    Notes: Summary Thirty-five patients (7 females and 28 males) with cerebral infarction and suspicion of cerebral infarction of 4 h to 27 months duration were studied 45 times with magnetic resonance (MR) imaging using Bd-DTPA. Spin echo (SE) images were obtained before and after the administration of Gd-DTPA (0.1 or 0.15 mmol/kg) and compared with the enhanced CT. MR imaging using Gd-DTPA was more sensitive than enhanced CT and very useful for detecting a new focus of cerebral infarction, especially in the cases with multiple infarcted areas and for showing the extent of cortical and subcortical infarction. In most cases the MR enhancement was obvious in the subacute stage, especially after cerebral embolism, and the signal intensity of the lesion tended to show a gradual increase. The diagnosis of embolism was accepted on the basis of acute onset without prior TIA, coupled with angiography showing the embolus itself and/or a capillary blush and a wide area of infarction.

    Notes: Summary The muscarinic cholinergic innervation of the rat gastric mucosa was investigated by localizing the muscarinic receptors using a tritiated muscarinic antagonist, pirenzepine. Radioautography was performed by freeze drying stomach tissue, which was then embedded in Epon and wet sectioned with ethylene glycol, and dry mounting on emulsion film by the wire-loop method to prevent loss of the labelled substance during fixation and the radioautographic procedure. Light and electron microscopy showed that the specific pirenzepine-binding sites were localized predominantly on parietal cells, chief cells and perivascular plexuses. Analysis of the grain distribution on parietal cells revealed that the silver grains corresponding to the pirenzepine-binding sites were mainly on the basolateral plasma membrane. On the other hand, the surface mucous or mucous neck cells had few pirenzepine-binding sites.

    Notes: Abstract Interactions of the mixed surfactant solution of dodecylamido propyl dimethyl aminoacetate and sodium dodecyl sulfate with the liposomal membrane were studied. Lytic activities of the surfactants were measured as a function of the concentrations of surfactant and phospholipid and the composition of mixed surfactants. The solubilization limits of phospholipid by surfactants were determined from the change of their aggregation behavior in suspensions at equilibrium by means of quasi-elastic light scattering. The mixed surfactant solutions showed lower lytic activity than single component surfactant solution in spite of the strong adsorption onto the liposome surface. This was attributed to low solubilization power of binary mixture for phospholipid.

    Notes: Summary When microspheres are injected into the circulation, they are trapped in the arteriolar or capillary system within various organs. It has been confirmed in animal experiments that the number of microspheres in a myocardial sample approximately follows a Poisson distribution, under adequate experimental conditions. On the basis of this result, we arrived at the following hypothesis: When regional blood flow is measured under a steady state by the reference sample method, the 95% relative error can be approximated by $$ \pm 196\sqrt {1/v + 1w} $$ , wherev andw represent the number of microspheres in blood sample and myocardial sample, respectively. The equation is valid ifv andw are greater than 400 and 49, respectively. We obtained an expression of the relation among the percent of increase or decrease in regional blood flow being verified, the probability of increase or decrease, as a statistically significant variation and the number of microspheres in a myocardial sample. An investigator can work out an approximate experimental design using this expression. For instance, when the increase in regional blood flow required for verification is expected to be 20% and he wants this increase to be verified by 90%-probability and as a statistically significant increase, he can predict from this expression that the number of microspheres in a myocardial sample should be 472 and 567 before and after the experimental intervention on the coronary circulation, respectively. The expression is useful as index for experiments involving use of the microsphere method.

    Notes: Summary Outer diameter of the isolated pig coronary artery was measured under isotonic conditions using a sonomicrometer, and tension development of a ring preparation was assessed isometrically by a strain gauge transducer. Potassium chloride (KCl), acetylcholine (ACh) and 5-hydroxytryptamine dose-dependently reduced the coronary outer diameter. There was a direct linear relation between the reduction of diameter and tension development by these agents (r=0.91, n=15, p〈0.001). KCl was one of the most potent vasoconstrictors tested and reduced the outer diameter by 35±1% (n=15, mean ± SEM) at 120 mM. ACh produced a transient increase in isometric tension and the reduction of the coronary diameter by ACh was smaller than that seen with KCl, even with a dose which produced the similar peak level of isometric tension by KCl and ACh. It is concluded that changes in the coronary diameter do closely correlate with those in tension development, however, the reduction of coronary diameter after exposure to vasoconstrictive agents is less in phasic than tonic contraction. The latter evidence suggests the importance of both the sterngth and duration of active vasoconstriction in eliciting coronary spasm.
