    Description / Table of Contents: Summary Comparative studies on the effect of incubation of EDTA-plasma at 37° C on platelet aggregation are presented. The aggregation of platelets was determined simultaneously by a microscopic and a fotometric method. After incubation of platelet rich plasma at 37° C for at least 10 min no aggregates can be detected by the fotometric method. Microscopical investigation reveals loose aggregates with altered formation kinetics. Because of chronological formation of the aggregates platelets from different blood-donors can be classified in different groups. The behaviour of platelets towards temperature is defined as “incubation resistance”. Parallel with the investigation of incubation of EDTA-plasma tests were done with recalcified EDTA-plasma. In this case aggregates can be found with both methods. The kinetics of aggregation are those of platelets which have not been incubated prior to addition of Calcium.
    Notes: Zusammenfassung Es werden vergleichende Untersuchungen über den Einfluß der Inkubation von EDTA-Plasma bei 37° C auf die Plättchenaggregation mit Hilfe der Rühr- und Kammermethode beschrieben. Nach Inkubation bei 37° C zeigte die Rührmethode keine Aggregation mehr an. In der Kammer hingegen fanden sich lockere Aggregate, deren Bildung eine gegenüber den Kontrollen veränderte Kinetik aufwies. Auf Grund des zeitlichen Aggregationsverhaltens bei der Inkubation lassen sich die Plättchen verschiedener Versuchspersonen in Gruppen einteilen. Das Verhalten der Plättchen gegenüber Temperatureinflüssen wird als „Inkubationsresistenz“ bezeichnet. Parallel zu den Inkubationsversuchen mit EDTA-Plasma wurden Inkubationsversuche mit recalcifiziertem EDTA-Plasma durchgeführt. Dabei lassen sich sowohl mit Hilfe der Rühr- als auch Kammermethode Aggregate nachweisen. Die Aggregationskinetik entspricht der nicht inkubierter Plättchen.

    Description / Table of Contents: Summary The effect of incubation of platelet rich plasma at 37° C on the aggregation of platelets is reported. The determination of aggregation is done by the fotometric method. Platelet aggregation is inhibited by incubation at 37° C. The inhibition is increased by incubation times from 2 to 10 min from 0 to nearly 100%. Platelet rich plasmas can be classified in 2 groups, the inhibition of which is significantly different at incubation times between 3 and 4 min. The significance of an appropriate reference system for investigations of this kind is commented.
    Notes: Zusammenfassung Es wird über den Einfluß der Inkubation von plättchenreichem EDTA-Plasma bei 37° C auf die Plättchenaggregation und ihre Bestimmung mit Hilfe der photometrischen Methode (Rührmethode) berichtet. Die Plättchenaggregation wird durch die Inkubation gehemmt. Die Hemmung nimmt während eines Inkubationszeitraumes von 2 bis etwa 10 min stetig von 0 auf nahezu 100% zu. Die plättchenhaltigen Plasmen der untersuchten Versuchspersonen lassen sich in 2 Gruppen einteilen, deren Hemmkurven bei Inkubationszeiten zwischen 3 und 4 min einen signifikant unterschiedlichen Verlauf zeigen. Auf die Bedeutung eines geeigneten Bezugssystems für derartige Untersuchungen wird hingewiesen.

    Notes: Summary About 50% of the circulating platelets can be taken from healthy donors by means of the cell separation technique. In order to investigate the effect of cell separation on thrombopoesis bone marrow samples were taken from 12 voluntary donors up to 5 times. The megacaryocytes were analyzed before and after cell separation by morphometric measures and determinations of platelet count and platelet function (adhesion, aggregation, spreading, retraction) were carried out. Removing of large amounts of platelets form the circulation by stimulation of thrombopoesis resulted in an increase of the peripherical platelet count already 48 h after cell separation. Of the platelet functions tested only aggregation induced by collagen was found to be increased after cell separation as compared to that before the procedure.

    Description / Table of Contents: Summary A method for the determination of the aggregation of blood platelets is reported, using a phase contrast microscope. The temporal course of the aggregation is followed in a counting chamber. The decrease of the count of free platelets as a measure for the count of aggregates formed proves to be the most sensitive criterion of platelet aggregation. Additionally the count of aggregates per mm3 is determined and the aggregates differentiated for loose, intermediary and compact aggregates. Further a modified photometric method for the determination of the aggregation of platelets is given, using a commercial photometer with an attachment which may easily be built. The two methods are compared for their applicability on the determination of platelet aggregation.
    Notes: Zusammenfassung Es wird über eine Methode zur Erfassung der Plättchenaggregation berichtet, bei der die Aggregation in einer Zählkammer mit dem Phasenkontrastmikroskop beurteilt und in ihrem zeitlichen Verlauf verfolgt wird. Die Abnahme der Zahl der freien Plättchen als Maß für die Zahl der gebildeten Aggregate erweist sich dabei als das empfindlichste Kriterium für die Beurteilung der Plättchenaggregation. Weiter werden die Zahl der Aggregate pro mm3 bestimmt sowie eine Differenzierung in lockere, Übergangs-und kompakte Aggregate vorgenommen. Es wird gleichzeitig über eine Modifikation der photometrischen Methode zur Messung der Aggregation berichtet, die es ermöglicht, ein serienmäßiges Photometer mit Hilfe eines leicht herzustellenden Zusatzes zur Messung zu verwenden. Beide Methoden werden hinsichtlich ihrer Aussagefähigkeit miteinander verglichen.

    Description / Table of Contents: Zusammenfassung Die Aktivität des Selenenzyms Glutathionperoxidase (EC 1.11.1.9) wurde in den Blutplättchen von 15 Patienten mit frischem Herzinfarkt und 13 Kontrollpersonen gemessen. Die Blutplättchen der Patienten zeigten signifikant niedrigere Enzymaktivitäten (P(t)〉0.99). Möglicherweise steht dies mit der Erkrankung in pathogenetischem Zusammenhang.
    Notes: Summary The activity of the selenoenzyme glutathione peroxidase (EC 1.11.1.9) was determined in platelets of 15 patients with acute myocardial infarction and 13 control subjects. The platelets of the patients had significantly lower activities of the enzyme (P(t)〉0.99). This may be related to the pathogenesis of the disease.

    Notes: Summary Delayed potassium channels were studied in internally perfused neurone somata from land snails. Relaxation and fluctuation analysis of this class of ion channels revealed Hodgkin-Huxley type K channels with an average single channel conductance (γ K) of 2.40±0.15 pS. The conductance of open channels is independent of voltage and virtually all K channels seem to be open at maximum K conductance (g K) of the membrane. Voltage dependent time constants of activation ofg K, calculated from K current relaxation and from cut-off frequencies of power spectra, are very similar indicating dominant first-order kinetics. Ion selectivity of K channels was studied by ion substitution in the external medium and exhibited the following sequence: T1+〉K+〉Rb+〉Cs+〉NH 4 + 〉Li+〉Na+. The sequence of the alkali cations does not conform to any of the sequences predicted by Eisenman's theory. However, the data are well accommodated by a new theory assuming a single rate-limiting barrier that governs ion movement through the channel.
