    Notes: Summary Immunohistochemical identification of human epidermal growth factor (hEGF) was carried out in a total of 152 cases of salivary gland tumours, consisting 107 pleomorphic adenomas and their variants, 13 adenolymphomas and 32 adenoid cystic carcinomas. A high percentage of pleomorphic adenomas revealed markedly positive hEGF staining of the luminal surface cells of tubuloductal structures and of modified or neoplastic myoepithelial cells. Clear cells of the tumour showed various reactivities from very slight to strong. Eosinophilic epithelial cells of adenolymphoma gave a positive reaction for hEGF in all the cases, whereas most adenoid cystic adenoma lacked hEGF staining; however some cases showed positive staining of the tumour cells. The immunohistochemical detection of hEGF in most salivary gland tumours suggests this factor to be a possible new marker of salivary glands tumours, and to have a biological role in tumour proliferation.

    Notes: Abstract Hereditary cerebellar vermis defect (CVD) rats, a new neurological mutant, developed both cerebellar vermis defect and cerebellar dysplasia. Developmental alterations in the cerebellum of the CVD rats were studied chronologically and immunohistochemically. The earliest architectural abnormality was a maldevelopment of the inferior cerebellar peduncle from embryonic day 17 (E17), leading to an indistinct separation between the cerebellum and the pons. From E19, the CVD rats lacked vermis development and, therefore, the cerebellar hemispheres were fused. After birth, Purkinje cells and external granule cells (EGCs) penetrated into the pontine tissue, but retained their normal position until postnatal day 10. Cerebellar lamination began to be disturbed due to abnormal perivascular aggregations of the EGCs, resulting in convoluted and occasionally perivascular lamination. There were no Bergmann glia in the heterotopic cerebellum of the pons, and abnormally arranged Bergmann glia were observed in the mildly disorganized cerebellar hemispheres. Immunohistochemistry for calbindin revealed that abnormal orientation of the Purkinje cells might be related to the perivascular EGCs. Parvalbumin-immunopositive microneurons were seen only in the disarranged molecular layers, and synaptophysin-immunopositive cerebellar glomeruli were present in the afflicted internal granular layers. These findings suggest that perivascular EGCs may play an important role in cerebellar dysplasia and the developmental plasticity in the altered cerebellogenesis.

    Notes: Abstract The cerebellar vermis defect (CVD) rat is a new neurological mutant characterized by a cerebellar vermis defect and dysplasia in the cerebellum, especially at the cerebellopontine junctions. In this study, the cytokinetics of glia in terms of the development of cerebellar dysplasia in the CVD rat was investigated using glial fibrillary acidic protein (GFAP) and vimentin immunohistochemistry. In the cerebellar hemispheres, dislocation of the Bergmann glia was observed from postnatal day 5 (P5) in lesions with abnormally aggregated external granule cells (EGCs). Rearranging Bergmann glia were often seen around the EGCs penetrating into the white matter. In the cerebellopontine junctional areas, Bergmann glia were induced after penetration of the Purkinje cells, identified with calbindin immunohistochemistry, and EGCs into the pons from P10. Bergmann fibers were frequently arranged perivascularly. In the clusters of Purkinje cells without EGC settlement in the pons, a small number of Bergmann fibers were observed and their alignment was completely disturbed. These findings suggest that morphological changes in the Bergmann glia depend on their contact with Purkinje cells, but that the orientation of their processes may be influenced by EGC settlement. These glial fibers in the CVD rat may play an important role in the aberrant migration of EGCs, resulting in the development of cerebellar dysplasia.

    Notes: Construction of a gene expression system in tobacco cultured cells (BY2) was studied. A 925 bp promoter fragment of a heat-shock protein gene (HSP18.2) of Arabidopsis thaliana showed clear heat-shock response of expression of the β-glucuronidase (GUS) reporter gene in BY2 cells. Similar results were observed in a 500 mL flask and 3-L jar fermentor.Isolation of strong promoters in BY2 cells was tried. cDNA clones, in which the mRNA level is high in log-phase cells and the copy number in the genome is low, were isolated. These clones showed high homology with F1-ATPase (mitochondria type), elongation factor 1-α, and a gene with an unknown function of A. thaliana (clone 27), respectively. A 5′-flanking region of clone 27 showed 6.2 times the promoter activity of the CaMV35S promoter in BY2 cells.Three cDNA clones, which are expressed in the stationary growth phase of BY2 cells, were isolated by a differential screening. These clones showed high sequence homologies to alcohol dehydrogenase, pectin esterase, and extensin.     Notes: An emulsion polymerization of styrene in the presence of an amphoteric emulsifier of amino-acid type, N-lauroyl-N,N-dimethyllysine (DMLL), has been studied at pH 7.0. The polymerization rate increased exponentially with the concentration of DMLL. The concentration of latex particles was proportional to the DMLL concentration to the 0.6 power. The latex particle size decreased with increasing concentration of DMLL. The molecular weight of polystyrene latexes increased with an increase in DMLL concentration. Furthermore, the zeta potentials of latex particles were significantly dependent on the pH, and showed the existence of an isoelectric point at about pH 4.0.

    Notes: To produce a bone-bonding polymer surface that is capable of inducing deposition of a hydroxyapatite (HA) layer in the body fluid, an organophosphate polymer was covalently immobilized onto a high-density polyethylene film by surface graft polymerization of a phosphate-containing monomer. The grafted film was immersed in simulated physiologic solution (SPS). The chemical composition and structure of the formed apatite layer as well as its bonding strength to the polymer surface were investigated. To distinguish the effect of phosphate groups on the deposition of apatite layer from the simple calcium absorption by the anion, a comparative study was done using a polyethylene film with surface immobilized carboxylic groups. Calcium phosphate deposition was observed on all the materials investigated, but the kinetics, composition, deposit amount, and bonding strength of the new phase were found to be significantly different among the modified materials, depending on the density and chemical nature of the surface immobilized ionic groups. It was found that the polymeric materials modified by surface graft polymerization of a phosphate-containing monomer produce a carbonated HA layer firmly bonded with the material upon immersion in SPS. Carboxyl groups in the grafted layer was not enough to activate bonding with the HA layer. © 1994 John Wiley & Sons, Inc.

    Notes: Surface modification and functionalization of polyaniline (PAN) and polypyrrole (PPY) films were achieved by Ar plasma treatment, near-UV-induced graft copolymerization with acrylic acid (AAc) and covalent immobilization of an enzyme (trypsin). The compositions and structures of the modified and functionalized polymer surfaces were characterized by angle-resolved XPS. Argon plasma treatment results in minor oxidations of the carbon atoms but is accompanied by a significant decrease in the intrinsic oxidation state ([=N-]/[-NH-] ratio) of the polymers. Graft copolymerization with AAc readily gives rise to self-protonated PAN and PPY surface structures. The surface concentration of the grafted AAc polymer increases with increasing monomer concentration and near-UV irradiation time during grafting but decreases with increasing plasma pretreatment time. Covalent immobilization of trypsin on the AAc-polymer-grafted PAN and PPY films is facilitated by the use of water-soluble carbodiimide intermediate (WSC). The effective enzyme activity increases initially with increasing surface concentration of the grafted AAc polymer, but becomes saturated at moderate to high AAc polymer concentration.
