    Description / Table of Contents: Zusammenfassung In menschlichen Erythrozytenfraktionen (hämoglobinfreie Erythrozytenmembranen; membranfreies Hämolysat; durch DEAE-Cellulose von Hämoglobin befreite Enzym-Protein-Fraktion) wurde mit Hilfe eines NADH-abhängigen optischen Tests mit Asparaginyl1-Angiotensin II-amid als Substrat die Aminopeptidaseaktivität untersucht. 1. Durch fraktionierte Ammoniumsulfatfällung wurden sechs Unterfraktionen aus der Enzym-Protein-Fraktion gewonnen. Mit Hilfe von enzymkinetischen Untersuchungen bei drei verschiedenen pH-Werten (pH 5,0; 7,0; 8,0) mit und ohne Zusatz von zwei verschiedenen Effektoren (Na2EDTA und Ca++) gelang der Nachweis von sechs verschiedenen Enzymen. 2. Durch die Inhibitoren Diisopropylfluorphosphat und p-Chloromercuribenzoat wurde bei verschiedenen pH-Werten (pH 5,0; 6,5; 7,0; 8,0 bzw. 6,5; 7,0; 8,0) eine signifikante Inaktivierung der Aminopeptidaseaktivität des membranfreien Hämolysats beobachtet. 3. Das Temperaturoptimum für die Aminopeptidaseaktivität des Hämolysats lag bei 47°C. Bei 37°C trat nach einer Inkubation von 2 Stunden ein Aktivitätsverlust von 20% ein.
    Notes: Summary Aminopeptidase activity of three fractions of human erythrocytes (membranes free of hemoglobin; hemolysate free of membranes; enzyme protein fraction made free of hemoglobin by DEAE-cellulose) was measured by a NADH dependent optical test using asparaginyl1-angiotensin II-amide as substrate. 1. From the enzyme protein fraction 6 subfractions were obtained by (NH4)2SO4 precipitation. By measuring enzyme kinetics at three different pH-values (pH 5,0; 7,0; 8,0) with and without addition of the effectors Na2EDTA and Ca++ the existence of 6 different enzymes could be demonstrated. 2. The aminopeptidase activity of the hemolysate made free of membranes could be inhibited by diisopropylfluorphosphate and p-chloromercuribenzoate at three different pH-values (pH 5,0; 6,5; 7,0; 8,0 and 6,5; 7,0; 8,0 respectively). 3. A reduction of enzymatic activity of 20% was found after incubation at 37°C for two hours.

    Notes: Abstract Monte Carlo simulation and “series expansion” shows the radius of gyration of large clusters withs sites each to vary ass ρ withρ≃0.56 in two andρ≃0.47 in three dimensions at the percolation threshold, and withρ(d=2)≃0.65 andρ(d=3)≃0.53 for random lattice animals (zero concentration). Clusters up tos=100 were used. The perimeter of random animals approaches 2.8s for larges on the simple cubic lattice. Monte Carlo simulation of the Eden process (“growing animals”) up tos=5,000 indicates a systematic variation of about ±0.05 for the effective exponentρ=ρ(s) and thus suggests that the true asymptotic exponents may be compatible with the predictions of “hyper”-scaling.

    Description / Table of Contents: Zusammenfassung Bericht über die Beobachtung einer neuen HbM-Variante bei einem 4jährigen Jungen mit kongenitaler Cyanose. Die Strukturaufklärung ergab Identität mit HbM Saskatoon = β63 His → Tyr. Bei keinem der untersuchten Familienangehörigen war eine Blutfarbstoffanomalie vorhanden, so daß es sich um eine neue Mutation handelt. HbM Erlangen weist eine normale Sauerstofftransportfunktion auf. Es ist kombiniert mit einer kompensierten Hämolyse, die wahrscheinlich durch eine erhöhte Instabilität des abnorm strukturierten Hämoglobins verursacht wird. Gleichzeitig fand sich eine verminderte Aktivität der NADH-Methämoglobinreduktase.
    Notes: Abstract Report on a further hemoglobin M variant, discovered in a boy with congenital cyanosis. Chemical analysis of hemoglobin structure revealed indentity with HbM Saskatoon (= β63 His → Tyr). The abnormal hemoglobin is characterized by normal oxygen transport function. The observed slight constant hemolysis is thought to be due to increased instability of the abnormal hemoglobin molecule. In addition to the hemoglobin anomaly, the activity of NADH-methemoglobin reductase was reduced by approximately 40% of normal. None of the parents presented with an abnormal hemoglobin, indicating that the occurrence of hemoglobin M Erlangen is most likely the result of a new mutation.

    Description / Table of Contents: Zusammenfassung An 6 normalen männlichen Freiwilligen im Alter von 20–26 Jahren wurden Plasmacortisol und -renin stündlich, Plasmaaldosteron, -angiotensinogen und -angiotensinasen alle 3 Std über jeweils 24 Std unter Kontrollbedingungen und anschließend unter Suppression der ACTH-Freisetzung durch Dexamethason gemessen. Die höchsten Cortisolspiegel fanden sich gegen 7 Uhr, die niedrigsten zwischen 21 und 1 Uhr. Die Gabe von Dexamethason führte zu konstant niedrigen Cortisolkonzentrationen. Aldosteron war unter Kontrollbedingungen und unter Dexamethason gegen 4 Uhr am höchsten und zeigte niedrigste Werte zwischen 16 und 22 Uhr. Zwischen den mittleren Aldosteronkonzentrationen entsprechender Zeitpunkte der Kontroll- und der Dexamethasonperiode bestanden keine signifikanten Unterschiede. Ähnlich dem Aldosteron zeigte das Plasmarenin Maximalwerte gegen 4 Uhr. Alle Mittelwerte entsprechender Zeitpunkte zwischen 7 und 23 Uhr und die jeweiligen 24 Std-Mittelwerte jedes einzelnen Probanden waren unter dem Einfluß von Dexamethason signifikant erhöht. Für die Existenz circadianer Rhythmen des Angiotensinogens und der Angiotensinasen konnte kein Anhalt gewonnen werden. Dexamethason bewirkte keine signifikanten Veränderungen dieser Parameter. Die Ergebnisse deuten darauf hin, daß die circadianen Rhythmen von Aldosteron und Renin miteinander vergleichbar, jedoch nicht exakt mit dem des Cortisols synchronisiert sind. Unter der Hemmung der ACTH-Freisetzung durch Dexamethason steigt die Reninaktivität an, der Aldosteronspiegel bleibt unverändert. Angiotensinogen und die Angiotensinasen, Parameter, die die aktive Konzentration des Angiotensin II beeinflussen können, scheinen an der Regulation des Aldosterons nicht beteiligt zu sein.
    Notes: Summary Plasma cortisol and renin were estimated in 1 h intervals, plasma aldosterone, angiotensinogen and angiotensinases in 3 h intervals over periods of 24 h in six normal volunteers (age 20–26) under control conditions and subsequently under suppression of ACTH released by dexamethasone. Highest cortisol levels were found around 7 a.m., minimum levels between 9 p.m. and 1 a.m. Dexamethasone reduced cortisol to constantly low concentrations. Aldosterone was highest around 4 a.m. under control conditions and under dexamethasone, and showed lowest concentrations between 4 and 10 p.m. There were no significant differences between mean aldosterone concentrations at corresponding time points of the control and the dexamethasone period. Similar to aldosterone, renin showed peak values around 4 a.m. All mean values at corresponding time points between 7 a.m. and 11 p.m. and the 24 hour mean values of each subject were significantly increased under the influence of dexamethasone. No evidence could be achieved for the existence of circadian rhythms of angiotensinogen and angiotensinases. Dexamethasone did not cause significant changes of these parameters. The data suggest that circadian rhythms of aldosterone and renin are similar to each other but not exactly synchronized to that of cortisol. When ACTH release is inhibited by dexamethasone renin increases to maintain normal aldosterone levels. Angiotensinogen and angiotensinases, parameters which may influence the active concentration of angiotensin II, do not seem to be involved in the regulation of aldosterone.
