    Notizen: The treatise “De morbis artificum diatriba” (Modena, 1700) is considered to be the first text to specifically deal with occupational illnesses. It was also the last for over 150 years. Written by Bernardino Ramazzini (Carpi, 1633-Padua, 1714), a professor at the University of Padua from 1700 to 1714, the book highlights the importance given at the time to headache as an occupational symptom. Among the 69 professions described, accounting for the majority of the occupations of the period, 12 were found to lead to headache as an important symptom caused by work. Ramazzini appears to have paid more attention to this than we do today. Ramazzini's work opens up a wide view on social conditions in the 18th century, as his sensitivity for occupational hazards was exceptional. His remarks on headache are typical of his way of collecting first-hand experience of working conditions, and they underline the importance of occupational hazards in the assessment of headache, today just as in 1710.

    Notizen: The linear Kelvin–Helmholtz stability regime of two radiating fluids in relative motion is discussed. The study is carried out in plane geometry and in the vortex-sheet approximation. Attention is focused on the effects of radiation losses on the instability pattern: it is found that the principal effect is the suppression of the cutoff at Mach number M=2(square root of)2, typical of the adiabatic Kelvin–Helmholtz mode; in particular radiation effects are small on the instability below the cutoff, while they become dominant above. Radiating fluids are also typically affected by thermal instability. In fact thermal modes are found also in the present case: they may have growth rates comparable to the Kelvin–Helmholtz instability above the cutoff.

    Notizen: Concanavalin-A-activated T cells and their crude supernatant were assayed for suppressivc activity un an IgE-producing U-266 cell line. Delectable and comparable degrees of suppression were obtained with the co-culture and the supernatant protocols. Separation of the effector population into T4+ and TS+ subsets showed the most effective cells in the T8+ fraction. Control experiments demonstrated that the IgE down-regulation was selective, since parallel measurement of β2-microglobulin synthesis showed no effect of T cells or T-cell-derived supernatants. In addition, several human T-cell lymphoma-leukaemia virus I-transformed T-cell lines were explored for their capacity to produce factor(s) able to suppress IgE synthesis in the U-266 cell line, and four out of 25 cell lines could be shown to do this in a constitutive manner. Kinetic studies suggested that the inhibition occurred at a transcriptional level, The results indicate that the T-cell-mycloma system is an interesting model to define belter the regulation of IgE in the human.

    Notizen: In patients with Crohn's disease (CD) we investigated the C3 conversion of zymosan-activated scrum (ZAS) and looked for the occurrence of chemotactic factor inactivation (CFI). We also studied the cell-directed inhibitory effect (CDI) of the CD patients' plasma and, in the same group, complement activation and complement-mediated deactivation. The mean value of ZAS C3 conversion in CD was no different from that of healthy controls, but in steroid-treated patients it was lower than in untreated CD. CFI occurred in 1 of the 23 CD sera tested, and CDI was observed in 6 out of the 22 patients tested. EDTA C3 conversion was present in 12 patients, and complement-mediated deactivation was associated with high values of EDTA C3 conversion. Our findings indicate that complement dysfunction and inhihitory factors of neutrophil chemotaxis are present in CD. These findings could explain the defective neutrophil migration into skin windows. Whether they are relevant to the pathogenesis of tissue injury or of infectious complications and are specific for CD, however, remains to be established.

    Notizen: Mononuclear cells isolated from peripheral blood of normal donors produce free oxygen radicals (FR), delectable by chemiluminescence (CL). when interacting with target cells during natural killer (NK) cell lysis. FR-producing cells were found to have monocyte characteristics and gave a positive CL reaction when mixed at low concentration (0.5%) with purified NK cells. No correlation was found between susceptibility to NK cell lysis and capacity to induce CL with different target cell lines. Using high and low molecular FR scavengers, no NK cell inhibition was seen with superoxide dismutase, cytochrome c, and catalase, whereas some inhibition was seen with 4,5-dihydroxy-in-benzcnedisulphonic acid (Tironø) and 2,3-dihydroxybenzoatc. These compounds, however, required higher concentraiions than used for inhibition of CL, suggesting an alternative action of these compounds. Normal levels of NK cell activity were found in two patients with chronic granulomatous disease, who were genetically incapable of producing detectable amounts of FR. As a result, it is concluded that human NK cells do not produce large amounts of FR during killing and that FR are unlikely to be the lytic end product. Nevertheless, neither a low degree of FR formation in NK cells nor a more subtle signal-transmitting role of FR during NK cell triggering can be excluded.

    Notizen: Human natural killer (NK) cells were tested in the presence of several fatty acid oxygenase inhibitors such as nordihydroguaiaretic acid (NDGA), 5,8,11,14-eicosatetraynoic acid, 3-amino-l-m-(trinuoromethyl)-phenyl-2-pyrazoline (BW 755C). and indomcthacin. All drugs inhibited NK lysis at the post-target cell-binding level at concentrations that also suppressed lipoxygenation of arachidonic acid, suggesting that such reactivity may be required for effector cell triggering. NDGA gave a 50% NK cell inhibition in the range of 10-30 μM and also suppressed antibody-dependent and lectin-dependenl systems. Further evidence of the involvement of arachidonic acid lipoxygenation was found as NK activity could be reconstituled to NDGA-suppressed effector cells with several metabolites such as LTB4. LTB4 analogues, and hydroxyeicosatetraenoic acids lipoxygenated at C-5, C-12, and C-15. Cyclic nucleotides such as cGMP and cAMP could also reconstitute activity with optimal effects at approximately 10−8 M. The combined evidence is compatible with a model for triggering lysis in which lipoxygenation products have a second messenger function. Whether arachidonic acid lipoxygenation is necessary for effector cells at all different activation/differentiation stages and whether the lipoxygenation products activate guanylic cyclase, protein kinase C, or some other target molecule remain to he further investigated.

    Notizen: The chemotactic and random mobility functions of twelve selectively IgA-deficient patients were evaluated by a method using agarose gel. A severe polymorphonuclear cellular chemotactic defect was found in ten out of twelve patients, but only five of them also showed a marked associated impairment of random locomotory function. Furthermore, in one subject, levamisole therapy resulted in a dramatic improvement of both chemotactic and random mobility functions. These results are discussed in the paper with respect to the possible pathogenetic implications.
