    Notizen: The Cretaceous Abukuma metamorphic terrane consists of the oceanic Gosaisyo Series overthrust onto the terrigenous Takanuki Series. Although the dominant mineralogy defines one of the classic areas of andalusite–sillimanite type progressive metamorphism, there are several lines of evidence suggesting an earlier higher-pressure (up to c. 12 kbar) history of the Takanuki Series and the nearby Gosaisyo Series. These are: (1) the occurrence of rare although widespread relic kyanite in sillimanite+K-feldspar zone-grade pelitic rocks; (2) the high grossular content of garnet interiors (up to c. 30 mol %) overgrown by Ca-poor rims (c. 2 mol % grossular) in pelitic rocks containing Al2SiO5 minerals (sillimanite±relic kyanite±retrograde andalusite), plagioclase and quartz; (3) the occurrence of rutile as inclusions in garnet in pelitic rocks; and (4) the occurrence of relic corundum+almandine association in silica-poor and Al–Fe-rich rocks. Garnet in the Takanuki Series pelitic rocks commonly shows textural sector zoning and preserves growth zoning despite the high metamorphic grade, suggesting rapid changes in P–T  conditions and a relatively short duration of high-temperature conditions. Combined with radiometric dating, these observations suggest that the Abukuma sillimanite+K-feldspar zone-grade rocks underwent a clockwise P–T  path with very fast (〉4 mm y−1) average burial and exhumation rates.

    Notizen: Background It is well-known that FK506 strongly inhibits cytokine production by T cells in vitro. However, less evidence is available from in vivo studies of ocular allergy.Objective To study the anti-inflammatory effect of FK506 eye drops on late and delayed-type responses in several animal models of ocular allergy.Methods Rats and guinea-pigs were sensitized with egg albumin (EA) in adjuvant and later challenged by topical EA application to their eyes to examine the late response. Biopsy specimens of conjunctiva were stained with haematoxylin–eosin or stained for T cells and eosinophils. In addition, rats, rabbits and guinea-pigs were sensitized with complete Freund's adjuvant and later challenged by injecting purified protein derivatives for the delayed-type response. Bulbar conjunctival oedema and hyperaemia were graded by score in rabbits, and Evans blue (EB) extravasation was measured in rats and guinea-pigs. FK506 (0.01–1%) and steroid (0.1%) eye drops were instilled in the eyes of animals several times, before and after challenge.Results FK506 eye drops inhibited T cell and eosinophil infiltration in the late response and EB extravasation in the delayed-type response in rats. Also, they inhibited conjunctival oedema, hyperaemia and ocular mucus in the delayed-type response in rabbits. These effects were similar to those of steroid eye drops (betamethasone sodium phosphate, fluorometholone). FK506 eye drops also inhibited inflammatory cell infiltration, the loss of conjunctival epithelium and decrease of goblet cells in the late response as well as EB extravasation in the delayed-type response in guineapigs, a steroid-resistant species.Conclusion FK506 eye drops inhibit late and delayed-type responses in animal models of ocular allergy.

    Notizen: Abstract. Objective and Design: Recently, Ogawa et al. [17] reported that the peritoneal mast cells (PMCs) of rats can release histamine by substance P (SP) in a receptor-dependent manner. In the present study, we confirmed and extended their findings.¶Material: PMCs were isolated from six strains of rats. In some experiments, peritoneal cells in the non-MC fraction were used.¶Methods: PMCs were incubated with SP, neurokinin (NK) receptor agonists or antagonists, and histamine content in the supernatant was measured. In the binding assay, PMCs were incubated with [125I]BH-SP together with SP or NK receptor antagonists. NK-1 receptor mRNA was detected using a reverse transcription-polymerase chain reaction (RT-PCR) assay.¶Results: PMCs from Slc:Wistar and F344/NSlc were highly sensitive to SP, leading to histamine release, whereas those from Slc:SD and three other strains were not. PMCs from Slc:Wistar and F344/NSlc also released histamine in the presence of an NK-1 agonist. The histamine release induced by SP and the NK-1 agonist was inhibited by the NK-1 receptor antagonists, FK888 and CP-99,994. [125I]BH-SP binding experiments revealed that PMCs from Slc:Wistar rats possessed a single high affinity binding site for SP and that the binding was blocked by NK-1 receptor antagonists. Peritoneal cells in the non-MC fraction exhibited no appreciable binding. In the RT-PCR assay, expression of NK-1 receptor mRNA was evident in Slc:Wistar PMCs, but not in the non-MC fraction from Slc:Wistar or Slc:SD PMCs.¶Conclusion: These data demonstrate the existence of functional NK-1 receptors on freshly isolated PMCs in at least some strains of rats.

    Notizen: Abstract The alteration of bone metabolism in the femur of rats with skeletal unloading for 4 days was investigated. Skeletal unloading was designed using the model of hindlimb hang in rats. Skeletal unloading caused a significant decrease in femoral weight, calcium, and phosphorus contents in the metaphysis but not diaphysis. Also, the unloading induced a significant decrease of zinc content, alkaline phosphatase activity, and deoxyribonucleic acid (DNA) content in the femoral diaphysis and metaphysis. When the femoraldiaphyseal and metaphyseal tissues from normal and skeletal-unloading rats were cultured in the presence of insulin (10-9 and 10-8 M) for 24 hours in vitro, the hormonal effect to increase alkaline phosphatase activity and DNA content in the diaphysis, but not metaphysis, was lost in the bone tissues from unloading rats. However, the culture with insulin-like growth factor-I (IGF-I; 10-8 and 10-7 M) produced a significant increase of alkaline phosphatase activity and DNA content in both the diaphyseal and metaphyseal tissues from normal and unloading rats. These results demonstrate that skeletal unloading causes an impairment of insulin effect, but not IGF-I effect, on bone metabolism in femoral tissues.

    Notizen: Summary Serial passage of bovine ephemeral fever virus in hamster lung cells, HmLu-1, deprived rapidly the virus of virulence for cattle with simultaneous loss of the ability to produce neutralizing antibody. The attenuated virus thus obtained, however, could prime cattle to produce neutralizing antibody rapidly to high titers following a subsequent inoculation of formalin-inactivated, AIPO4 gel-adsorbed vaccine (K). Although one subcutaneous dose of live vaccine (L) prepared from the attenuated virus produces no detectable amounts of neutralizing antibody unless as heavy a dose as 107 TCID50 was used, the priming effect of L vaccine was shown even with 10 TCLD50. Similar priming effect was also shown in mice by intracerebral inoculation of L vaccine. Maximal antibody response of cattle was obtained by one subcutaneous L vaccine dose of ≥ lO5]TCID50 followed by 3 ml of K vaccine administered intramuscularly at intervals of 2 to 4 weeks. This LK method induced seroconversion in nearly all of initially seronegative cattle, and the neutralizing antibody thus produced persisted much longer than that following two doses of K vaccine given at one-month intervals. The LK method exerted little side effects in cattle. Cows vaccinated during pregnancy delivered healthy calves in term. No adverse effect on milk production was shown. The vaccine virus could not be passaged serially in calves by intravenous inoculation of the blood. Lyophilized L vaccine was stored at 4

    Notizen: Summary.  In order to determine in what condition and by what mechanism gp 120 can deplete not only CD4 but also CD8 T cells, an in vitro system was established in which peripheral blood lymphocytes from healthy donors were treated with recombinant gp 120. We found that gp 120 can deplete both CD4 and CD8 T cells when they have recently been activated and are exposed to IL-2-deficient conditions. Bioassay of the Fas ligand (FasL) demonstrated augmented expression and release of soluble FasL by CD4 T cells in the supernatant of this culture. The administration of anti-FasL mAb and anti-Fas mAb, both of which exhibit neutralizing activity, completely abolished the depletion of these two T cell populations in culture. Based on these findings, we concluded that FasL depletes Fas antigen expressing CD4 and CD8 T cells by programmed cell death.
