    Notizen: Abstract: Ganglioside analysis of human motor and sensory nerves revealed that ceramide compositions of sensory nerve GD1a, GD1b, and GMl differed apparently from those in the motor nerve. These gangliosides from sensory nerve contained a large amount of long-chain fatty acids and d18:1 as a major long chain base. On the contrary, the motor nerve gangliosides contained C16–18 fatty acids and a large amount of d20:1 besides d 18:1. Furthermore, these gangliosides were enriched more in the axon fraction than in the myelin fraction. LM1, which was a major ganglioside in myelin from human peripheral nerve, was composed of similar ceramide compositions in the two nerves. The present findings suggest that the characteristic ceramide species of nerve gangliosides may reflect in part properties of their own neurons.

    Notizen: Abstract: Myelins of the PNS were isolated from human motor and sensory nerves of cauda equina, and their ganglioside compositions were compared. The predominant ganglioside in the human PNS myelins, both from motor and sensory nerves, was LM1 (sialosylneolactotetraosylceramide). Sialosyl-nLc6Cer and disialosyl-nLc4Cer, GD3, GM3, and GDlb were detected as common components of the two nerve myelins. Furthermore, it was revealed that the motor nerve myelin contained GM1 (about 15% of total gangliosides), whereas sensory nerve myelin contained only a trace amount of GM1 (less than 5%), by TLC analyses together with TLC immunostaining using anti-GM 1 antibody. As for the disialoganglioside fraction, the content of GD1 a, as well as that of GM1, differed in motor and sensory nerves. Thus, the different contents of the ganglioseries gangliosides in human motor and sensory nerve myelins were demonstrated.

    Notizen: Abstract: Compositions of neutral and sulfated glucuronyl glycosphingolipids purified from human motor and sensory nerves and myelins were studied. Higher neutral glycosphingolipids (fraction B), which were separated from GazlCer (fraction A), were analyzed by TLC and TLC-immunostaining. Both nerve myelins contained paragloboside (nLc4Cer) and nLc6Cer dominantly as major higher glycosphingolipids and very little globoside (Gb4Cer), whereas both nerves contained Gb4Cer and nLc4Cer. Besides these major glycosphingolipids, a neutral glycolipid containing asialoGMI (Gg4Cer) epitope and other minor components such as ceramide trihexoside and ceramide dihexoside were detected in both nerves and their myelins. Furthermore, sulfated glucuronyl nLc4Cer and n Lc6Cer, which were monoclonal antibody HNK-1 reactive glycolipids, were detected in both nerves and myelins.

    Notizen: Four hydrogen pellets, two of 3 mm(large-closed-square)×3 mml pellets and two of 4 mmF(Slashthrough accent mark)×4 mml pellets, with the velocity of ∼2.3 km/s, were injected to OH and NB heated plasmas by a four-barrel pneumatic injector on the JT-60 tokamak. The pellet ablation profile was estimated by the measurement of the Hα emissions originating from the ablation cloud using a fiber optics array combined with the Hα -filtered multichannel photodiode. The fiber optics array consisting of ten channel bundles views the pellet pass in the vacuum vessel. The optical fiber was developed to tolerate baking up to 300 °C. The measured ablation profiles were compared with the calculated one by the neutral gas shielding model with a self-limiting effect. A good agreement was obtained between measured and calculated ablation profiles in OH plasmas; however, the fast-ion-induced ablation needed to be taken into account in the calculation for NB-heated plasmas.

    Notizen: A broadband reflectometric system in O-mode operation has been developed for the density profile determination, MHD and turbulent fluctuation measurement, and evaluation of the particle diffusion coefficient in the JT-60 tokamak. For the profile determination, full Ka-Q band frequencies of BWOs have been swept in 750 μs, in order to minimize the effect of doppler shift due to the fluctuation of reflection layers. The heterodyne fixed-frequency reflectometer has unveiled its effectiveness to probe MHD activities, diagnosing the m=1 tearing mode oscillations. It has also enabled the observation of the dramatic suppression of edge plasma density fluctuations at the L- to H-mode transition in the LHCD limiter plasma. Furthermore, the propagation delay of density pulses has been observed by different fixed-frequency channels during a series of sawteeth, from which the particle diffusion coefficient was evaluated, with a newly proposed method applicable to fully diffusive plasmas.

    Notizen: Summary Progressive multifocal leukoencephalopathy (PML) is a demyelinating disease caused by polyomavirus JC (JCV). In the majority of cases of PML the cerebrum is mainly affected (cerebral PML) but on rare occasions lesions are restricted to the cerebellum and brain stem (cerebellar PML). We report a rare cerebellar PML case which occurred in a Japanese patient undergoing prolonged hemodialysis treatment. To understand the molecular basis of the viral tissue tropism, we molecularly cloned JCV DNA and compared it with those of cerebral PML. Of ten clones analyzed nine showed identical fragment patterns after digestion with various restriction endonucleases, and we designated these clones Sap-1. It could be shown that the basic structures of the regulatory regions are similar between Sap-1 and isolates from cerebral PML. Restriction endonuclease mapping analysis was used to examine the genetic relationship between Sap-1 and urine-derived isolates containing the archetypal regulatory sequence. We found that Sap-1 was genetically related to an archetypal JCV isolate in Japan.

    Notizen: Summary Viral infection, especially by reactivation of herpes simplex virus (HSV) has been considered to be a possible explanation for the pathogenesis of idiopathic peripheral facial nerve palsy (Bell's palsy). We investigated whether the geniculate ganglia of man contain latent HSV type 1 (HSV-1), and compared the frequency of HSV-infected ganglia and that of latently infected neurons in human geniculate ganglia and in trigeminal ganglia. From autopsy specimens of eight adults 15 geniculate ganglia and 16 trigeminal ganglia were examined by means of in situ hybridization and immunohistochemical staining. The HSV-1 genome was detected in 11 of the 15 (71%) geniculate ganglia and in 13 of the 16 (81%) trigeminal ganglia. No HSV antigen was noted in any of the ganglia. The incidence of latently infected neurons was 0.9% in the trigeminal ganglia and 5.3% in the geniculate ganglia. The difference in percentages between the two types of ganglia was significant. Our results suggest that reactivation of latent HSV in the geniculate ganglia is a probable cause of some cases of herpetic stomatitis and of idiopathic peripheral facial nerve palsy.
