    Notes: Summary In rats chronic hypoxia causes extramedullary haematopoiesis mainly localized in the spleen. It is not known how splenic erythropoiesis develops and how it regresses after termination of hypoxia. In this study the spleen of rats exposed to chronic hypoxia was studied by light and electron microscopy; the findings were compared to relevant peripheral blood values. Splenic erythropoiesis begins almost immediately after exposure to hypoxia and reaches its maximum after 24 weeks. It occurs mainly in the splenic cords drawing upon local erythroblasts and is accompanied by an increase in splenic weight as well as a decrease in splenic iron stores. After termination of hypoxia marked phagocytosis by splenic cord macrophages diminishes the number of erythroblasts and of erythrocytes with a concomitant increase in splenic iron stores. Thus, splenic erythropoiesis appears to be inhibited as part of a rebound phenomenon and returned to normal by phagocytosis of erythroid cells within 4 weeks after cessation of the hypoxic stimulus.

    Description / Table of Contents: Summary A selective staining method for eosinophils and their precursors in smears and paraffin sections is described. The reaction bases on tryptophane like substances within eosinophilic granules, which can be demonstrated by a modified p-dimethylaminobenzaldehyde-nitrite method after Adams. By this means eosinophilic granules are stained distinctively in a blue colour.
    Notes: Zusammenfassung Es wird eine Methode zur selektiven Anfärbung der Eosinophilen und ihrer Vorstufen in Paraffinschnitten und Ausstrichen beschrieben. Aufgrund des Reichtums der eosinophilen Granula an tryptophanhaltigen Substanzen gelingt es, sie mit Hilfe von p-Dimethylaminobenzaldehyd-Nitrit nach Adams darzustellen. Die eosinophilen Gra nula erscheinen dabei distinkt blau gefärbt.

    Notes: Summary Fifty consecutive AIDS autopsy cases were evaluated. All subjects showed one or more opportunistic infections and malignancies included in the AIDS case definition with cytomegalovirus and Kaposi's sarcoma being most prevalent. Mycobacterial and cryptococcal infections occurred only infrequently. Most patients of our series after successful treatment ofPneumocystis carinii pneumonia or cerebral toxoplasmosis later succumbed to less treatable conditions like disseminated cytomegalovirus or fungal infections or malignant lymphoma. In the absence of specific treatment for the HIV infection leading to these lethal complications special emphasis must be put on the prevention of HIV transmission and spread.

    Description / Table of Contents: Summary A cytological and enzyme cytochemical (α-naphthyl-acetate-esterase, naphthol-AS-acetate-esterase, naphthol-AS-D-chloroacetate-esterase, acid and alkaline phosphatase) analysis of human spleen cells in smear preparations was performed with special reference to littoral cells of splenic sinuses. These littoral cells show strong naphthol-AS-acetate-esterase and none or little α-naphthyl-acetate-esterase activity. Thus they can be differentiated from reticular cells and other cellular elements to be found in splenic smears. Morphology and enzyme cytochemistry of littoral cells render them to be a distinct cell type of human spleen.
    Notes: Zusammenfassung In Ausstrichpräparaten menschlicher Milzen wurden cytologische und fermentcytochemische Untersuchungen (α-Naphthyl-acetat-Esterase, Naphthol-AS-acetat-Esterase, Naphthol-AS-D-Chlor-acetat-Esterase, saure Phosphatase und alkalische Phosphatase) durchgeführt. Dabei wurde besonders das Verhalten der Sinuswandzellen beobachtet. Es gelang, morphologische und enzymcytochemische Kriterien aufzuzeigen, aus denen ersichtlich ist, daß die Sinuswandzellen weder mit den Reticulumzellen noch mit anderen im Milzausstrich anzutreffenden Zellen identisch sind, sondern eine besondere, für die menschliche Milz sehr typische Zellform darstellen.

    Description / Table of Contents: Zusammenfassung Bei der zytophotometrischen Bestimmung von Kernproteinen an gemischten Zellpopulationen wie Lymphknoten, Knochenmark oder Milz ist die Zuordnung der jeweils zu untersuchenden Einzelzelle zu einer bestimmten Zellrasse oft schwierig. Durch diesen Unsicherheitsfaktor kann die Aussagekraft derartiger Messungen stark eingeschränkt werden. Um diese Fehlermöglichkeit zu vermindern, wird ein Verfahren beschrieben, welches durch Anwendung zytochemischer Enzymnachweismethoden eine zuverlässige Klassifizierung der zu messenden Einzelzelle vor der anschließend vorzunehmenden eigentlichen quantitativen Analyse derselben Zelle erlaubt, ohne die Meßergebnisse zu beeinträchtigen.
    Notes: Summary In cytophotometric analyses of nuclear proteins of mixed cell populations such as lymph nodes, bone marrow or spleen it is frequently difficult to place a single cell to be explored into a precise category of cell types. This factor of uncertainty concerning the classification of cells may substantially reduce the value of those measurements. In order to eliminate or diminish such sources of error a method is described which by means of cytochemical enzyme tracing allows for a reliable classification of single cells to be examined prior to the real quantitative analysis of the same cell without impairment of the results of the measurements.

    Description / Table of Contents: Zusammenfassung Ein Verfahren zur Hexazotierung von Triamino-tritolyl-methanchlorid (Neufuchsin) wird beschrieben und die Anwendungsmöglichkeit des Hexazotats in der Fermenthistochemie geprüft. Dabei erwies sich Hexazonium-Neufuchsin in Verbindung mit Substraten der Naphthol-AS-Reihe als ausgezeichneter und den bislang üblichen Diazoniumsalzen hinsichtlich der Lokalisationsschärfe sowie der Eigenschaften des resultierenden Azofarbstoffs weit überlegener Kupplungspartner beim Nachweis der alkalischen Phosphatase.
    Notes: Summary A method for hexazotizing triamino-tritolyl-methanchloride (Neufuchsin) is described, and the use of this compound in enzyme histochemistry investigated. The results show that hexazonium-Neufuchsin in combination with substrates of the naphthol-AS group is an excellent reagent for the determination of alkaline phosphatase; as regards the precision in localizing the enzyme and the quality of the resulting azo-dye it is by far superior to the customary diazo compounds.
