    Notizen: A case of secondary localized cutaneous amyloidosis in solar elastosis was studied by light and electron microscopy. Amyloid deposition was restricted to areas with elastotic changes, and in some areas both changes were intermingled with each other. The amyloid was permanganate-sensitive, and proved to be protein AA. Ultrastructurally it was composed of fine tubular filaments. It is suggested that the amyloid deposition in this case was related to sunlight damage of the skin.

    Notizen: Background  Recently, the novel antimicrobial peptide named dermcidin (DCD) was reported in human eccrine sweat glands.Objectives  We investigated the expression of DCD in a variety of cutaneous tumours in order to assess the usefulness of the monoclonal antibody (G-81), which recognizes a fragment of DCD.Patients/methods  We studied the immunoreactivity of the G-81 antibody on 197 cutaneous tumours.Results  A total of 13 of 26 cutaneous mixed tumours showed substantial immunoreactivity. In contrast all the following cases were completely unreactive: (i) epithelial tumours (seborrhoeic keratosis, squamous cell carcinoma, Bowen's disease, actinic keratosis, genital Paget's disease); (ii) follicular tumours (basal cell carcinoma, trichilemmoma, trichoepithelioma, trichoblastoma, keratoacanthoma, proliferating trichilemmal tumour, pilomatricoma); (iii) melanocytic tumours (malignant melanoma, naevus cell naevus, Spitz naevus, blue naevus); (iv) neural tumours (schwannoma, neurofibroma, Merkel cell neoplasm); (v) mesenchymal tumours (soft fibroma, dermatofibroma, dermatofibrosarcoma protuberans, vascular leiomyoma, leiomyosarcoma, lipoma, juvenile xanthogranuloma, angiomyoma); and (vi) other sweat gland tumours (poroid neoplasms, syringoma, cylindroma, clear cell hidradenoma, spiradenoma, syringoid eccrine carcinoma, mucinous carcinoma, apocrine cystadenoma, syringocystadenoma papilliferum, apocrine adenocarcinoma). Twenty-six cutaneous mixed tumours were considered from histopathological findings to be the apocrine type, but 13 of 26 mixed tumours contained some DCD-immunopositive cells that possibly differentiate into eccrine secretory glands.Conclusions  We found the expression of DCD in tubular structures of 50% of cutaneous mixed tumours with apocrine differentiation. These results suggest that a number of cutaneous mixed tumours show both eccrine and apocrine differentiation in the same neoplasm.

    Notizen: Enhanced expression of decay-accelerating factor and CD59/homologous restriction factor 20 in intestinal metaplasia, gastric adenomas and intestinal-type gastric carcinomas but not in diffuse-type carcinomas Aims: Variable expression of the complement regulatory proteins, decay-accelerating factor, CD59/homologous restriction factor 20 (HRF20) and membrane cofactor protein has been shown in human gastrointestinal malignancies, but their expression in gastric cancer has not been fully described. Thus, we immunohistochemically defined the distribution of these proteins in human normal gastric mucosa, intestinal metaplasia, adenomas and gastric cancers. Methods and results: Gastric tissues were obtained by endoscopic biopsy or surgical resection and stained with mouse monoclonal antibodies to decay-accelerating factor, CD59/HRF20, and membrane cofactor protein. In the normal gastric mucosa, membrane cofactor protein was diffusely stained on the basolateral surface of epithelial cells, whereas the expression of decay-accelerating factor and CD59/HRF20 was inconspicuous. In intestinal metaplasia, adenoma and intestinal-type gastric carcinoma cells, decay-accelerating factor and HRF20 were intensely stained on the apical surface; membrane cofactor protein retained its location on the basolateral surface. In diffuse-type gastric carcinomas, the expression of decay-accelerating factor, CD59/HRF20 was lost, but membrane cofactor protein was present on the tumour cell surface. Conclusions: These findings suggest that membrane cofactor protein plays a primary role in the regulation of complement activation in normal and neoplastic gastric cells and that the expression pattern of the complement regulatory proteins is closely related to gastric carcinoma development.

    Notizen: The surface structure of altered elastic libers and elastotic material in solar elastosis has been described using a scanning electron microscope. There were 3 different areas in the dermis, depending on the degree of changes in elastic fibers: areas of (1) normal-looking fibers, (2) thick, cylindrical fibers, and (3) a large mass of markedly changed fibers. They were mainly located in the deep, middle, and upper dermis, respectively. The normal-looking fibers were the same as the normal elastic fiber. The thick, cylindrical fibers showed various findings ranging from relatively normal to heavily damaged surface. The large mass was composed of a cord-like element and a fine net-like one. There was a transition between the thick, cylindrical fibers and the large mass. It is suggested that the thick, cylindrical fiber is an early event and the mass is more damaged, as one of the elaslolic changes in solar elastosis. It is also suggested thai the thick, cylindrical fiber corresponds to the fibrous form ami the mass to the amorphous form of the elastotic material.

    Notizen: Abstract Fluorescent microscopy has been used to separate autotrophic from heterotrophic microorganisms in environmental studies. We report on an improvement on this method for measuring standing stock of phytoplankton. Our method employs a membrane filter and can measure numbers of living phytoplankton including fragile flagellates and size of phytoplankton cells. This method was successfully used in Tokyo Bay and Kuroshio regions.

    Notizen: Evaluation method of nano-scale internal cracks by ultrasonic atomic force microscopy (UAFM) is proposed based on two approaches. The first one is a linear vibration analysis of the contact stiffness calculated from a finite element method analysis of a model including a subsurface gap. The second one is a nonlinear vibration analysis of a stiffening or softening spring representing the opening-and-closing behavior of the gap. These methods were verified by obtaining the resonance frequency mapping, the load dependence of the resonance frequency and the resonance spectra in UAFM on a subsurface gap in highly oriented pyrolytic graphite. As a result, it was proved that the proposed method is useful for evaluating the opening-and-closing behavior of the gap. Although the present study is focused on a nano-scale gap, this method is applicable to larger scale cracks using a larger tip and more stiff support than those used in AFM

    Notizen: Abstract Epidemiologic studies have indicated the association between tobacco smoking and skin aging, but the exact mechanism of tobacco smoke-induced premature skin aging is currently unknown. In this study, we investigated the alterations of collagen, matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) in human fibroblasts treated with tobacco smoke extract. Human fibroblasts were exposed to different concentrations of water-soluble extract from tobacco smoke. Human fibroblasts irradiated with ultraviolet A1 (UVA1) were used as positive controls because the mechanism of UVA1-mediated MMP expression has been well characterized. The expression of MMP and TIMP was analyzed semiquantitatively following reverse transcriptase-polymerase chain reaction. Production of type I and type III collagens was detected by Western blotting and biosynthesis of new collagen was assessed by 3H-proline incorporation. Upon treatment with tobacco smoke extract or UVA1 irradiation, the expression of MMP-1 and MMP-3 mRNA was significantly increased in a dose-dependent manner. Maximum induction was observed with 25 μl/ml tobacco smoke extract. In contrast, the expression of TIMP-1 and TIMP-3 mRNA remained unchanged. Western blotting of the supernatant revealed that type I and type III collagens were decreased as compared with untreated controls. Collagen biosynthesis was significantly reduced by 40.1% following treatment with ¶25 μl/ml tobacco smoke extract. Sodium azide, l-ascorbic acid and Trolox (a water-soluble vitamin E) prevented both the UVA1- and the tobacco-induced alteration of MMP-1. These observations suggest that the imbalance of connective tissue matrix components might contribute to the molecular basis for premature skin aging in smokers. They also suggest that reactive oxygen species including singlet oxygen mediate this process.

    Notizen: Summary A rare case of operatively verified intracranial adenoid cystic carcinoma is reported. In this case, haematogeneous metastasis to the brain parenchyma was suspected. The computer tomography showed a large hypodense mass in the left frontal lobe. The lateral wall of the tumour was irregularly enhanced with contrast media.
