    Description / Table of Contents: Summary 1. Three different methods are described for the visualisation of the Thomsen-Friedenreich (TF) antigen on cell suspensions, formalin-fixed and paraffin embedded or frozen tissue sections: a) Rosette formation with chicken and sheep erythrocytes. b) Fluorescence-microscopy with fluorescein labelled peanut agglutinin. c) Autoradiography with3H-labelled peanut agglutinin. 2. The TF antigen was shown, as far as presently investigated, to be exposed on various blood cells, glomerula of the kidney and normal mammary gland after neuraminidase treatment. Mammary gland was also shown to possess TF receptors without prior treatment with neuraminidase. 3. The exposure of this cryptantigen can be brought about by bacterial or viral neuraminidase and is followed by an antigen/antibody reaction, which can lead to possible pathological consequences.
    Notes: Zusammenfassung 1. Drei verschiedene Methoden zur Darstellung von sog. Thomsen-Friedenreich (TF)-Antigenen in Zellsuspensionen, an Formalin-fixiertem und in Paraffin eingebettetem Gewebe sowie in Gefrierschnitten werden beschrieben: a) Rosetten-Bildung mit Hühner- und Schaferythrocyten. b) Fluorescenzmikroskopie mit Fluorescein-markiertem Erdnußagglutinin. c) Autoradiographie mit3H-markiertem Erdnußagglutinin. 2. Nach Neuraminidasebehandlung konnten TF-Antigene — soweit bisher untersucht — auf verschiedenen Blutzellen, in Nierenglomerula und in normalem Brustdrüsengewebe nachgewiesen werden. Ein Teil derartiger Antigene lag im Brustdrüsenparenchym auch bereits als freie Rezeptoren vor, d. h. waren nicht von Neuraminsäure bedeckt. 3. Die Freilegung dieser Kryptantigene kann im Rahmen von Infektionen durch bakterielle und virale Neuraminidase erfolgen. Dabei kommt es zu einer Antigen/Antikörper Reaktion mit unter Umständen klinisch relevanten Folgen.

    Description / Table of Contents: Zusammenfassung Bei 100 gesunden Probanden sowie bei 50 Patienten, die sich Operationen unterschiedlichen Schweregrades unterzogen hatten, wurden quantitative Bestimmungen fragmentierter Erythrocyten (Schistocyten) im peripheren Venenblut (Ausstrichpräparate und Thoma-Zählkammer) durchgeführt. 1. Bei den Normalpersonen wurden durchschnittliche Schistocytenzahlen von 1–2‰ ermittelt. Der Mittelwert betrug in der Zählkammer 1,4‰ (maximale Streubreite: 0–5‰), in den Ausstrichpräparaten 2,03‰ (maximale Streubreite: 0–6‰). 2. Bei den operierten Patienten wurden 2 Std post operationem erhöhte Schistocytenzahlen festgestellt, die offenbar in Abhängigkeit von dem Schweregrad des Eingriffs und der Gabe von Bluttransfusionen im Mittel zwischen 6 und 10‰ lagen. Bereits nach 24 Std war es in vielen Fällen wieder zu einem Abfall auf Normwerte gekommen. 3. Zwei splenektomierte Patienten wiesen höhere Schistocytenzahlen auf (18 bzw. 35‰), die über einen längeren Zeitraum persistierten. 4. Der Durchschnittswert fragmentierter Erythrocyten in 12 unterschiedlich alten Blutkonserven lag bei 2,5‰ 5. Beide angewandten Untersuchungsmethoden eignen sich in der Praxis als Auswertungsverfahren. Die Schistocytenwerte liegen lediglich in der Regel bei der Auszählung in den Ausstrichpräparaten etwas höher als bei der Bestimmung in der Thoma-Zählkammer. Durch die quantitative Bestimmung fragmentierter Erythrocyten ist ein empfindlicher Gradmesser für Störungen der Mikrozirkulation gegeben.
    Notes: Summary Fragmented erythrocytes (schistocytes) were counted in blood smears and Thoma chambers of venous blood from 100 healthy subjects and from 50 patients who underwent various surgical procedures. The results showed to be as follows: 1. The average number of schistocytes in healthy controls of both sexes was 1–2‰ (medium in Thoma chamber 1.4‰, in blood smears 2.03‰, with a range from 0–5‰ and 0–6‰, respectively). 2. There was an increase in the number of schistocytes two hours after a surgical procedure (average 6‰), the degree of which correlated with the extent of the operation. Higher values of fragmented erythrocytes were counted after intraoperative blood transfusion (average 10‰). In most cases schistocyte values returned to normal at about 24 hours after surgery. 3. The highest schistocyte counts (18–35‰) were observed in two patients undergoing splenectomy with persistence of elevated counts for a longer time period. 4. The average schistocyte number in stored blood of various age was 2.5‰. 5. Both counting of schistocytes in Thoma chambers and in blood smears are applicable, though the values obtained with the latter technique are slightly higher. Quantitative estimation of schistocytes proves a useful method for evaluating disturbances in microcirculation.

    Notes: Summary Serum levels of antibodies against the Thomsen-Friedenreich (TF) antigen were determined for a group of patients with malignancies of the breast, as well as a control group of healthy persons and patients with no breast disease. Despite recent claims of anti-TF levels being severely depressed in patients with breast cancer, we were unable to show any significant differences between the two groups. Standardised anti-TF containing sera or solutions of the TF specific agglutinin from peanuts (PNA) were absorbed with membrane preparations from normal and cancerous breast tissue, but again no differences in absorption were noted. Histological examination, by contrast, showed both free and sialic acid-covered receptors to be present on both normal and malignant breast epithelium. Based on these findings it was concluded that determination of serum anti-TF is an unsatisfactory method of diagnosing breast cancer and that the TF antigen cannot be considered to be carcinoma associated.

    Description / Table of Contents: Zusammenfassung Das Ziel der vorliegenden Untersuchungen war es, den Einfluß des Geschlechts der tumortragenden Mäuse und der Ovariektomie auf das Wachstumsverhalten eines soliden transplantablen Mammacarcinoms der C3H-Maus zu prüfen. Dabei konnte durch Wachstumskurven ermittelt werden, daß das Wachstum des Tumors bei Transplantation auf ovariektomierte und männliche Tiere gegenüber dem bei weiblichen Tieren verzögert war. Autoradiographisch ließ sich hierbei eine längere Generationszeit der Tumorzellen ermitteln, die vor allem durch eine längere präsynthetische Ruhephase (T G1) bedingt war. In bezug auf die Wachstumsfraktion und den Zellverlustfaktor wurden keine wesentlichen Unterschiede gefunden. Insgesamt ließ sich nur eine eingeschränkte hormonelle Beeinflussung des Wachstums der Tumorzellen feststellen, was u.a. mit dem niedrigen Gehalt an Oestrogenreceptoren zusammenhängen könnte.
    Notes: Summary The aim of the present investigation was to study the influence of sex of tumour bearing mice and of ovariectomy on the proliferation kinetics of a solid, transplantable mammary carcinoma (C3H-mouse). According to the growth curves the tumour growth was delayed in spayed female and in male mice compared with normal female animals. This was due to a longer cell cycle time of tumour cells in the former, mainly caused by a prolonged presynthetic gap (TG1). Concerning the growth fraction and the cell loss factor, essential differences could not be detected. Thus, the solid tumour displayed a distinct though partial disengagement of growth from hormonal dependence.

    Notes: Summary Normal tissue as well as various benign and malignant lesions of the breast were histochemically examined for the presence of the Thomsen-Friedenreich (TF)-antigen. Fluorescein- or 3H-labelled peanut agglutinin was used for this purpose, a lectin that is known to have a high affinity for the TF-antigen. The occurrence of this TF-antigen seemed in all cases, even in the carcinoma lobulare in situ that is regarded as being derived from myoepithelial cells by some authors, to be associated with a secretory condition. Its presence (free and neuraminicacid covered) in normal, hyperplastic and malignant breast tissue, however, cannot be considered a specific tumour associated antigen as has been previously assumed. Furthermore the investigations have shown that the intensity of fluorescence for peanut agglutinin (PNA)-receptors was generally stronger in differentiated carcinomas than in undifferentiated carcinomas of the breast. The histochemical findings are discussed with regard to diagnostical and immunotherapeutical aspects.

    Description / Table of Contents: Zusammenfassung Humanseren mit Toxoplasma-Antikörpern werden — vor und nach Gelfiltration (Sephadex G 200) — mit dem IgG- und dem IgM-Immunfluorescenztest sowie der Toxoplasma-Komplementbindungsreaktion untersucht. — Bei Seren von Patienten mit „latenter“ Infektion und IgG-IFT-Antikörpertitern zwischen 1∶50 und 1∶200 lassen sich nach Gelfiltration im 19 S-Bereich weder mit dem IgM-IFT noch mit der KBR Antikörper nachweisen. — In den 19 S- bzw. 7 S-Fraktionen von Seren aus der „akuten“ Phase der Infektion findet man mit dem IFT regelmäßig erregerspezifische IgG- und IgM-Antikörper. Komplementbindende Toxoplasma-Antikörper sind nicht nur in den 19 S (IgM)-Fraktionen, sondern auch — und in einigen Fällen ausschließlich — in den Fraktionen des 7 S (IgG)-Bereichs nachweisbar. — Aus diesen Beobachtungen ergibt sich, daß IgM-IFT- und KBR-Antikörper immunologisch differenziert werden können. Die Bedeutung der verschiedenen Antikörper für die Serodiagnose der Toxoplasmose beim Menschen wird diskutiert.
    Notes: Abstract Human sera with antibodies against Toxoplasma gondii were investigated—before and after gel filtration (Sephadex G 200)—by IgM- and IgG-specific immunofluorescent reactions as well as by complement fixation tests.—In sera from patients with “latent” infections IgM-antibodies could not be demonstrated neither by the IgM-IFT- nor the CFT technique, although IgG antibodies with IFT titres between 1∶50 qnd 1∶200 were present.—In sera from patients in the “acute” stage of infection both IgM and IgG antibodies were always found. In some cases the IgG fraction had complement fixing properties only, although IgM antibodies were present as demonstrable by the IFT technique.—These observations leed to the possibility of immunologic differentiation between IgM-IFT and CF-antibodies. The role of the different antibodies in the serodiagnosis of human toxoplasmosis is discussed.
