    Notes: Abstract A simple, sensitive, non-stimulated assay was developed to measure the superoxide anion concentration in whole blood, using an ultra-sensitive chemiluminescence (CL) analyzer, and lucignin amplification. The assay system can be performed without leukocyte isolation or stimulant administration. The blood CL levels of healthy males (362.8±337.7 counts/10 sec) were not different from those of females (335±308.7 counts/10 sec) (p=0.64), whereas the CL levels in whole blood in patients with acute pancreatitis (2522±2014 counts/10 sec) were significantly higher than those of healthy controls (p〈0.001). This assay system may be valuable in the future for quantitative measurement of reactive oxygen species in various disorders.

    Notes: Abstract. Oxygen-derived free radical injury has been associated with several cytopathic conditions. Oxygen radicals produced by chondrocytes is an important mechanism by which chondrocytes induce matrix degradation. In the present study, we extend these observations by studying oxidative processes against osteoblasts. Osteoblasts were mixed in in vitro culture with 200 μM menadione. The cytotoxic effect of menadione-induced oxidative stress was monitored by lucigenin- or luminol-amplified chemiluminescence, tetrazolium assay and immunocytochemical study. Results showed that adding menadione induces an oxidative stress on osteoblasts, via superoxide and hydrogen peroxide production, that can be eradicated by superoxide dismutase (SOD) and catalase in a dose-dependent manner. Catalase and the appropriate concentration of dimethyl sulfoxide have a protective effect on cytotoxicity induced by menadione, whereas SOD does not. Menadione-treated osteoblasts have a strong affinity for annexin V, and the nuclei are strongly stained by TUNEL (TdT-mediated dUTP nick-end labelling). The results suggest that menadione-triggered production of reactive oxygen species leads to apoptosis of osteoblasts.

    Notes: Abstract  HLA-B*0801 is unique among HLA-B allotypes in having dominant amino acid anchors at positions 3 and 5 of the peptide-binding motif. HLA-B*0802 is a variant of HLA-B*0801 in which the Bw6 sequence motif is replaced by a Bw4 sequence motif. This change, involving substitutions at positions 77, 80, 81, 82, and 83 of the B*08 heavy chain, is probably the result of a single evolutionary event of interallelic conversion. Moreover, the difference between B*0802 and B*0801 is sufficient to stimulate a cytotoxic T-cell response. To assess further the functional impact of the Bw4 motif on a B8 background, we compared the peptide-binding specificity of the B*0801 and B*0802 allotypes by sequencing the mixture of peptides endogenously bound to B*0802 and 12 individual peptides purified from that mixture. The HLA-B*0802 allotype, while able to bind some peptides bound by B*0801, has a broader repertoire of endogenously bound peptides than B*0801: the peptides bound by B*0802 are more variable in length and exhibit greater diversity in the carboxyl-terminal amino acid which interacts with the F pocket.

    Notes: The extracellular form of glutathione peroxidase from human plasma has been crystallized by the sitting-drop vapour-diffusion method. The crystals belong to tetragonal space group I41 with cell dimensions of a = b = 83.1 and c = 131.0 Å. They diffract beyond 2.9 Å resolution and have one dimer in the asymmetric unit. A self-rotation function analysis shows the possible (222) symmetry for the tetramers of this enzyme.

    Notes: A method for detecting surface plasmon resonance with high resolution (∼10−5 degrees or ∼10−8 refractive index units) and fast response time (1 μs) is described. In the method, light is focused through a prism onto a metal film on which molecules to be detected are adsorbed. The total internal reflection of the incident light is collected with a bicell photodetector instead of a single cell or an array of photodetectors that are widely used in previous works. The ratio of the differential signal to the sum signal of the bicell photodetector provides an accurate measurement of shift in surface plasmon resonance angle caused by the adsorption of molecules onto the metal films or by conformational changes in the adsorbed molecules. Using the method, we have studied subtle conformational changes in redox protein, cytochrome c, due to an electron transfer reaction. © 1999 American Institute of Physics.

    Notes: Variable angle spectroscopic ellipsometry (VASE) technique is used to measure the optical constants of InGaAs/GaAs multiple quantum well (MQW) structures for the purpose of designing tunable optical modulators. The VASE measurements also include field-induced changes in index of refraction and absorption. These measured changes in MQWs are in agreement with theoretical computations. In addition, the design and simulated performance of an InGaAs/GaAs MQWs Fabry–Perot modulator using the VASE data is presented. © 1997 American Institute of Physics.

    Notes: In aquaculture, controls of size (length and weight) and production of fish are two important tasks to meet the market demands, and increasing the stocking density is a way of dealing with the problem of land shortage. In this study, tilapia fry were stocked at the densities of 0.1, 0.2, 0.4, 0.8, 1.6 and 3.2 fry I−1 for an experimental period of 10 weeks. The size, size variation, percentage survival and production were found to be significantly affected by stocking density, but not for condition factor. The simple linear regression model, after the logarithmic transformation, was appropriate for the relationship between size and experimental period. The percentage survivals at all stocking densities were high (〉 95%), especially at the stocking density of 0.4 fry I−1 and less (100%). At the higher densities, percentage survival and experimental periods were anti-logistically related. At the end of the experiment, there was a negative relationship between the logarithms of size and the logarithms of stocking density, but a positive relationship between the logarithms of production and the logarithms of stocking density.

    Notes: We present theoretical calculations of the magneto-optic polar Kerr effect in bcc iron, fcc cobalt, and hcp cobalt. The Kerr angle and ellipticity are derived from the optical conductivity tensor, which is computed using wave functions obtained from full potential local spin-density electronic structure calculations. Predicted Kerr angles and ellipticities are computed as a function of magnetization direction. In the case of iron, the calculated Kerr spectra are found to be in good agreement with experimental results and previous calculations. However, in the case of cobalt, the results of previous theoretical studies differ significantly. Our results are in close agreement with experiment and with one of these previous theoretical studies. © 1996 American Institute of Physics.

    Notes: First principles electronic structure calculations are used to predict the energy dependence of the polar Kerr effect in MnBi, MnSb, and a Mn2BiSb alloy. The compounds all have respectable Kerr angles and thus are potential data storage media. The technique used in this work is a full-potential linearized augmented Slater orbital approach. The Kerr angle is obtained from the conductivity tensor that is calculated, within linear response theory, using the Kubo formula. There are several desirable features in this approach. First, unlike methods based upon muffin-tin potentials, open crystal structures present no problems. Second, since the electronic wave functions are defined over the crystal unit cell rather than over space filling overlapping spheres, difficulties in handling the interstitial part of the required matrix elements are avoided. Finally, the small basis set (25 orbitals per atom) leads to reasonable computational times. Results of these calculations show that the Kerr angle in MnBi is above 1° at 1.6 eV and that its variation over 2–4 eV is linear and decreasing. MnSb shows a smaller rotation, about 0.5°, that is almost constant up to 3 eV and then also decreases linearly. The Kerr rotation of Mn2BiSb is approximately the average of MnBi and MnSb.
