Notes: Abstract Two fusion experiments using the heteromyeloma cell line CB-F7 and splenocytes from two diabetesprone BB (BioBreeding) rats at the onset of diabetes resulted in 128 islet cell reactive autoantibodies primarily detected with permeabilized insulin-producing rat insulinoma cells (RIN) by a cellular enzyme-linked immunosorbent assay. Seventy-nine (62%) of 128 RIN cell reactive supernatants exhibited a cross-reactivity with rat splenic lymphocytes. Six stable hybridomas secreting monoclonal ICSA (islet cell surface antibodies) were established, but only one monoclonal antibody, R4B10, showed preferential beta-cell binding. Six monoclonal antibodies showed a dual reactivity as ICA (islet cell cytoplasmic antibodies) detected by immunostaining of pancreatic islet cryosections and as ICSA on the surface of viable islet cells, whereas two reacted only with an ICA-like pattern. One monoclonal ICSA was specifically displaced from the RIN cell surface by sera of type 1 diabetic patients.

Description / Table of Contents: Summary Intravenous application of N1,n-Butylbiguanide made an decrease of the plasma level of adrenalin in diabetic patients when was given a dosis of 6 mg/kg, and made an increase by a dosis of 8 mg/kg; normal persons show no changes. Correspond with clinical observations of the biguanidetherapy these results explained the “smoothing effect” as an inhibition and the “hyperglycemic effect” as a stimulation of adrenalin output.

Description / Table of Contents: Summary According to Negus and Pressman the sphincter systems of the vocal folds and the ventricular folds form a respiratory ”laryngeal double valve function”. Correspondingly, we found a physiological phonation system of the glottis and a pathological-compensatory one of the supraglottis. They appear to be regulated through an automatic phonatory control system with the glottal phonatory function evidently acting as sensor level. In order to confirm this hypothesis, objective voice analyses with glottal-relevant parameters of 26 voice-rehabilitated patients after minimally invasive laser surgery of glottal carcinomas are presented and integrated into a ”hoarseness diagram” with the coordinates roughness and breathiness. Using statistically deliminated acoustic clusters, our data show a qualitative hierarchy of different postoperative phonation mechanisms. They demonstrate the influence of the vibratory capacity of glottal and supraglottal structures on the quality of the vibratory closure. Both functional parameters evidently determine the resulting voice quality in the sense of our hypothesis.
Notes: Zusammenfassung Nach Negus bzw.

Notes: Summary The effect of a 30 min infusion of nicotinic acid on the blood levels of glucose, pyruvate, lactate, FFA, glycerol, catecholamines, and 11-OH-corticosteroids was investigated over 180 min in 73 insulin-dependent diabetics with different clinical states of metabolism and in 7 healthy subjects. The action of nicotinic acid on insulin secretion was measured in vivo in some diabetic and some healthy subjects by means of ILA. Studies on insulin secretion in vitro were done by measuring the insulin release from isolated islets of mouse pancreas. The insulin content of the incubation medium was estimated with the radio-immunological method. — It was shown that nicotinic acid in vitro stimulates insulin release from B cells significantly. In vivo the rise of serum ILA after nicotinic acid infusion was only detectable in healthy subjects and in diabetics of the stable type. Furthermore, a negative correlation was demonstrated between the peak of the FFA-rebound induced by nicotinic acid and the stimulatory effect of the drug on insulin secretion. The behaviour of blood glucose during infusion of nicotinic acid together with the daily insulin dosage permitted a metabolic distinction to be made between diabetics of the stable and unstable (‘brittle’) type.

Notes: Conclusion The rat insulinoma line RIN-5AH-T2 has several advantages over the normal rat islet cells to detect ICSA:1. The RIN cell population is homogenous, whereas PRI cells contain other cell types in addition to B-cells; 2. large quantities of RIN cells can be easily prepared; 3. the rat insulinoma line should make it possible to compare more readily results from different laboratories.
