    Notes: Abstract Since Staphylococcus aureus is an important human pathogen, and infection of the lungs in characterized by neutrophil infiltration we studied the role of a staphylococcal toxin, enterotoxin A (SEA) on the synthesis and secretion of IL-8 by human alveolar macrophages. As SEA concentration was increased, the IL-8 accumulation in the macrophage conditioned medium increased. The concentration of mRNA encoding IL-8 was also elevated in the macrophage in response to increases in SEA concentration. Although the monocytic cell line U937 was able to respond to SEA and secrete IL-8, treatment with PMA prior to SEA stimulation increased the IL-8 accumulation around fifty fold indicating that maturation of the undifferentiated cell to a more macrophage-like cell facilitated IL-8 accumulation. Stimulating human alveolar macrophages with high concentrations of SEA caused an increase in IL-1 accumulation. However, when the cells were incubated with SEA in the presence of IL-1 receptor antagonist, there was no decrease in IL-8 accumulation. Addition of a neutralizing anti-IL-8 monoclonal antibody to the culture medium of SEA-stimulated macrophages significantly reduced the neutrophil chemotactic activity of the medium. These studies showed that IL-8 is a major neutrophil chemotaxin from human alveolar macrophages stimulated with SEA.

    Notes: Summary  The long-term effects of post-ischaemic hypothermia are controversial. The purpose of this study was to examine the long-term effects of post-ischaemic hypothermia on neuronal survival in gerbils in terms of morphology and function. Hypothermia was induced at 32° C for 4 h immediately after ischaemia. Examination was performed at 1 week and at 1 month after ischaemia. Post-ischaemic hypothermia prevented CA1 neuronal damage 1 week after ischaemia. At 1 month after ischaemic insult, however, the degree of the protective effect of post-ischaemic hypothermia was reduced in the lateral and medial CA1 areas. DNA fragmentation was also observed at 1 month. The errors in the 8-arm radial maze trial were increased at 1 month. These data may indicate that cells in the CA1 area are very vulnerable to ischaemia and die after post-ischaemic hypothermia, and that their death is associated with apoptosis.

    Notes: We have studied both continuous-wave and time-resolved photoluminescence from type-II GaP/AlP/GaP quantum wells with thickness of 1, 2, 3, 5, and 8 monolayers. Highly efficient no-phonon luminescence was observed at low temperatures, indicating long-lived temporal behavior. Photoluminescence results indicated that the lowest confined electron states in the AlP wells were the Xz states. Nonexponential time decay of the no-phonon line suggested that the high efficiency of luminescence was due to the localization of indirect excitons by fluctuations in the potential at the interfaces. The effective interface roughness, which gave rise to the in-plane localization of the excitons, was much less than 1 atomic layer. A fit to the observed type-II transition energies gave a value for the conduction band offset of 0.38 eV for the GaP/AlP heterointerface. © 1997 American Institute of Physics.

    Notes: Abstract The efficacy of the epipodophyllotoxins VP-16 and VM-26 is limited by the occurrence of drug resistance in the tumor cell population. Cellular insensitivity to drugs that stabilize the cleavable complex is frequently expressed as multidrug resistance (MDR). In some cell lines, overexpression of MDR-1/P-glycoprotein or the multidrug resistance associated protein (MRP) has been demonstrated and implicated as the mechanism of resistance. Typically, these cells have reduced drug accumulation, secondary to increased drug efflux. In other cell lines, an atypical MDR phenotype has been identified, with the predominant mechanism of resistance shown to be qualitative and/or quantitative changes in the levels and activity of topoisomerase II. For VP-16, increased expression of MDR-1 or MRP and alterations in topoisomerase II α have been shown to confer tolerance. To further understand resistance to VP-16, T98G-VP(1000) was initially isolated as a single clone from parental cell, T98G, by exposure to VP-16. Subsequently, a population of cells from this subline was exposed to three-fold higher drug concentration allowing stable sublines to be established at higher extracellular drug concentration. Characterization of the resistant sublines demonstrates the adaptation that occurs with advancing drug concentrations during in vitro selections. Reduced topoisomerase II mRNA and protein levels were observed in the initial isolate. This reduction was accompanied by a decrease in topoisomerase II activity and cellular growth rate and was associated with 47-fold resistance to topoisomerase II poisons. With advancing resistance, MRP expression increased, with increased VP-16 efflux and reduced accumulation. This adaptation allowed for partial restoration of topoisomerase II activity secondary to increased expression and hyperphosphorylation, with a resultant increase in growth rate. In this cell line, hyperphosphorylation coincided with increased casein kinase II mRNA protein levels, without increased PKC protein levels, suggesting a role for this kinase in the acquired hyperphosphorylation. In this cell line, hyperphosphorylation mediated the increased activity despite a fall in topoisomerase IIα protein levels secondary to an acquired 615 bp deletion in one topoisomerase IIα allele, which resulted in reduced protein levels. In this subline, high levels of resistance were attained as a result of synergism between the reduced topoisomerase IIα levels and MRP overexpression. These studies demonstrate how cellular adaptation to increasing drug pressure occurs and how more than one mechanism can contribute to the resistant phenotype when increasing selecting pressure is applied. Reduced expression of topoisomerase II is sufficient to confer substantial resistance early in the selection process, with synergy from additional mechanisms helping to confer high levels of resistance.

    Notes: Abstract Geochemical association of137Cs and239,240Pu in the surface sediments from two oligotrophic and mesotrophic lakes in Japan was studied by sequential chemical extractions. The fractions separated were exchangeable and bound to carbonate, bound to oxides, bound to organic mattersulfides, and residual. The137Cs and239,240Pu were mainly associated with the organic matter-sulfides and residual fractions but exhibited the different vertical profiles at both lakes. The differences in association patterns between the lakes may be controlled by transport of these nuclides by organic settling particles to the bottom sediments and decomposition of the organics in the sediments.
