    Notes: Summary A defined medium was developed which, by means of a specific fed-batch mode, allows growth of the recombinant Escherichia coli strain TG1 (pBB210) up to a cell density of 60 g dry weight/l. Apart from glucose and aqueous ammonia fed as carbon and nitrogen sources, it was unnecessary to supply other nutrients or O2-enriched air. Aqueous ammonia also served for pH control. The pO2 level was kept at 20% saturation via closed-loop controls operating the two output variables of stirrer speed and glucose feeding rate. This fed-batch method prevented significant accumulation of acetate and other metabolic by-products. The recombinant E. coli expressed interferon alpha 1 more efficiently at a lower specific growth rate (μPr ≈ 0.15 h−1) than at the maximum specific growth rate (μmax = 0.45 h−1). Therefore, fermentation in the batch phase at μmax was only allowed to continue up to a medium cell density. In the succeeding fed-batch phase, the specific growth rate was reduced to μPr by increasing the stirrer speed according to an empirically developed time scale.

    Notes: Zusammenfassung 4989 Carcinomfälle unter 25147 Sektionen der Jahre 1920–1953 werden hinsichtlich ihrer Verteilung auf Luiker und Nichtluiker nach Häufigkeit, Organlokalisation, Alter und Geschlecht eingehend statistisch bearbeitet. 1. Unter den Luikern findet sich eine signifikant geringere Häufigkeit des Carcinoms (12,7%) als bei den Nichtluikern (20,2%). 2. Organverteilung und Alterszusammensetzung der Carcinome bei Lues unterscheiden sich nicht grundsätzlich von der der Nichtluiker und der Carcinome insgesamt. Das Geschlechtsverhältnis dagegen weicht mit einer stärkeren Beteiligung der Männer (1,5∶1) von dem der Nichtluikercarcinome ab (1∶1,1) und entspricht genau dem der Gesamtluesfälle. 3. Nur eine einzige, dem Umfang nach kleine Sektionsstatistik zur Untersuchung der Frage des Zusammentreffens von Lues und Carcinom liegt in der Literatur vor; sie kommt zu einem ähnlichen Ergebnis wie wir. Die Fälle der zahlreichen, einschlägigen, ganz vorwiegend klinischen und kasuistischen Veröffentlichungen, die meist für einen krebsbegünstigenden Einfluß der Lues eintreten, sind einseitig ausgewählt und bei kritischer Durchsicht nach statistischen Gesichtspunkten in ihren Relationen nicht auf die personelle Gesamtheit übertragbar. 4. Bisher bestehen weder für eine kanzerogene noch für eine krebsverhütende Wirkung der Lues eindeutige theoretische oder experimentelle Grundlagen. 5. Die statistischen Ergebnisse geben zunächst keinen genügenden Anhalt für einen direkten Zusammenhang zwischen Lues und Carcinom. Ob tatsächlich das Carcinom bei Luikern etwas seltener vorkommt als bei Nichtluikern, bedarf erst noch der Bestätigung durch weitere vielfältige entsprechende Erhebungen an großem Material. Sicher aber könnte dafür nicht die geringere Lebensdauer der Luiker verantwortlich gemacht werden, die sie das durchschnittliche Krebsalter nicht erreichen ließe; denn Luiker ohne und mit Carcinom zeigen die gleiche Altersverteilung.

    Notes: Abstract Development of efficient methods for synthesis of oligonucleotides and oligonucleotide analogs has opened up the possibility of designing a broad spectrum of affinity reagents for specific modification of nucleic acids and proteins. These affinity reagents are used for investigation of the topology of ribosomes and nucleic acid polymerases. Oligonucleotides and their analogs are already used for suppression of specific gene expression and for elucidation of the physiological role of their products.     Notes: By investigation of a model of benzylpenicillin fermentation it was demonstrated that high fermentation productivity is connected with low stability. The increasing instability is not the result of external disturbances but of peculiarities inherent to the system. The main process variables and the essential parameters were selected by analysis of data obtained from fermentation runs in a pilot plant. By simulation on an analog computer it was shown that the proposed model gives a sufficient description of the kinetic behavior of the fermentation runs. By varying the initial conditions it was possible to obtain two types of fermentation runs. One type is distinguished by a high productivity connected with low initial concentrations of sucrose. An increase of the initial sucrose concentration also causes an increase of productivity. When a certain value of this concentration is exceeded a changeover to a faster growth of biomass takes place combined with the breakdown of productivity. The same phenomenon occurs at a defined initial concentration of sucrose if a disturbance of oxygen transfer rate takes place. Whether the culture stays in the one or in the other state after the disturbance has passed depends on the duration, the strength, and the beginning of the disturbance. In the pilot plant similar behavior can be observed. The sensitivity of fermentation processes to disturbances increases with the increase of productivity.
