases, and oral habits including betel quid chewing, alcohol drinking and smoking in the OSCC group.Conclusion:  The results of the present study raise the possibility that soluble c-erbB-2 protein levels in serum is a useful parameter for monitoring the disease status as well as the effect of therapy on patients with OSCC.
