    Notes: In fourteen patients with food allergy, intragastral provocation under endoscopical control (IPEC) was performed. In all patients positive immediate-type reactions of the gastric mucosa were observed consisting of oedema, erythema and petechial bleeding. Microscopically, mast cell degranulation was observed and measured by mast cell counts using the o-phthaldialdehyde technique. Concomitantly, tissue histamine content in gastric mucosa decreased significantly after allergen provocation, while there was no change in normal volunteers. Plasma histamine concentration increased in most patients; the increases were most evident in four patients showing mild systemic reactions (urticaria and bronchospasm). The technique described might prove to be useful in establishing the diagnosis in doubtful cases of food allergy.

    Description / Table of Contents: Zusammenfassung Es wurden die neutralen Proteasen (Elastase, Kathepsin G und Kathepsin B1) aus den Granula menschlicher Leukocyten isoliert und der dosisabhängige Einfluß von Glykosaminoglykanpolysulfat (GAGPS, Luitpoldwerke München) auf die Aktivität dieser Enzyme mit chromogenen Substraten getestet. Eine 70 %ige Hemmung von Elastase, wie in der Literatur beschrieben, konnten wir bestätigen. Dagegen wird die enzymatische Aktivität von Kathepsin G und Kathepsin B1 von GAGPS nicht beeinflußt. Da eine Hemmung von Elastase bereits mit Inhibitorkonzentrationen von 1 μg/ml erreicht wird, erklären diese Befunde die therapeutische Wirksamkeit von Arteparon.
    Notes: Summary Neutral proteases (elastase, cathepsin G and cathepsin B1) from human leukocytes were isolated and the dose-dependent influence of glycosaminoglycanpolysulfate (GAGPS, Luitpoldwerke München) on the activity of these enzymes was tested using chromogenic substrates. The inhibition of elastase up to 70 %. (as reported in the literature) could be confirmed, whereas cathepsin G and cathepsin B1 were resistant to inhibition under the applied conditions. Because the inhibition of elastase has already been achieved at concentrations of 1 μg/ml, these results may account for the therapeutic efficiency of Arteparon.

    Description / Table of Contents: Zusammenfassung Die antithrombogene Qualität der Neointima in implantierten Gefäßprothesen wurde charakterisiert durch Messung der PGI1α-Freisetzung aus der Gefäßwand ins Lumen mittels einer speziellen Versuchsanordnung und einem 6-Keto-PGF1α- Radioimmunoassay. In die A. carotis von Schafen interponierte Gefäßprothesen (Microvel Doppelvelour Dacron, l = 10 cm, d = 6 mm) erreichten 20 Wochen postoperativ in den anastomosennahen Abschnitten (30 mm), wo endothelähnliche Zellen vorherrschten (REM), eine PGI2-Syntheseleistung von 0,3–0,6pg/mm2/5 min — entsprechend 10% der normalen Gefäßwand (Fritsche 1980). In mittleren Prothesenabschnitten mit unspezifischen Oberflächenstrukturen (REM) wurde nur etwa halb so viel antiaggregatorisches PGIZ pro Zeiteinheit freigesetzt.
    Notes: Summary To characterize the non-trombogenicity of the newly formed endothelium in vascular prostheses, we have developed a device to measure PGIZ release from the vascular wall into the lumen using a 6-Keto-PGF1α-radio-immunoassay. Vascular grafts (Microvel Double Velour Dacron, l = 10 cm, d = 6 mm) were placed in the carotid arteries of sheep. Twenty weeks post-operatively the synthesis capacity of the neo-intima for PGI2 approximates 0.3–0.6pg/mm2/5 min (equivalent to 10% of the normal vascular wall; Fritsche 1980) in the vicinity of the anastomoses (30 mm), where endothelial-like cells predominate (SEM). Half as much was released from the disc-haned. non-snecific cells lining the medial sections of the grafts (SEM).

    Description / Table of Contents: Zusammenfassung Zur Prüfung des Nutzens von Extremitäten-Perfusionen mit einem O2-Träger vor Replantationen wurden hintere Kaninchenextremitäten amputiert, 4 h bei 4°C gelagert, 30 min mit Fluosol 43 (Pfrimmer, Erlangen) perfundiert und nach Anschluß an die Halsgefäße gesunder Tiere für 2 h rezirkuliert. Verglichen mit nichtperfundierten Kontrolltieren wurden höhere finale CP-Werte (111 % gegenüber 67 % der Anfangswerte) erreicht. Weiter wurden der arterielle Zufluß und der periphere Widerstand der Beine sowie der arterielle Blutdruck. Serum-K+ und Säure-Basen-Status der Empfäneertiere günstig beeinflußt.
    Notes: Summary The benefit of perfusion using an oxygen carrier was investigated in extremities to be replanted. Lower extremities of rabbits were amputated, stored for 4h at 4°C, perfused with oxygenated Fluosol 43 (Pfrimmer, FRG) and recirculated for 2 h after connection with the neck vessel of a healthy rabbit. Compared to non-perfused control legs, higher final CP-values were recorded (111 % vs 67 % of the normal values). Furthermore, the arterial flow and the peripheral resistance of the legs, as well as the arterial blood pressure, the serum K+ and acid base balance of the recipient animal were positively influenced.

Aprotonin verhindert ferner bei Ratten signifikant den Übertritt von Proteinen (1317-HSA) aus dem Bauchraum in die Zirkulation.
    Notes: Summary Peritoneal proteinases during acute pancreatitis deplete active endogenousα2M in peritoneal exudates (p. e.) in patients (2.8 ± 1.2 nMol/g protein vs. 41.3 ± 6.7 in patient's plasma, despite an immunological reactivity of 63 %) and rats (15.4 ± 3.0 nMol/g protein vs. 68.8 + 27.1 in plasma). Proteolytic activities of p. e. on fibrin plates (14.5 ± 8.7 μg trypsin equivalents/ml) could be inhibited byα2M to a rate of 70 %, by aprotonin (Trasylol, Bayer AG) to 80 %. Furthermore, in rats, intraperitoneal aprotinin reduces significantly the movement of proteins (1317-HSA) from the peritoneal space to the circulation.

    Description / Table of Contents: Zusammenfassung In experimentellen Untersuchungen wurden Zusammenhänge zwischen posttraumatischen leukocytären Infiltrationen des Lungeninterstitiums und ödematösen Lungenveränderungen aufgezeigt. Aus menschlichem Frischblut wurden Leukocytenkonzentrate hergestellt, deren Lysosomen isoliert und Enzyme extrahiert. Vier neutrale Proteinasen konnten nachgewiesen und durch ihre spezifischen Substrate Bowie ihr Inhibitorspektrum charakterisiert werden. Die leukocytäre Kininogenase ist durch den therapeutisch anwendbaren polyvalenten Proteinaseninhibitor Trasylol®, hemmbar. In Tierversuchen wurde die pathogenetische Bedeutung der Kinine für die Ödembildung nachgewiesen. Der polyvalente leukocytäre Proteinaseninhibitor ist nur in geringer Menge nachweisbar.
    Notes: Summary Experimental investigations were carried out to correlate posttraumatic leukocyte infiltration of lung interstitium and edema formation. From human leukocyte lysosomes, four different neutral proteinases could be extracted and characterized by means of their specific substrates and their inhibitor spectra. Granulocyte kininogenase was sensitive to inhibition by Trasylol®, a polyspecific therapeutic proteinase inhibitor. In morphologic experiments on rabbits, the participation of kinins in the events leading to pulmonary edema is made obvious. The existence of small amounts of a polyvalent proteinase inhibitor of leukocyte origin could be shown.
