    Notes: The analysis of ultraviolet-absorbing constituents in urine was undertaken by anion-exchange and reversed-phase liquid chromatography. A newly developed anion exchanger (TSK-IEX540 DEAE) was used for ion-exchange mode liquid chromatography and its high polarity and physical strength greatly reduced the analysis time. Up to 80 peaks were resolved within 30 min. Hydrophobicity values were used to predict the retention times of acids in reversed-phase mode liquid chromatography.

    Notes: The C1 -C6 fatty acids present in various types of heterogeneous animal wastes were isolated by steam distillation and determined as benzyl esters by high resolution gas chromatography. Glass capillary columns, deactivated with barium carbonate and coated with UCON HB 5100, allowed the analysis of benzyl formate, acetate, propionate, isobutyrate, butyrate, isovalerate, valerate, and caproate in less than 20 minutes. Recovery tests, performed on more than 50 different samples, indicated a benzylation yield and an overall efficiency higher than 99%, with a standard deviation of 2.6% (C5) to 6.0% (C1). The absolute detection limit ranged from 0.3 picomole (caproic acid) to 1.0 picomole (formic acid).

    Notes: An integrated gas chromatography/Fourier transform infrared spectrometry (GC/FT-IR) system developed for the analysis of environmental pollutants is described. The versatility of the system allows the utilization of many different techniques of sample introduction and manipulation during analysis. The sample can be introduced by direct injection or thermal desorption from an adsorbent cartridge, and can then be separated on one of two capillary columns and detected by FT-IR or an FID. Cold traps and collection cartridges incorporated in the system permit recovery and additional fractionation of samples. Recovered sample and sample fractions can then be re-analyzed by GC/FT-IR or subsequently analyzed by GC/MS or other methods.

    Notes: Direct in situ determination of PGE1 in dioxane/water (1:1) at concentrations between 50 μg/ml and 1 mg/ml is described. HPTLC of PGE1 was carried out on HPTLC Kieselgel 60 o.F. plates; application (200 nl) was with a Nano-Appllcator and ethyl acetate/formic acid (400:5, v/v) served as mobile phase. The spots were located by dipping the plates into a 3% cupric acetate solution in 15% aqueous phosphoric acid, followed by heating at 120°C for 15 min. The plates were then evaluated directly at λ = 345 nm by chromatogram spectrometry. The coefficient of variation is 3.6%.

    Notes: Two glass capillary gas chromatographic systems were equipped with inert effluent splitters which allowed simultaneous data acquisition using nonspecific and element-specific detectors. Simultaneous detection was achieved using the nonspecific flame ionization detector (FID) and the Hall® electrolytic conductivity detector (HECD) operated in either the sulfur-or the nitrogen-specific mode. Typical application of the simultaneous detection system as applied to analysis of petroleum residues is briefly described. The Hall electrolytic conductivity detector can be made element specific for halogen-, sulfur-, or nitrogen-containing compounds. Simultaneous detection enhances the information yield from a single sample injection and proves to be a powerful complementary technique when used with computerized gas chromatography/mass spectrometry.

    Notes: A gas chromatographic method developed for the simultaneous determination of residues of pirimiphos-methyl (O-[2-(dimethyl-amino)-6-methyl-4-pyrimidinyl] O,O-dimethyl phosphorothioate) and malathion (diethyl mercaptosuccinate S-ester with O,O-dimethyl phosphorodithioate), in whole peanuts, peanut hulls, peanut meats, and peanut oil gives good recoveries and has a limit of detection for pirimiphos-methyl and malathion of 0.04 ppm. Quantitative analysis for the two insecticides with electronic integration was reliable to 0.1 ppm.

    Notes: The measure of the distribution of a mixture of substances in TLC with a binary mobile phase is expressed as ΔRM1,2i,j which represents the difference between the RM values of substances i and j on use of the binary solvent 1 + 2 as the mobile phase. The possibility of determining its maximum value at an optimum composition of the binary mobile phase is demonstrated in this paper. This value can calculated from experimental and theoretical functions RM1,2 = f(Φ1) of substance i and j. More simply, ΔRM1,2i,j can be calculated theoretically from easily measurable adsorption parameters of excess adsorption isotherms and from chromatographic parameters obtained for pure solvents. The ΔRM1,2i,j value calculated theoretically can be utilized in a pilot technique for determination of the optimum composition of the mobile phase in gradient liquid chromatography.

    Notes: Discrimination may occur when injecting samples onto gas chromatography capillary columns, whereby peak areas for higher boiling point compounds are smaller than they should be compared to lower boiling compounds. This problem is most important in quantitative work on solutes having a wide range of volatility. An all-glass inlet system was used. Injections were made with a 10-μl syringe onto a column without and injection point heater and with the column oven about 20°C below the boiling point of the solvent used for the solutes. Discrimination did not occur in a range of C12 to C36 n-alkanes.

    Notes: The use of gradient elution in RP-HPLC is increasing. Aqueous buffers, such as phosphate at pH 1.7, with acetonitrile as the organic modifier are particularly advantageous as eluents with linear elution strength (LES). In the isocratic mode, under the experimental conditions used, fairly good linearity exists between logk' and xB (xBº is the volume ratio of the organic modifier, i.e. acetonitrile) with a correlation coefficient of 0.9995. The relationship between isocratic and gradient elution parameters, such as xB, xBº (starting composition in gradient elution), m (slope of the linear gradient), is transparent and the parameters of interest can be predicted fairly accurately using simple equations. The proposed system could be used in elaboration of a general retention index system for organic molecules in RP-HPLC.

    Notes: This paper describes the screening for metabolites of butoprozine, a new anti-anginal drug, in dog bile by means of reverse-phase HPLC. Although it does involve a simple clean-up step to remove a substantial amount of endogenous bile compounds, this screening method nevertheless avoids extraction of metabolites and thus allows all metabolites to be introduced into the chromatographic system. A single run of 100 minutes from 100% water to 100% methanol in a linear gradient effects adequate separation of the great majority of metabolites without interference from remaining endogenous compounds.Two methods of differentiating between metabolite peaks and endogenous peaks have been worked out. The first one makes use of 14C-labeled butoprozine by measuring the amount of radioactivity in the column effluent while simultaneously recording the UV absorbance. The second method compares continuous gradient chromatograms of bile recorded before and after butoprozine administration under very similar conditions. The latter method can be applied to both radioactive and non-radioactive materials.

    Notes: A method is described for the preparation of a Porous Layer Open Tubular (PLOT) fused silica column coated with submicron particles of aluminum oxide for the analysis of light hydrocarbons. The column is thermally stable and provides reproducible analytical data over a long period of time. The column can be operated at temperatures easily controlled by commercially available gas chromatographic apparatus. 1,3-Butadiene is retarded much more than its principal impurities: thus the polarity of aluminum oxide is extremely useful for butadiene analysis because none of the contaminants is obscured by the butadiene peak. The analysis of the 16 most common C1-C4 hydrocarbons is achieved within 5 min at 60°C with helium as the carrier gas. Conditioning of the carrier gas with water vapor from CuSO4.5H2O to decrease the activity of the aluminum oxide is described. Some applications of a 30 m × 0.32 mm aluminum oxide PLOT column are given.

    Notes: This contribution comprises two parts, the one covering some variables in the dry-packing of columns for liquid chromatography, the other the use of an automatic dry-packer in liquid chromatography. A high-pressure stainless steel column was dry-packed using a 20-44 μm size, irrregularly shaped, silica gel support. It was found that, when support was added in discrete amounts, lateral tapping produced a column of higher efficiency (HETP = 0.69 mm) than was the case for vertical tapping (HETP = 1.37 mm). Simultaneous vertical and lateral tapping produced an intermediate result. Bulk filling of the column followed by tapping was inferior to adding support in discrete amounts. An apparatus constructed to perform the vertical and lateral tapping operations automatically confirmed the above conclusions. It was concluded that commercially available dry-packing apparatus do not offer the best action for packing an efficient column since they involve predominantly vertical tapping.

    Notes: In the approach described in this paper, the layer of adsorbent is divided into n sublayers. The radiation density in each sublayer is given by the sum of the transmittance and the reflectance of the adjacent sublayers. Absorption of light by the layer or the substance will diminish this radiation density. Depending upon the factors of reflectance and transmittance some light will pass to the adjacent sublayers. Two polynomials are derived from this readily understood formulation to calculate the reflectance or the transmittance of a layer of finite thickness. The algorithms given describe the reflectance and transmittance encountered in practical measurements with large amounts of substance per spot, as well as with small amounts. There are no restrictions with respect to nonideal scattering or reflection by the adsorbent layer support.

    Notes: A simple, quick method is described for GC head space evaluation of herbs and drugs. The proposed procedure consists of milling a weighed sample of a certain herb in a glass device like a Braun MXK coffee grinder, modified in such a way that a special gas-tight valve allows the direct sampling of the vapors. Glass capillary columns are used to analyze the head space volatiles. Some preliminary applications of the method are given.

This is accomplished by injecting the extractable organics, then applying a cryogenic focus to the front of the column, and desorbing the volatile components. The column utilized for this procedure is a fused silica open tubular (FSOT) column coated with the liquid phase SE-54. Minimum detection limits of this procedure by electron impact GC/MS are shown to be≤ 1 μg/liter in tests conducted on actual water samples.

    Notes: The utility of Rekker's hydrophobic fragmental constant has been examined for optimization of reversed-phase mode liquid chromatographic separations. The chromatographic behavior of about 60 non-ionic compounds was measured in different acetonitrile/water mixtures and the logarithm of their capacity factors (log k) was correlated with their calculated hydrophobicities (log P). Linear relations were found in each case between log k and log P. The slope of the various lines was related to the percentage concentration of acetonitrile in the mobile phase. It was shown that, by using nine stand ard compounds and measuring their capacity factors in five eluents with different acetonitrile concentrations, the retention time could be predicted for 60 compounds. Calculation of the concentration of the organic modifier was also possible in a system of well coated octadecyl bonded packings with acetonitrile/water mixtures as eluent. Prediction of the capacity factor was accomplished to within 5% error.

    Notes: Analytical results are often obtained from reflectance or fluorescence measurements in TLC or HPTLC with the aid of calibration lines. Curve fitting may not be done by conventional least squares methods for both the independent and the dependent variables are erroneous; errors occur in the volumes spotted on to the plate and in the reflectance/fluorescence measurements. Therefore the sum of the squares of the distances to the calibration line has to be minimized instead of the squared sum of the errors of the dependent variable. The algorithms used are derived and explained. Implications of error propagation for the analytical result are also given.

    Notes: Summary The effects of pimozide, a dopamine receptor-blocking agent, were studied in the pars intermedia of the rat. The animals received 100 μg/100 g pimozide daily for 2, 5, 10, 15, and 20 days. Pimozide induces ultrastructural changes after 5 days of treatment. About 50% of the MSH-cells display characteristics of stimulation. Their cytoplasm is partially or totally depleted of secretory granules. The rough endoplasmic reticulum displays a network of interconnecting cisternae and ribbon-like structures. The well-developed Golgi complexes exhibit numerous dilatations of their cisternae, which contain electron-dense material. The nerve endings are not altered. Twenty days after treatment, the above-described changes have not decreased in magnitude. The present findings suggest that pimozide stimulates the mechanism of synthesis and release in some MSH-cells, most probably the elements underlying an inhibitory dopaminergic control.

    Notes: Summary By use of the PAP-immunohistochemical staining technique with serial sections, somatostatin-immunoreactive fiber projections into the brain stem and the spinal cord are described. These projections originate in the periventricular somatostatin-immunoreactive perikarya of the hypothalamus and form three main pathways: (1) along the stria medullaris thalami and the fasciculus retroflexus into the interpeduncular nucleus; (2) along the medial forebrain bundle into the mammillary body; and (3) via the periventricular gray and the bundle of Schütz into the midbrain tegmentum. Densely arranged immunoreactive fibers and/or basket-like fiber terminals are observed within the following afferent systems: somatic afferent systems (nucleus spinalis nervi trigemini, substantia gelatinosa dorsalis of the entire spinal cord), and visceral afferent systems (nucleus solitarius, regio intermediolateralis and substantia gelatinosa of the sacral spinal cord). These projections form terminals around the perikarya of the second afferent neuron. Perikarya of the third afferent neuron are influenced by somatostatin-immunoreactive projections into the auditory system (nucleus dorsalis lemnisci lateralis, nucleus corporis trapezoidei). Furthermore, a somatostatin-immunoreactive fiber projection is found in the ventral part of the medial accessory olivary nucleus, in nuclei of the limbic system (nucleus habenularis medialis, nuclei supramamillaris and mamillaris lateralis) and in the formatio reticularis (nucleus Darkschewitsch, nuclei tegmenti lateralis and centralis, nucleus parabrachialis lateralis, as well as individual perikarya of the reticular formation). Targets of these projections are interneurons within interlocking neuronal chains.

    Notes: Summary The axon hillock (AH) and initial segment (IS) of 10 Golgi neurons and 6 basket cells in the cerebellar cortex of the rat were investigated by electron microscopy using serial sections. An average of 10.4 and 11.3 synaptic terminals were observed to establish synaptic contact with the axon hillock region of Golgi and basket cells, respectively. Most of these terminals were identified as the varicosities of the ascending parallel fibers. It is suggested that the focal innervation of AH regions represents an excitatory input pattern which is basically different from the randomly distributed, huge, parallel-fiber input onto the dendritic trees of Golgi and basket cells. In contrast to Golgi and basket neurons, no accumulation of parallel-fiber synapses was observed around the AH of stellate cells. The IS proper of the three neuronal types were devoid of true axo-axonal synapses.

    Notes: Summary The development of neuro-muscular junctions (mouse, rat) from the time of first contact between neurons and myotubes in culture and the changes which lead to the formation of functional synaptic contacts have been investigated using light microscopy and ultrastructural techniques. An extensive basal lamina was present when the neuronal cell population was added to the developing myotubes in culture. The nerve cells were initially strongly attracted to each other and nerve cell aggregates formed rapidly. It was only when nerve fibres began to grow out of these aggregates to contact developing myotubes that changes within the cytoplasm of the two adjacent cells were observed. These developments included accumulations of filaments, membrane densities, mitochondria and large clear vesicles within both cells in the region of contact. In addition, collections of glycogen granules and an extensive membrane reticular complex were found within myotubes, and an extensive granular material filled many of the nerve processes. The basal lamina within the intercellular space appeared more electron-dense than elsewhere and was traversed by strands linking the two cell membranes. These features all appeared to be stages in the initial formation of neuro-muscular junctions. It was only after these events had occurred that presynaptic vesicles gradually appeared within the future nerve terminal. The results of this paper therefore support the view that synaptic transmission at developing mammalian neuromuscular junctions is not necessarily dependent on the presence of presynaptic vesicles.

    Notes: It has been reported that glass wool packed tightly into the glass liner of a vaporizing injector used in the splitting mode considerably reduces the standard deviation of the results obtained, because of improved evaporation of the sample prior to reaching the split point at the capillary column entrance. This finding could not be reproduced on using the same sample composition as reported in the literature, i.e. methanol/2-ethyl-1-hexanol (1:1).The standard deviations obtained were between 3 and 10% (depending on the conditions selected) and were not influenced significantly by the introduction of glass wool.The peak area ratio (methanol/2-ethyl-1-hexanol) was found to depend on a number of parameters, such as: injector temperature; glass liner internal diameter; syringe handling technique; the relative position of the syringe needle exit and capillary column entrance; the sample volume injected; and the packing of the glass liner. Generally, the area ratio deviated further from the correct one (determined by cold on-column sampling) when the glass liner was packed with glass wool.On the basis of the results, it is speculated that either a complete evaporation of the sample should be achieved (which appears to be impossible under the conditions we used) or, alternatively, the sample should be given the least possible opportunity to evaporate, thus allowing it to enter the column in the form of droplets. The results were worse in terms of precision and accuracy the greater the partition of sample components between the liquid (droplet) and vapor phase. It is concluded that the use of evaporation aids such as glass wool cannot be generally recommended.

    Notes: The separation of Digitalis cardenolides has been carried out by HPLC on an adsorbent column. By choice of suitable mobile phase, isocratic elution permitted resolution of mixtures of (a) aglycones, (b) secondary glycosides, and (c) primary glycosides, while gradient elution provided a means of resolving more complex mixtures of these cardiac steroids. HPLC could therefore be used in the quality control of cardiotonic drugs replacing the TLC tests for related compounds currently used, and by suitable calibration could replace the colorimetric assay procedure normally used for such drugs.
