    Notes: Abstract Single tolerated doses of a lyophilizate fromAmanita phalloides (APL) failed to increase the survival rate of mice after the LD90 of the mushroom. Also, pretreatment with large doses of ethanol did not protect against lethal doses of APL. However, with combined administration of single tolerated doses of APL and of ethanol, the survival rate of mice after a lethal challenge with APL was increased from 13% to up to 100%. Both ethanol and APL potentiated the effect of barbiturate narcosis, but the combined pretreatment was not more effective. The protection against APL afforded by the combined regimen may be due to inhibition of activating hepatic microsomal enzymes, but biochemical studies are needed to support this hypothesis.

    Notes: Summary Survival of mice after lethal doses of a lyophilizate fromAmanita phalloides (‘death cap’) was markedly increased by single doses of ethanol applied 30 min before or 5 min after the mushroom. Hepatic histopathological damage (confluent necrosis) was largley prevented. Acute, but not chronic, consumption of ethanol may thus influence favorably the outcome of death cap poisoning and should be taken into consideration in the evaluation of therapeutic measures.

    Notes: Abstract Pretreatment with zinc aspartate increased the survival rate of mice poisoned with a lyophilisate from the mushroomAmanita phalloides (APL). In the livers of the zinc-treated animals, confluent necrosis was cleared earlier. Zinc also prevented the reduction of brain noradrenaline caused by the mushroom as well as the rise of brain dopamine and brain weight.d-Penicillamine also protected against the lethal effects of APL. The survival afforded by zinc pretreatment was marginally ameliorated by additional oxygenation.

Die Nachteile, die diesen Mitteln vorläufig noch anhaften, bestehen in ihrer mangelnden Elektivität, d. h. allgemeine Cytotoxicität und allgemeine Immunosuppression statt gezielter Hemmung der Transplantationsimmunität. Dennoch ist dank dieser Mittel die Organtransplantation in ihrem heutigen Ausmaß und Gelingen möglich geworden.
    Notes: Summary The immunosuppressive chemicals which are in use at present, when given in a dosage that will suppress rejection in practically every case, will not be tolerated by all patients. The drawback attached to these drugs for the time being is a lack of selectivity, that is a generalized cytotoxicity and immunosuppressive action instead of restricted inhibition of transplantation immunity. In spite of these imitations it is these drugs which made the success of organ transplantation possible.

    Notes: Summary Extended survival of skin homografts in mice was obtained by treatment with a methylhydrazine derivative (1-methyl-2-p-(isopropylcarbamoyl)-benzylhydrazine hydrochloride), representative of a new class of cytotoxic agents. Administration of 100 mg/kg prior to and continuously after the transplantation led to similar survival times as when 300 mg/kg was given daily only after the transplantation. In mice, the compound seems to be more effective than most of the drugs so far known to suppress transplantation immunity.
