Deshalb ist eng verbunden mit den stündig steigenden Anforderungen an Quantität und Qualität der Informationen aus den verschiedenen hierarchischen Niveaus biotechnologischer Prozesse die Entwicklung hoch instrumentierter leistungsfähiger Laborfermentorsyteme.Der Trend, biotechnologische Prozesse in ihrer Raum-Zeit-Ausbeute traditionellen chemischen Prozessen anzugleichen, führte zur Entwicklung von Fermentorsystemen mit neuen Wirkprinzipien zur Gewährleistung hoher Stoffäbertragungsleistungen. Mit der Realisierung derartiger Hochleistungsfermentoren in großtechnischen Dimensionen ist verbunden die Forderung nach leistungsfähigen Laborfermentoren für die Erarbeitung wissenschaftlicher Grundlagen als Basis für eine gesicherte Maßestabsäbertragung.Die Entwicklung eines 2,5 l Laborsterilfermentors mit hohen Stoffübertragungsleistungen stellt einen Beitrag zur Vervollständigung der gerätetechnischen Basis für die biotechnologische Grundlagenforschung dar.

    Notes: One of the most striking features of alkane-grown yeast cells is conspicuous appearance of peroxisomes in harmony with a high level of catalase. This unique phenomenon was first demonstrated in the authors′ laboratory, and the metabolic functions of peroxisomes in yeasts utilizing alkanes has been estabilished with intact peroxisomes isolated by density gradient centrifugation. The organelles participate in the degradation of fatty acids derived from alkanes to C2-units and the synthesis of gluconeogenic intermediates from C2-units. The abundant appearance of peroxisomes in alkane-utilizing cells has allowed successful production of several useful enzymes including catalase, D-amino acid oxidase, uricase, acyl-CoA oxidase etc. Yeast cells will be an excellent system for investigation the functions and development of peroxisomes because biogenesis of the organelles is induced only by transferring the cells into alkane medium from glucose or ethanol medium.

    Notes: A study of alkane degradation in Pseudomonas aeruginosa has been carried out over the last twelve years in the author's laboratory. This study has been aimed at the detailed characterization of the enzymes involved in alkane oxidation to fatty acids, alkane hydroxylase, alcohol dehydrogenase and aldehyde dehydrogenase. Here are summarized the main results of this study and described with some more detail the points which have not yet been reported. Some general conclusions which can be drawn from the evidence obtained are presented.

    Notes: Some aspects of the nonsteril yeast production are shown by a process with the alkane utilizing yeast Lodderomyces elongisporus D EH 15. Thereby an accompanying flora developes besides the yeast culture. The accompanying flora is using a lot of biogenic substances which are lysis products or extracellular metabolites. The dynamics of the development of the accompanying flora are shown, some remarks are made concerning the interactions between production strain and bacteria, the stability of the yeast strain and the recirculation of process water streams after the separation.

    Notes: A thermotolerant yeast species of Lodderomyces elongisporus EH 60 was isolated and physiologically characterized. This yeast possesses a high specific growth rate with μmax = 0.61 h-1. The dependence of the specific growth rate and cell yield on temperature, dilution rate, sucrose concentration and pH-value is investigated.     Notes: The influence of various concentrations of lysine and threonine on the growth rate (μ), homoserine- deficient and prototroph revertant at their joint growth in liquid fermentation medium has been investigated.It has been established that even relatively small concentrations of threonine (25 mg/l) at the background of lysine concentration exceeding 10 g/l reduce μ of prototrophs, but do not influence μ of auxotrophs.It is supposed that the addition of threonine at the background of comparatively large concentrations of lysine is one of the factors creating selective conditions under which an auxotroph producer has advantage over prototroph revertant.

    Notes: There is treated the combined apply of chemical and biological plant protection preparations as well as their reciprocal effect. Purpose of this work is to investigate the effect of the chemical preparation Fekamatribufon EC-50 (butylat-0,0-dimethyl-II-butyryloxy-2,2,2-trichlorethylester of phosphonic acid) on the spores of Bacillus thuringicnsis, var. dendrolimus and var. Berliner. The electronic-microscopical investigations determine that the chemical insecticid Fekamatribufon EC-50 accelerates the lysying of the spore structural elements of investigated Bacillus strains. The results of these investigations prove the incompatibility of the preparation Fekamatribufon EC-50 in concentrations of 0.6-1.2% (Water solution) when it is combined with the microbial preparations Dendrobacillin and Türicid.The above mentioned results present that the combined apply of chemical in ecticids with microbial preparations as plant protection means have obligatory to be preseded bysexact determination of their reciprocal effect.

The procedure comprises concentration, sedimentation, ethanol and sodiumsulfate precipitation and drying in laboratorial and semitechnical scale.The elimination of rape seed oil which impairs the salt sedimentation was specially taken into account. A certain sedimentation procedure using caoline or a aqueous solution of CaCl2 and Na2HPO4 or filtration was elaborated to eliminate the lipid components in the concentrated solution.The method is convenient to get good yields of the thermostable protease.

    Notes: The activity of methanol dehydrogenase of Protaminobacter thiaminophagus ATCC 21371 and its 10 mutants which where able to overproduce amino acids from methanol was studied. It was found that the activity of methanol dehydrogenase depended on the used strain varied from 56.1 to 100.6 mU/mg d. w. of cells. Specific production of amino acids was between 4.3 × 10-7 and 13.0 × 10-7 μg/CFU.Statistical analysis confirmed expected high positive correlation (r = 0.93) between activity of methanol dehydrogenase and specific production of amino acids.Based on the measurement of methanol dehydrogenase activity a rapid method of estimation of amino acid production ability of Protaminobacter thiaminophagus mutants was developed.

    Notes: Millet amylase formed during malting was used for teh saccharification of cassava (Manihot esculenta) into glucose syrup. The resulting syrup was also fermented to ethanol and citric acid. The maximum α-amylase activity occurred within 30 hours of millet germination at an optimum pH 8.0 and 40°C. The T. reesei enzymes enhanced the rate of cassava hydrolysis and paper chromatography revealed the presence of maltose, glucose and galactose in the cassava hydrolysate. The low cost of millet coupled with the fact that it is readily available makes it an economic source of amylase.

    Notes: Electrophysical devices with standard sensors were used for measuring the principal electrophysical parameters of raw materials, microbiological suspension and biopreparations: their dielectric constant, tangent of dielectric loss angle, specific conductivity and duration of dielectric relaxation of the medium. The obtained characteristics proved ot be rather informative for monitoring different technological parameters of the biosynthesis. The applicability of electrophysical methods in monitoring the process of biosynthesis was substantiated with examples.The electrophysical methods provide a simple and reliable way of monitoring the technological parameters in technical microbiology.

    Notes: The kinetics of enzymatic hydrolysis of different lignocellulosic materials (wheat straw, newspaper and microcrystalline cellulose Avicel PH 101) was studied using the cellulase complexes from Trichoderma reesei QM 9414 and its mutants M 5, M 6, MHC 15 and MHC 22. The maximum yields of hydrolysis were obtained with wheat straw partially delignified with 1% NaOH as substrate, and using the enzyme from the mutants T. reesei M 6 and MHC 22. The possibility of simultaneous enzymatic hydrolysis and ethanol fermentation of wheat straw using the enzyme complex from M 6 and yeasts of the genus Candida and Torulopsis was also investigated. A good conversion of liberated glucose and cellobiose to ethanol was obtained, however, xylose was not fermented.

    Notes: At the microbial oxidation of long-chain alkanes such monocarbonic acids at a monoterminal oxidation are formed which correspond to the chain length of the alkanes. At diterminal oxidation the corresponding dioic acids are produced.Under the influence of cerulenin a cerulenin a direct incorporation of fatty acids into cell lipids increases. Undecanoic acid cannot be metabolized byMortierella isabellina. It causes an inhibition of alkane oxidation. A main effect of undecanoic acid is an inhibition of the elongation of other fatty acids. The de novo fatty acid biosynthesis has not been inhibited by undecanoic acid.

    Notes: Up to the year 2000 the total demand of protein for animal feeding will rise by 146%. This demand can not be covered by agriculture alone. According to FAO data it will be an expected deficit of some 400 million tons of cereals at this time all over the world. Therefore the large scale industrial production of SCP from alkanes, methanol and other raw materials will undoubtely become soon an absolute necessity. On the other hand, a new process of solid state fermentation (ORSTOM/IRCHA process) having special interest for the developing countries has been recently set-up in France.Based on dried and crushed potatoes or cassava (manioc or tapioca) protein enrichment by fermentation of feeds of foods (PEFF) in a simple manner without aseptic conditions with the filamentous fungus Aspergillus hennebergii is carried out. The process is presently optimized and scaled-up to the size of a 700 kg (dry weight) fermenter.Nutrititional and toxicological assays have demonstrated complete safety and excellent nutrititional value of the product, which is applicable as a feedstuff for breading of e.g. pigs or chickens.

    Notes: The kinetics of the bacterial extracellular protease synthesis (neutral and alkaline protease of Bacillus mesentericusstrain 76, R-form) in batch and chemostat cultures under conditions of glucose limitation were investigated. When the medium was supplemented with casein the production of the proteases was significantly higher. Optimal dilution rates for obtaining of two proteases are fixed. The synthesis of both alkaline and neutral proteases is controlled by catabolite repression and induction.

    Notes: Investigations of a wide range of methane- and methanol-utilizers showed a striking versatility of their metabolism dependent on the genotype and growth conditions. A correlation between pathways of carbon and nitrogen metabolism was found. It was most stringent in obligate methane-utilizers: the hexulosephosphate pathway bacteria assimilated NH3 by the reductive amination of α-ketoglutarate or pyruvate whereas the serine pathway bacteria used the glutamate cycle (glutamine synthetase + glutamate-oxoglutarate aminotransferase).Multiple enzymic lesions were found in central metabolism of obligate methylotrophs, i.e. the absence of the enzymes of glycolytic and pentosephosphate pathways, gluconeogenesis, citric acid cycle and glyoxylate shunt. These metabolic blocks were not so profound and could be compensated in restricted and facultative methylotrophs during heterotrophic growth.The average levels of exogenous CO2 fixation in methylotrophic bacteria with the hexulosephosphate, serine and ribulosebisphosphate pathways were found to be 10, 30 and 80% of their total cell carbon, respectively.These results served as a basis for biotechnological applications of metabolic potential of methylotrophs (production of biomass, polysaccharides and enzymes as well as for microbiological treatment of industrial waters containing toxic C1- and Cn-compounds).

    Notes: It is demonstrated that aliphatic hydrocarbons (alkanes) penetrate into bacteria cells by the way of passive diffusion. The mechanism of this process is different for several bacteria species. A hydrophobic cell wall is essential for that process. In saprophytic Mycobacteria hydrocarbons are solubilized in the thick hydrophobic cell wall. During the process of absorption hydrocarbons pass through the whole cell wall up to the membrane. In the case of Arthrobacteria the hydrocarbons might pass not through the whole cell wall, but through special lipophilie canals.Mobile hydrocarbon-oxidizing bacteria g. Pseudomonas form peptidoglycolipid and excrete it into the medium. The peptidoglycolipid emulsifies hydrocarbon substrate.

    Notes: The free monosaccharide content of C. lipolytica (strain 4 124) cells grown on n-hexadecane was identified and found to be only glucose. The chromatographic analysis of the hydrolysate of intracellular cell wall polysaccharides indicated the presence of glucose: mannose: galactose: xylose in a ratio of 1 : 1.32 : 1.07 : 0.35.Paper and dise electrophoresis of extracellular polysaccharid from the culture broth was found to be heterogeneous.Ethanol fractionation separated it to a major component F (I) 81.99% and a minor one F (II) 13.04%.Analysis of the major fraction showed that it consisted of galactose and mannose only while the minor polysaccharide consisted of galactose, glucose and mannose.Thus it was concluded that the predominant sugar in both extracellular and intracellular polysaccharides was mannose. Xylose was detected in the intracellular polysaccharide only.

    Notes: A nonisothermal flow calorimeter operating directly in the fermenter was used for heat flow measurements of aerobic microbial growth processes with high biomass productivities. The measuring arrangement makes it possible to describe transitional stages of aerobic yeast cell growth by the ratio of heat production to oxygen consumption (oxy-caloric coefficient). The oxy-caloric coefficient was not constant under the described conditions. The results refer to the existence of an additional energy-delivering mechanism in microbial systems with aerobic carbon source utilization. The mechanism can involve polyphosphate bond division coupled to biomass synthesis.

    Notes: This review deals with different aspects concerning the genus Thiobacillus as an obligat chemoautotroph able to oxidize reduced anorganic sulphur compounds. There are given the following topics: physiology of the genus (nutritional requirements, enzymatic complexes for sulphur oxidation, energy generation, reductive CO2 assimilation and growing in presence of organic compounds).The ecological significance of Thiobacilli in the sulphur cycle in Nature is presented. The industrial applications of Thiobacilli in respect to the leaching processes that allow the recovery of metals through oxidation and solubilization from low-grade ores are discussed, with special reference to copper and uranium. A critical revision is done of acid mine drainage impact on the environment, and the use of adequate countermeasures is stressed.

    Notes: Fermentation kinetic, thermokinetic and morphological investigations of biological carbonsource conversions into microbial biomass and other reaction products can be used for improving the efficiency of biotechnical processes. In this way biorhythmic processes on a macrosocopic scale could be found in transitional stages of microorganism cultivation. These biorhythmic processes can be explained by the occurence of different cell states in the cell cycle. In order to characterize these cell states synchronized microorganism growth was investigated with the aid of the phased culture method. Two states could be observed in the case of yeast growth: the single cell state and the budding cell state. The duration and efficiency of the single cell state is dependent on the growth limitation and the carbon substrate feeding.Thus the reduction of carbon substrate feeding in states of synchronized populations which are characterized by a high percentage of single cells and by energy metabolite production which, in turn, is influenced by carbon substrate concentration can cause increased material and energetic efficiencies of biological carbon-source conversions.

    Notes: In all bacteria growing on methane or methanol the methanol is oxidised by way of methanol dehydrogenase. This unusual quinoprotein interacts with the electron transport chain at the level of cytochrome c. All methylotrophs having methanol dehydrogenase contain 2 different types of soluble cytochrome c. One of these, but not the other, reacts with methanol dehydrogenase; no intermediate electron carrier is required for this reaction. The P/O ratio for methanol oxidation is 1 although the midpoint redox potential of the methanol/formaldehyde couple suggests that a higher ATP yield might be theoretically possible. Curves are presented that allow the prediction of cell yields, and the effects of altering the system for methanol oxidation, on these cell yields.

    Notes: The mechanism and the kinetic of the assimilation of mixed substrates during the organism growth and the product excretion by strain of Saccharomycopsis lipolytica have been studied. The assimilation of citric acid for the organism growth was prevented when glucose and/or n-paraffins are present as substrates. Citric acid concentrations higher than 30 g/l in the fermentation medium decrease the growth rate on the substrate glucose. Kinetic studies of the mixed substrate assimilation by means of a n-tetradecane-1-614C-labelled paraffin fraction proved that in discontinuous as well as in continuous 1-stage-processes for microbial production of citric acid glucose as substrate is only used for the organism growth whereas the n-paraffin fraction is only used for the acid excretion.

    Notes: A new approach for the investigations of biotechnical processes is presented in this article on the basis of an integrating strategy, incorporating a joint analysis of biological and engineering aspects. Thereby, the experimental work can be carried out on the macroscopic level, taking into account the behavior of bioprocesses from measurements in the liquid phase. Mathematical modeling uses the concept of analogies instead of speculative mechanisms. A predictive character of these “formalkinetics” supplementary to the descriptive nature is achieved with the aid of deductive research methodology. This general theoretical concept is illustrated in the special case of modeling the diauxic growth of yeast. Formalkinetics based on analogies are shown to be not only able to be successfully applied to elucidate bioprocessing but also to form a more realistic and simple approach than pure mechanistic investigations needed for the realization of bioprocessing in technical scale.

    Notes: Cytochrome P-450 is found in soluble as well as in particulate fractions of n-alkane-induced cells of Acinetobacter calcoaceticus EB 104 as shown by differential centrifugation and; gelfiltration experiments. It is reduced by NADH in the presence of at least two soluble proteins. Particulate cytochrome P-450 was extracted by Triton X-100 and purified to homogeneity. The 80 fold enrichment resulted in a content of 19.0 nmol cytochrome P-450/mg protein. The maxima of the absolute spectra are 417 nm for the oxidized, 408 nm for the reduced and 448 nm for the CO complex of reduced cytochrome. A ferredoxin, identified by maxima of 415 and 460 nm in the absorption spectrum of the oxidized form and by the occurrence of acid-labile sulfur, as well as a corresponding NADH-dependent ferredoxin reductase were isolated independently from the same bacterial strain. Enzymatic reduction of purified cytochrome P-450 is mediated by these proteins, which are discussed as in vivo components of the cytochrome P-450 system of A. calcoaceticus EB 104.

Independently, a rubredoxin and a NADH-dependent rubredoxin reductase were isolated from this strain. Rubredoxin is induced by n-alkanes. The occurence of a cytochrome P-450 was observed only in other strains of Acinetobacter.A. calcoaceticus69/V contains a particulate pyridine nucleotide-independent and a soluble NADP-dependent alcohol dehydrogenase; the particulate aldehyde dehydrogenase is also NADP-dependent. The particulate dehydrogenases are inducible.The results concerning the regulation of enzymes of the citrate and glyoxylate cycle as well as of the malic enzyme indicate that the corresponding fatty acids formed from the alkanes are metabolized via β-oxidation.

    Notes: Asia is rich in resources which can contribute widely to food processing and production over the next 207-50 years as world population reaches 6 billion. The world needs low cost methods of providing nutritious proteinrich meat analogues for its thousands of millions of consumers. The Indonesian tempe fermentation will serve as a model. A bacterium present in commercial tempe can be used to add vitamin B-12 to other vegetarian foods. Fuel requirements for cooking can be decreased by applying a fungal fermentation of the tempe/ontjom type to legume substrates. The world needs high quality meat-flavors derived from vegetable protein. The soy sauce (kecap)/miso (tauco) processes and the fish/shrimp sauce and paste processes can be modified to yield a wide variety of meat-like flavors for use in formulating new foods. The protein content of high starch substrates can be increased by applying the Indonesian tape fermentation. Leavened sour-dough bread-like products can be produced without the use of wheat or ryeflours using the Indian idli/dosai fermentation. Coconut protein, if extracted without denaturing, can serve as a valuable base for a new type of puddings and related foods. Finally Asia is an almost endless source of cultures of edible microorganisms that, with further study of their synthetic abilities, particularly regarding amino acids and vitamins, could lead to new industries.

    Notes: The ability of Aspergillus nidulans (EIDAM) WINT to grow and sporulate at various temperatures and to degrade soluble and insoluble forms of cellulose were studied. A. nidulans was found to grow and sporulate best at 37°C in continuous light and alternating light-darkness respectively. The fungus was able to cause losses in the dry weights of filter papers on incubation and made appreciable growth on CMC and hemicellulose. The culture filtrates contained cellulases which hydrolysed filter papers and CMC to reducing sugars, and were only able to produce these enzymes in the presence of cellulose or its derivatives in the growth medium. The CM-cellulases had peak activity at pH 5.2 and at 50°C while optimal FP-activity occurred at a pH of 5.5 and at 45°C. The participatory role of A. nidulans in composting is discussed.

Using an estimated A factor for A2, we find \documentclass{article}\pagestyle{empty}\begin{document}$$\begin{array}{l} k_2 ({\rm benttransitionstate}) = 1.6 \times 10^{ - 11 - 4.3/\theta } \\ k_2 ({\rm lineartransitionstate}) = 4 \times 10^{ - 12 - 3.5/\theta } \\ \end{array}$$\end{document} suggesting that a best choice is E2 = 3.9 ± 0.4 kcal/mol. The relation of these values to ΔHf0 (HO2) is discussed.

    Notes: Rate constants for the reactions of O3 and OH radicals with furan and thiophene have been determined at 298 ± 2 K. The rate constants obtained for the O3 reactions were (2.42 ± 0.28) × 10-18 cm3/molec·s for furan and 〈6 ×10-20 cm3/molec·s for thiophene. The rate constants for the OH radical reactions, relative to a rate constant for the reaction of OH radicals with n-hexane of (5.70 ± 0.09) × 10-12 cm3/molec·s, were determined to be (4.01 ± 0.30) × 10-11 cm3/molec·s for furan and (9.58 ± 0.38) × 10-12 cm3/molec·s for thiophene. There are to date no reported rate constant data for the reactions of OH radicals with furan and thiophene or for the reaction of O3 with furan. The data are compared and discussed with respect to those for other alkenes, dialkenes, and heteroatom containing organics.

    Notes: Vibrationally excited OH in v = 9 [designated OH†(9)] was generated by the reaction of hydrogen atoms with ozone in a fast-flow discharge system at 300 ± 3 K and a total pressure of 1.1 ± 0.1 torr, with argon as the carrier gas. The addition of a species X, which can deactivate the OH†(9) or react with it, led to a decrease in the Meinel band chemiluminescent emission intensities at both 626 nm (9 → 3 band) and 519 nm (9 → 2 band), which were monitored as a function of the concentration of X. Application of the kinetic scheme developed previously for this chemical system gave the relative rate constant for the removal of OH†(9) by X. The relative rate constants determined in this study, taking O2 as the reference deactivator (kO2 = 1.0), are as follows: He ≤ 0.02; H2 ≤ 0.05; SF6 0.09 ± 0.01; CF4 0.19 ± 0.01; N2O 3.5 ± 0.4; NO 17.7 ± 1.5; H2O 74.3 ± 2.9; D2O 57.6 ± 2.0; NH3 61.3 ± 1.9; ND3 58.7 ± 1.6; SO2 7.1 ± 1.4; COS 8.4 ± 1.7; H2S 33.7 ± 8.4; CH4 1.56 ± 0.03; CD4 1.06 ± 0.06. Application of these relative rate constants to conditions in the upper atmosphere (60-100 km) suggests that OH†(9) is removed primarily by deactivation by O2, and at altitudes ≳90 km, possibly by O(3P). However, since O2 is unusually efficient for a homonuclear diatomic in deactivating OH†(9), it may not be the primary deactivator for the lower (v ≤ 8) vibrational levels. These results are compared to earlier studies of OH†(9), and possible mechanisms of interaction of OH†(9) with these molecules are discussed.

    Notes: The rate of formation of toluene from ethylbenzene was studied at atmospheric pressure in the temperature range of 600-725°C in conventional flow equipment. Residence times were 0.55-1.7 s, and initial partial pressures of ethylbenzene (H2O diluent) were 0.05-0.17 atm. The rate of appearance of toluene measures [1] the rate of the reaction C6H5C2H5 → C6H5CH3· + CH3·, with log k = 15.70 - 74.49 ± 3.2/θ. From the activation energy, E0 for the decomposition is estimated to be 72.2 ± 3.2 kcal/mol. The results are compatible with those of Esteban et al. [2], for which regression analysis gives log k1 = 14.79 - 70.76/θ. A composite, log k = 15.10 - 72.0/θ, fits both sets of data and is experimentally indistinguishable from Robaugh and Steina's [3] recent estimate by the VLPP technique or other estimates in the literature [4-6]. In light of the high overall conversion of ethylbenzene studied (2-80%), the industrial-type flow equipment, and the widely differing methods of characterization and analysis, the excellent agreement with the results of [2,3,5] is worthy of note. The data are compatible with current estimates of the heat of formation of benzyl radicals [6,7].

    Notes: The kinetics of the pyrolysis of n-hexane was studied in a conventional static reactor over a temperature range of 650-840 K. The overall reaction is essentially first order with the kinetic parameters A = 1013.92 s-1 and EA = 260.3 kJ/mol. The distributions of the main products were analyzed by gas chromatography. A reaction model involving 240 elementary reactions was developed to describe the experimental rate data. The agreement of the model with experimental data was surprisingly good over a wide range of temperatures and pressures and up to medium extents of conversion. Methods for sensitivity studies based upon the quasi-stationary-state assumption (QSSA) were developed, and for a number of more detailed effects, such as self-inhibition, explanations could be given. It was also shown that the hexyl isomerization reactions influence strongly the product distribution. The outstanding capability of kinetic modeling with computer simulations in handling complex kinetic systems is demonstrated.

    Notes: The pressure dependence of the first-order rate coefficient of oxetan and oxetan-2,2-d2 decomposition has been studied in the pressure range from about 7 kPa down to 0.01 kPa at various temperatures between 673 and 758 K. Experimental data were analyzed using RRKM theory. Interpretation of the fall-off curves lends support to the high-pressure Arrhenius parameters A = 1015.42s-1 and EA = 259.5 kJ/mol derived from measurements made in the pressure-independent range. Decomposition of oxetan is found to occur via biradical intermediates. Data for the kinetic isotope effect were used to derive kinetic parameters for the ring-opening elementary steps in oxetan and oxetan-d2 decomposition.

    Notes: A study has been made both of secondary reactions occurring during the reaction of I2 with GeH4, and of the direct reaction between I2 and GeH3I. Both these studies show that the abstraction reaction \documentclass{article}\pagestyle{empty}\begin{document}$$ {\rm I}^ \cdot + {\rm GeH}_{\rm 3} {\rm I} \to {\rm \dot GeH}_{\rm 2} {\rm I + HI} $$\end{document} occurs about 30 times faster than the reaction \documentclass{article}\pagestyle{empty}\begin{document}$$ {\rm I}^ \cdot + {\rm GeH}_{\rm 4} \to {\rm \dot GeH}_{\rm 3} {\rm + HI} $$\end{document} in the temperature range of 425-446 K. This information is used to show that iodine substitution weakens Ge-H bonds by 14.4 ± 2.5 kJ/mol and that D(H2IGe—H) = 332 ± 10 kJ/mol (79.3 kcal/mol). Possible reasons for the effects of halogen substituents on Ge—H and Si—H bond strengths are discussed.

    Notes: The rate constants of gas-phase reactions of the hydroxyl radical with β-dimethylstyrene and acetone have been determined by a relative method at 298 K. The values obtained are β-dimethylstyrene (3.3 ± 0.5) × 10-11 cm3/molecule·s and acetone (6.6 ± 0.9) × 10-13 cm3/molecule·s. A simplified kinetic treatment of the experimental data shows that β-dimethylstyrene is stoichiometrically converted to benzaldehyde and acetone. In the photooxidation study of benzaldehyde, carbon dioxide was the only detected product. The ratio between carbon dioxide produced and benzaldehyde reacted was ≥1.

Interestingly the oxidation of phenylacetylene and diphenylacetylene does not undergo catalysis by Ru(III). The mechanism involving the metal-arene π complex is discussed.

    Notes: A new approach, the method of polynomial approximations (PAM), to the sensitivity analysis in chemical kinetics is presented. The method is based on first dividing the time domain of interest into subintervals, and then, within each subinterval, using low-degree interpolation polynomials to mimic the system temporal behavior. This procedure forces all parametric dependences of the system to reside in the expansion coefficients and transforms the differential sensitivity equations into a set of algebraic ones. The major computational effort of PAM is proportional to the number of components in the system, not to the number of parameters. In addition, higher order sensitivity coefficients in PAM can be generated quite readily once first-order ones are known. The information required to divide the time domain comes from a preliminary simulation study of the system temporal behavior, which is always available in any kind of modeling studies. Typically, for an interpolation polynomial of degree 3-4, only 10-20 subintervals are needed to attain satisfactory accuracy. The application of PAM is well suited to large-scale kinetic models, especially when an inexpensive scanning of the system sensitivity behavior is desired. The extremely high computational speed of PAM in securing sensitivity informations was demonstrated by two illustrative kinetic examples. Furthermore the problem of utilizing sensitivity information to unravel the functional dependence of a species concentration upon rate coefficients, to simplify a complex reaction model, and to elucidate mechanistic details of a reaction process was examined in detail.

    Notes: Shearing of ghosts in a French pressure cell produces three classes of microvesicles that differ from endocytic vacuoles, exocytic vacuoles, and inside-out vesicles. It was thought that an analysis of these vesicles might provide some clues about the assembly of proteins within the human erythrocyte membrane. The microvesicles were separated into three visible bands, labeled top, middle, and bottom, and assayed for activity of Mg++-ATPase, Na+, K+-ATPase, acetylcholinesterase, glyceraldehyde-phosphate dehydrogense, and NADH oxidoreductase. Their proteins were also characterized by polyacrylamide gel electrophoresis with both Coomassie blue staining, to assess total protein content and distribution, and PAS-staining, to characterize sialoglycopeptides. In order to minimize problems inherent in ghost preparation, Dodge or hypotonic ghosts and glycol or isotonic ghosts were used in all studies. Middle membrane vesicles most resembled intact ghosts. Top vesicles had reduced levels of NADH oxidoreductase and more PAS-2 at the expense of PAS-1. The bottom vesicle class was very much enriched with PAS-1 at the expense of PAS-2, and PAS-3 was completely absent. In addition bottom vesicles had highest NADH oxidoreductase activity but lowest activity of all the other enzymes measured. These vesicle classes could not have been produced by tangential shearing through the membrane, nor could radial shearing through a membrane in which all proteins were free to move laterally have accounted for the three discrete vesicle classes or for their different patterns of enzymes and proteins. The analysis of the microvesicles produced by shearing is most consistent with radial shearing through membranes where there may be fixed domains superimposed on the basic fluid-mosaic structure.

    Notes: A lactose-binding lectin previously purified from embryonic chicken muscle and adult chicken liver, and here referred to as chicken-lactose-lectin-I (CLL-I), was added to sections of various adult chicken tissues to detect available binding sites. Both the sites of binding of added CLL-I as well as the tissue distribution of endogenous CLL-I were determined by indirect immunofluorescence using a rabbit antibody to CLL-I followed by fluorescent goat anti-rabbit IgG. Some tissues such as intestine and kidney showed abundant extracellular binding sites for the lectin, primarily between cells, in basement membrane, and in material on the luminal surface. In contrast, adult heart showed no significant binding sites for CLL-I. Adult pancreas showed considerable endogenous CLL-I in an extracellular site surrounding exocrine lobules, but added CLL-I did not bind substantially. The distribution of CLL-I binding sites in intestine were mimicked by those of purpurin, another lactose-binding lectin. CLL-I binding sites were also detected on the surface of cultured chick embryo skin fibroblasts. The factors controlling the specific distribution of occupied and unoccupied CLL-I binding sites are not known.

    Notes: A specific in vitro assay was developed for the protein carboxyl methyltransferase that is involved in the chemotactic behaviour of Salmonella typhimurium. This cytosolic enzyme catalyzes an S-adenosyl-L-methionine-dependent methyl esterification of glutamyl residues on a class of 60,000-dalton inner-membrane proteins. The activity was found to display a pH optimum of 6.5 and be sensitive to the concentration of salts in the assay medium. No detectable activity was found towards a variety of other proteins which serve as substrates for mammalian and other bacterial carboxyl methyltransferases. This assay was used to quantitate the methylation of the 60,000-dalton methyl-accepting proteins in response to chemoeffectors. Small but reproducible concentration-dependent changes in the initial rates of in vitro methylation were observed with chemotactic attractants and repellents. The specific methyltransferase activity was found to be absent in several mutants in flagellar synthesis (fla-), suggesting that the synthesis of this enzyme is coordinately regulated with that of flagellin and basal bodies. The hydrodynamic properties of the enzyme in crude extracts were determined by gel filtration and sucrose velocity gradient centrifugation, and a native molecular weight of 41,000 was calculated from these data.

    Notes: Plasma contains a number of insulin-like activities (ILA) of molecular weights 7,000 to 90,000 (somatomedins and insulin-like proteins) which stimulate cellular metabolism and may function as growth factors. We have found evidence for the presence of an 800 Dalton peptide in human plasma which markedly stimulates the metabolism of chick chondrocytes.This peptide was extracted from human Cohn fraction IV-1 by procedures similar to those used for somatomedin isolations. At the Sephadex G-50 column separation step, the fraction with molecular weights of 300-1,000 was found to markedly stimulate chick chondrocyte metabolism. Rechromatography on Sephadex G-25 concentrated activity in peptides of molecular weight of about 800. An HPLC separation on a silica C-18 reverse phase column gave elution of the active peptide at 18% acetonitrile in water. This bioactivity appears to be a peptide which is free of lipids, carbohydrates, nucleic acids, metal ions, and immunoreactive insulin. This factor markedly increased the metabolism of cultured chick chondrocytes, but had only marginal activity on rat chondrocytes. When added at 1 μg/ml to chick chondrocytes cultured in F-12 medium plus 1.5% fetal calf serum, the HPLC-purified activity increased DNA synthesis 7.3-fold, lipid synthesis 10.2-fold, and lactate production 2.9-fold after 48 h incubation.However, unlike somatomedins A and C, this factor did not displace insulin from placental membranes. These results suggest that low-molecular-weight peptides, which are smaller than the somatomedins, may contribute to the total ILA of human plasma.

    Notes: We have recently described the affinity chromatography purification of the turkey erythrocyte β-adrenergic receptor. The minute amounts obtained initially precluded extensive biochemical characterization. To improve the yield of the receptor, the erythrocyte membranes have been prepared by a new method. This procedure resulted in a 10-fold higher receptor density in comparison with the membrane preparation used previously. The new membranes also contained a catecholamine-sensitive guanine triphosphatase and an adenylate cyclase sensitive to Gpp(NH)p and l-epinephrine. Solubilization by a double digitonin extraction resulted in a preparation containing 4-6 pmoles of 3H-dihydroalprenolol binding sites per mg of membrane protein.A single step of affinity chromatography on alprenolol-sepharose of the soluble digitonin extract resulted in an additional 1,000-fold purification of the receptor. The overall purification factor was 20,000 relative to the binding activity of the crude membrane preparations.Electrophoresis in SDS-polacrylamide of iodinated purified β-receptors revealed, after autoradiography, the presence of four major components. Three of these, corresponding to molecular weights of 170,000, 33,000, and 30,000, respectively, were not affected by reduction with β-mercaptoethanol and were not observed when the digitonin extracts were loaded on the affinity gel in the presence of an excess of l-propranolol. A fourth 52,000-dalton component (60,000 daltons after reduction with β-mercaptoethanol) remained apparent even when affinity purification was prevented by addition of l-propranolol.Our results suggest that the β-adrenergic receptor is composed of at least three subunits that interact by noncovalent bonds.

    Notes: An Mg2+-dependent low ATPase activity can be detected in erythrocyte “white membranes,” in addition to that of the well known (Ca2+ + Mg2+)-ATPase. The thiol oxidizing agent diamide affects both activities. The oxidation of neighboring thiols seems to leave the mechanism of the (Ca2+ + Mg2+)-ATPase amplification system evoked by Ca2+ largely unaffected. The perturbation caused by diamide in the membranes seems to affect primarily a step of the ATP hydrolysis mechanism that is common to both ATPase activities. The effectiveness of diamide seems to be the same when either Ca2+ and Mg2+, or Mg2+ alone are present during the reagent action. Reduction of disulfide bonds by DTE after diamide treatment restores the (Ca2+ + Mg2+)-ATPase activity but is unable to take the Mg2+-ATPase activity back to the original level.The hypothesis is discussed that the redox state of one (or more than one) couple of —SH close to each other and possibly connected to the active site, may be an important factor in optimizing the efficiency of Ca action on the (Ca2+ + Mg2+)-ATPase.

    Notes: The cationic local anaesthetics carbocaine and unpercaine were found to increase the fluoride-stimulated adenylate cyclase up to a maximum level; above this maximum level further increases in drug concentration inhibited the enzyme. At concentrations where this activity was stimulated, a fatty acid spin label detected an increase in bilayer fluidity, which, it is suggested, is responsible for the activation of the enzyme. A solubilized enzyme was unaffected by the drugs, a finding consistent with this proposal.These cationic drugs began to inhibit the glucagon-stimulated activity at concentrations where they activated the fluoride-stimulated activity. It is suggested that this is due to their effect on the coupling interaction between the receptor and catalytic unit.The anionic drugs, phenobarbital, pentobarbital, and salicylic acid, all inhibited the fluoride-stimulated enzyme. This may be due in part to a direct effect on the protein and in part to the interaction of the drugs with the bilayer. The drugs had small inhibitory effects on the lubrol-solubilized enzyme.The glucagon-stimulated enzyme was initially inhibited by the anionic drugs at low concentrations, then activated, and finally inhibited with increasing drug concentration. The reasons for such changes are complex, but there was no evidence from electron spin resonance studies to suggest that the elevations in activity were due to increases in bilayer fluidity.

    Notes: Calcium has been suggested as an internal second messenger when lymphocytes are stimulated by mitogens to enter the cell cycle. We have assessed the effect of 2 lymphocyte stimulants, the plant lectin phytohemagglutinin (PHA) and the calcium ionophore A23187, on human lymphocyte nucleic acid synthesis, total cell calcium content, and 4 5Ca labeling. We have used an ultrasensitive method for the measurement of total cell calcium in the same samples used for radiolabeling. Mitogenic concentrations of A23187 (∼ .25 μ mole/liter) caused an increase in both total cell calcium and 4 5Ca labeling. These increases were almost completely blocked by inhibitors of mitochondrial respiration, suggesting that the calcium increment after ionophore treatment was located in the mitochondria. In contrast, total cell calcium was not altered at optimal mitogenic PHA concentrations (0.1 μg/ml and above). However, at the minimum PHA concentrations that caused stimulation (0.025 to 0.1 μg/ml), the dose response of 4 5Ca uptake was very similar to that of DNA sysnthesis. Importantly, we could not stimulate DNA synthesis with PHA without increasing lymphocyte 4 5Ca labeling. Thus, an increase in total cell calcium is not essential for mitogenesis; however, an increase in 4 5Ca exchange is closely associated with the mitogenic effects of A23187 and PHA.

    Notes: The role of hematopoietic microenvironments in the regulation of maturation and differentiation of hematopoietic cells, although heavily debated, remains uncertain. Several investigators have suggested that the adherent “stromal” cell populations, which grow as colonies in cultures of lymphomyeloid tissues, include the cells involved in such regulatory processes. Grossly, the colonies described by several investigators appear similar morphologically, and the cells giving rise to them have been variously termed (1) fibroblast colony forming cells (FCFC), (2) plaque forming units-culture (PFU-C), (3) macrophage colonies, and (4) marrow stromal cells. FCFC have been reported to re-establish their parent microenvironment when transplanted in an allogeneic system. In this study, cloned and enriched cell populations obtained from such colonies in cultures of murine lymphomyeloid tissues have been characterized by their growth in culture and using morphological, histochemical, and electron microscopic techniques. The results demonstrated that, although the initial stromal colonies appeared to be identical, the constituent cell types varied considerably. Some colonies were comprised primarily of macrophages, while others appeared to contain predominantly fibroblasts; two additional cell types that established colonies have not yet been satisfactorily identified. These results demonstrate the heterogeneity of lymphomyeloid stromal colonies. There is a need for caution in the analysis of experiments in which uncharacterized stromal cell colonies are transplanted or employed as supporting monolayers in culture systems in experiments designed to evaluate the origins and functions of lymphohematopoietic stroma.

    Notes: An anti-T cell serum raised in allotype congenic mice recognizes the product of a new locus coding for a heavy chain-linked polypeptide found on a subpopulation of T cells. Anti-Tsd raised in BALB/cAnN mice against selected C.AL-20 T cells reacts with a cell surface antigen in virgin animals that is found on 25% of mature thymocytes and Lyt-bearing T cells, but not on prothymocytes, Lyt1 T cells or B cells. The antigen is restricted to strains bearing the Ig-1d and Ig-1e heavy chain allotype haplotypes, and is expressed in the F1 animal. The antigen is unlinked in expression to the Lyt2, H-2, or kappa light chain loci. The antigen is not detected in the hematopoietic cells in the bone marrow and appears to mark only the mature peripheral pool of T cells. As previously reported, the antiserum blocks the binding of suppressor T cells to the cross-reactive idiotype for arsonate, while reagents specific for Fab, Fc and Ig were ineffective. It seems probable that the marker may represent a T cell constant region marker analogous to the Igh products on immunoglobulin. Antiserum against this marker induces in vivo triggering of Ts cells for a wide variety of T-dependent antigens. All subclasses of anti-hapten antibodies are suppressed; no affinity restrictions or clonotype specificity is observed in suppressed adult mice. Results suggest that precursor T cells regulating major serum idiotypes regulate individual idiotypes.

    Notes: Polypeptide growth factors are substances that stimulate an increase in cell size and/or cell number during embryonic development. In some cases, they have a similar effect on tissues in the mature organism where they function as “maintenance” factors to sustain cell viability. While their profound impact on cell behavior is well recognized, their relationship to other regulators of cell function has remained generally ill-defined. However, the developing appreciation of their hormone-like behavior suggests that they may be conveniently grouped with many other endocrine agents to form a broader group of secondary hormones. The utility of the classification is illustrated by the insulin-related family of molecules. It also serves to emphasize the similarities in function shared by many of these substances including trophic stimulation and modulation of gene expression. Internalization, though, appears to be another common feature. However, whether the uptake of the growth factor mediates an intracellular action or is designed solely to regulate responsiveness at the cell surface and/or degradation remains an important unanswered question. A brief review of two growth factors (nerve growth factor and epidermal growth factor) serves to outline the possible functions that may be served by this endocytotic process.

    Notes: Thy-1.2 negative progenitors give rise to Thy-1.2 positive colony cells when mouse bone marrow is cultured in vitro. The bone marrow cells are immobilized in a viscous medium containing methyl cellulose; discrete colonies are identifiable at 2 days and contain 30-60 cells by day 3 of culture. Colonies are tightly packed spheres (raspberries) and grow suspended in the gel. Growth of the raspberry colonies is absolutely dependent upon the presence of the appropriate serum (horse or human; not fetal calf) and conditioned medium from pokeweed mitogen-stimulated mouse spleen cells. As little as 0.1% of the conditioned medium is sufficient to promote raspberry colony growth. Under these conditions, nude mouse bone marrow yields as many colonies (1 per 1,000 nucleated cells plated) as normal marrow. Thymus, lymph node; and spleen (normal or nude) do not form colonies. Colony precursors are predominantly in S phase of the cell cycle, as determined by tritiated thymidine suicide of fresh bone marrow. Their numbers fall with age. Because the cells in colonies are Thy-1 positive, peanut agglutinin-positive, and active in a pre-T cell synergy assay, we conclude that their precursors are early committed T cell progenitors, and propose that they be called CFU-preT.
