    Description / Table of Contents: Zusammenfassung Bei 40 Kranken mit Multipler Sklerose (M.S.) und verschiedenen Kontrollgruppen von insgesamt 55 Personen wird die Lymphocytentransformation (14C-2-Thymidininkorporation) durch eine Anzahl von Antigenen untersucht. — M.S.-Kranke haben eine signifikant erhöhte Transformationsrate durch basisches Markscheidenprotein. Es besteht eine Korrelation zum Prozeßstadium und zum Schweregrad der Erkrankung. Der Befund zeigt immunologische Vorgänge an, die auf den Untergang von Markscheidengewebe zurückgehen, ist aber nicht für M.S. spezifisch, denn er läßt sich auch bei Kranken mit akuten cerebralen Prozessen erheben. Mit dem Lymphocytentransformationstest kann der Nachweis einer Autoaggressionskrankheit für die M.S. nicht erbracht werden. — Die Transformationsraten durch PHA, Human-Gammaglobulin und Tuberkulin, sowie die Spontantransformation bei M.S.-Kranken entsprechen denen der Kontrollen. Vaccinia-Antigen führt zu einer signifikant erhöhten Lymphocytentransformationsrate bei M.S.-Kranken und bei frisch gegen Pocken geimpften Personen. Dieser Befund bedarf weiterer Abklärung als Hinweis auf eine mögliche chronische Virusinfektion bei M.S. — Durch eine Langzeitbehandlung mit Azathioprin kommt es für alle geprüften Antigene bei Kranken mit M.S. nicht zu einer signifikanten Erniedrigung der Transformationsraten.
    Notes: Summary Lymphocyte transformation (14C-2-thymidin incorporation) is tested with a number of antigens on 40 patients with Multiple Sclerosis (M.S.) and different control groups totalling 55 subjects. M.S. patients have a significantly increased transformation by myelin basic protein. The transformation indexes correlate with the stage and the degree of severity of M.S. The finding indicates immune reactions induced by the destruction of myelin. This is, however, non specific to M.S. alone, since this same finding arises also in patients with other acute cerebral diseases. The lymphocyte transformation cannot proof that M.S. is an autoimmune disease. — The transformation by PHA, human gamma-globulin and tuberculin, as well as the spontaneous transformation in M.S. patients matches those of the control groups. Vacciniaantigen leads to a significantly increased lymphocyte transformation in M.S. patients as well as in subjects recently inoculated against smallpox. This finding needs further clarification and points to a possible chronic virus infection in M.S. A prolonged therapy with Azathioprin does not significantly reduct the transformation for any tested antigen in M.S. patients.

    Notes: Abstract We used the short-lived radionuclide, 82Br− to follow γ-aminobutyrate (GABA) receptor-mediated halide exchange into membrane vesicles from rat cerebral cortex in millisecond and second time regions using quench-flow technique. The radioisotope was prepared by neutron capture [81Br−(n,γ)82Br−] on irradiation of a natural isotope of bromine, 81Br− in a neutron flux. 82Br− decays by β-emission with secondary γ-emission. Possible advantages of 82Br− over 36Cl− in anion tracer measurements include, (a) a short lifetime (t1/2 = 35.3 hr), which alleviates contamination and disposal problems, (b) high counting efficiency (1.54) due to the secondary radiation, (c) measurement with a γ-counter as well as a β-counter, (d) a simple preparation not requiring subsequent purification steps giving a specific activity depending on the irradiation time. With 6 hr irradiation time the specific activity was sufficient to make measurements with 〈1 mm Br−, which is less than the bromide concentration known to affect the properties of GABAA receptor. The radiotracers, 82Br− and 36Cl− could be compared with the same solution composition. In conditions where a direct effect of binding of halide to receptor does not contribute to a difference in measured ion-flux, 82Br− was translocated only marginally faster than 36Cl−. The effect of chlordiazepoxide (CDPX) (2–250 μm) on the progress of GABA (10 μm)-mediated 82Br− uptake was measured in a time range of 200 msec to 20 sec using quench-flow technique. The two phases of anion exchange previously reported in this experimental model with GABA alone were observed. The rate of 82Br− exchange was increased 2.3-fold at 30–60 μm CDPX and was not further increased with increasing [CDPX]. The rate of halide exchange is a measure of open channel concentration. The isotope exchange rate constant, J, in a membrane vesicle preparation, is a measure of the membrane permeability per internal volume/surface area, J = PmA/V. Receptor desensitization rate was also increased by CDPX, but unlike the isotope exchange rate, it continued to increase up to at least 250 μm CDPX.

    Description / Table of Contents: Summary The humoral and cell-mediated immune response was investigated in 90 patients suffering from chronic posttraumatic osteomyelitis. In 10–20% of the patients the serum IgG and IgM was increased, in 5–10% the levels were decreased. The values were compared with those of a control group of 35 normal healthy volunteers. Of 55 patients with staphylococcal infection only 25 had a positive antistaphylolysin titer. Eleven out of 25 patients with staphylococcal osteomyelitis showed a positive in vitro lymphocyte transformation after stimulation with α-staphylolysin (mean transformation rate 5.4 ± 0.9). The phytohemagglutinin and the pokeweed-mitogen response were normal. A comparison of the immunological reactivity of the patients and the clinical severity of the disease showed that patients with established delayed-type hypersensitivity phenomena suffered a significantly worse clinical course of the disease.
    Notes: Zusammenfassung Neunzig Patienten mit chronischer post-traumatischer Osteomyelitis wurden immunologisch untersucht. Die Ergebnisse wurden mit einer Kontrollgruppe von 35 gesunden Freiwilligen verglichen. In 10–20% der Patienten fanden sich erhöhte Serum-IgG- und IgM-Konzentrationen; in 5–10% waren diese erniedrigt. Unter 55 Patienten mit Staphylokokkeninfektion zeigten nur 25 einen positiven Antistaphylolysintiter. Elf von 25 untersuchten Patienten mit Staphylokokken-Osteomyelitis zeigten eine positive in-vitro-Lymphozytentransformation nach Stimulation mit α-Staphylolysin (durchschnittliche Transformationsrate 5,4 ± 0,9). Dabei war die Phytohämagglutinin- sowie die Pokeweed-Mitogen-Antwort normal. Ein Vergleich der immunologischen Reaktionslage der Patienten mit dem Schweregrad der klinischen Erkrankung ergab, daß die schwersten Verläufe in der Gruppe der Patienten mit nachweisbarer zellulärer Überempfindlichkeit gefunden wurden.

    Notes: Abstract The ultrastructural and biochemicalphysiological aspects of postfloral greening have been studied in hypsophylls of Heliconia aurantiaca Ghiesbr., Guzmania cf. x magnifica Richter and Spathiphyllum wallisii Regel. In all three species the greening of the hypsophylls is due to plastid transformation, chloroplast formation proceeding from the initially different types of plastids. The degradation process of the original plastid structures and the mode of thylakoid formation are distinct in each case. In none of the species do the transformed plastids look identical to the chloroplasts of the corresponding foliage leaves. On a chlorophyll basis, the rate of photosynthesis of the greened hypsophylls surpasses the rate of the leaves considerably in Spathiphyllum, but is much lower in Heliconia (no data for Guzmania). In all species, anatomy, plastid structure, pigments, 77° K-fluorescence emission, ribulose-1,5-bis-phosphate carboxylase activities and short-term photosynthesis 14CO2-assimilation patterns prove the greened hypsophylls to be capable of providing additional carbon to the developing fruits, thus supplementing the import of organic matter from the foliage leaves.

    Description / Table of Contents: Summary The lymphocyte transformation test was applied to 15 persons vaccinated against smallpox and to 10 unvaccinated persons. It is shown that vacciniaantigen results in a specific stimulation of sensitized cells. It is not yet possible to explain the high transformation indexes to encephalitogenic protein (basic protein) in four vaccinated persons.
    Notes: Zusammenfassung Bei 15 Pocken-geimpften und 10 Pocken-ungeimpften Personen wird die Lymphozyten-transformation durch Vaccinia-Antigen untersucht. Es wird gezeigt, daß Vaccinia-Antigen eine spezifische Stimulation sensibilisierter Zellen bewirkt. Die dabei in vier Fällen beobachtete Erhöhung der Transformationsrate von Lymphozyten durch enzephalitogenes Protein (basisches Protein) kann noch nicht geklärt werden.

    Notes: Abstract Polypeptide microspheres containing polycysteine crosslinked with polylysine were prepared and radiolabeled with186Re and188Re. High labeling yields with the microspheres with both186Re and188Re (97%) were obtained, and above 99% retention of radiolabels in water in 24 hours was obtained. Rhenum-186 labeled polycysteine and polylysine microspheres (1∶1 ratio, 20 μm as mean diameter) were injected intra-articularly into the rear stifes (knee joints) of normal New Zealand white rabbits. About 87% of injected dose was retained in rabbit stifles and adjacent tissues in 96 hours after injection, while most of the activity lost from the joints was excreted in the urine. Due to its simplicity of preparation and radiolabeling, versatility, and biodegradability, this type of conjugate system may become the therapeutics of choice for radiation synovectomy.

    Notes: Abstract The principal objective of this study was to determine if the use of a stable enriched tracer of Se-76 could be used to determine the delay time between a dietary intake of selenium and its appearance in fingernails and toenails. Selenium is an essential trace element in human nutrition. It has been studied at the Missouri University Research Reactor (MURR) for the past 15 years using an Instrumental Neutron Activation Analysis (INAA) technique. The principal route of human exposure to selenium is through the diet. Selenium concentrations of nails, blood, hair, and urine have been used as indicators of dietary selenium intake. In this study, a cohort consisting of seven men and five women ingested three selenium supplements of 150 μg each over a three day period. The selenium was enriched in Se-76 (96.48%) and ingested as selenite in orange juice following an overnight fast. Fingernails and toenails were collected prior to the selenium supplementation and for several months afterward to be used as biochemical indicators. The peak76Se concentration in the fingernails and toenails occurred at 19–23 and 16–32 weeks after supplementation, respectively.
