Notes: Abstract. Objective and Design: Recently, Ogawa et al. [17] reported that the peritoneal mast cells (PMCs) of rats can release histamine by substance P (SP) in a receptor-dependent manner. In the present study, we confirmed and extended their findings.¶Material: PMCs were isolated from six strains of rats. In some experiments, peritoneal cells in the non-MC fraction were used.¶Methods: PMCs were incubated with SP, neurokinin (NK) receptor agonists or antagonists, and histamine content in the supernatant was measured. In the binding assay, PMCs were incubated with [125I]BH-SP together with SP or NK receptor antagonists. NK-1 receptor mRNA was detected using a reverse transcription-polymerase chain reaction (RT-PCR) assay.¶Results: PMCs from Slc:Wistar and F344/NSlc were highly sensitive to SP, leading to histamine release, whereas those from Slc:SD and three other strains were not. PMCs from Slc:Wistar and F344/NSlc also released histamine in the presence of an NK-1 agonist. The histamine release induced by SP and the NK-1 agonist was inhibited by the NK-1 receptor antagonists, FK888 and CP-99,994. [125I]BH-SP binding experiments revealed that PMCs from Slc:Wistar rats possessed a single high affinity binding site for SP and that the binding was blocked by NK-1 receptor antagonists. Peritoneal cells in the non-MC fraction exhibited no appreciable binding. In the RT-PCR assay, expression of NK-1 receptor mRNA was evident in Slc:Wistar PMCs, but not in the non-MC fraction from Slc:Wistar or Slc:SD PMCs.¶Conclusion: These data demonstrate the existence of functional NK-1 receptors on freshly isolated PMCs in at least some strains of rats.

Notes: Abstract. We described the influence of a type of gas and its pressure upon the size distribution of Si nanoparticles fabricated by laser ablation in an ambient gas and the plume dynamics during the synthesis. The plume dynamics was investigated by laser-induced fluorescence and ultraviolet Rayleigh scattering. Based on the results, the importance of the gas flow within the ablation plume in the formation of the nanoparticles is understood.

Notes: Abstract. We describe the deposition and characterization of Bi12SiO20 (bismuth silicon oxide; BSO) thin films on Y-stabilized zirconia (YSZ) and SiO2 glass substrates by pulsed-laser deposition (PLD) for the application of an electric field sensor. It was found that all films deposited on YSZ substrates heated at 400 °C and more were crystallized and the (310) plane was perpendicular to the substrate normal. The highly (310) oriented crystallized films were even deposited on SiO2 glass substrates, and this will make it possible to grow the crystallized films on the end surface of a SiO2 glass fiber.

Notes: Abstract. The aim of this study was to assess the upper airway status of sleep apnea syndrome (SAS) patients with low-field MR fluoroscopy. Twenty patients with clinically diagnosed SAS underwent upper airway monitoring using MR fluoroscopy for 5 min while awake and for 30 min while asleep. A 0.064-T permanent-magnet MR imaging system was used for the study. No patients required any sedative drugs because of the very small gradient noise, except in one case. No occlusion was observed while patients were awake. Nine patients showed repeated occlusion at retropalatal (Rp) pharynx, whereas 11 demonstrated both simple Rp occlusion and combined retropalato–retroglossal (Rp + Rg) occlusions (complex occlusion). The mean frequency of occlusion in complex cases was significantly higher than that in simple Rp cases (p 〈 0.05). Low-field MR fluoroscopy was useful in determining the occlusion level while asleep in patients with SAS because of its quiet gantry and long-term monitoring capability. The MR fluoroscopy technique should prove to be a valuable clinical tool for the diagnosis and for determining the appropriate therapy in patients with SAS.

Notes: Abstract The phenotypes of chromosomal 22q11.2 microdeletion are quite variable among individuals and hypoparathyroidism (HP) constitutes a definite portion of the clinical spectrum. For the correct diagnosis and pertinent follow up of the HP children due to del22q11.2, we tried to delineate the clinical characteristics of such patients. By employing fluorescence in situ hybridization (FISH) to all the patients diagnosed as HP in our clinic, ten possessed the 22q11.2 microdeletion. Among them, the incidence of cardiac defect (5/10), recurrent infection (1/10) and cleft palate (1/10) was modest. Additionally, seven of them had been diagnosed as HP during the infantile period, when their facial abnormality and intellectual problem had not become evident. Notably, two patients were complicated by Graves disease, while the association of idiopathic thrombocytopenic purpura was also observed in two girls. Conclusion HP due to del22q11.2 may be misdiagnosed as idiopathic, especially in an infant who lacks apparent complications like cardiac anomaly. They should be closely followed up for auto-immune complications.

Notes: Abstract We report two sporadic cases of a hitherto undescribed skeletal dysplasia with short stature and characteristic facies. The present patients, a 6-year-old girl and a 15-year-old boy, were almost equally affected. Craniofacial anomalies included a sloping forehead, bitemporal bulging, sparse medial eyebrows, a prominent nasal bridge, hypertelorism, proptosis, a beaked nose, hypoplastic alae nasi and a pointed chin. Shallow orbits, short anterior cranial fossae and bitemporal bossing found on skull radiograph corresponded with the facial dysmorphism. Thickening of the dorsum sellae was another hallmark in the skull. Skeletal survey revealed mild osteopenia, interpediculate narrowing of the lumbar spine with short neural arches and, most important, osteopathia striata of the long tubular bones. There was no sclerosis of the craniofacial bones. The clinical and radiological findings in the present patients were overall inconsistent with those of previously known skeletal dysplasias and congenital malformation syndromes, which possess osteopathia striata as a cardinal feature. Conclusion The unique clinical and radiological constellation of our patients constitutes a hitherto unknown bone dysplasia.

Notes: Abstract We have applied atomic force microscopy (AFM) to the measurement of BAL 31 nuclease activities. BAL 31 nuclease, a species of exonuclease, is used to remove unwanted sequences from the termini of DNA before cloning. For cutting out only the appropriate sequences, it is important to know the nuclease properties, such as digestion speed and the distribution of the lengths of the digested DNA. AFM was used to obtain accurate measurements on the lengths of DNA fragments before and after BAL 31 nuclease digestion. We analyzed 4 DNAs with known number of base pairs (288, 778, 1818, and 3162 base pairs) for correlating the contour length measured by AFM with the number of base pairs under the deposition conditions used. We used this calibration for analyzing DNA degradation by BAL 31 nuclease from the AFM measurement of contour lengths of digested DNAs. In addition, the distribution of digested DNA could be analyzed in more detail by AFM than by electrophoresis, because digested DNA were measured as a population by electrophoresis, but were measured individually by AFM. These results show that AFM will be a useful new technique for measuring nuclease activities.

Notes: Summary Surface chemical states of57Co2+ and119Sb5+ ions adsorbed on γ-Fe2O3 and NiFe2O4 were studied in the presence of the aqueous phase using the magnetic interaction between the substrates and57Fe2+ or119Sn4+ arising from the adsorbed species. Two chemical forms were observed for the57Co2+ adsorbed on γ-Fe2O3; one giving a peak in the high-field region and another giving a broad distribution in the lower fields, which were attributed to57Co2+ at surfaceB sites of the spinel structure of γ-Fe2O3 and57Co2+ species weakly bonded to the substrate, respectively. In case of the NiFe2O4 substrate, the latter species was dominant and was converted to the former by heating. Most of the119Sb5+ ions adsorbed on the γ-Fe2O3 and NiFe2O4 particles were found to have a certain magnetic interaction with the magnetically ordered ions of the substrates.

Notes: In order to investigate the el led of occlusion on the skin, the flexor sides of both upper arms were covered with column-shaped closed chambers, 30 mm in outer diameter. 20 mm in inner diameter. and 5 mm in height, which were made of polyethylene loam. The tops of the chambers were sealed be plastic films with various levels of water vapor permeability to control moisture in each chamber. The raised chamber walls prevented direct contact between the skin and the plastic film. After 24 h of application, morphological changes of the skin surface were observed microscopically by the nitrocellulose-replica method. Although no visual alterations were found on all areas of occluded skin, microscopic evaluation showed that simple occlusion by films induced an increase in the number of deepened skin furrows on the skin surface. This increase was associated with lower water vapor permeability of the films, as well as with higher values of both temperature and humidity on the test day. Thus, since conditions which facilitate perspiration from the skin lend to cause skin irritation, prolonged exposure of the skin to sweat by simple occlusion may act as a primary skin irritant
