    Notes: We have developed a new compact retarding-potential Mott spin polarimeter and achieved an efficiency of 1.9×10−4 for gold target operating in 25 keV. A novel design of the retarding field electron optics with 0.59 sr collection solid angle for scattered electrons was adopted based on Monte Carlo calculations for the spin-dependent electron scattering process and electron beam ray-tracing calculations. We have combined the new spin polarimeter with an angle-resolved photoelectron spectrometer and measured the spin- and angle-resolved photoelectron spectra and studied the spin-dependent electronic structure of Ni(110) along the Γ¯S¯ line of its surface Brilluoin zone. © 1997 American Institute of Physics.

    Notes: A cell and gas-handling system of the supercritical material for x-ray absorption measurement are designed. The cell was made of stainless steel with a pair of Be windows of 3 mm thickness. The x-ray path can be controlled by changing the distance between two Be windows. Electric cartridge heaters are embedded in the cell to control the temperature. With this system we have studied the supercritical state of CF3Br, whose critical point is 67 °C and 39.1 atm. The results of x-ray absorption fine structure measurements for Br K edge for gaseous, liquid, and supercritical states of this material are presented together with the performance of the gas-handling system. © 1995 American Institute of Physics.

    Notes: Ion-molecule reactions of silicon-carbon binary cluster ions SiC+n (n=1–6) and SiC−n (n=2–4) with O2 molecules were studied by using an rf ion trap. For all selected positive and negative parent ions, only SiCnO+ or SiCnO− were produced, respectively. The rate constants for the oxidation of these clusters were determined for the first time. The size dependence of the reactivities are discussed, and the presence of the ring form positive ions for n=3 and 5 are suggested. © 1995 American Institute of Physics.

    Notes: Subclasses of lymphocytes can be separated on gradients of non-toxic polyvinylpyrrolidone-coated colloidal silica (Percoll) by virtue of differential densities. Such gradients can yield functionally active lymphocyte populations after brief centrifugation. Gradients can he generated in a discontinuous step fashion and centrifuged in standard table-top laboratory centrifuges or as self-generating gradients duriny ultracentrifugation. The density medium has low viscosity and can be made isotonic for virtually any use. Gradients have proved useful in both human and experimental animal studies, and high percentage yields allow for separations from small cell numbers. Methods are described for separation of whole blood and lymphotd subpopulations. including blasts stimulated with mitogens or in mixed lymphocyte reactions. The cytotoxic capability of various density fractions was evaluated for mixed lymphocyte culture-induced allogeneic killing and spontaneous, so-called ‘natural’ killer cell activity. The lower density associated with blast transformation allows for significant enrichments of stimulated cells from in vitro cultures. Higher thymidine incorporation, restimulation in mixed lymphocyte reactions, and greater cytotoxic capacity are associated with these‘blast’fractions.

    Notes: Two new TCRVβ coding region polymorphisms were identified: Vβ6.9a/b and Vβ21.4a/b. In both cases, a single nucleotide difference gives rise to an amino acid exchange. Genomic typing by the PCR/sequence-specific oligonucleotide probing technique was performed to study a possible contribution of these two new polymorphisms in susceptibility to autoimmune diseases. However, there was no association with insulin-dependent diabetes mellitus, rheumatoid arthritis, juvenile rheumatoid arthritis, multiple sclerosis, myasthenia gravis or coeliac disease. On the other hand, significant differences were found between Caucasoid and Oriental populations in frequencies of the Vβ6.9 and Vβ21.4 alleles.

    Notes: Upon in vivo alloactivation of Ig-anti-Ig column-purified splenic ‘T’ cells in lethally irradiated allogeneic recipients, a variable proportion of donor-derived cytotoxic T lymphocytes (CTLs) are able to bind IgG antibody-coated erythrocytes through surface Fc receptors (FcR) and form rosettes. The use of fractionation procedures based on the ability of these cells to form rosettes has enabled us to separate FcR-positive CTLs from FcR-negative CTLs and to examine the ability of these two cell populations to perform as effector cells in direct T-cell-mediated killing and in antibody-dependent cellular cytotoxicity. A series of experiments, either by direct isolation of the two cell populations or by deletion of the FcR-positive population by filtration through complexed immunoglobulin columns (Ig-anti-Ig), has shown both populations to be efficient in direct T-cell mediated cytotoxicity against the relevant target cell. The striking difference between the two populations is the exclusive ability of the FcR-positive population to function as effector cells in antibody-dependent cellular cytotoxicity (ADCC). Purification steps before in vivo alloactivation of our responding cells for the removal of ‘B’ cells and FcR-bearing cells with‘K’-cell activity, followed by procedures to remove phagocytic and adherent cells in the resulting immune spleen cell preparation and, finally, by velocity sedimentation of the rosetting and nonrosetting blasts from the small lymphocyte population, has resulted in a population of FcR-positive cells 98% positive for the Thy 1.2 alloantigen. These fractionation steps and immnunofluorescence criteria of purity strongly favor the contention that the ADCC activity within the FcR-positive T-cell population is indeed a property of the CTL itself.

    Notes: A multi-allergen dipstick enzyme immunoassay ‘Quidel Allergy Screen’ (QAS) has recently been developed commercially for measuring IgE antibodies against nine allergens (house dust 1, house dust 2, Dermatophagoides pteronyssinus, D. farinae, Japanese cedar, ragweed, cat dander, sweet vernal grass, and egg white) at one time. To assess whether this assay is useful in screening allergen-specific IgE antibody, we compared the titres of IgE antibodies against the nine allergens measured by QAS to those in the skin-prick test and by RAST in 93 atopic asthmatics and 22 normal subjects. We found a good overall agreement between the results of the skin-prick test and the results of QAS (sensitivity = 47.1-81.4%, specificity = 84.5-100%, and agreement =78.9-88.9%). The sensitivities against house dust I, D. pteronyssinus. and D. farinae ranged from 77.2 to 81.4%. However, the sensitivities against house dust 2, Japanese cedar, ragweed, and cat dander were low (47.1-68.8%). We also found a good overall agreement between the results of RAST and the results of QAS, except for egg white (sensitivity = 46.2-94.4%, specificity = 87.4-100%, and agreement = 77.4-96.5%). The sensitivities against house dust 1 and 2, D, pteronyssinus, D. farinae, and Japanese cedar ranged from 86.0 to 94.4%. The sensitivities against ragweed, cat dander, and sweet vernal grass were low (46.2-52.6%). There were strong correlations between the titres of RAST and the titres of QAS except cat dander and egg white (r= 0.701 0.924 for the seven allergens). Thus, we conclude that QAS is useful in screening IgE antibodies against multiple allergens at one time. However, because the sensitivities against some allergens tested were low, further improvement of some allergen preparations seems to be necessary in the assay.
