Summary Studies of delta-aminolaevulic acid (Ala) metabolism during almost physiological conditions (incubation in concentrated crude haemolysate of erythrocytes, steady flow of small quantities of Ala running with an Ala-level of 230 μg%) showed in 19 healthy adults, that non-heme-pathway of Ala somewhat surpasses erythrocytic heme-pathway (52.3 ∶ 47.7%). Hemolytic anemias demonstrate due to enhanced count of reticulocytes (100 to 400‰) a still more enhanced non-heme-pathway (62.6 ∶ 37.4%), which leads to the conclusion, that erythroid cells of bone marrow may use less than about 1/5 of present Ala for heme synthesis. Non-heme-pathway was slight induced in iron deficiency (no significance), but normal in one case of Addisonian anemia. Investigations of inhibition of Ala-pathways under these conditions with proto-, copro- and uroporphyrins I and III, haemoglobin and haemiglobin pointed out only a slight inhibition of Ala-Dehydrase (Ala-D., 30% decrease) and non-heme-pathway of Ala by protoporphyrin (800 μg%) and a moderate significant inhibition of Ala-D. (70% decrease) and non-heme-pathway by haemiglobin (12g%). High rates of coproporphyrinogen-III synthesis in incubations did not point out an inhibition of Al-D. There was no distinct feedback regulation of erythroid heme synthesis on the step of Ala-D. demonstrated with products of heme synthesis investigated in this study.

Zusammenfassung Inkubationsstudien des Delta-Aminolävulinsäure (Ala)-Metabolismus unter weitgehend physiologischen Bedingungen (konzentriertes, natives Erythrozytenhämolysat, konstante Zufuhr kleiner Ala-Mengen entsprechend einem Anfangsspiegel von 230 μg%) zeigten bei 19 gesunden Probanden, daß der Ala-Metabolismus außerhalb der Hämsynthese (non-heme-pathway) relativ intensiv ist und die erythrozytäre Ala-Utilisation für die Hämsynthese (heme-pathway) noch etwas übertrifft (52,3 ∶ 47,7%). Im retikulozytenreichen Blut (hämolytische Anämien) war der Anteil des non-hemepathway der Ala noch größer (62,6 ∶ 37,4%), so daß in jungen, zur Hämsynthese befähigten Blutzellen die Ala-Utilisation zur Hämsynthese weniger als 1/5 des gesamten Ala-Metabolismus betragen dürfte. Eisenmangel-Anämien ließen eine leichte Zunahme des non-heme-pathway erkennen (nicht signifikant), ein Fall von Perniciosa wies einen normentsprechenden Ala-Metabolismus auf. Hemmversuche unter diesen Inkubationsbedingungen mit Protoporphyrin, Kopro- und Uroporphyrin I und III, Hämoglobin und Hämiglobin ergaben nur eine leichte Hemmung der Ala-Dehydrase (30% Aktivitätsminderung) und des non-hemepathway der Ala durch Protoporphyrin (800 μg%) sowei eine mäßige signifikante Hemmung der Ala-Dehydrase (ca. 70% Aktivitätsminderung) und des non-hemepathway der Ala durch Hämiglobin (12 g%). Hohe endogene Koproporphyrinogen-III-Syntheseraten führten nicht zur Ala-Dehydrase-Hemmung. Eine deutliche Autoregulation der erythrozytären Hämsynthese durch die untersuchten Hämstoffwechselprodukte war auf der Stufe der Ala-Dehydrase nicht nachweisbar.
