    Remarkable Depression of CD4+2H4+ T Cells in Severe Chronic Active Epstein-Barr Virus Infection (1987)

    Notizen: In order to better understand the features of chronic active Epstein-Barr (EB) vims infection, we employed two-colour immunofluorescence staining with monoclonal antibodies and flow cytometry analysis to study the lymphocyte phenotypes of two patients with severe symptoms of this disorder as well as four patients with mild symptoms. We found an increased number of activated T cells, as characterized by CD4+ la+, CD8+Ia+, or CD4+Tac+ phenotypes, and a markedly decreased CD4+2H4+ T cell subpopulation, previously characterized as a suppressor-inducer subset, in the patients with severe symptoms. In contrast, the four patients with mild symptoms showed only a slightly elevated number of activated T cells and a normal CD4+2H4+/CD4+ ratio. These phenotypic differences may suggesst heterogeneity in this disorder. Also, a failure in ihe suppressor-inducer population could contribute to changes in the host-virus relationship and the degree of the decrease in this population may correlate directly with ihe severity of the disease.

    Notizen: Adenine nucleotides and their related compounds in white and dark muscles of yellowtail Seriola quinqueradiata were measured and compared during ice storage. In white muscle, adenosine 5′-triphosphate was degraded rapidly to inosine 5′-monophosphate, which then underwent gradual build-up of inosine (HxR). In dark muscle, HxR accumulated very rapidly and the K value, a freshness index, thus increased at a greater initial rate than in the white muscle (62 vs 3%/day). Urate occurred in a small amount only in the white muscle stored for over 40 days (0.2 μmole/g), while little was found in the dark muscle during the entire storage period. Nicotinamide adenine dinucleotide, which existed in both the fresh white and dark muscles (0.8 vs 0.4 μmole/g), decreased rapidly in the early stage of storage.

    Notizen: Abstract The promoting effects on cell proliferation of the 105K glycoprotein (105Kgp), purified from sera of chickens to which a Marek's disease (MD) lymphoblastoid cell line, MDCC-MSB1-41C (MSB1-41C), had been transplanted, were examined using culture cells from various sources. The MSB1-41C line as well as the other chicken lymphoblastoid cell lines examined were sensitive to the 105Kgp. The growth-promoting effects of 105Kgp showed a biphasic pattern depending upon the amount of 105Kgp added into the culture medium.These findings indicate that the 105Kgp may be a promoting factor for chicken growing cells, especially lymphoblastoid cell lines.

    Notizen: Summary Attempts to isolate the molt-inhibiting hormone (MIH) of crustaceans from crab eyestalks (ES) resulted in the characterization of xanthurenic acid as an inhibitor of ecdysone biosynthesis in the cultured Y-organ-complex (YOC) homogenate. It was also found that 3-hydroxyl-l-kynurenine present in the ES is transformed into xanthurenic acid in the YOC and body fluid. Its mode of inhibitory action in ecdysone biosynthesis is probably inactivation of cytochrome P-450.

    Notizen: Summary Rat brain tumor was used as a model to evaluate radiation therapy with and without misonidazole. BD-IX rats were implanted intracerebrally with an ethylnitrosourea-induced glioma. Three series of experiments were performed, with radiation given 14 days after inoculation of the glioma clone. In each series, the following radiation doses were given: 500 rads once, 1,000 rads once; and 1,000 rads twice, every time with or without two different doses of misonidazole. Radiation therapy significantly prolonged survival when compared to the longevity of the control group. The dose of 1,000 rads given twice was highly effective and the life-span of tumor-bearing rats increased from 72% to 121%. Misonidazole plus irradiation negated the prolongation of survival, achieved with radiation therapy alone.

    Notizen: Summary In order to know the mutagenic effects of synthetic auxins (NAA, 2,4-D, and 2,4,5-T) and a cytokinin (kinetin) in vitro, sister chromatid exchanges (SCEs) were analyzed in cultured cells of a hexaploid wheat (Triticum aestivum L.). In the MS medium supplemented with 2.0 mg/l 2,4-D, the mean number of SCEs per cell was 15.2, and per pg of DNA, 0.42. No significant effect was found in the treatments of NAA or 2,4-D at concentrations of 0.5–10.0 mg/l, whereas more than 2.0 mg/l of 2,4,5-T induced dramatic increases of SCEs. Kinetin itself had no significant effect on SCE induction, but there was a tendency that SCEs induced by 2,4,5-T were suppressed by kinetin.

    Notizen: Abstract The electrophoretic mobility of polystyrene — latex (PSL) of diameter 870 Å was determined using a Laser Zee System 3000. This instrument enables automatic measurement of the electrophoretic mobility of fine particles. Effect of pH and ionic strengthI on the PSL mobility was analyzed. It was found that the mobility as a function of pH has a minimum around pH 3–4 atI=0.1, showing no isoelectric point, and that the mobility decreases with increasing ionic strength up toI=0.1 but is almost constant forI=0.1–0.2 at pH=7.34. From the mobility date, we estimated the zeta potentialζ of PSL. We used an approximate mobility formula derived by Ohshima, Healy and White, which is considerably more accurate than Smolchowski's and Henry's formula and is applicable for fine particles with smallχ a (χ a≳10), whereχ is the Debye-Hückel parameter anda is the particle radius. Further, we calculated the surface charge densityσ of PSL using an approximateσζ relationship. It was found that there are 2000–4000 negative charges on the PSL surface at pH=7.34. Comparison is made with the results on synaptic vesicles (SV) from brain cerebrum cortex and brush-border-membrane vesicles (BBMV) from the small intestine.
