    Notes: Summary Large granular lymphocytes (LGL) represent a morphologically distinct mononuclear cell type. In healthy individuals an average of 12.5±5.2% of peripheral mononuclear cells exhibit this morphology. Structurally they are characterized by azurophilic granules, light cell density, low surface adherence and expression of Fc-IgG receptors. Functional analyses on LGL suspensions enriched by means of density gradient fractionation and adherence procedures included testing of T-cell and monocyte-specific in vitro reactions as well as evaluation of their natural cytotoxicity (NC). Results indicated a close correlation between the percentages of LGL and the capacity to bind and to lyse NC-susceptible tumor targets. In agreement with others we would thus conclude that LGL embrace the majority of NK-effector cells. The morphological definition of LGL provides an additional tool for the clinical evaluation of this cell mediated immune reactivity.

    Notes: Summary This study aimed to investigate high molecular weight surface glycoprotein (S-GP) patterns on various types of human leukocytes. S-GP were externally labelled by the Galactose-oxidase-NaB3H4 technique. Results based on the analysis of 120 samples derived from different types of normal and malignant leukocytes indicate that (i) the relative expression of high molecular weight S-GPs changes during haemopoietic cell differentiation and (ii) to some extent these changes enable the classification of human leukocytes.

    Description / Table of Contents: Summary Investigations on the cellular composition of “round cell infiltrates” of the liver in chronic and persistent hepatitis were performed using various histochemical methods. (Nonspecific esterase using naphthol-AS-acetate as substrate with and without added NaF, naphthol-AS-D-chloroacetate-esterase, acid and alkaline phosphatase, Sudanblack B-stain, periodic acid-Schiff reaction and methylgreen-pyroninstain.) The following results were obtained: The central parts of the infiltrates are mainly composed by small round cells exhibiting a cytochemical pattern similar to that of lymphocytes. The infiltrates enlarge by peripheral growth replacing necrotic acinar cells; the v. Kupffer cells persist and are included in the peripheral parts of the infiltrates. In these regions few neutrophils and rarely monocytes are seen. Plasma cells are also mainly localized in this region. Many venoles and biliary ductuli were identified within the infiltrates. The methods used did not permit an unequivocal identification of the role of the fibroblasts in infiltrate formation.
    Notes: Zusammenfassung In den „Rundzelleninfiltraten“ bei chronischen und persistierenden Hepatitiden läßt sich auf Grund histochemischer Untersuchungen (unspezifische Esterase mit Naphthol-AS-Acetat mit und ohne NaF-Hemmung, saure und alkalische Phosphatase, Naphthol-AS-D-Chloroacetat-Esterase, Sudanschwarz B-Färbung, Perjodsäure-Schiff-Reaktion (PAS) und Methylgrün-Pyronin-Färbung) folgender Befund erheben: In den Zentren der entzündlichen Infiltrate liegen Ansammlungen von Lymphocyten, welche den hauptanteil der Infiltrate ausmachen. Die Infiltrate wachsen peripher durch eine Abschmelzung von Leberzellen. Die v. Kupfferschen Zellen bleiben bestehen und werden in die Randbezirke der Infiltrate einbezogen. In diesen Bezirken finden sich häufig auch einige Neutrophile und wenige Monocyten. Auch Plasmazellen lassen sich überwiegend in diesen Bereichen nachweisen. Schließlich können noch Venolen und Gallecapillaren identifiziert werden. Eine Beteiligung von Fibroblasten läßt sich nicht sicher erfassen.

    Description / Table of Contents: Summary In patients with acute monocytic leukemia the cellular layer on the converslips taken from skin windows during the first 6 hours is increased as compared with normal and leukemic controls, and is characterized by an excessive content of leukemic monocytes, thus differing from acute myeloblastic leukemia. This findings yield a new aid for the differentiation of acute leukemias.

Diese Tatsache deckt einen neuen Aspekt in der Differenzierung akuter, myeloischer Leukämien auf.     Description / Table of Contents: Zusammenfassung Bei 23 Patienten mit verschiedenen Leukämieformen wurde in Verlaufsbeobachtungen bis zu einem Jahr gleichzeitig die Muramidaseaktivität (MA) von Cytolysaten peripherer Leukocyten, Serum und Harn gemessen. Die Werte der leukocytären MA schwankten bei Normalpersonen nur gering (1,0–2,8 µg/106), bei Patienten mit Leukämien zwischen kaum meßbaren Werten und 26,0. Die Leukocyten-MA-Änderungen im Krankheitsverlauf waren sowohl leukämischen Monocyten, Promyelocyten, Myelocyten als auch reifen Neutrophilen bei Patienten in Remission zuzuordnen. In den meisten untersuchten Fällen waren Leukocyten-MA und Serum-MA gleichsinnig verändert, wenn auch in unterschiedlich starkem Ausmaß. Lediglich bei je cinem Patienten mit myelomonocytärer und chronisch myeloischer Leukämie wurden extrem erhöhte Serum- und Harn-MA-Werte trotz wiederholt normaler Leukocyten-MA beobachtet. Erniedrigte MA-Werte wurden regelmäßig beim Auftreten von Stammzellen und Lymphoblasten in der Peripherie beobachtet, wobei die Leukocyten-MA empfindlicher als die Serum-MA reagierte. Patienten mit chronisch myeloischer Leukämie wiesen eine geringere Serum-MA auf als aus der Berechnung der peripheren leukocytären MA zu erwarten gewesen wäre. Eine MA im Harn wurde bei Patienten mit Monocytenleukämie, Promyelocytenleukämie, myelomonocytärer Leukämie und auch vereinzelt bei Patienten mit chronisch myeloischer Leukämie gefunden. Eine Beziehung zwischen peripherer Blastenzahl und Harn-MA konnte nur bei Patienten mit Monocytenleukämie und myelomonocytärer Leukämie erstellt werden. 10 Patienten mit einer Überlebenszeit von weniger als 3 Monaten zwischen Diagnosestellung und Tod wurden einer Gruppe von 6 Patienten mit einer Überlebenszeit von mehr als 6 Monaten gegenübergestellt. In der ersten Gruppe war der mittlere Serum-MA-Spiegel signifikant höher als in der zweiten Gruppe.
    Notes: Summary The activity of muramidase (MA) of lysates from peripheral blood leukocytes, serum and urine from 23 patients with various forms of leukemia were measured (by serial determinations up to one year). The values of the leukocytic MA from normal subjects varied only within a small range (1.0–2.8 µg/106 cells), whereas in patients with leukemia, high (up to 26 µg/106) as well as almost undetectable rates were observed. Variations in leukocytic MA during the course of the disease could be ascribed to the number of leukemic monocytes, promyelocytes, myclocytes and mature neutrophiles from subjects in remission. In most cases, the MA of leukocytes and serum was accordingly altered although to different degrees. Only one patient with myelomonocytic and one with chronic granulocytic leukemia had extremely high serum and urine MA although the leukocytic MA remained within the normal range. Low MA values were seen when stem cells and lymphoblasts appeared in the peripheral blood. This was more accurately observable in the leukocytic than in the serum MA. Patients with chronic granulocytic leukemia showed lower serum MA levels than those calculated from the total peripheral leukocytic MA. — Urinary MA was only observed in patients with monocytic, promyelocytic, myelomonocytic and also in a few patients with chronic granulocytic leukemia. A correlation was found only between the number of peripheral leukemic cells and the amount of urinary MA in patients with monocytic and myelomonocytic leukemia. 10 patients with a survival time of less than three months between diagnosis and death had significantly higher serum MA than 6 patients with a survival period of six months or more.
