Summary In 5 patients with acute and 1 patient with chronic monocytic leukemia histochemical studies were performed on smears from blood and bone marrow, on kryotom-sections from various organs, on inflammatory exudates from skin windows, and on blood cells cultured in vitro with and without addition of phytohemagglutinin. The following histochemical techniques were used: PAS, Sudanblack B, methylgreenpyronin, esterase with α-naphthyl-acetate and naphthol-AS-acetate as substrates — with and without addition of NaF —, naphthol-AS-D-chloroacetateesterase, alkaline and acid phosphatase, ATP-ase, naphthylamidase, NAD-H-diaphorase, succinic dehydrogenase and peroxidase. Leukemic monocytes accorded fairly in their cytochemical pattern to normal monocytes. They were characterized by a high activity of naphthol-AS-esterase completely inhibited by 1,5 mg/ml NaF. Furthermore a large amount of acid phosphatase, naphthylamidase and NAD-H-diaphorase could be demonstrated within these cells. As a rule the contents of Sudanblack-positive lipids, peroxidase and naphthol-AS-D-chloroacetate-esterase were low. Cytochemical investigations give strong support against the theory of different types and point to the myelogenous origin of true monocytic leukemia. These being furthermore characterized by a specific functional behaviour of the leukemic cells as studied by the skin window technique and by the in vitro culture of peripheral blood cells. The developmental potencies of these leukemic cells are similar to that of normal blood monocytes.

Zusammenfassung Bei 6 Fällen von Monocytenleukämie, darunter einer chronischen Form, wurden Ausstriche von Blut und Knochenmark, sowie Gewebsschnitte, Hautfensterpräparate und in vitro kultivierte Blutzellen untersucht. Neben bausteincytochemischen Untersuchungen wie PAS, Sudanschwarz B- und Methylgrün Pyronin-Färbung, wurden α-Naphthyl- und Naphthol-AS-Acetat-Esterase—mit und ohne Zusatz von NaF — Naphthol-AS-D-Chloroacetat-Esterase, alkalische und saure Phosphatase, ATP-ase, Naphthylamidase, NAD-H-Diaphorase, Succinodehydrogenase und Peroxydase nachgewiesen. Die leukämischen Monocyten sind durch einen hohen Gehalt an unspezifischer Esterase gekennzeichnet, die bei Verwendung von Naphthol-AS-Acetat als Substrat durch NaF (1,5 mg/ml) fast vollständig gehemmt wird. Sie enthalten außerdem reichlich saure Phosphatase, Naphthylamidase und NAD-H-Diaphorase. Charakteristischerweise ist der Gehalt an Sudanschwarz-positiven Phospholipoiden, an Peroxydase und an Naphthol-AS-D-Chloroacetat-Esterase gering. Leukämische Monocyten stimmen also in ihrem cytochemischen Verhalten mit normalen Blutmonocyten überein. Cytochemische Untersuchungen sprechen gegen das Vorkommen verschiedener Typen monocytärer Leukämien und unterstreichen die Auffassung der Monocytenleukämie als myelogene Leukose. Leukämische Monocyten zeigen bei Hautfensterversuchen und bei in vitro-Kulturen ein völlig abweichendes Verhalten von den Zellen anderer akuter Leukämien und ähneln in ihren Entwicklungspotenzen normalen Monocyten.
