    Notes: Abstract Objectives: To determine whether continuous venovenous hemofiltration leads to extraction of tumor necrosis factor alpha (TNFα) and cytokines from the circulation of critically ill patients with sepsis and acute renal failure and to quantitate the clearance and the removal rate of these cytokines and their effect on serum cytokine concentrations. Design: Prospective, controlled study in patients with continuous venovenous hemofiltration (24 l/24 h) using a polysulphone membrane in patients with acute renal failure. Patients: 33 ventilated patients with acute renal failure of septic (n = 18) and cardiovascular origin (n = 15) were studied. Interventions: Hemodynamic monitoring and collection of blood and ultrafiltrate samples before and during the first 72 h of continuous hemofiltration. Measurements and main results: Cardiovascular hemodynamics (Swan-Ganz catheter), Acute Physiology and Chronic Health Evaluation II score, creatinine, electrolytes, and blood urea nitrogen were recorded daily. Cytokines (TNFα, TNFα-RII, interleukin (IL) 1β , IL1RA, IL2, IL2R, IL6, IL6R, IL8, IL10) were measured in prefilter blood and in ultrafiltrate immediately preceding and 12, 24, 48, and 72 h after initiating continuous venovenous hemofiltration (CVVH). Septic patients showed elevated cardiovascular values for cardiac output (7.2 ± 2.1 l/min), cardiac index (4.2 ± 1.3 l/min per m2), and stroke volume (67 ± 23 ml) and reduced values for systemic vascular resistance (540 ± 299 dyn · s · cm− 5). All hemodynamic values normalized within the first 24 h after initiating CVVH treatment. TNFα was 1833 ± 1217 pg/ml in septic patients and 42.9 ± 6.3 pg/ml in nonseptic patients (p 〈 0.05) prior to CVVH. TNFα was detected in ultrafiltrate but did not decrease in blood during treatment with CVVH. There was no difference in IL 1β between septic (3.8 ± 1.9 pg/ml) and nonseptic patients (1.7 ± 0.5 pg/ml). No significant elimination of cytokines was achieved in the present study by CVVH treatment. Conclusions: These findings demonstrate that CVVH can remove TNFα and special cytokines from the circulation of critically ill patients. Cardiovascular hemodynamics seemed to improve in septic patients after induction of hemofiltration treatment, although there was no evidence that extracorporeal removal of cytokines achieved a reduction in blood levels. The study indicates that low volume continuous hemofiltration with polysulphone membranes in patients with acute renal failure is not able to induce significant removal of cytokines.

    Notes: This study estimates the impact of reduced demand for U.S. corn during the 2000/01 marketing year caused by the StarLink-contamination event in the year 2000. A partial equilibrium model is constructed that separates both the domestic and foreign demand functions for corn into food and non-food demand curves. The complexities arising from the Loan Deficiency Payment (LDP) Program are also incorporated. The empirical results indicate that the reduction in Japanese corn demand alone would have reduced the average price received by U.S. corn producers in 2000/01 by between U.S. 4.17¢ to U.S. 5.18¢ per bushel if LDP payments did not partially offset the market impact. However, after adjusting for LDP payments, the average price received by U.S. corn producers dropped by only between U.S. $0.50¢ and U.S. 0.83¢ per bushel over the 2000/01 marketing year. This translates into a loss in revenue of between U.S. $48 and U.S. $78 million.

    Notes: We compare the current Canadian supply management regime in which producers and importers benefit from rent-seeking activities that set production quota and import quota levels with those under a tariff, in which producers partakes in rent-seeking activities in order to induce the government to introduce a favorable tariff regime. We explore two different quota-setting games: (1) the import quota and production quota are set at a level that arises from a Cournot-Nash equilibrium between producers and importers; and (2) the producer marketing board acts as a Stackelberg leader, taking into account the importers' reaction to its production quota level. We compare these quota-setting games with two different tariff-setting games: (1) A non-cooperative game in which the government sets the tariff at a level that maximizes tariff revenue; and (2) A cooperative game in which producers, through rent-seeking activities, induce the government to set the tariff at a level that maximizes joint government and producer rents.

    Notes: The sharp rise in energy prices in the 1980s triggered a strong interest in the production of ethanol as an additional energy component. Economists are divided as to the payoffs from ethanol derived corn in part because of the complex interrelationship between energy produced from ethanol and energy from fossil fuels. Using a welfare economic framework, we calculate that there can be treasury savings from ethanol using tax credits as these subsidies can be smaller than direct payments to corn farmers which are essentially eliminated from the expansion of ethanol. Also, to the extent that ethanol dampens fuel prices there can be a net welfare gain from ethanol production in the presence of ethanol subsidies.

    Description / Table of Contents: Zusammenfassung Die kombinierte Chemotherapie mit Vincristin (VCR) und 5-Fluorouracil (5-FU) war nach Festlegung der LD50 im humanmedizinischen Dosierungsbereich bei 150 kongenital thymusdysplastischen “nude”-Mäusen ohne Komplikationen durchführbar. Unter spf-Bedingungen durchliefen ca. 5 Wochen alte syngene Balb/c-“nude”-Mäuse einen 9 tägigen Therapiezyklus. Die Teilsynchronisation wurde mit VCR am Tag 1 eingeleitet und 5-FU jeweils vom 1. bis 3. Tag intraperitoneal appliziert. Pathohistologisch entstanden unter VCR bei einer Dosis von 20% der LD50 bei allen Tieren Milzatrophien und schwere Jejunitiden. Die durch Verminderung überwiegend segmentkerniger Zellen entstandenen Leukopenien waren erwartungsgemäß reversibel. Nach 5-FU-Applikation entstanden bei einer 20% igen Dosierung der LD50 hauptsächlich Zystitiden, bei 67% Bronchitiden und Lobärpneumonien. In der Kombination mit humanmedizinisch relevanten Dosierungen entwickelten die Tierkollektive regressive Veränderungen an Harnblasen- und Zottenepithelien des Jejunums. Neben Hepatitiden mit Gruppennekrosen fanden sich Pneumonien bei 50% der Tiere; Milzatrophien waren mäßig ausgeprägt. Die Lymphknoten zeigten eine Tendenz zur Strukturauflösung der Follikel ohne spezifische Veränderungen der paracorticalen Region. Hämatologisch war nach reversibler Leukopenie mit Granulozytopenie die Einleitung eines 2. Zyklus am 9. Tag möglich. Nach Kenntnis dieser Befunde ist die Möglichkeit gegeben, die Wirksamkeit von VCR und 5-FU auf das Wachstum humanen kolorektalen Tumorgewebes nach erfolgter Transplantation auf “nude”-Mäuse in vivo zu testen.
    Notes: Summary Five-week-old syngeneic, female BALB/c nude mice were injected i. p. with vincristine (VCR) and 5-fluorouracil (5-FU) to determine the LD 20 of these drugs under spf-conditions. One hundred fifty animals were given dosages similar to those administered clinically, and a complete autopsy was performed on all nude mice. Whereas VCR caused splenic atrophy and regressive changes in the jejunal epithelium, the application of 5-FU was followed mainly by regressive changes in the transitional epithelium of the bladder, and 67% of the animals developed pneumonia. These pathohistological changes were also seen after a combined chemotherapy, and a temporary leucopenia was reversible. These experiments showed that chemotherapy of nude mice with VCR and 5-FU is possible provided they have been successfully xenotransplanted with human colorectal adenocarcinomas.
