    Notes: Abstract  The anatomical distribution of the GABAB receptor (GBR) splice variants GBR1a and 1b in the CNS has not previously been studied. In the present study, distribution of the splice variants was mapped using immunohistochemistry. Polyclonal antibodies against splice variant unique epitopes were raised in rabbits. Affinity purified antibodies were used according to routine immunohistochemical procedures in sections from the rat CNS or dorsal root ganglia (DRG). The staining intensity was high in the cerebral cortex but lower in basal ganglia and the hippocampus. In the cerebellum, there was a marked difference in the distribution of GBR1a- and 1b-like immunoreactivity (LI). GBR1a-LI was preferentially localised in the granule cell layer whilst GBR1b-LI was mostly found in Purkinje cells and in the molecular layer. Cell bodies of the deep cerebellar nuclei stained for the GBR1a antibody while terminals surrounding the cell bodies were strongly labelled with the GBR1b antibody. A similar pre- vs postsynaptic pattern was seen in several nuclei ventral or caudal to the cerebellum (e.g. the cochlear nucleus, the facial nucleus, the spinal cord) but not in regions rostral to the cerebellum. In the spinal cord, strong labelling for both antibodies was seen in the dorsal horn. The GBR1b but not the GBR1a antibody stained tanycytes in the epithelium of the 3rd ventricle and in the central canal at the brain stem level. DRG neurons were positive for both the GBR1a and 1b antibody, but the former stained the cells much more intensely. Satellite cells were labelled with the GBR1b antibody. The most important aspect of these findings is that in some nuclei, GBR1b may mediate inhibition of transmitter release while in the same regions, GBR1a may mediate postsynaptic inhibition. Further, the observations support previous findings that GBR1b is the predominant splice variant in Purkinje cells.

    Notes: Summary The occurrence of abdominal aortic aneurysms (AAAs) and intracranial aneurysms (IAs) in the same patient and in the same family was studied among 89 patients with AAAs and 485 patients with IAs. Among the AAA-patients two had IAs themselves and five had IAs in the family, whereas three IA-patients had AAAs themselves and eight had AAAs in the family. Moreover, one of the patients with both AAA and IA had a blood relative with AAA, and in six of the families with both types of aneurysms there were more than two subjects with aneurysms. The results indicate, that AAAs and IAs may have a common aetiologic factor.

    Notes: The epidermal response to 2 different irritants, nonanoic acid (NAA) and sodium lauryl sulfate (SLS), was investigated with 2 different methods. NAA 80% and SLS 4% were applied under occlusion for up to 24 h. Elemental changes were determined in cryosections by x-ray microanalysis. Compared to unexposed skin a significantly higher sodium/potassium ratio was found after 6 h in NAA-exposed skin and a lower ratio in SLS-exposed. At 24 h both substances had induced similar changes, compatible with a cell injury. The findings demonstrate a time-dependent NAA and SLS response, With reverse transcription potymerase chain reaction, the mRNA expression of interleukin-1 alpha (IL-1α;), -1beta (IL.-1β), -6 (1L-6), and -8 (IL-8). tumor necrosis factor alpha (TNFα) and granulocyte macrophaye colony stimulating factor (GM-CSF) in shave biopsies from irritated and unexposed skin was studied at 0. 4, 8 and 24 h NAA, but not SLS. induced an increase in mRNA expression for IL-6. mRNA-expression for GM-CSF was increased after SLS exposure, but not after NAA. These findings indicate a time and substance dependent different in the up-regulation of m-RNA different cytokines in epidermis during the first 24 h of the irritant reaction. This might he the effect of differences in the irritants action on the cell membranes, which is also reflected by the differences found in the elemental content at 6 h.

    Notes: Aims:  The aims were to study health-related quality of life (HRQL) in patients with hand eczema, and to compare two different instruments for assessment of HRQL.Methods:  100 consecutive hand eczema patients (51 females and 49 males) at an occupational dermatology clinic completed the generic questionnaire Short Form-36 (SF-36), and the dermatology specific Dermatology Life Quality Index (DLQI). To compare the instruments factor analysis, with a polychoric correlation matrix as input, was performed, thus taking the ordinal aspect of the data into account.Results:  HRQL was affected by hand eczema, measured with both SF-36 and DLQI. The mean (SD) sum scores of DLQI was 7.4 (5.8), no difference between genders. However, the SF-36 showed more impaired HRQL for females than for males in the mental health dimension. There was a high correlation between the instruments for physical health, but lower for mental health.Conclusions:  Hand eczema has an impact on HRQL, and both SF-36 and DLQI are suitable instruments. Our conclusion from the factor analysis is that the SF-36 measures mental health better than the DLQI. The choice of instrument to be used in a specific study depends on the purpose of the study. A generic instrument has the advantage of making comparisons with other diseases possible. The SF-36 also appears suitable for use in hand eczema studies where gender differences in HRQL are of interest.

    Notes: Aim:  To investigate the risk of adult hand eczema and possible influence on later working life in persons with childhood atopic dermatitis.Method:  After reviewing medical records from the school healthcare in Stockholm regarding individuals born 1960–69, 600 individuals where signs of atopic dermatitis were noted (“cases”) and 600 matched controls without eczema or allergic disease (“controls”) were identified. 405 cases and 378 controls answered a questionnaire regarding past and present skin disease, choice of job, exposure at work and possible change of job due to eczema.Results: Hand eczema was more than 3 times more common among cases, 42%, compared to controls, 13%(p 〈 0.001). Hand eczema during the past 12 months was reported by 24% of the cases and 9% of the controls (p 〈 0.001). The proportion of individuals working in jobs with high-risk for hand eczema was equal between the groups, as well as exposure to water, detergents, chemicals and hand washing. Among the cases 9% reported change of job due to eczema compared to 2% of controls (p 〈 0.001) and sick leave 10% compared to 2%(p 〈 0.001).Conclusions: Childhood atopic dermatitis is a risk factor for hand eczema. As the proportion of individuals in jobs with high-risk for hand eczema and the exposure was the same in cases and controls measures preventive measures are important to reduce consequences like sick leave and change of job.

    Notes: Occupational skin diseases are among the most frequent work-related diseases in industrialized countries. Good occupational skin disease statistics exist in few countries. Questionnaire studies are needed to get more data on the epidemiology of occupational skin diseases. The Nordic Occupational Skin Questionnaire Group has developed a new questionnaire tool – Nordic Occupational Skin Questionnaire (NOSQ-2002) – for surveys on work-related skin disease and exposures to environmental factors. The 2 NOSQ-2002 questionnaires have been compiled by using existing questionnaires and experience. NOSQ-2002/SHORT is a ready-to-use 4-page questionnaire for screening and monitoring occupational skin diseases, e.g. in a population or workplace. All the questions in the short questionnaire (NOSQ-2002/SHORT) are included in the long version, NOSQ-2002/LONG, which contains a pool of questions to be chosen according to research needs and tailored to specific populations. The NOSQ-2002 report includes, in addition to the questionnaires, a comprehensive manual for researchers on planning and conducting a questionnaire survey on hand eczema and relevant exposures. NOSQ-2002 questionnaires have been compiled in English and translated into Danish, Swedish, Finnish and Icelandic. The use of NOSQ-2002 will benefit research on occupational skin diseases by providing more standardized data, which can be compared between studies and countries.

    Notes: Contact allergy to and allergic contact dermatitis from methyldibromo glutaronitrile (MDBGN) have frequently been reported. As there has been no agreement on which MDBGN test preparation to use, a study was initiated to help determine the optimal patch test preparation for MDBGN. 2661 consecutively patch tested patients at 11 test clinics representing 9 European countries participated. Petrolatum preparations with MDBGN at 1.0%, 0.5%, 0.3% and 0.1% were inserted in the standard series. Contact allergy rates were noted in the range 4.4–1.1% following decreasing test concentrations. Reactions not fulfilling all criteria to be classified as allergic reactions could represent either weak allergic or irritant reactions, and such reactions were noted in the range 8.2–0.5% with decreasing concentrations. A significant number of these reactions represented weak allergic reactions, as allergic reactions were obtained to higher patch test concentrations in the same individual. Morphologically irritant reactions were noted only for the highest test concentrations. In summary, the contact allergy rates and frequencies of doubtful and irritant reactions vary with the patch test concentration. The final decision on patch test concentration for MDBGN should not only rely on these factors but also include information on patch test concentrations required to diagnose individual cases with allergic contact dermatitis from MDBGN as well as results of repeated open application tests.

    Notes: Questionnaire-tools for surveying occupational skin diseases and exposure are needed for comparable epidemiological research, workplace assessments, and evaluation of workplace interventions. The Nordic Occupational Skin Questionnaire Group supported by the Nordic Council of Ministers has developed a standardized questionnaire-tool for surveys on work-related skin diseases and skin exposures to environmental factors.Nordic Occupational Skin Questionnaire (NOSQ-2002) includes two questionnaires designed for separate purposes. NOSQ-2002/short is a 4-page questionnaire for screening skin problems at workplaces. NOSQ-2002/long is an in-depth survey tool for research purposes. The questionnaire covers occupational history, atopic symptoms, self-reported hand or forearm eczema, exacerbating factors, consequences and life impact of dermatoses, self-reported contact urticaria on hands or forearms, skin symptoms, skin tests, exposures, and protective glove use. For the time being, NOSQ-2002 is available in English, Danish, Swedish, Finnish and Icelandic. Further translations are welcomed.The NOSQ-2002 report includes a review of pertinent literature on questionnaire methods for skin disease studies. Questions on work and exposure can be tailored to specific populations or occupational groups, according to the instructions and recommendations given in NOSQ-2002/INFO version of the questionnaire.The NOSQ-2002 questionnaire files can be downloaded from /NOSQ. The Nordic Council of Ministers has the copyright to the NOSQ-2002 questionnaires. Use of the questionnaires is free of charge. The NOSQ-2002 questionnaires and their present and possible future translations cannot be used commercially.
