    Notizen: . A State loan program was analyzed for its effects on groundwater development in Wyoming. The analysis focused on the effects of low-interest State financing on the economic feasibility of investments in center-pivot sprinkler irrigation systems. The feasibility analysis indicated that investments in sprinkler systems for production of cash crops are highly profitable, and would frequently be carried out whether or not the State loan program were available. Investments in sprinklers for production of forage crops are perhaps frequently made attractive by the loan program. Thus, while the program appears to have accelerated the pace of private water resource development in Wyoming, it has also subsidized some investments that would have been carried out regardless of the program's existence. Further analysis indicated that sprinkler investments carried out with State financing contribute substantially to firm growth when cast crops are grown, but have less marked, and sometimes negative, effects on growth when forage crops are produced. Some positive effects of the loan program on southeast Wyoming's regional economy were noted, though a complete empirical analysis of regional impacts was beyond the scope of the study.

    Notizen: Summary By the use of indirect immunofluorescence it is shown that “early” functions of human cytomegalovirus induce a loss of microtubuli in human foreskin fibroblasts within 12 hours postinfection (p.i.) which persists until about 60 hours p.i. At later times p.i. microtubular structures are eventually reestablished. Following depression during the initial 48 hours p.i. tubulin synthesis in infected cells is significantly enhanced during the late phase of the infectious cycle suggesting that the mechanisms regulating tubulin synthesis are not impaired.

    Notizen: Abstract   Objective: To describe how patients cope with the propos-al of treatment with intermittent artificial ventilation after acute respiratory failure due to progressive respiratory muscle weakness. Design: Case series, follow-up study. Setting: Neurological intensive care unit (ICU). Patients: 7 consecutive patients with metabolic myopathy treated for acute respiratory failure between 1983 and 1992. interventions: Intermittent positive pressure ventilation (IPPV) via tracheostomy. Measurements and results: Symptoms of chronic hypoventilation preceded acute respiratory failure for months. With one exception, patients were mainly disabled from respiratory muscle weakness and sleep-related breathing disorders. IPPV was recommended to prevent recurrent respiratory failure. Two of 28.8nthree patients who accepted home IPPV returned to full-time jobs. One patient, who decided against IPPV, died from CO2 narcosis several months after discharge. All patients adhered to the respiratory regimen once instituted. Conclusions: Acute respiratory failure in chronic myopathy is heralded by daytime drowsiness. IPPV, or at least regular monitoring of waking and sleeping partial pressure of carbon dioxide, is highly recommended even if weaning is successful. IPPV improved quality of life. The treatment strategy at discharge from the ICU should be optimal, as patients are reluctant to modify regimens.

    Beschreibung / Inhaltsverzeichnis: Summary The influence of variing ratios of collagen to meat protein on bodyweight and carcass collagen of growing male rats was investigated. 1. Addition of sinew protein up to 5 % (28 % of total protein) to a feed mixture containing 13 % muscle protein yielded slight improvements on daily gain and feed conversion. In a ration containing 18 % muscle protein, 2 % grain protein and 2 % protein from fishsolubles and dried whey, the addition of 10 % sinew protein (31 % of total protein) depressed gain and feed conversion. 2. Raw sinew protein was utilized as good as cooked sinew protein. Autoclaving and drying meat and sinews together did not influence the protein quality of the mixture. 3. Analysis of total carcasses of 100 rats showed no influence of high collagen intake on collagen content.
    Notizen: Zusammenfassung In drei Fütterungsversuchen wurde der Einfluß von Kollagenzulagen zu schierem Fleisch auf Entwicklung und Bindegewebsansatz wachsender männlicher Albinoratten untersucht. 1. Durch Zulagen von Sehnenprotein bis zu 5 % (28 % am Gesamtprotein) zu einem Futter mit 13 % Muskelprotein wurden die Gewichtszunahmen und die Futterverwertung noch etwas verbessert. Zulagen von Sehnenprotein in Höhe von 10 % (31 % des Gesamtproteins) zu einem Futter mit 18 % Muskelprotein, ca. 2 % Getreideprotein und ca. 2 % Protein aus Fishsolubles und Molkenpulver bewirkten bereits eine Depression der Gewichtsentwicklung und der Futterverwertung. 2. Zerkleinerte rohe Sehnen wurden von den Ratten ebenso gut verwertet wie gekochte Sehnen. Das Erhitzen der zerkleinerten Sehnen in Mischung mit zerkleinertem schierem Muskelfleisch beeinträchtigte die Proteinqualität der Mischung nicht. Die Ratten entwickelten sich mit diesem Futter ebenso gut wie mit Rationen aus einer nachträglichen Mischung aus getrocknetem Fleisch und getrockneten Sehnen. 3. Ganzkörperanalysen von insgesamt 100 Ratten ließen keine Beeinflussung des Bindegewebsansatzes durch hohe Kollagenfütterung erkennen. Im Mittel aller Untersuchungen (als Kriterium diente der Oxyprolin- und Glycingehalt) enthielten 40 Ratten, die ohne Sehnenzulagen gefüttert wurden, 20,8 % Bindegewebseiweiß im Gesamtkörperprotein. 60 Ratten mit 24–31 % kollagenem Eiweiß im Protein der Gesamtration enthielten 20,5 % Bindegewebe im Protein des Gesamtkörpers.

    Notizen: Abstract Endotoxin-induced early inflammatory reactions may inhibit the function and survival of isolated cells or cell aggregates after transplantation. By the chromogenic Limulus amebocyte lysate assay we found rather high but variable endotoxin concentrations in the chemicals used for islet isolation, i.e. collagenase and Ficoll. Liberase, a special collagenase preparation from Boehringer, was nearly endotoxin-free. Correlating to the endotoxin content, collagenase and Ficoll had the capacity to induce interleukin-1β release from human peripheral blood mononuclear cells. Because collagenase and density gradient media are needed in most cell isolation procedures from solid organs, each lot of these chemicals should be tested for endotoxin contamination. In pancreatic islet transplantation, the use of endotoxin-free chemicals may diminish early local inflammatory reactions at the graft site and thereby reduce the number of islets needed for successful islet transplantation.

    Notizen: Abstract The macrophage derived cytokines interleukin-1 beta (IL-1β), and tumor necrosis factor alpha (TNFα), and the T-cell derived cytokine interferon gamma (IFNγ) have been implicated to play an important role in early attack on islet cells during human islet transplantation (ITx). Therefore, the aim of this study was to investigate the influence of the current immunosuppressive induction therapy in clinical islet transplantation on mRNA expression of these cytokines in blood cells, compared to lipopolysaccharide (LPS) induced cytokine release in vitro and to plasma levels. The cytokine release correlated to lymphocyte counts and significantly decreased after ATG, and partially recovered 2 weeks after ITx. Unexpectedly, there was no correlation between mRNA expression for IL-1β in total blood and the number of lymphocytes and monocytes remaining after anti thymocyte globulin (ATG)-therapy. Even when the blood was nearly totally depleted from mononuclear cells, high amounts of IL-1β mRNA could be detected. However, IL-1β secretion could not be stimulated in vitro. Our results show that application of ATG during ITx might contribute to graft survival during the early posttransplant period by suppression of the synthesis of monocyte derived cytokines IL-1β and TNFα.
