    Description / Table of Contents: Summary In 7 female dogs a constriction of the left renal artery was performed according to the Goldblatt technique. Four to 6 weeks after the operation the blood pressure of the dogs was increased. The total renin activity and the osmolality in the urine from the ischemic kidney were enhanced when compared to the urine from the contralateral untouched kidney. The sodium excretion of the clamped kidney was diminished. These results suggest that determination of urine renin activity might be useful in split-renal function studies for the diagnosis of human renovascular hypertension.
    Notes: Zusammenfassung Bei 7 weiblichen Hunden wurde mit einer Goldblatt-Klemme die linke Nierenarterie cingeengt. Der Blutdruck war 4–6 Wochen nach der Operation angestiegen. Im Urin der gedrosselten linken Niere fand sich eine niedrigere Natriumausscheidung als im Urin der rechten Niere. Die tierexperimentellen Untersuchungen lassen eine Bestimmung der Reninaktivität im Urin bei Patienten mit Verdacht auf eine hämodynamisch wirksame Nierenarterienstenose für sinnvoll erscheinen und könnten diagnostische Hinweise geben.

    Description / Table of Contents: Zusammenfassung Es wird ein einfaches, empfindliches Verfahren zur Bestimmung der Plasma-Renin-Konzentration beim Menschen beshrieben, das auf der von Boucheret al. [Canad. J. Physiol. Pharmacol.45, 881 (1967)] angegebenen Methode beruht. 1,0 ml Plasma, 1.0 ml Dowex 50WX2-(NH4)+ und Schaf-Angiotensinogen im Überschuß — gelöst in 2,0 ml EDTA- und NaN2-haltigem Trisphosphatpuffer — werden 15 Std bei 37°C inkubiert. Die Umsetzung des Human-Renins mit dem Schafsubstrat folgte einer Michaelis-Menten-Kinetik und ergab eine 1,5–2fach höhere Angiotensinausbeute als bei der entsprechenden Reaktion mit homologem Substrat. Die Präzision bei Doppelbestimmungen betrug 7,0±6,2% des kleineren Wertes. Die Angiotensin II-Wiederfindungsrate war in dem untersuchten Bereich von 25–200 ng linear und betrug im Mittel 84,4±10%. Bei gesunden Erwachsenen unter freier Kochsalzzufuhr wurde folgender Normbereich ermittelt: 0,9±0,6 ng Angiotensin/ml/h in Ruhe bzw. 2,1±1,0 ng/ml/h nach aktiver Orthostase. Bei gesunden Säuglingen betrug die Plasma-Renin-Konzentration unter ambulanten Bedingungen 3,4±1,6 ng/ml/h und war damit signifikant höher als die Normalwerte der Erwachsenen in Ruhe und nach Orthostase (P〈0,001).
    Notes: Summary A simple, sensitive method for determination of human plasma renin concentration is described. The procedure is based on the method of Boucheret al. [Canad. J. Physiol. Pharmacol.45, 881 (1967)]. One ml plasma and 1.0 ml Dowex 50WX2-(NH4)+ are incubated for 15 hours at 37°C with 150 mg sheep angiotensinogen in 2.0 ml trisphosphate buffer containing EDTA and NaN3. The reaction of the human renin with the sheep angiotensinogen followed the Michaelis-Menten kinetics and yielded a 1.5 to 2 fold amount of angiotensin compared to the reaction with homologous substrate; the recovery of angiotensin was 84.4±10% being linear in the range studied (25–200 ng angiotensin II). The normal values of 22 healthy adults during a free diet were 0.9±0.6 ng angiotensin/ml/h in recumbent and 2.1±1.0 ng/ml/h in upright posture. During ambulatory conditions the plasma renin concentration in healthy babies was 3.4±1.6 ng/ml/h. These values were significantly higher than those in adults in recumbent and upright posture (p〈0.001).

    Notes: Zusammenfassung Nach einseitiger Drosselung der Nierenarterie beim Hund fanden sich höhere Renin-Aktivität und Norepinephrin-Konzentration im Urin der ischämischen Niere als im Urin der Gegenseite. In der Rinde der gedrosselten Niere waren Renin-Aktivität und Granulationsindex der juxtaglomerulären Zellen höher als in der Rinde der ungeklemmten Niere, während der Natriumgehalt niedriger war.

    Notes: Abstract We have recently shown that the angiotensin I converting enzyme (ACE) gene is linked to NaCl-loaded blood pressure in the stroke-prone spontaneously hypertensive rat (SHRSP), and that high-NaCl loading selectively stimulates ACE in the aorta of SHRSP but not in normotensive Wistar-Kyoto (WKY) rats. We therefore investigated the relationship between cardiac ACE and the development of hypertension and left ventricular hypertrophy in response to normal- and high-NaCl diet in these rats. ACE mRNA and ACE activity were measured in left ventricular tissue after completion of hemodynamic characterization of the animals. While SHRSP rats increased blood pressure (P〈0.0001) and heart rate (P〈0.005) in response to high NaCl, blood pressure remained unchanged in WKY. Similarly, relative left ventricular weight increased only in SHRSP after high NaCl (P〈0.002). A significant two- to threefold increase of cardiac ACE mRNA and fourfold stimulation of ACE enzyme activity in response to high NaCl was found in both WKY and SHRSP rats (P〈0.005). The induction of ACE gene expression was significantly more pronounced in SHRSP compared to WKY (P〈0.02), whereas no significant strain differences in left ventricular ACE activity were found after either normal- or high-NaCl diet. Thus, arterial blood pressure and left ventricular weight remained unchanged in the WKY rats despite the activation of left ventricular ACE activity after high-NaCl exposure. These results demonstrate that left ventricular ACE activity is equally upregulated in response to high-NaCl in the normotensive and hypertensive strain, independently from the development of hypertension. We conclude that the pretranslational induction of left ventricular ACE with high-NaCl loading may be important both for the regulation of cardiac angiotensins and kinins and for local therapeutic ACE inhibition in the heart during high-salt status.

    Notes: Abstract To assess the renal haemodynamic response to manipulations of the nitric oxide (NO) system, we examined subtotally nephrectomized (SNX) rats and control rats (CON) 28 days after their operation. Bolus infusions of the NO synthase inhibitor N G-nitro-l-arginine (l-NA) were given intravenously at doses of 2 mg/kg and 10 mg/kg. Blood pressure was measured intra-arterially, glomerular filtration rate was measured by inulin clearance and fractional changes in renal blood flow (RBF) were determined by a Doppler flow probe. Both doses of l-NA caused a similar and dose dependent increase in mean blood pressure in both SNX and CON rats. In contrast, the decrease in RBF and the increase in the renovascular resistance index (RVRI) was less in SNX rats as compared to CON rats (RBF = −70.1±2.2% of baseline vs −52.7±5.2%, P〈0.01; RVRI = +177±9% of baseline vs +243±24%, P〈0.05). These changes were not affected by autonomic blockade (hexamethonium), or by blockade of the angiotensin II receptor (Losartan). The exogenous NO donor sodium nitroprusside (0.5 and 1.5 μg · kg−1 · min−1) lowered mean blood pressure to a similar degree in SNX and CON rats; in contrast, RVRI decreased less in SNX rats (86.9±9.2% of baseline) than in CON rats (68.2±4.6%, P〈0.05). We conclude that the reaction of the renal vasculature to manipulations of the NO system is altered in the SNX rats. The data suggest that in the remnant kidney, renovascular resistance is less dependent on endogenous NO and the vascular bed is less sensitive to exogenous NO.
