    Notes: Abstract It was to be shown whether during the biogenesis of microbodies some of their components were already present in the cell prior to the organelle's assembly. To this end, the occurrence and properties of catalase in soluble and particular fractions of ripening cucumber seeds were examined. Homogenates of seeds from ripening fruits were fractionated by isopycnic density gradient centrifugation, and thus catalase was found in three different fractions: as a soluble enzyme in the gradient supernatant, as a membrane fraction at density d=1.18 kg l-1, and in association with microbodies. In the early steps of seed formation, catalase was detected at density d=1.18 kg l-1 and in the gradient supernatant. At a later stage of seed maturation, however, catalase was primarily associated with microbodies which exhibited an equilibrium density of d=1.23 kg l-1. M r as well as subunit M r of catalase were determined, and their close immunological relationship to leaf peroxisomal catalase and glyoxysomal catalase was demonstrated. Biosynthesis of catalase at different stages of seed maturation was investigated by in vivo labeling with l-[35S]methionine, l-[14C]leucine and δ-[3H]aminolaevulinic acid. Electrophoretic analysis of de novo synthesized catalase subunits revealed the occurrence of a heavy form (M r 57,500) in the soluble fraction; this form was preferentially labeled. A light form, M r 53,500, was detected in microbodies and also in the soluble fraction. The findings lend support to the hypothesis that the rate of catalase synthesis is highest in an early stage of seed formation, when globulins have already been formed, but before de novo synthesis of malate synthase has commenced. Prior to microbody assembling, a cytoplasmic pool of catalase was labeled.

    Description / Table of Contents: Zusammenfassung Behandlungsergebnisse des Morbus Hodgkin der Stadien I und II werden durch Vergleich der Überlebensdaten der Literatur nach verschiedenen Strahlentherapieprogrammen und nach zusätzlicher Chemotherapie diskutiert. Im Stadium I A bleiben 90–97% und im Stadium II A 75–80% der Patienten nach ausgedehnter Strahlentherapie rezidivfrei. Für das Stadium IIB werden 0–80% langdauernde Remissionen nach alleiniger Strahlentherapie berichtet. Zusätzliche Chemotherapie verbessert die Rezidiv-, aber nicht die Überlebenszahlen der Stadien I und II. Gruppen mit hohem Rezidivrisiko (große Mediastinaltumoren, E-Befall der Lunge und lymphozytenarme Histologie) werden dargestellt.
    Notes: Summary The results obtained in the treatment of Hodgkin's disease, stages I and II, are discussed comparing survival data of the literature after various radiotherapy programs and after combined modality using additional chemotherapy. In stage I A 90 to 97% and in stage II A 75 to 80% of patients are not prone to relapse after extended-field irradiation. In stage IIB 0 to 80% long-lasting remissions are reported after radiotherapy. Additional chemotherapy improved relapse-free survival, but not overall survival in stages I and II. Subgroups are discussed which bear a high risk of relapsing disease (big mediastinal masses, E-lesions of the lungs, histological findings with lymphocyte depletion).

    Description / Table of Contents: Summary L-asparaginase therapy is often limited by allergy or toxicity and probably in some cases by antibody mediated inactivation of the enzyme. These problems can be avoided by extracorporeal application of l-asparaginase. As the enzyme is a stable tetramer with high molecular weight it cannot pass through the dialysis membrane in contrast to the amino acid l-asparagin which is destroyed. The resulting l-asparagin depletion of the plasma is sufficient for therapeutic success. Effective extracorporeal l-asparaginase therapy is demonstrated in two patients with ALL who were resistant to other chemotherapy and could not be treated intravenously because of allergy and toxicity.
    Notes: Zusammenfassung Allergie mit der Gefahr der Anaphylaxie, Toxizität und möglicherweise Inaktivierung durch Antikörper begrenzen den Einsatz von L-Asparaginase in der Leukämiebehandlung. Diese Probleme können durch die extrakorporale Anwendung umgangen werden. Sie ist möglich, weil das Enzym als stabiles Tetramer mit höherem Molekulargewicht Dialysemembranen nicht passiert, während die Aminosäure L-Asparagin aus dem Körper entfernt werden kann und dann abgebaut wird. Für den Therapieeffekt ist allein die L-Asparagindepletion notwendig. Am Besipiel von zwei Patienten mit ALL, die gegenüber anderer Chemotherapie résistent und wegen Allergie bzw. Toxizität parenteral nicht mit L-Asparaginase behandelt werden konnten, wird das Verfahren beschrieben.

    Description / Table of Contents: Summary ESR investigations of lyophilized blood of patients with acute lymphatic leukemia exhibit an increase in spin concentration and an additional peak not present in control samples. This peak disappears almost completely concomittantly with a reduction in spin concentration after treating the patients with prednisolone, vincristine, and doxorubicin or after addition of 5 mM of CuCl2 to blood samples. The results show that the ESR spectra of lyophilized blood samples can be utilized to follow the effect of therapy. Moreover, they suggest that the leukemic blood possesses a high concentration of antioxidants.
    Notes: Zusammenfassung ESR-Untersuchungen an lyophilisiertem Blut von Patienten mit akuter lymphatischer Leukämie zeigten eine Zunahme in der Spinkonzentration. Außerdem trat ein zusätzliches Signal auf, welches in Kontrollproben nicht vorhanden ist. Wenn man die Patienten mit Prednisolen, Vincristin und Doxorubicin behandelt, verschwindet gleichzeitig mit einer Abnahme der Spinkonzentration dieses Signal fast vollständig. Ein ähnlicher Effekt wurde erhalten, wenn entsprechende Blutproben mit 5 mM CuCl2 versetzt wurden. Die Ergebnisse zeigen, daß die ESR-Spektren von lyophilisierten Blutproben als Indikator für den Therapie-Verlauf benutzt werden können. Diese Ergebnisse deuten darüber hinaus darauf hin, daß leukämisches Blut eine hohe Konzentration an Antioxidantien besitzt.

    Notes: Summary In a recently published review of the literature [40] we came to the conclusion that the Ann-Arbor staging classification is of limited prognostic value for chemotherapy of Hodgkin's disease (Table 2). Four risk factors accounted for impaired complete remission rates: stage IVB, lymphocyte depletion or not classifiable histologic type, previous chemotherapy, and older age. Fifty-eight evaluable patients were treated with COPP; 23 reached a complete remission (40%). Disease-free survival was 31%, overall survival 49% after five years [33]. Besides the known risk factors, impaired bone marrow function (leucocyte counts 〈4×109/1, platelet counts 〈100×109/1) at the start of therapy was associated with poor treatment results: none of six patients achieved a complete remission [41]. Eleven of 16 patients with no and 11 of 23 patients with one risk factor achieved a complete remission, as did only one patient with more than one risk factor. Survival rates after 30 months were; 87% with no, 66% with one, 36% with two, and 13% with more than two risk factors. We can conclude from our results that the prognosis of patients undergoing chemotherapy for Hodgkin's disease depends on the number of risk factors.
