    Notes: Zusammenfassung Es werden Normtonsillektomien unter Verwendung von Adrenalin mit Mandelausschälungen, die unter Hexamethoniumwirkung durchgeführt wurden, verglichen. Auf die nicht unwesentliche rolle psychischer Faktoren bei der Blutdruckgestaltung intra operationem wird hingewiesen. Eine ausreichende Blutarmut läßt sich bei Verwendung von 2,5 mg Hexamethonium intramuskulär pro kg Körpeergewicht erzielen. Es ist wünschenscwert, den Blutdruck auf Werte, die bei etwa 60–50 mm Hg liegen, zu senken. Vorteile des Verfahrens sind die zumeist geringe Blutung während der Operaation sowie fast trockene Wundverhältnisse nach beendeter Tonsillektomie bei häufig geringer Gewebsreaktion und unter dem Durchschnitt liegenden postoperativen Beschwerden. Nachteilig wirken sich der größere technische Aufwand und ein durch die Hirnanoxie hervorgerufener inkonstanter, aber nicht immer unerheblicher Würg- und Brechreiz aus.

    Notes: Summary Carcinoid tumours (bronchial and intestinal) were analyzed by immunoblotting for the presence of chromogranin A, B and secretogranin II. In all tumours an antigen corresponding in electrophoretic behaviour to adrenal chromogranin A was present. Lung carcinoids (3 out of 5) contained a relatively high concentration of a proteoglycan form of this antigen in addition. Chromogranin B was found in all tumours. In one and two dimensional immunoblotting it appeared identical to the corresponding adrenal antigen. Secretogranin II was also present, however concentrations (especially in intestinal carcinoids) were low and variable. Furthermore, in intestinal tumours it differed from the adrenal antigen by having a slightly higher molecular size and a more alkaline pI. Immunohistochemistry revealed that the tumour tissues stained positively for all three antigens. For secretogranin II the staining in intestinal tumours was relatively weak and quite variable. These results should provide a defined basis for immunohistochemical screening of carcinoids for the chromogranin/secretogranin antigens.

    Notes: Summary GAWK is a recently discovered peptide isolated from extracts of human pituitary gland and subsequently shown to be identical to sequence 420–493 of human chromogranin B. The distribution of this peptide was studied in human gut, pancreas, adrenal and pituitary glands using antisera to two portions of the 74 amino acid peptide (sequences 1–17 and 20–38). In addition, the co-existence of GAWK immunoreactivity with other peptides and chromogranin B was investigated using comparative immunocytochemistry. In the gut, GAWK was localised mainly to serotonin-containing cells of the mucosal epithelium, where electron microscopy showed it to be stored in typical electron-dense (250 nm diameter) granules, and to a moderate population of nerve fibres in the gut wall. Considerable quantities of GAWK-like immunoreactivity were measured in the gut, up to 36.3±18 pmol GAWK 1–17/g wet weight of tissue (mean±SEM) and 12.4±2.9 pmol GAWK 20–38/g. Chromatography of gut extracts revealed several GAWK-like immunoreactive peaks. GAWK-like immunoreactivity was also detected in endocrine cells of pancreas, pituitary gland and adrenal medulla, where the highest concentrations of GAWK-like immunoreactivity were measured (GAWK 1–17 2071.8±873.2 and GAWK 20–38 1292.7±542.7 pmol/g). Endocrine cells containing GAWK-like immunoreactivity were found also to be immunoreactive for chromogranin B. Our results define a discrete distribution of GAWK immunoreactivity in human endocrine cells and nerves and provide morphological support for the postulated precursor-product relationship between chromogranin B and GAWK. Details of the functions of this peptide are awaited.

    Notes: Summary The co-localization of various antigens in rat chromaffin granules was investigated by the immunogold staining procedure. In ultrathin serial sections staining of chromaffin granules was obtained with antisera against chromogranin A, chromogranin B, secretogranin II and neuropeptide Y. These results indicated that these antigens are costored within chromaffin granules. To further corroborate this point a double immunogold staining procedure was used. This method unequivocally established that chromogranin A, chromogranin B, secretogranin II and neuropeptide Y are co-localized in the same chromaffin granules. These results are relevant for studies demonstrating changes in the level of these peptides in adrenal medulla. The co-localization makes it likely that such changes lead to a different relative composition of the secretory quanta of chromaffin granules.

    Notes: Summary Antisera against chromogranin A and B and secretogranin II were used for analysing the bovine pancreas by immunoblotting and immunohistochemistry. All three antigens were found in extracts of fetal pancreas by one dimensional immunoblotting. A comparison with the soluble proteins of chromaffin granules revealed that in adrenal medulla and in pancreas antigens which migrated identically in electrophoresis were present. In immunohistochemistry, chromogranin A was found in all pancreatic endocrine cell types with the exception of most pancreatic polypeptide-(PP-) producing cells. For chromogranin B, only a faint immunostaining was obtained. For secretorgranin II, A-and B-cells were faintly positive, whereas the majority of PP-cells exhibited a strong immunostaining for this antigen. These results establish that chromogranins A and B and secretogranin II are present in the endocrine pancreas, but that they exhibit a distinct cellular localization.

    Notes: Abstract UV-visible and 13C NMR measurements described in the literature and our 31P NMR measurements support the following mechanism of proton transfer reactions in aqueous solutions of pyridoxamine phosphate: Only the tautomeric equilibrium between neutral form, A N, and zwitterion, A Z, which is analogous to the tautomeric equilibrium of 3-hydroxypyridine in aqueous solution, is important, and that equilibrium does not change upon the dissociation of the second phosphate proton. With these simplifying assumption, we have simulated the relaxation spectrum of the proton transfer reactions of pyridoxamine phosphate in water using parameters from analogous reactions and compared it with our ultrasound and temperature jump measurements. We have found that the relaxation process measured by the temperature jump experiment is mainly caused by the overall reaction A N=A Z (or A N - =A Z - ) and the ultrasound absorption at the isoelectric point between pK2 and pK3 is mainly caused by the overall reaction $$A^{\text{ + }} + {\text{x}} A_N^ - + \left( {1 - {\text{x}}} \right)A_Z^ - = {\text{y}}A_N + \left( {2 - {\text{y}}} \right)A_Z , 0 \leqq {\text{x}} \leqq {\text{1,}} {\text{0}} \leqq {\text{y}} \leqq 2$$ .

    Notes: Abstract The electronic structure of Fe(II) substituting Zn in Horse Liver Alcohol Dehydrogenase was investigated by Mössbauer spectroscopy at various temperatures and applied magnetic fields and by spin Hamiltonian analysis of the results. The novelty we found, is an unusually weak spin coupling of Fe(II) with a diradical (S=1). From ESR results and biochemical findings we conclude, that the corresponding chemical species is triplet oxygen (O2). Oxidation experiments, followed by Mössbauer spectroscopy, show that the spin-coupled species is an outer-sphere Fe(II)… O2 complex occuring as an intermediate of the dioxygen activation reaction, catalysed by Fe(II). A second Fe(II)−O2 complex could be detected, which corresponds to an inner-sphere complex with O2 directly bound to iron. The spin hamiltonian parameters in the coupled system describing the electronic properties of iron are presented. The results are compared with those of iron in other nonheme iron proteins.

    Notes: Summary Antisera against chromogranin A, B and C were used to study the distribution of these acidic proteins in bovine endocrine and nervous tissues. The three chromogranins occur together in several endocrine organs (adrenal medulla, anterior pituitary, endocrine pancreas) and in sympathetic ganglion cells. In the posterior pituitary, only chromogranin C and in the intermediate lobe only A and C are found. The parathyroid gland contains only A, and enterochromaffin cells are immunoreactive for A and B. Cells of the thyroid gland and some cells of the anterior pituitary apparently do not contain any chromogranins. It is concluded that the three chromogranins are not always stored together and that they are not present in all endocrine cells. This distinct localization of the chromogranins indicates some special, although still undiscovered, function for these proteins.
