    Notes: Summary The eyes and urinary bladder of non-diabetic, prediabetic and diabetic Chinese hamsters were evaluated by radioimmunoassay and immunocytochemistry to determine the content and distribution of vasoactive intestinal peptide (VIP). The average concentration of VIP was increased in the eyes of all diabetic (pmol/g = 68%, pmol/organ = 50%) and prediabetic (pmol/g = 152%, pmol/organ = 115%) hamsters compared with age-matched non-diabetic animals. Immunocytochemistry showed that the elevation of VIP was primarily related to greater intensity of fluorescence of the nerve fibres in the vasculature of the choroid. The average content of VIP in the urinary bladder was greater in diabetic animals only on the basis of pmol/organ (135%) and in prediabetics on the basis of pmol/g (87%) compared with non-diabetic animals. Qualitative immunocytochemistry suggested that the elevated level of VIP was related to a larger distribution of nerve fibres in the urinary bladder of diabetic hamsters. The high level of VIP in the eyes and urinary bladder of diabetic and prediabetic hamsters is an interesting observation which should receive further study to determine whether it is an aetiological agent underlying the pathogenesis of ophthalmic complications and neurogenic bladder or the result of some pathological process which affects these organs.

    Notes: Abstract Two nonpenetrating membrane probes, p-azidobenzylphlorizin (p-AzBPhz) and 4,4′-diisothiocyano-2,2′-stilbene disulfonate (DIDS), have been shown in earlier studies to induce dose-dependent changes in red blood cell (RBC) shape and volume at the same low concentrations that inhibit anion transport. In the present work, these ligand-induced morphology and rheology changes were studied using video digital image morphometry (VDIM) and microfiltration techniques. The results of these experiments corroborate our earlier investigation. RBCs were filmed using a Nomarski optics microscope with video camera attachment and cell size and shape changes were computer analyzed using VDIM. Low μm p-AzBPhz or DIDS levels caused collapse of the cell's biconcave structure and cell flattening occurred within 1–2 sec after drug exposure. Higher doses of either agent converted cells to a new steady-state in which a concurrent limited increase in erythrocyte volume and blunt membrane protrusions were produced. These changes were reversed in less than 2 sec by washing the drug from the membrane. Both ligands increased the deformability of RBCs in a dose-dependent manner as determined by filtration through Nuclepore polycarbonate filters (3 μm pore diameter). The improvement in deformability of drug-treated sickle cells was much more dramatic than for normal cells at low p-AzBPhz concentrations. These results support our earlier conclusions that the ligands, through a common interaction with band 3, induce volume-associated cytoskeletal alterations which lead to changes in morphology and flexibility.

    Notes: Summary In dilute suspensions of sperm cells there is electrode noise due to the changes in the electrical double layer during contacts of the suspended cells with the electrode surface. The noise frequency, the number of current fluctuations above one nanoampere per unit time, depends upon the concentration of cells, the potential at the electrode surface and the presence of adsorbed material at the electrode. We have studied the electrode noise due to suspensions of rabbit sperm cells, and have obtained information about the properties of the cell membranes and their ability to adsorb or interact with substances in the environment. The observed differences in the noise frequency between ejaculated and epididymal sperm cells indicate that the ejaculated sperm cell does not adsorb material as readily. The titration of sperm cell suspensions with Zn++ ions shows a lower negative surface charge on the epididymal sperm cell, in agreement with published values on cell electrophoresis. The experiments reported here demonstrate that there are significant changes in the membranes of sperm cells at the later stages of maturation which are independent of membrane charge and affect the ability of the cells to adsorb material. Such adsorption processes could be the first stages of physiological processes (e.g. “sperm capacitation”, “acrosome reaction”) that are known to occurin vivo, and that play important roles in fertilization.

    Notes: Summary When mammalian sperm cells are exposed to solutions of albumin there are changes in the membranes of some species that resemble those that normally occur in the uterus prior to fertilization. We have shown that albumin molecules adsorb on to the membranes of ejaculated rabbit sperm cells, and that the equilibrium binding constant,K, (1) varies inversely with the albumin concentration, (2) is independent of the sperm cell concentration in the range 106–107 per ml, (3) is independent of the time of exposure of the sperm cells to the albumin solution, and (4) decreases in the presence of Ca++ and Mg++ ions. An unusual aspect of the adsorption is that if the albumin concentration is given the symbol [A],K[A] is a constant in our measurements. This means that for virtually the entire range of [A] studied, the sperm cells bind albumin so that half of the available surface is coated and half remains uncoated. This situation is rather remarkable and it suggests a role that adsorption could play in the physical processes preceding fertilization. In purely physical systems, the optimum for the bridging and flocculation of particles that are coated with adsorbed macromolecular films occurs when half of the available surface is covered. The sperm cell appears to provide the optimal situation for interacting with itself or with another surface.

    Notes: Studies were carried out on the fractionation and analysis of egg lipid by several new techniques employing thin-layer chromatography. The fatty acid composition of the major lipid classes and subfractions thereof was determined by a combination of alkali-isomerization and gas-liquid chromatography. The positional arrangements of the fatty acids in the glycerides and lecithin fraction were determined. The fatty acids in lecithin were distributed mainly in one type, the α-saturated β-unsaturated type, in contrast to the fatty acids of triglycerides.
