    Notes: Objective To establish reference ranges for the levels of alpha-fetoprotein, albumin, prealbumin (transthyretin) alpha-1-antitrypsin, transferrin, ceruloplasmin and total protein in the plasma of normal human fetuses and newborn babies.Design Prospective study of individual normal cases to fulfil objectives.Setting Pathology laboratories of the University of Edinburgh and the biochemistry laboratories of the University of Keele.Subjects Twenty-two normal fetuses 13 to 22 weeks of gestation and 66 babies born between 24 and 41 weeks gestation.Results Albumin is the predominant plasma protein throughout gestation. The levels of alpha-fetoprotein and prealbumin fell significantly with increasing gestation, whereas the concentrations of the other proteins studied increased. The ratios of individual proteins to total protein demonstrated similar trends.Conclusions This study provides developmental profiles of normal human fetal plasma proteins to serve as possible reference data for abnormal fetuses. Declining levels of prealbumin (transthyretin) were unexpected and suggest a functional role for this protein in early pregnancy.

    Notes: Abstract: Polyunsaturated fatty acids are needed for normal neonatal brain development, but the degree of conversion of the 18-carbon polyunsaturated fatty acid precursors consumed in the diet to their respective 20-and 22-carbon polyunsaturates accumulating in the brain is not well known. In the present study, in vivo 13C nuclear magnetic resonance spectroscopy was used to monitor noninvasively the brain uptake and metabolism of a mixture of uniformly 13C-enriched 16-and 18-carbon polyunsaturated fatty acid methyl esters injected intragastrically into neonatal rats. In vivo NMR spectra of the rat brain at postnatal days 10 and 17 had larger fatty acid signals than in uninjected controls, but changes in levels of individual fatty acids could not be distinguished. One day after injection of the U-13C-polyunsaturated fatty acid mixture, 13C enrichment (measured by isotope ratio mass spectrometry) was similar in brain phospholipids, free fatty acids, free cholesterol, and brain aqueous extract; 13C enrichment remained high in the phospholipids and cholesterol for 15 days. 13C enrichment was similar in the main fatty acids of the brain within 1 day of injection but 15 days later had declined in all except arachidonic acid while continuing to increase in docosahexaenoic acid. These changes in 13C enrichment in brain fatty acids paralleled the developmental changes in brain fatty acid composition. We conclude that, in the neonatal rat brain, dietary 16-and 18-carbon polyunsaturates are not only elongated and desaturated but are also utilized for de novo synthesis of long-chain saturated and monounsaturated fatty acids and cholesterol.

    Notes: The incorporation of [3H]arachidonate ([3H]AA) and [14C]eicosapentaenoate ([14C]EPA) into glycerophos-pholipids was studied in isolated brain cells from rainbow trout, a teleost fish whose lipids are rich in (n-3) polyunsaturated fatty acids (PUF As), EPA was incorporated into total lipid to a greater extent than AA, but the incorporation of both PUF As into total glycerophospholipids was almost identical. The incorporation of both AA and EPA was greatest into phosphatidylethanolamine (PE). However, when expressed per milligram of individual phosphoglycerides, both AA and EPA were preferentially incorporated into phospha-tidylinositol (PI), the preference being significantly greater with AA. On the same basis, significantly more EPA than AA was incorporated into phosphatidylcholine (PC). When double-labelled cells were challenged with calcium ionophore A23187, the 3H and 14C released from the cells closely paralleled each other, peaking at 10 min after addition of ionophore.The 12-monohydroxylated derivative was the predominant lipoxygenase product from both AA and EPA with a rank order of 12-hydroxyeicosatetraenoic acid (12-HETE) 〉 leukotriene B4 (LTB4) 〉 5-HETE 〉 15-HETE for the AA products and 12-hydroxyeicosapentaenoic acid (12-HEPE) 〉 5-HEPE 〉 LTB5 〉 15-HEPE for EPA products. The 3H/14C (dpm/dpm) ratios in the glycerophospholipids, total released radioactivity, and the lipoxygenase products suggested that PC rather than PI was the likely source of eicosanoid precursors in trout brain cells.

    Notes: Spectral-based signal-processing techniques for characterizing fluctuation data from plasma discharges in the Advanced Toroidal Facility fusion experiment are described. The analysis process moves from wave form display and spectral analysis of single discharges to coherence filtering and database techniques for identifying coherent fluctuations. These analysis techniques are used to distill large volumes of spectral data from many signals per discharge (for many discharges) into simple forms suitable for comparison with the global variation of plasma parameters.

    Notes: Recent experiments in the Advanced Toroidal Facility (ATF) [Fusion Technol. 10, 179 (1986)] have been directed toward investigations of the basic physics mechanisms that control confinement in this device. Measurements of the density fluctuations throughout the plasma volume have provided indications for the existence of theoretically predicted dissipative trapped electron and resistive interchange instabilities. These identifications are supported by results of dynamic configuration scans of the magnetic fields during which the magnetic well volume, shear, and fraction of confined trapped particles are changed continuously. The influence of magnetic islands on the global confinement has been studied by deliberately applying error fields which strongly perturb the nested flux-surface geometry, and the effects of electric fields have been investigated by means of biased limiter experiments.

    Notes: Measurements of electrostatic fluctuations on the edge of the Advanced Toroidal Facility (ATF) torsatron [Fusion Technol. 10, 179 (1986)] are used to study the role of the edge turbulence in the particle transport in this current-free magnetic configuration. Spatial profiles of the plasma electron density ne, temperature Te, and fluctuations in density (ñe) and in the plasma floating potential (φ˜f ) are measured at the edge in electron cyclotron heated plasmas using a Langmuir probe array. At the last closed flux surface (LCFS), r/a≈1, Te≈20–40 eV, and ne≈1012 cm−3 for a line-averaged electron density n¯e=(3–6)×1012 cm−3. The relative fluctuation levels decrease as the probe is moved into the core plasma. For Te(approximately-greater-than)20 eV, ñe/ne≈5%, and eφ˜f /Te≈2ñe/ne at r/a=0.95. The measured fluctuation spectra are broadband (40–300 kHz) with k¯ρs≈(0.05–0.1), where k¯ is the average wave number of the fluctuations and ρs is the ion Larmor radius at the sound speed. Near the LCFS, the density fluctuations can be approximated by ñe/ne ≈0.4/k¯Ln, where Ln is the gradient scale length of ne. The propagation direction of the fluctuations reverses to the electron diamagnetic direction at r/a〈1. The phase velocity of the fluctuations and the electron drift velocity are comparable. The particle flux estimated from the fluctuations is consistent with fluxes obtained from the particle balance using the Hα spectroscopic measurements. Many of the features seen in the ATF edge fluctuations resemble those of Ohmically heated plasmas in the Texas Experimental Tokamak (TEXT) [Phys. Rev. Lett. 62, 1844 (1989)].

    Notes: Experiments involving plasma improvement, confinement scaling, bootstrap currents, and edge fluctuations have been carried out in the Advanced Toroidal Facility (ATF) torsatron [Fusion Technol. 10, 179 (1986)]. Average densities n¯e≤9×1019 m−3 have been obtained, with global energy confinement times τ@B|E≤20 msec. Confinement times generally follow the stellarator/torsatron empirical scaling law, τSL =0.17×P−0.58n0.69eB0.84a2R0.75 (with τSL in seconds, power P in megawatts, density ne in 1020 m−3, and plasma radius a and major radius R in meters). Gas injection during neutral beam injection (NBI) causes increases in ne, so that τ*E does not decrease during NBI. Edge plasma fluctuations are found to exhibit a mode change near the peak of the energy confinement time. Plasma currents observed during electron cyclotron heating have been identified as bootstrap currents.
