    Notes: Summary The Maudsley Reactive and Non-Reactive strains have been developed as a model for the study of individual variations in stress-reactivity, and many differences in biobehavioral systems have been found between them. This review discusses limitations of the ‘emotionality’ construct in accounting for differences between the Maudsley strains and offers an alternative, theoretical approach. Amaral and Sinnamon have proposed that the locus ceruleus (LC) plays a stress-attenuating role in mediating behavioral, physiological and neuroendocrine response to prepotent, emergency-provoking stimuli and, building upon this formulation, it is proposed that the LC has been an important focus for gene action in the Maudsley model. It is suggested that the LC of the Non-Reactive strain is more strongly activated by stressful stimuli than the LC of Reactive rats, and is the basis of many of the behavioral and physiological differences between them. Behavioral and biochemical evidence consistent with this proposition is reviewed. Identification of the LC as a target for gene-action in the Maudsley model has an important advantage. It substitutes variations at a specific anatomic location in the brain for a loosely defined construct like emotionality, and the hypothesis is amenable to empirical tests by a variety of experimental approaches.

    Notes: Summary The subject of heart development has attracted the interest of many embryologists over the last two centuries. As a result, the main morphologic features of the developmental anatomy of the heart are already well established. Although there are still some controversial points, and there is probably much descriptive work yet to be done, emphasis is currently being placed on developmental mechanisms rather than simply on descriptive facts. The availability of new techniques and the overall advances in biological research are placing heart embryology in a new perspective. Today, we do not simply ask whether one or another embryonic structure arises further right or further left; instead, we are studying how cells, tissues, and their microenvironment interrelate at the several levels of biological organization (from the gene upwards) so as to give rise to a mature organ with a distinct shape and well-established functions. This paper attempts to review some of the basic aspects of the developmental anatomy of the heart. Descriptive embryology is used here as a tool. Emphasis is placed on developmental mechanisms, and on the present knowledge of how these mechanisms are related to the structural development of the heart.

    Notes: Summary Cardiac development involves a complex integration of subcellular processes into multicellular and, finally, whole organ effects. Until recently it has been difficult to investigate the genetic control of this organ level differentiation of the heart. The proliferation of molecular biology methodologies has provided mechanisms to directly investigate the control of these processes. This article focuses on molecular lines of research on two key areas in cardiac development: the regulation of expression of sarcomeric contractile and regulatory proteins, and atrial natriuretic factor. Molecular approaches are described which have allowed investigators to begin to determine the tissue and stage-specific expression of genes, to locate those genes in the genome, determine their sequences, and to directly investigate the mechanisms controlling their expression.

    Notes: Summary The variation in immune responses to standard inoculation of the hepatitis-B virus vaccine suggest that host factors influence response in ways that are not presently understood. We studied 25 low/nonresponding health care workers (anti-HBs titer 〈50 IU/l) after the third inoculation of an experimental hepatitis-B vaccine to determine their immune status (through lymphocyte phenotypes) and HLA type. After application of a fourth inoculation, the seroconverting subjects showed only low anti-HBs levels; three male subjects remained anti-HBs negative. Twelve months after the fourth inoculation only 9 of 25 subjects (36%) maintained anti-HBs titer 〉10 IU/l. Almost all subjects had normal B-cell and CD-4 and CD-8 counts and ratios. Relative to other European populations HLA-A-10 (P〈0.05), B-12 (P〈0.025), CW-5 (P〈0.05), DR-3 (P〈0.025), and DR-5 (P〈0.025) were increased, whereas DR-2 (P〈0.05) was decreased. However, after correction of theP-values for the number of HLA antigens determined, these differences were no longer significant. Furthermore, these HLA types were not the same as those reported in other studies (except for DR-3). We suggest that larger sample sizes or even not yet available immunogenetic markers will be required to prove an “immunogenetic background” in low/nonresponders, if it exists.

    Notes: Summary In the last five years, genetic markers for a large number of diseases have been localised using linkage analysis of DNA polymorphisms in affected families. The site of the genetic defect or defects leading to Type 2 (non-insulin-dependent) diabetes mellitus, a common illness with a major genetic component, remains unknown. This is due, at least in part, to the lack of large well-defined Type 2 diabetic pedigrees suitable for linkage analysis. There are several features of the disease which make large pedigrees difficult to find. The late age of onset of most probands means that informative older generations are often dead, while there is difficulty in detecting disease in younger generations. The diagnostic criteria for diabetes are, as yet, dependent on an arbitrary cut-off along a continuum of plasma glucose. The high prevalence of the disease may also produce problems as, in any given family, diabetogenic genes may be contributed by more than one parent. Varieties of the disease with a well-defined inheritance, such as maturity onset diabetes of youth, are more suitable for linkage analysis but might be due to defects at a different gene locus. Despite these difficulties, once large well-defined pedigrees have been found, linkage analysis using both candidate genes and random highly polymorphic markers is the strategy most likely to find genetic markers for the disease.

    Notes: Summary The possibility of linkage between the human insulin receptor gene locus and diabetes was examined in three Type 2 (non-insulin-dependent) diabetic families and one family with maturity onset diabetes of the young. Insulin receptor gene haplotypes were established using BglII, Rsal and Sstl restriction enzyme digests of genomic DNA from all available family members. The digested DNA was subjected to agarose gel electrophoresis, Southern blotted, and hybridised to 32P-labelled human insulin receptor gene cDNA. In the pedigree with maturity onset diabetes of the young, formal linkage analysis allowed exclusion of close linkage between the insulin receptor locus and diabetes (logarithm of the odds for linkage versus non-linkage was −5.35 at recombination fraction of 0.01). This confirms the absence of linkage between insulin receptor and diabetes which has been reported in two similar pedigrees. In the three Type 2 diabetic families there were a minimum of 4 recombinants between the insulin receptor locus and diabetes, which makes a direct role for insulin receptor defects unlikely. The importance of using realistic estimates of penetrance when performing linkage analysis in a disease with a late age of onset is emphasised. In contrast to the one previous linkage analysis study of the insulin receptor gene, no specific association of diabetes with the rare Sstl Sl(-) allele was observed in either the maturity onset diabetes of the young or the Type 2 diabetic families.

    Notes: Summary Protamines were extracted from the sperm of fertile and infertile human males and the relative proportion of protamines 1, 2, and 3 were determined by scanning microdensitometry following electrophoresis of total protamine in polyacrylamide gels. The proportion of the three protamines was found to be similar in sperm obtained from different normal males. The distribution of protamines in sperm obtained from a select group of infertile males producing an elevated level of large sperm heads, in contrast, was different from that of the fertile males.

    Notes: Abstract Two siblings with typical features of the Cockayne syndrome were studied at autopsy. Many glomeruli revealed a paucity of capillary loops and had thickened capillary walls. Some glomeruli with advanced lesions showed collapse of the glomerular tufts or complete hyalinization. Atrophy of tubules and interstitial fibrosis were also observed. There were no significant arteriosclerotic changes in the vessels. Ultrastructural studies demonstrated thickened glomerular basement membranes with bends and folds. These histopathological findings are different to those previously reported with the exception of the 1966 report by Ohno and Hirooka.

Characteristic findings in this autosomal recessively inherited pterygium syndrome are, in addition to multiple pterygia, short stature, cleft palate, vertebral fusion defects and minor facial anomalies. The adult height in the two male siblings was below the third centile. Secondary sexual development and testicular size were normal, in contrast with the cryptorchidism and pubertal delay documented in most young patients.

    Notes: Abstract C57BL/6J and DBA/2J mouse strains have been characterized as morphine preferrers and avoiders, respectively (Horowitz et al. 1977). Previously, sweetened morphine solutions were presented with a water alternative, primarily with male subjects. Because sweetness may affect the endogenous opioid system and rodents have shown strain and sex differences in taste preferences, this study looked for strain- and gender-related taste preferences that might have affected opiate consumption. Preference for sweetened and unsweetened morphine and etonitazene was compared across gender and strain. In all choice tests, the control was a similar tasting quinine sulphate solution. Under these conditions, C57BL/6J mice continued to show strong preference for morphine. However, DBA/2J mice drank approximately equal amounts of morphine and quinine solutions, rather than avoiding morphine as when water was the alternative. Both strains appeared surprisingly indifferent to the synthetic opioid etonitazene, compared because it is potent at concentrations having barely perceptible bitterness. This raises the possibility of unexpected differences in post-ingestional effects between morphine and etonitazene. Contrary to reports of gender differences in sweet preference in rats, none were found in either strain of mouse. Neither were there any significant sex differences in opiate preference in either strain. C57 mice preferred sweetness more than did DBA mice.

    Notes: Abstract Four strains of mice (C57, BALB, DBA, C3H) were used to determine whether genetic factors influence the effects of lithium on hypoactivity induced by a low dose of the alpha2-adrenoceptor agonist clonidine (0.2 mg/kg). Lithium was administered in the diet for 3–4 weeks at a dosage that produced average serum lithium levels of 0.58–0.66 mmol/l. Locomotor activity was reduced by either clonidine or by lithium given alone. When combined, however, lithium attenuated the activity-suppressant effects of clonidine, and that action was influenced by genetic factors. The findings suggest that genetic differences in alpha2-adrenoceptors play a role in behavioural effects of lithium.

    Notes: Abstract Low doses of ethanol (EtOH) stimulate activity in an open field in many strains of laboratory mice. We are selectively breeding two lines of mice to exhibit a large (FAST) response on this test, and two other lines to exhibit a small (SLOW) response (Crabbe et al. 1987). The lines initially diverged in response to EtOH, but despite continued selection pressure, the difference between each pair of FAST and SLOW lines has not increased over generations as much as expected. Our practice has been to test animals on the 1st day after saline injection, and repeat the test after EtOH injection 24 h later. Lister (1987) recently demonstrated that the order in which an animal was exposed to EtOH and saline influenced the magnitude of the response to EtOH, with animals tested initially after EtOH having greater stimulation. Middaugh et al. (1987) recently demonstrated that the magnitude of EtOH stimulation was greater under conditions of relatively bright light than under dim light. Using non-selected Swiss mice, the current experiments essentially confirmed Lister's findings. Using FAST and SLOW mice, the predictions of both groups were tested. Both hypotheses were confirmed. Additionally, these experiments demonstrated that the magnitude of the difference between FAST and SLOW mice was greater under bright light than under dim light. The line difference was also greater when tested in the EtOH-Saline order. In experiments with Swiss mice, the possible role of peritoneal irritation in the EtOH effect was eliminated, and the optimal dose and time for demonstrating the effect was determined. These experiments confirm the importance of lighting condition, order of testing, dose, and route of administration in eliciting EtOH-stimulated open field activity in mice. They demonstrate a genotype-environment interaction, since the magnitude of difference between genetically selected lines varied as a function of the testing parameters chosen. Finally, they indicate that the differences between FAST and SLOW lines in sensitivity to EtOH generalizes to several environmental conditions. We interpret this to mean that the various EtOH-induced activation traits represented by these different environmental and testing conditions are genetically correlated.

    Notes: Summary Within the broader context of our investigations into the heredity of the human EEG, we analysed the EEGs of 28 pairs of monozygotic and 21 pairs of dizygotic twins who were separated as infants and reared apart. The principal goal of this study was to determine the degree to which environmental factors possibly influence the development of a person's EEG. Monozygotic twins reared apart were, with respect to their EEGs, only slightly less similar to each other (if there is any difference at all) than the same person is to himself over time. For dizygotic twins reared apart, we verified the findings of our previous study, namely, that the average within-pair similarity of EEGs estimated from a sufficiently representative sample of fraternal twins was significantly higher than the average inter-individual similarity of EEGs obtained from unrelated persons. The results on both monozygotic and dizygotic twins, yielded conclusive proof that the individual EEG pattern is predominantly determined by hereditary factors.

    Notes: Summary A 4-year-search for spinal muscular atrophies (hereditary motor neuropathies, HMN) in Benghazi, Libya, yielded a total of 24 patients, among whom 18 were index cases. This group comprised 6 acute infantile, 12 chronic childhood, and 3 each with adult-onset proximal, and distal forms of the disorder. Distal HMN constituted 12.5% of the total cases. The crude average annual incidence of acute infantile HMN was 0.3/100,000 total population and 1/12,500 births in Benghazi. The crude prevalence rates of chronic childhood, adult-onset proximal, and distal types of HMN were 2.3, 0.6, and 0.6/100,000 respectively. The segregation ratios, 0.26 for acute infantile HMN and 0.24 for chronic childhood HMN, suggested autosomal recessive inheritance. The consanguinity rates among parents of cases and the population did not differ significantly.

    Notes: Summary After castration, the rat dorsolateral prostate M-40 mRNA initially decreased then rebounded to precastrated levels. The cellular site of M-40 expression and its renewed expression after castration was defined by in situ hybridization histochemistry. In situ hybridization with either a 32P-labeled or biotin-labeled M-40 cDNA probe demonstrated that M-40 mRNA levels were higher in the lateral than dorsal prostate. A second androgen regulated gene, RWB, also was highly expressed in the lateral prostate. The biotinylated cDNA probes provided microscopic resolution of the expressing cells, revealing two distinct morphologies of lateral epithelium which expressed both the M-40 and RWB mRNA. These morphologies appeared in ducts which contained either epithelial cell sheets that were highly convoluted or thinner epithelial cells with a minimal degree of convolution. The RWB mRNA decreased in both cell populations in response to androgen withdrawal. The decline and reappearance of M-40 mRNA also appeared in both epithelial cell types. These data demonstrated that after castration the M-40 mRNA initially decreased as expected for an androgen sensitive gene and then progressed to a fully inducible state. The mechanism of this progression remains to be elucidated.

    Notes: Abstract The molecular basis for the genetic control of variable proportions of the two cat hemoglobins Hb A(α2β 2 A ) and Hb B (α2β 2 B ) was investigated. Ratios of Hb A/Hb B vary between 50/50 and 90/10 among members of the mongrel cat population, with clusters around 50, 35, and 10% Hb B. Genomic DNA from cats of 50/50, 70/30, and 90/10 phenotypes were cut by restriction endonucleasesHindIII,EcoRI,BamHI,Bg1II,and Pst1 and hybridized to a fragment of the human β-globin gene. The results of the Southern blots suggested a pattern of homozygote, heterozygote, homozygote for the respective cat phenotypes, 50/50, 70/30, and 90/10. Therefore, the cat hemoglobin polymorphism seems to result from the possible combinations of an allelic gene pair.

    Notes: Abstract Full-length cDNA clones for barley chymotrypsin inhibitor 1 (CI-1) have been isolated from an endosperm-specific cDNA library. Hybridization and nucleotide sequence analyses indicate that these cDNAs represent two distinct types of CI-1 mRNA which we have called CI-1A and CI-1B. Both mRNAs encode polypeptides of 83 residues (M r=8790 and 8960) which differ at eleven positions. The full-length cDNA sequences do not predict N-terminal signal peptide extensions indicating that CI-1 is synthesized in the mature form in contrast to the homologous proteinase inhibitors of tomato and potato. Northern hybridization experiments show that the CI-1 genes are under strict developmental and organ-specific control. CI-1 transcripts were first detected in the developing barley endosperm between 12 and 14 days after anthesis but no CI-1-related sequences were detected in the RNA preparations from shoots, leaves or roots. The expression of CI-1 was also studied in the high-lysine barley mutants Hiproly, Risø 56 and Risø 1508. Approximately 15-fold (Hiproly) and 4-fold (Risø 56 and 1508) higher levels of CI-1 mRNA were detected in the mutant endosperms compared to normal barley. These results correlate well with the increased deposition of CI-1 in the high-lysine lines and show that the differential expression is controlled mainly at the level of transcription or stability of the mRNA. Using Southern-blots of barley DNA we estimate that there are three copies of CI-1 per haploid genome in both normal and mutant barley lines.

    Notes: Abstract A histone H3 gene was isolated from a dicotyledonous plant, alfalfa (Medicago sativa). The sequence analysis of this gene revealed no obvious GC preference in its codon usage. Apart from containing most of the typical consensus sequences found in both animal and plant histone genes, the alfalfa H3 gene exhibits distinct structural features such as (1) the unusual location of two GATCC motifs in its 5′ flanking sequence, (2) the existence of a CGCGGATC on the nonsense strand at position −232, (3) the existence of a long palindromic structure, and (4) several polyadenylation signal-like sequences in the 3′ flanking region. There are about 160 copies of histone H3 gene in alfalfa tetraploid genome. Using the alfalfa H3 gene as a probe to study the pattern of histone H3 transcripts in the alfalfa, we found that the H3 RNAs are undetectable in leaves, more in stems than in roots, and highest in somatic embryos. Moreover, the RNA products of H3 genes in all alfalfa tissues tested show unusually long nontranslated region compared to those of animal histone genes. An additional high molecular weight species of H3 transcript was detected only in somatic embryos.

    Notes: Abstract A 3.4-kilobase genomic DNA fragment from Pisum sativum L. containing the LegA gene, which encodes a major legumin storage protein, was transferred to Nicotiana plumbaginifolia using an Agrobacterium tumefaciens strain containing the Bin 19 binary vector system. Northern hybridisation analysis of legA-transformed plants demonstrated that legumin-specific RNA was present in developing seeds but not in developing leaves. Legumin protein was immunologically detected in the mature seeds of legA-transformed plants, and was present as the correct-size protein composed of disulphide-bonded polypeptides. It is concluded that the transferred pea genomic fragment contains all the information necessary for seed-specific expression of the legA gene, and for correct processing of the primary transcript and the precursor legumin protein.

    Notes: Abstract Tuberization in potato is a complex developmental process involving the expression of a specific set of genes leading to the synthesis of tuber proteins. We here report the cloning and analysis of mRNAs encoding tuber proteins. From a potato tuber cDNA library four different recombinants were isolated which hybridized predominantly with tuber mRNAs. Northern blot hybridization experiments showed that three of them, pPATB2, p303 and p340, can be regarded as tuber-specific while the fourth, p322, hybridizes to tuber and stem mRNA. Hybrid-selected in vitro translation and nucleotide sequence analysis indicate that pPATB2 and p303 represent patatin and the proteinase inhibitor II mRNA respectively. Recombinant p322 represents an mRNA encoding a polypeptide having homology with the soybean Bowman-Birk proteinase inhibitor while p340 represents an mRNA encoding a polypeptide showing homology with the winged bean Kunitz trypsin inhibitor. In total, these four polypeptides constitute approximately 50% of the soluble tuber protein. Using Southern blot analysis of potato DNA we estimate that these mRNAs are encoded by small multigene families.

    Notes: Abstract Work using a full-length cDNA clone has revealed that the plastid-located glutamine synthetase (GS) of Phaseolus vulgaris is encoded by a single nuclear gene. Nucleotide sequencing has shown that this cDNA is more closely related to a cDNA encoding the plastidic GS of Pisum sativum than to cDNAs encoding three different cytosolic GS subunits of P. vulgaris. The plastid GS subunits are initially synthesized as higher M r (47000) precursors containing an N-terminal presequence of about 50 amino acids which is structurally similar to the presequences of other nuclear-encoded chloroplast proteins. The precursor has been synthesized in vitro and is imported by isolated pea chloroplasts and processed to two polypeptides of the same size as native P. vulgaris chloroplast GS subunits (M r 42000). Experiments with fusion proteins show that the N-terminal 68 amino acids of this precursor allow the cytosolic GS subunit β also to be imported and processed by isolated chloroplasts. Polyadenylated mRNA specifically related to the plastidic GS gene is most highly abundant in chloroplast-containing organs (leaves and stems) but is also detectable in roots and nodules.

    Notes: Abstract Cell cultures of Corydalis sempervirens adapted to growth in the presence of 5 mM glyphosate [N-(phosphonomethyl)glycine] display a 30- to 40-fold increase in the cellular content of 5-enolpyruvylshikimic acid 3-phosphate (EPSP) synthase, the target enzyme of the herbicide. Translatable mRNA activity as well as transcript levels for EPSP synthase were increased 8-to 12-fold in the adapted (glyphosate-tolerant) as compared to the non-adapted (glyphosate-sensitive) cultures. Northern blot analysis revealed a single 1.8 kb transcript after hybridization with an oligonucleotide probe deduced from the N-terminal amino acid sequence of the enzyme. No significant differences in the relative abundance of EPSP synthase-specific DNA sequences could be detected, however, in Southern and dot blot analyses of restricted DNA isolated from the two cultures. We conclude that the overproduction of EPSP synthase in glyphosate-tolerant C. sempervirens cells is not based on the amplification of the corresponding gene.

    Notes: Abstract Abscisic acid (ABA) inhibits the gibberellic acid induced synthesis of α-amylase in barley aleurone layers, yet ABA itself induces more than a dozen polypeptides (Lin & Ho, Plant Physiol 82: 289–297, 1986). As part of our effort to elucidate the molecular action of ABA in barley aleurone layers, we have isolated and characterized an ABA-induced cDNA clone, pHV A1. This cDNA clone hybridizes to an RNA species of approximately 1.1 kb from ABA-treated barley aleurone layers. The level of this mRNA is tripled within 40 minutes after ABA treatment, reaches a peak at 8–12 h, and is present up to 48 h. The induction of this mRNA responds to concentrations of ABA as low as 10-9 M, but higher ABA concentrations induce higher expression of this mRNA. The products of hybrid-select translation and in vitro transcription/translation with pHV A1 comigrate on SDS gel as a 27 kDa polypeptide. However, the sequence of pHV A1 indicates that it has an open reading frame encoding a 22 kDa protein. This size discrepancy is probably due to the high content of the basic amino acid, lysine. This notion has been confirmed by two-dimensional gel electrophoresis showing that this polypeptide is one of the most basic proteins in ABA-treated barley aleurone layers. The deduced amino acid sequence of pHV A1 contains nine imperfect repeats 11 amino acids long which share homology with cotton Lea 7 protein (Baker, Steele & Dure, Plant Mol Biol, in press). The identity and function of the encoded product of pHV A1 is under investigation.

    Notes: Summary Restriction Fragment Length Polymorphisms (RFLP) have been identified between widely distant cultivars (‘Minsoy’ and ‘Noir 1 ’) of soybean Glycine max (L.) Merrill. Using as probes randomly chosen clones of DNA, one in five probes revealed a polymorphism. More than half of these polymorphisms appear to result from rearrangements of the genomic DNA. Twenty seven markers were analyzed for linkage in F2 plants. Eleven of these markers were contained in four linkage groups. Five cultivars were compared in a search for new alleles. When RFLP markers corresponding to low copy DNA were used to analyze three other cultivars — ‘Sooty’, ‘Forrest’ and ‘Mandarin (Ottawa)’ — few new alleles were found. Using these probes, five different markers could be used to differentiate the five cultivars. Complex probes, which correspond to repeated DNA, revealed different polymorphisms in different cultivars and a single such probe could be used to distinguish the five cultivars from each other.

    Notes: Summary Little is known about the biochemical correlates of selection for growth in farm or laboratory animals, or the identity of the gene products affected or produced by ‘trait-genes’. Modern broiler chickens have about 8-fold greater breast muscle mass than layer chickens at 7 weeks of age and over 2-fold greater breast muscle mass than their 1972 counterparts. This increase in muscle mass is associated with over 20-fold higher levels of ornithine decarboxylase (ODC) in broiler chickens at 1 week of age as compared with layer strain chickens; there is a comparable increase in a relaxed-selection strain of broilers. The increase in ODC levels is larger than the differences in muscle or body weight between broilers and layers at 7 weeks of age, occurs at an age when there is no difference in weights between the strains and precedes the major growth spurt. Increases in ODC levels and hence polyamine synthesis have been associated with, and usually precede, rapid growth and cell proliferation in a wide range of cell types and organisms in response to many different stimuli. Therefore, the correlation of ODC levels with genetic differences in muscle growth make it worth investigating the control of ODC gene expression in these strains.

    Notes: Summary Separations of kafirin and alcohol soluble glutelin proteins by reversed-phase high-performance liquid chromatography (RP-HPLC) from 7 inbreds and one hybrid of sorghum [Sorghum bicolor (L.) Moench] and one source of Johnsongrass [Sorghum halapense (L.) Pers.] were compared. Objectives were to assess the stability of protein profiles for seed sources produced at different locations and in different environments to examine the potential of RP-HPLC to provide genotypic profiles for sorghum. Analyses of variance data showed that levels of variation due to environments and locations were small; the majority of variation (93%) was among genotypes. Associations among inbreds revealed by multivariate and cluster analysis showed similarity with those that would be expected on the basis of pedigree. A chi-square analysis showed no deviation in the hybrid profile from the expected 2∶1 ratio of peaks from the female and male inbred parents, respectively. Improvements in the ability to correctly assign common peaks are necessary before associations among numerous sorghum genotypes can be reliably demonstrated by analysis of data from reversed-phase high-performance liquid chromatography (RP-HPLC).

    Notes: Summary The effect of genotype on maternal haploid plant production in maize was studied. The frequency of gynogenetic plants when “Stock 6” was used as pollinator varied according to the female parent genotype. No simple relation was observed between genotypic aptitudes for gynogenetic and androgenetic development, which occured after pollination of “W23” plant carrying the “indeterminate gametophyte” gene. Furthermore, the population NS, a favorably responsive genotype to anther culture, does not exhibit exceptional ability for in vivo gynogenesis. The effect of inbreeding and the influence of maternal haploid origin suggest that specific genes control maternal haploid initiation and development. However, gynogenetic development is not limited to a particular genotype. The frequency of maternal haploids may be increased by using specific pollen parents. Attempts were made to select for a high haploidyinducing trait and the present study reports the successful development of lines that can be utilized as pollen parents to induce haploids for experimental purposes and breeding programmes. When an inbred line “WS14”, derived from the cross W23 x Stock 6, was used as pollen parent, 2%–5% maternal haploids were obtained according to the female parent genotype. A high haploidy-inducing potential is a heritable trait and may be controlled by a limited number of genes. Genetic determination of the haploidy-inducing character was examined in relation to the efficiency of the selecting method and the mechanisms involved in the origin of maternal haploids.

    Notes: Summary A total of 70 cnx mutants have been characterized from a collection of 211 nitrate reductase deficient (NR-) mutants isolated from mutagenized Nicotiana plumbaginifolia protoplast cultures after chlorate selection and regeneration into plants. They are presumed to be affected in the biosynthesis of the molybdenum cofactor since they are also deficient for xanthine dehydrogenase activity but contain NR apoenzyme. The remaining clones were classified as nia mutants. Sexual crosses performed between cnx mutants allowed them to be classified into six independent complementation groups. Mutants representative of these complementation groups were used for somatic hybridization experiments with the already characterized N. plumbaginifolia mutants NX1, NX24, NX23 and CNX103 belonging to the complementation groups cnxA, B, C and D respectively. On the basis of genetic analysis and somatic hybridization experiments, two new complementation groups, cnxE and F, not previously described in higher plants, were characterized. Unphysiologically high levels of molybdate can restore the NR activity of cnxA mutant seedlings in vivo, but cannot restore NR activity to any mutant from the other cnx complementation groups.

    Notes: Abstract The human-type ABO blood groups were determined for 94 families of the squirrel monkey which included 151 animals. Four phenotypes of ABO blood groups (A, B, AB, and O) were detected. Family analysis revealed that the human-type ABO blood groups in this species were governed by three alleles, codominantA andB and silentO. There were intraspecific differences in the distribution of phenotypes and gene frequency among three populations imported by different routes at different times. The usefulness of ABO blood groups for defining the genetic variability of a squirrel monkey breeding colony through successive generations is discussed on the basis of the difference in distribution of ABO blood groups between wild-originated parental and its first colony-born populations.

    Notes: Abstract In order to understand more fully chloroplast genetic systems, we have determined the complete nucleotide sequence (155, 844 bp) of tobacco (Nicotiana tabacum var. Bright Yellow 4) chloroplast DNA. It contains two copies of an identical 25,339 bp inverted repeat, which are separated by 86, 684 bp and 18,482 bp single-copy regions. The genes for 4 different rRNAs, 30 different tRNAs, 44 different proteins and 9 other predicted protein-coding genes have been located. Fifteen different genes contain introns. Twenty-two genes for components of the photosynthetic apparatus have so far been identified. Most of the genes (except the gene for the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase) code for thylakoid membrane proteins. Twenty of them are located in the large single-copy region and one gene for a 9-kd polypeptide of photosystem I is located in the small single-copy region. The gene for the 32-kd protein of photosystem II as well as the gene for the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase have strong promoters and are transcribed monocistronically while the other genes are transcribed polycistronically. We have found that the predicted amino acid sequences of six DNA sequences resemble those of components of the respiratory-chain NADH dehydrogenase from human mitochondria. As these six sequences are highly transcribed in tobacco chloroplasts, they are probably genes for components of a chloroplast NADH dehydrogenase. These observations suggest the existence of a respiratory-chain in the chloroplast of higher plants.

    Notes: Abstract From studies conducted in both our laboratory and by Gibson, Tabita and colleagues, as well as drawing on the recent studies with Alcaligenes eutrophus, we describe two genetic regions which have been identified on the chromosome of Rhodobacter sphaeroides which code for a number of enzymes involved in CO2 fixation. One region was found to contain the genes coding for fructose 1,6-bisphosphatase (fbpA), phosphoribulokinase (prkA), a 37 kDa polypeptide (cfxA), and form I ribulose 1,5-bisphosphate carboxylase/oxygenase (rbcL, S). These genes appear to be expressed in the same transcriptional direction and are tandomly arranged. A second, apparently unlinked region of the chromosome contains a duplicate (with respect to functionality of gene products) but not identical set of these same four genes. Although the gene order in both regions is apparently identical, there is approximately 4 kb of DNA separating the 3′-end of prkB and the beginning of cfxB. The specific genetic organizations and proposed roles of these two genetic regions are discussed.

    Notes: Abstract Three CF1 and three CF0 subunits of the chloroplast H+-ATP synthase are encoded on the chloroplast genome. The chloroplast atp genes are organized as two operons in plants but not in the green alga, Chlamydomonas reinhardtii. The atpBE or β operon shows a relatively simple organisation and transcription pattern, while the atpIHFA or α operon is transcribed into a large variety of mRNAs. The atp genes are related to those of cyanobacteria and, more distantly, to those of non-photosynthetic bacteria such as E. coli, suggesting a common origin of most F1F0 ATP synthase subunits. Both the chloroplast and cyanobacterial ATP synthases have four F0 subunits, not three as in the E. coli complex. The proton pore of the CF0 is proposed to be formed by the interaction of subunits III and IV.

    Notes: Abstract The synthesis in Escherichia coli of both the large and small subunits of cereal ribulose bisphosphate carboxylase/oxygenase has been obtained using expression plasmids and bacteriophages. The level and order of synthesis of the large and small subunits were regulated using different promoters, resulting in different subunit pool sizes and ratios that could be controlled in attempts to optimize the conditions for assembly. Neither assembly nor enzyme activity were observed for the higher plant enzyme. In contrast, cyanobacterial large and small subunits can assemble to give an active holoenzyme in Escherichia coli. By the use of deletion plasmids, followed by infection with appropriate phages, it can be demonstrated that the small subunit is essential for catalysis. However, the small subunit is not required for the assembly of a large subunit octomer core in the case of the Synechococcus enzyme; self-assembly of the octomer will occur in an rbcS deletion strain. The cyanobacterial small subunits can be replaced by wheat small subunits to give an active enzyme in Escherichia coli. The hybrid cyanobacterial large/wheat small subunit enzyme has only about 10% of the level of activity of the wild-type enzyme, reflecting the incomplete saturation of the small subunit binding sites on the large subunit octomer, and possibly a mismatch in the subunit interactions of those small subunits that do bind, giving rise to a lower rate of turnover at the active sites.

    Notes: Abstract Theunc (or atp) operon ofEscherichia coli comprises eight genes encoding the known subunits of the proton-translocating ATP synthase (H+-ATPase) plus a ninth gene (uncI) of unknown function. The subunit stoichiometry of the H+-ATPase (α 3β3γ1δ1ε1a1b2c10–15) requires that the respectiveunc genes be expressed at different rates. This review discusses the experimental methods applied to determining how differential synthesis is achieved, and evaluates the results obtained. It has been found that the primary level of control is translational initiation. The translational efficiencies of theunc genes are determined by primary and secondary mRNA structures within their respective translational initiation regions. The respective rates of translation are matched to the subunit requirements of H+-ATPase assembly. Finally, points of uncertainty remain and experimental strategies which will be important in future work are discussed.

    Notes: Summary Primary cultures of neonatal cardiocytes expreses the gene for atrial natriuretic peptide (ANP). In general the levels of expression follow the rank order: atrial 〉 ventricular ≫ nonmyocardial cells. Following the initial dispersion cardlocytes require 48 to 72 hours before ANP seeretion and ANP mRNA accumulation approach a new steadystate level. In situ hybridization analysis indicates that ANP gene expression is concentrated in a subpopulation of cardiocytes in both the atrial and the ventricular cell cultures. These findings suggest that these primary cultures may be of value in defining the faciors governing the expression of the ANP gene in the cardiac cell.

    Notes: Abstract Mesembryanthemum crystallinum responds to high salinity in the soil by shifting the mode of carbon assimilation from the C3 mode to Crassulacean acid metabolism (CAM). Several enzymes of carbon metabolism have increased apparent activities in the CAM mode, including phosphoenolpyruvate carboxylase (PEPcase) and pyruvate orthophosphate dikinase (PPDK). We have identified cDNA clones for PEPcase and PPDK by immunological screening of a cDNA library constructed in the protein expression vector lambda gt11. The clones were characterized by immunoblotting and RNA blotting techniques. RNA blotting showed that during CAM induction the steady-state level of mRNAs for both PEP case and PPDK increased.

    Notes: Abstract The puf operon of the purple nonsulfur photosynthetic bacterium, Rhodobacter sphaeroides, contains structural gene information for at least two functionally distinct bacteriochlorophyll-protein complexes (light harvesting and reaction center) which are present in a fixed ratio within the photosynthetic intracytoplasmic membrane. Two proximal genes (pufBA) specify subunits of a long wavelength absorbing (i.e., 875 nm) light harvesting complex which are present in the photosynthetic membrane in ≃15 fold excess relative to the reaction center subunits which are encoded by the pufLM genes. This review summarizes recent studies aimed at determining how expression of the R. sphaeroides puf operon region relates to the ratio of individual bacteriochlorophyll-protein complexes found within the photosynthetic membrane. These experiments indicate that puf operon expression may be regulated at the transcriptional, post-transcriptional, translation and post-translational levels. In addition, this review discusses the possible role(s) of newly identified loci upstream of pufB which may be involved in regulating either synthesis or assembly of individual bacteriochrlorophyll-protein complexes as well as the pufX gene, the most distal genetic element within the puf operon whose function is still unknown.

    Notes: Stereoisomeric Sinensiaxanthins and Sinensiachromes: Separation and Absolute ConfigurationThe so-called sinensiaxanthins and sinensiachromes, important apocarotenols from various fruits, have been separated into 2 and 4 stereoisomers, respectively, and their absolute configurations have been determined: (3S,5R,6S)-5,6-epoxy-5,6-dihydro-10′-apo-β-carotene-3,10′-diol (2), its (9Z)-stereoisomer 7, the (8R)- and (8S)-epimers of (3S, 5R)-5,8-epoxy-5,8-dihydro- 10′ -apo-β-carotene-3, 10′-diol (4 and 5), and their (9Z)-stereoisomers 3 and probably 6. Thus, sinensiaxanthins are cleavage products from (Z/E)-isomeric antheraxanthins or violaxanthins (scission at C(9′)-C(10′)) and sinensiachromes analogously from mutatoxanthins or auroxanthins.

The Re enantiotopic acetate group appears to be saponified preferentially, as proved by the X-ray crystal structure analysis of three camphanoates 4d, 6d, and 7d. Elimination of H2O or AcOH from the hydroxy acetates thus available gives derivatives of nitro-allylic alcohols (see 20-24, 27, and 29) which are subjected to diastereoselective Michael additions or SN2′ substitutions.

These are attacked by electrophiles at the N or C terminus. Some notable reactions of imidazoisoquinolone 2 are those with maleic and acrylic acids to form the tetracycles 48 and 51, respectively. With propiolic acid, 5 underwent an interesting reaction to form the benzimidazonaphthyridine 53. An equally interesting behaviour was elicited from 2 in its reaction with formaldehyde, when in addition to the expected methylene-bridged molecule 59, the novel spiro derivative 60 was formed by the dimerisation of a presumed azadiene intermediate 63.

In general, most of the groups are compatible with the conditions for the cyclization and do not interfere even when directly attached to the divinyl ketone. The influence of substituents on the rate of the cyclization has been addressed and is consistent with a simple mechanistic picture. O- and N-Containing functions are tolerated except when attached to the α-vinyl C-atom of the divinyl ketone. The diastereoface-directing effect of a fused cyclobutane is discussed.

    Notes: A 1:1 mixture of the racemic trans- and cis-1-p-menthene-3,8-diols ((±)-3 and (±)-4, resp.) was readily prepared in 3 steps starting from 3-methyl-2-cyclohexen-1-one. The relative configuration of the diols, purified via the corresponding cyclocarbonates, was assigned by 1H-NMR spectroscopy and found to be at variance with tentative claims in the literature. Optically active samples of 3 and 4 were prepared by resolution of the racemates with (R)-1-phenylethylamine. The absolute configuration of the resulting diols was determined by chemical correlation with standards of known absolute configuration.

After double deprotonation with LDA, the corresponding dianionic derivatives could be diastereoselectively alkylated with electrophiles (MeI, C6H5CH2Br, C6H5CHO, CH3CHO). Acid hydrolysis led to the two enantiomeric 2-methyl- and 2-benzyllysines and to the enzyme inhibitor (S)-2-methylornithine. Several α-alkylated tryptophan derivatives were obtained through alkylation of the heterocycles derived from various amino acids with 1-(tert-butyloxycarbonyl)-3-(bromomethyl)indole (26). Alkaline hydrolysis of the five-membered auxiliary ring of 30b followed by treatment with HCl afforded (S)-2-methyltryptophan (31).

    Notes: Single crystals of the N,N,N′,N′-tetramethylethylenediamine (TMEDA) complex 6 of the title compound have been isolated. Compound 6 decomposes in the crystalline state above -20°. From the bond lengths and angles obtained by X-ray crystal-structure analysis (data collected at -70°), compound 6 is best described as a (E)-1-(tert-butylthio)-1-lithio-2-butene with the double bond acting as an additional ligand on lithium (unsymmetrical allylic group). The S-atom is in a cisoid arrangement in a common plane with the four C-atoms of the butenyl system. The t-Bu group and the Li-atom are located above and below this plane. The structure is discussed with respect to the reactivity of 6 (α/γ reactivity). The gross structure is reproduced surprisingly well by an ab initio SCF MO calculation of the model lithiopropene-1-thiol 7(HS instead of t-BuS, CH2 instead of CHCH3, no solvation of Li). The prominent difference is the symmetry of the allyllic moiety in the calculated structure.

The sign of the Cotton effects associated with lowest-energy transition of 2-(dicyanomethylidene)-((-)-(1S,4S)-15), (E)-2-(methoxyimino)-((+)-(1S,4S)-16), (Z)-2-(methoxyimino)-5,6-dimethylidenebicyclo[2.2.1]heptane ((-)(1S,4S)-17), and 2,3,5-trimethylidenebicyclo[2.2.1]heptane ((-)-(1R,4S)-18) is opposite to the chirality constituted by the coupling of the electric transition moments of the two homoconjugated π-chromophores (Kuhn-Kirkwood dipole-coupling mechanism). When the substituents at C(2) are not π-functions, no general rule can be retained for the chiroptical properties of the 5,6-dimethylidenebicyclo[2.2.1]hept-2-yl systems as shown for dimethyl acetal (-)-(1S,4S)-19, ethylene acetal (+)-(1R,4R)-20, exo and endo methyl ethers (+)-(1R,2S,4R)-21 and (+)-(1R,2R,4R)-22, and for spirol[5,6-dimethylidenebicyclo[2.2.1]heptane-2.2'-oxiranes](-)-(1S,2S,4S)-23 and (-)-(1S,2S,4S)-24.

    Notes: Irradiation (λ 〉 340 nm) of the title compound 2a in t-BuOH affords 3,3,10,10-tetramethyl-2,8-dioxatricyclo-[4.3.2.01,6]undecane-5,9-dione (3) via photoextrusion of 2-methylpropene and subsequent photocycloaddition of 2a to the alkene. The same regioisomer 3 is formed selectively and in much higher yields on irradiating 2a in the presence of excess 2-methylpropene. Irradiation of 3 (λ = 300 nm) in t-BuOH in the presence of the same alkene gives a 6:1 mixture of spirooxetanes 7a and 7b but not α-cleavage products. In 2-propanol, 2a is photoreduced to a 2:1 mixture of diastereoisomeric hydrodimers 9.

    Notes: Enantiomerically pure methyl 5-bromo-5-deoxy-2,3-O- isopropylidene-β-D- (D-5b) and -β-l-ribofuranoside (l-5b) have been derived from (-)-(1R,2S,4R)-2-exo-cyano-7-oxabicylo[2.2.1]hept-5-en-endo,-yl (1′S)-camphanate (1) and (+)-(1S,2R,4S)-2-exo-cyano-7-oxabicyclo[2.2.1]hept-5-en-2-endo-yl(1′R)-camphanate (2), respectively, in 5 synthetic steps and 28% overall yield. Hydrolysis of D-5b and L-5b afforded methyl 2,3-O isopropylidene-β-D-ribofuranoside (D-5a) and methyl 2,3-O-isopropylidene β-L-ribofuranoside (L-5a), respectively. The intermediate (+)-(1R,4R,5R,6R) 5-exo,6-exo-(isopropylidenedioxy)-7-oxabicyclo[2.2.1]heptan-2-one ((+)-7) and its enantiomer(-)-7 were also obtained enantiomerically pure by resolution of (=)-7 by the Johnson-Zeller method. In bothe approaches, the chiral auxiliaries ((-)- and (+)-camphanic acids, or (+)-(S)-N,S-dimethyl-S-phenylsulfoximide) were recovered at an early stage of the synthesis.

    Description / Table of Contents: Solvolysis Mechanism of cis - and trans-2-Arylcylopentyl p-Toluenesulfonates. The Step: 1-Deuterium Isotope Effects, Basic Salt Effects, and Special Salt EffectWe have studied the first step of the solvolysis of cis and trans-2-arylcyclopentyl p-toluenesulfonates in HCOOH, AcOH, and EtOh. All substrates show a high kinetic 1-deuterium isotope effect (kH/kD(1) 〉1.15). This fact indicates that first step leads to classical intimate ion-pair Which dissociates to a solvet-separated ion-pair, without participation either of solvent, the 2-aryl group, or a H-atom at C(2). The slight influence of added basic ions on reaction rate allows us to exclude any direct solvent attack on the covalent substrate even in the most favorable case, i.e. ethanolysis of 2-(p-nitrophenyl)cylopentyl-p-toluenesulfonates. Furthermore, solvent-separated ion pair formation is indicated by the special salt effect induced by LiClO4.

    Notes: The synthesis and antifungal activity of a novel series of 3-aryl-5-[(aryloxy)alkyl]-3-[(1H-imidazol-1-yl)-methyl]-2-methylisoxazolidines and related compounds, are discussed. The synthesis of the title compounds was accomplished via a 1,3-dipolar cycloaddition of α-substituted ketonitrones with l-alkenyl phenyl ethers (Scheme 2 and 3). The compounds were evaluated for in vitro antifungal activity in solid agar cultures against a broad variety of yeast and systemic mycoses and dermatophytes. While antifungal activity was evident throughout the series, in general, derivatives having halogen atom(s) in either or both aryl rings demonstrated the highest potency, especially against Trichophyton rubrum and Candida albicans. The dichloro analog 20 (PR 967-248) was found to possess the most useful activity. Its minimum inhibitory concentration (MIC) values ranged between 0.2 and 2.0 μg/ml, as compared to 0.2-20.0 μg/ml for the standard drug ketoconazole (4).

    Notes: Olefins with captodative substitution are reactive dienophiles in Diels-Alder reactions with normal and inverse electron demand. This is shown for reactions of 2-(tert-butylthio)acrylonitrile (1) with various dienes and heterodienes, e.g. 1,3-cyclohexadiene, hexachloro-1, 3-cyclopentadiene, acrolein, methacrolein, and methyl vinyl ketone (Schemes 2 and 3). In case of the hetrodienes, 3,4-dihydro-2H-pyrans are formed beside small amounts of tetrahydrothiophenes; however, with methyl vinyl ketone, both reaction pathways are equally followed. The high reactivity of captodative olefins in Diels-Alder reactions are rationalized on the basis of Sustmann's FMO model under consideration of Viehe's concept of captodative substitution of alkenes.

    Notes: Short Total Syntheses of (±)-Sativene and (±)-cis-SativenediolOur approach to (±)-sativene (7) and (±)-cis-sdtivenediol (9) involves: (a) reaction of 3-methylbutanoyl chloride with Et3N/cyclopentadiene to give the endo-isopropyl-ketone 1 (here improved to 71%), (b) NBS bromination of 1 to a 5:1 mixture (87%) of the bromo-ketones 2 and 3, (c) NFD-reaction sequence initiated by the attack of 1,2-butadienyl titanate (complex of 15, obtained from 2-butine) on 2/3 to afford 52% of the brexenone derivative 4 (along with 8% of its epimer 16), (d) addition of dibromomethane to 4 forming 63% of the diene-alcohol 5 (along with 13% of the diene-carbaldehyde 38), and (e) carbenoid ring-expansion with MeLi applied to 5 resulting in 41% the diene-ketone 6 (along with 15% of a 1:3 mixture of the diene-ketones 32 and 33). Wolff-Kishner reduction of 6 led to 81% of (±)-sativene (7), when enough O2 was present, but to 97% of the diene 8 in the strict absence of O2. (±)-cis-Sativenediol (9) was obrained (86%) by OsO4 hydroxylation of 8. The brexenone derivatives 4 and 16 (6:1, 50%) were also produced when the NFD-reaction sequence was applied to the isomeric bromo-ketone mixture 13/13 (1:3). The latter was obtained by NBS bromination of 10, which in turn was available by base epimerization of 1, followed by destructive removal of unreacted 1 by repeated gas-flow thermolysis. An analogous (less convenient) route to (±)-sativene (7) passed through a series of dihydro compounds (the ene series) it started with the methylidene-ketone 36, which was the product (97%) of a partial hydrogenation of 4. Addition of dibromomethane to 36 led t 62% of the methylidene-alcohol 39 (along with a little tetracyclic ether 40). Carbenoid ring expansion of 39 with MeLi afforded ca. 42% of the methylidene-ketone 41 (along with 7% of the methylidene-ketone 43 or, under slightly different condition, along with 9% of the methylidene-ketone 42 and 10% of the methylidene-carabaldehyde 44). The methylidene-alcohol 39 and the methylidene-ketone 43 were also obtained by partial hydrogenation of 5 and 33, respectively. Wolff-Kisher reduction converted 41 into (±)-sativene (7 99%); the same conditons applied to 42 afforded only ca. 8% 7 (along with three other hydrocarbons, one of them (ca. 21%) probably being (±)-copacamphene (45)). In the diene series, the two succeeding reactions (4→5 and 5→6) competed with the same side reaction, a rearrangement leading to the brendene-aldehyde 38. In the ene series, the corresponding dihydro-by-product 44 was found in the reacton 39→41, but not during 36→39. These side reactons could largely be suppressed by keeping the reaction temperature low. An explanation is proposed.

    Notes: The reaction of the bromo-substituted naphthalene 1 with the alkanethiolate anions 2a-b and arenethiolate 2c in tetraglyme gave the corresponding 1-naphythyl thio-ethers 3a-c. Thio-ethers 3a-c were oxidized to the corresponding sulfones 4a-c with m- chloroperoxybenzoic acid. The reaction of the dichloro-substituted anthracene 5a with 2b gave the disubstutution product 6a. The reaction of 9-bromoanthracene 5c with the alkanethiolate 2b gave 6b, whereas the reaction of 5c with the arenethiolate 2c gave a mixture of substitution product 6c and anthracene 7. The observation of the formation of both 6c and 7 is explained by the competition between substitution (SnAr) and electron-transfer (ET) mechanisms. Consistent with this interpretation, the reaction of the monochloro-substituted 5b, which has a higher-energy σ* orbital, with 2c gave 6c without the formation of 7. Zn/KOH in tetraglyme was shown to reduce the aryl halides 5b-c and thio-ether 6c to 7.

    Notes: Diets supplemented with astaxanthin and canthaxanthin, respectively, and a control diet without carotenoid additions, were fed to 1½-year-old Atlantic salmon (Salmo salar, L.) for one year. The integuments were investigated as to their quantitative and qualitative carotenoid composition. Astaxanthin and canthaxanthin deposited in the skin amounted to 20 and 14% of the total carotenoids only. Seventy % must be considered as metabolites of astaxanthin and canthaxanthin and 10% as basic xanthophylls also present in the control groups. Astaxanthin apparently underwent the following metabolic pathway: astaxanthin→idoxanthin→adonixanthin→zeaxanthin→zeaxanthin 5,6-epoxides. Reduction of the 4′-carbonyl group was stereospecific leading to the (4′R)-idoxanthin. Canthaxanthin was obviously converted to β,β-carotene via 4′-hydroxyechinenone, echinenone, and 4-hydroxy-β,β-carotene.

    Notes: Four highly oxygenated polyphenols 1-4, have been isolated from the stem bark of Cordia goetzei Gürke (Boraginaceae) and their structures elucidated by a combination of one- and two-dimensional (including COLOC) NMR spectroscopic techniques, mass spectroscopy, and chemical methods. The first new compound, cordigone (1), is a tetrahydrofuran derivative and the second one, cordigol (2), a furobenzopyran derivative. The benzofurans 3 and 4 are orange pigments, responsible for the coloration of the stem bark of the tree. All four polyphenols are fungicidal against cladosporium cucumerinum.

    Notes: Base-modified octanucleotides derived from d(G1-G2-A-A-T-T-C-C-) (11) but containing 8-aza-7-deaza-2′-deoxyguanosine (2) instead of 2′-deoxyguanosine (1) have been prepared by solid-phase synthesis employing P(III) chemistry. Isobutyrylation of 2, followed by 4, 4′-dimethoxytritylation and subsequent phosphitylation yielded the methyl or the cyanoethyl phosphoramidites 6a or 6b, respectively. They were used as building blocks in automated DNA synthesis. The resulting octanucleotides 12-14 containing 2 showed increased Tm values compared to the parent oligomer 11. The oligomers 11-14 were employed as sequence-specific probes in endo-deoxyribonuclease Eco RI oligonucleotide recognition. Whereas displacement of dG-2 (enzymic cleavage site of 11) abolished phosphodiester hydrolysis, replacement of dG-1 enhanced the cleavage rate compared to 11.

    Notes: The title complexes have been derived from tricarbonyl [(1R,4S,5S,6R,)-C,5,6,C-η-(5,6-dimethylidenebicyclo[2.2.2]oct-2-ene)]iron (1.). The crystal structure and absoulte configuration of (+)-tricarbonyl (1S,2S,4R,5R,6S)-C,5,6,C,-η-(5,6-dimethylidenebicyclo[2.2.2]oct-2-yl p,-bromobenzoate)]iron ((+)-6) determined by X-ray diffrac-tion studies proved earlier attribution of the relative configuration (endo us. exo,) of the tricarbonyliron moieties in complexes 1 and 2 of 5,6-dimethylidenebicyclo[oct-2-ene. The structure of (+)-6 confirms the absolute configuration derived earlier from circular-dichroism spectra of 5,6-dimethylidenebicyclo[2.2.2]oct-2-yl esters and 5,6-dimethylidenebicyclo[2.2.2]octan-2-one.

    Notes: The [2 + 2] Photocycloaddition of 2-(trimethylsilyloxy)-1,3-butadiene to a number of 2-cycloalkenones proved to be quite a general reaction leading to good yields of the cycloadducts (Table). This finding is surprising since dienes, in general, are better known as quenchers of enone triplets rather than as photochemical reactants. Both the high substrate concentrations, which can be employed in these cycloadditions, and the remarkable regio and stereoselectivity of the processes qualify them as valuable for syntheses. In a first application, the photoproducts 1a, b were transformed in three steps into a viable precursor 4 of the pentalenolactone-G and -H antibiotics.

    Notes: The near-infrared luminescence of singlet oxygen (1O2) has been measured in order to determine the efficiency of 1O2 quenching by two carotenoid compounds, β-carotene and canthaxanthin. 1H-Phenalen-1-one and rose bengal have been used as photosensitizers in those steady-state luminescence experiments. Stern-Volmer analysis of the 1O2 luminescence in solutions of CCl4 and CD3OD, containing different concentrations of the carotenoids, has shown a very efficient quenching by canthaxanthin. The rate constants are about a factor of 2 below the diffusion limited values for the given solvents, confirming earlier results in benzene. In comparison, the efficiency of 1O2 quenching by β-carotene is slightly lower than that by canthaxanthin in non-polar solvents and is reduced by an order of magnitude in CD3OD, due to the aggregation of this quencher.

    Notes: The preparation of 3-azetidinones with different N-substituents and their transformation to 3-ethylideneazetidines has been studied in relation with a projected synthesis of 3-ethylideneazetidine-2-carboxylic acid (= polyoximic acid; 1). The stable crystalline 3-azetidinone hydrochloride (18), obtained by hydrogenolysis of the known 1-(diphenylmethyl)-3-azetidenone (4), is described for the first time, From this key intermediate, 3-oxoazetidine-derived amides, exemplified by benzamide 12,urethanes (e.g. 17), and ureas (e.g. 20) can be prepared in good yield (Scheme 3). The olefination of 3-azetidinones with alkylidene(triphenyl)phosphoranes is a perparatively useful process only for derivatives with a pyramidal N-atom, e.g. the diphenylmethyl derivative 4, and not for the amide 12 or the urethane 17 (Scheme 4). 3-Alkylidene-azetidines with an amide N-atom should, therefore, be prepared by exchange of the N-substituent after the introduction of the double bond.

    Notes: The absorption spectra, luminescence spectra, and luminescence lifetimes of the isomeric [M(bph)(bpy)] and [M(phpy)2] complexes M = Pt(II) or Pd(II), bph2- = 1,1′-biphenyl-2,2′-diyl dianion, phpy- = 2-phenylpyridine-2′-yl anion, and bpy = 2,2′-bipyridine have been investigated and compared with those of [M(bpy)2]2+ complexes and of the free protonated ligands H2bph, Hbpy+, and Hphpy. In the absorption spectra, the region below 320 mm is dominated by ligand-centered (LC) transitions, whereas metal-to-ligand charge transfer (MLCT) transitions are responsible for the bands present in the near UV/VIS region. The MLCT bands move to higher energies on replacing Pt with Pd and in going from [M(bph)(bpy)] to the [M(phpy)2] isomer. For the mixed-ligand complexes, evidence for both M → bph2- (at higher energies) and M → bpy bands is found. The structured luminescence observed at 77 K shows lifetimes of 4.0 and 1.1 μs for [Pt(phpy)2] and [Pt(bph)(bpy)], respectively, and 480 and 250 μs for the analogous Pd complexes. On the basis of the energy and lifetime data, the luminescence of the Pt(II) complexes is assigned to the lowest triplet MLCT excited state, whereas for the Pd complexes the luminescent state is thought to result from a mixture of MLCT and LC triplet levels.

Schemes 3, 5-7). Heating 6a and 6c in the presence of ethyl propiolate yielded ethyl quinoline-3-carboxylate (19) and ethyl pyridine-3-carboxylate(22), respectively (cf. Scheme 8). These results are rationalized on the basis of the intermediate formation of thio-substituted nitrile ylides of type 7 (cf. Scheme 2), which undergo regioselective 1,3-dipolar cycloadditions with reactive dipolarophiles. In the absence of such a dipolarophile, the nitrile ylides isomerize via a [1,4]-H shift to give 2-aza-1,3-butadienes of type 20. The latter are trapped in a Diels-Alder reaction with ethyl propiolate (cf. Scheme 8).

    Notes: The tautomerisation between proxibarbal (I) and the two diastereoisomers of valofan, IIX and IIY, was investigated in aqueous solutions, and various rate and equilibrium caonstants were calculated by compartmental analysis. The proportion of I at equilibrium was found to increase with pH, and indeed, the two equilibrium constants are linear functions of pH. In contrast, the equilibrium-concentratio ratio of IIY/IIX was close to 63:37 and remained constant in the pH range investigated. The rate constants were also determined as a function of temperature, allowing calculation of the tehrmolynamic parameters. Under physiological conditions, the difference in free energy favouring I vs. IIX and IIY is 6.7 and 5.4KJ · mol-1, respectively.

The rate constants of the two successive Diels-Alder additions of a given dienophile to 12 and 13 were nearly the same. Lewis-acid-catalyzed Diels-Alder addition of methyl vinyl ketone to 14 gave adduct 39 with high regio- and stereoselectivity. Lewis acids isomerized 39 into(1RS,5RS,6SR,7SR,11RS)-6,7-bis(chloromethyl)-3-oxo-2-oxatricyclo[7.4.0.01,5]tridec-8-en-11-yl methyl ketone (40) whose structure was determined by single-crystal X-ray crystallography. Heating 39 with CsF/Cs2CO3 in DMF/HMPT gave (1RS,5RS,11RS)-6,7-dimethylidene-3-oxo-2-oxatricycl. [7.4.0.01,5]tridec-8-en-11-yl methyl ketone (42). The exocyclic butadiene moiety of triene 42 added to methyl propynoate and 2,3-didehydroanisole with good regioselectivity giving polysubstituted anthracenyl and naphthacenyl derivatives, respectively.

The separation factors α range from1,1 to 2,4. Use of a Prepbar®-chromatography system allows injection of several grams at a time. The enantiomeric acetals thus obtained are fully characterized. First application to amino-acid synthesis are mentioned.

    Notes: The kinetics of the electron-transfer reaction between racemic or optically active [Co(bamap)H2O]+ and optically active Fe2+ complexes of the three new pentadentate ligands bamap, alamp, and valmp has been investigated. All the reacting species show C2 symmetry. With respect to aquo-Fe2+, the reaction rate for the Fe2+ complexes is enhanced by a factor of 104 to 105, and the observed kΔΛ/kΛΛ ratio is 1.0, 1,9, and 1.2, respectively. In all cases where stereoselectivity is observed, the reaction is faster between the complexes of opposite absolute configuration (ΔΛ or ΛΔ) than between species with the same configuration (ΛΛ or ΔΔ). The stereoselectivity effects are discussed of the basis of the structure of the transition state and the interatomic distances between the two metal centers at the moment of electron transfer.

Hydride reductions of (±)-7 were less exo-face selective than reductions of bicyclo[2.2.1]heptan-2-one and its derivatives with NaBH4, AlH3, and LiAlH4 probably because of smaller steric hindrance to endo-face hydride attack when C(5) and C(6) of the bicyclo-[2.2.1]heptan-2-one are part of an exo oxirane ring.

The isotropic coupling constants for \documentclass{article}\pagestyle{empty}\begin{document}$ 1^{\scriptstyle {{\relbar}\kern-4pt {.} }}$\end{document} are +3.14 mT for the two 14N Nuclei in the azo group, -0.336 mT for the two protons at the bridgehead C atoms, and +151 and +0.135 mT for the sets of four exo- and four endo-protons, respectively. MO calculations indicate that these values are consistent with an electron removal from an orbital represented by the antibonding combination (n_) of the lone-pair atomic orbitals at the two N atoms. The corresponding radical anion \documentclass{article}\pagestyle{empty}\begin{document}$ 1^{\scriptstyle {{\relbar}\kern-4pt {.} }}$\end{document} is persistent in fluid solution and its hyperfine data, combined with MO calculations, point to an electron uptake into the antibonding π orbital (π*) of the azo group. It is, thus, reasonable to classify \documentclass{article}\pagestyle{empty}\begin{document}$ 1^{\scriptstyle {{+}\kern-4pt {.} }}$\end{document} as a σ and \documentclass{article}\pagestyle{empty}\begin{document}$ 1^{\scriptstyle {{\relbar}\kern-4pt {.} }}$\end{document} as a π radical.

    Notes: An efficient synthesis of chiral semicorrin ligands is described (see 6-9, Schemes 2 and 3). Both enantiomers are readily obtained in enantiomerically pure form starting either from D-or L-pyroglutamic acid (1). Semicorrins of this type possess several features that make them attractive ligands for enantioselective control of metal-catalyzed reactions. Their structure is characterized by C2 symmetry, a conformationally rigid ligand system, and two stereogenic centers adjacent to the coordination sphere. In a metal complex, the two substituents at the stereogenic centers shield the metal atom from two opposite directions and, therefore, are expected to have a pronounced effect on the stereochemical course of a reaction occurring in the coordination sphere. The structure of these two substituents can be easily modified in a variety of ways. A series of (semicorrinato)copper(II) complexes (see 10-14, Scheme 4) has been prepared, and in one case (14), the three-dimensional structure has been determined by X-ray analysis (Fig. 1).

    Notes: Solid-liquid phase-transfer glycosylation (KOH, tris[2-(2-methoxyethoxy)ethye]amine ( = TDA-1), MeCN) of pyrrolo[2,3-d]pyrimidines such as 3a and 3b with an equimolar amount of 5-O-[(1,1 -dimethylethyl)dimethylsilyl]-2,3-O-(1-methylethylidene)-α-D-ribofuranosyl chloride (1) [6] gave the protected β-D-nucleosides 4a and 4b, respectively, stereoselectively (Scheme). The β-D-anomer 2 [6] yielded the corresponding α-D-nucleosides 5a and 5b with traces of the β-D-compounds. The 6-substituted 7-deazapurine nucleosides 6a, 7a, and 8 were converted into tubercidin (10) or its α-D-anomer (11). Spin-lattice relaxation measurements of anomeric ribonucleosides revealed that T1 values of H—C(8) in the α-D-series are significantly increased compared to H—C(8) in the β-D-series while the opposite is true for T1 of H—C(1′). 15N-NMR data of 6-substituted 7-deazapurine D-ribofuranosides were assigned and compared with those of 2′-deoxy compounds. Furthermore, it was shown that 7-deaza-2′deoxyadenosine ( = 2′-deoxytubercidin; 12) is protonated at N(1), whereas the protonation site of 7-deaza-2′-deoxyguanosine (20) is N(3).

The axial ligands of dichloro[tetra(15-crown-5)phthalocyaninato]silicon or -tin have been converted into dihydroxy derivatives by hydrolysis in aqueous Et3N. The catalytic effect of H2O-free CaC12 in quinoline is used for the polycondensation of dihydroxysilicon-phthalocyanine to cofacially arrayed polymers. The thermal stability of group-IV-A-metal phthalocyanines is confirmed by the higher initial decomposition points (290-440°) compared to those of the corresponding transition-metal phthalocyanines.

    Notes: The dimethyl acetals 4 of (E)-2,4-pentadienal and of (E,E)- and (E,Z)-2,4-hexadienals undergo regio- and stereospecific cycloaddition reactions with in-situ-generated acylnitroso dienophiles 5a and 5b, leading thereby to the corresponding dihydrooxazines 7a-d and 8c-d. cis-Glycolization of these latter adducts stereospecifically gave the dihydro derivatives 9a-d and 10d which, after sequential hydrogenolysis, deacetalization, and instant cyclization, led to the aminodeoxyribose derivatives 17a, 17f, and 18, and to the amino-dideoxyallose compounds 17c and 17h. These piperidino-deoxysugar derivatives exhibit a strong anomeric effect, i.e. OH—C(1) is always axial, which is explained in terms of a nN(π)-σ*(C—OH) orbital compression, as compared to the less pronounced one in the more classical pyranose series.

Starting from the aldehyde 1, the corresponding 4′-aza derivative 15 could be obtained by reductive amination with the appropriate and protected spermidine derivative 14 (Scheme 4). Enlargement of the carbocyclic ring in 15 by five members gave, after further transformations, the hydroxylactam 18. Transamidation of 18, the second ring-enlargement step, led to (±)-inandenin-10-ol (7;22.9% overall yield) and, after oxidation, to inandenin-10-one (8; 22.5%, overall yield). (±)-Oncinotine 6 was synthesized by two pathways (Scheme 6): protection of the terminal NH2 group by treatment with the Nefkens reagent and replacement of the OH group by Cl gave 24, which by thermal transamidation followed by direct ring closure led to the oncinotine derivative 26. The same intermediate could be obtained in higher yield via 28 by oxidation and protection of 18 followed by transamidation and reductive ring closure. Treatment of 26 with hydrazine finally gave (±)-oncinotine 6 in 15.9% overall yield.

    Notes: Diastereoselective Alkylation 3-Aminobutanoic Acid in the 2-PositionThe enantiomerically pure 3-aminobutanoic acids (R)- and (S)-6 are readily available by preparative HPLC separation of the two diastereoisomers 5 obtained from addition of (S)-phenethylamine to methyl crotonate and subsequent hydrogenolysis (Scheme 2). (S)-Methyl 3-(benzoylamino) butanoate ((S)-3) is also available by enzymatic kinetic resolution with pig-liver esterase. The N-benzyl- and N- benzyloxycarbonyl derivatives rac3,8, and 9 of 3-aminobutanoates are doubly deprotonated with LDA and alkylated or aminated in high selectivity (17 examples, relative topicity like; see Tables 1 and 2). The configuration of three of the products is assigned (Schemes 4-6), and in four cases, the free α-substituted β-amino acid is prepared by acidic hydrolysis (see Table 3). It is shown that the doubly lithiated β-amino-acid derivative is solubilized, and its reactivity may be strongly influenced by the presence of 3 equiv. of LiCl.
