    Notes: Abstract AHR-10037 is an anti-inflammatory compound possessing analgesic and antipyretic properties and a high therapeutic index. AHR-10037 was comparable to indomethacin in suppressing acute (Evans blue-carrageenan pleural effusion) and chronic (adjuvant-induced arthritis) inflammation. There was a delayed onset of antipyresis (yeast-induced hyperthermia in rats), analgesia (acetylcholine-induced abdominal constriction in mice) and inhibition of caster oil-induced diarrhea in rats. Antipyresis occurred 3 hours after administration of AHR-10037, 4 mg/kg, PO, vs 1 hour after administration of acetylsalicylic acid, 100 mg/kg, PO; maximum analgesic activity (ED50-4.1 mg/kg) occurred at 4 hours. AHR-10037 was inferior to indomethacin in suppressing castor oil-induced diarrhea in rats. The therapeutic index of AHR-10037 (relating acute anti-inflammatory potency to gastric toxicity potency relative to indomethacin) ranged from 56–91. The pharmacological profile suggests that AHR-10037 is a prodrug convertedin vivo to a cyclooxygenase inhibitor.

    Notes: Summary Hepatic glucose output is decreased by hyperglycaemia through an unknown mechanism. We hypothesize that free radicals generated by hyperglycaemic pseudohypoxia might cause glucose output to decrease by inhibiting glucose 6-phosphatase – a key enzyme of gluconeogenesis. To prove this a model experiment was performed on a microsome fraction of rat liver. One of the characteristic features of pseudohypoxia due to hyperglycaemia is an increase in the ratio of NADH/NAD + , so in the present study the changes in NADH – induced glucose 6-phosphatase activity were investigated as related to the release of inorganic phosphate (Pi) derived from glucose 6-phosphate. After incubation for 50 min, Pi release was significantly reduced by NADH (4.026 ± 0.189 vs 2.696 ± 0.429 μmol · l–1· mg protein–1, control vs NADH samples, p 〈 0.01). The decrease in the activity of glucose 6-phosphatase generated by NADH was prevented by using desferrioxamine, an irreversible ferric chelator, butylated hydroxytoluene and Trolox, two agents which inhibit lipid peroxidation, and reduced glutathione, a non-specific radical scavanger. Superoxide dismutase, catalase and the hydroxyl radical scavenger dimethyl sulphoxide proved to be ineffective. When the above investigations were carried out in the presence of a ferric-EDTA complex the inhibition of glucose 6-phosphatase was found to be inducible by hydrogen peroxide and/or hydroxyl free radicals. These investigations seem to indicate that pseudohypoxia due to hyperglycaemia can inhibit the activity of glucose 6-phosphatase both by lipid peroxidation and by inducing hydrogen peroxide and/or hydroxyl free radicals and thus it may play a part in the glucose-induced decrease of hepatic glucose output.     Notes: Summary We evaluated the influence of high ambient glucose on cellular expression of adhesion molecules, known to mediate endothelial interaction of leucocytes and monocytes. Paired cultures of individual isolates of human umbilical vein endothelial cells (HUVECs) were studied by fluorescence activated cell sorter analysis after exposure to 30 vs 5 mmol/l glucose. Incubation of HUVECs for 24 h in 30 mmol/l glucose increased ICAM-1 (intercellular adhesion molecule-1; 116.4±16.9% of control, p ≤ 0.05), but not PECAM (platelet endothelial cell adhesion molecule) expression, compared to cultures kept in 5 mmol/l glucose. Long-term exposure (13±1 days) of HUVECs to 30 mmol/l glucose increased expression of ICAM-1 to 122.5±32.2% (p〈0.002) and reduced that of PECAM to 86.9±21.3% vs the respective control culture in 5 mmol/l glucose (p〈0.02). Stimulation of confluent HUVECs, kept in 30 vs 5 mmol/l glucose for 13±1 days, with 20 U/ ml interleukin-1 for 24 h (ICAM-1) and 4 h (endothelial leukocyte adhesion molecule 1) resulted in reduced ICAM-1 (84.8±27.0%, p〈0.05) and endothelial leukocyte adhesion molecule-1 (87.6±22.4%, p〈0.05) expression vs control cells, while that of PECAM (t: 24 h) and vascular cell adhesion molecule-1 (t: 16 h) remained unchanged. In conclusion, it appears that differences in expression of adhesion molecules on HUVECs in response to high glucose reflects endothelial glucose toxicity, which may also induce endothelial dysfunction in diabetes.

    Description / Table of Contents: Zusammenfassung In einer multizentrischen „open label“ Studie mit IFN-α2b wurden 85 Patienten mit Haarzell-Leukämie behandelt und ausgewertet. Die Induktionsbehandlung bestand in 2 × 106 E IFN-α2b/m2, 3 × wöchentlich, s.c. Die Ergebnisse zeigen die hohe Effektivität der Substanz, wobei unter den verwendeten Dosierungen geringe, gut tolerierbare und passagere Nebenwirkungen im Sinne eines grippeähnlichen Syndroms auftraten. Die Prozentzahl der Remissionen (CR + PR + MR) nach sechsmonatiger Behandlungszeit lag bei 89%, der Anteil von CR betrug 4%, von PR 69%. An einer kleinen Gruppe (4 Patienten) wurde nach Erzielen einer CR oder PR der Effekt unterschiedlicher Formen der Erhaltungstherapie getestet. Die bisherigen, statistisch noch nicht auswertbaren Resultate sprechen dafür, daß bei Auftreten eines Rückfalles nach Aussetzen oder Verringerung der IFN-Dosis (3 × 106 I.E., s.c., 1 × wöchentlich) durch Wiedereinsetzen oder Erhöhung von IFN-α2 neuerlich Remissionen erzielt werden können. Zur Erhaltung der Remissionen dürfte eine IFN-Dauertherapie notwendig sein. Mittels eines Kurzzeit in vitro Tests werde der Effekt von Interferon α2 auf die Inkorporation von H3-Thymidin und H3-Uridin in Haarzellen bestimmt. Für beide Präkursoren ergab sich keine Hemmung des Einbaues. Allerdings zeigte eine Langzeitinkubation (48 h) einen signifikant vermehrten H3-Uridin Einbau, während der H3-Thymidin Einbau unbeeinflußt blieb. Diese Resultate sprechen gegen einen unmittelbaren antiproliferativen Effekt von IFN-α2 auf Haarzellen, sie weisen aber auf eine Induktion der RNS-Synthese hin. Eine Zellmarker-Analyse der Haarzellen mit monoklonalen Antikörpern vor und während einer 7tägigen in vitro Behandlung mit IFN-α2 ergab keine Änderung des Phänotyps, so daß eine differenzierende Wirkung des IFN auf Haarzellen nicht beobachtet werden konnte.
    Notes: Summary Eighty-five patients with hairy-cell leukemia were treated in a multicentric “open label” study with IFN-α2b and evaluated. Induction therapy was 2 × 106 U IFN-α2b/m2, 3 times a week, s.c. The results show this regimen to be highly effective with only a few tolerable and transient side effects consisting mainly of flu-like symptoms. After 6 months of therapy 4% CR, 69% PR, and 16% MR, were noted. In a small group of four patients who had achieved CR or PR, we tested the effect of varying doses for maintenance therapy. Our preliminary results indicate that a relapse caused by interruption of IFN therapy or dose reduction to 3 × 106 U given once a week, o.c. could be successfully treated by readministration, or escalating the dosage of IFN. It seems that remission maintenance requires long-term treatment with IFN. In a short-term in vitro test we studied the effect of IFN-α2 on the incorporation of3H-thymidine and3H-uridine into hairy cells of five patients. Fort both precursors no appreciable effect was detected. However, after prolonged incubation for 48 h, a significant enhancement of3H-uridine incorporation was observed, while3H-thymidine incorporation remained unaffected. Cell marker analysis performed with monoclonal antibodies before and after incubation of hairy cells with IFN-α2 for up to 7 days did not reveal any change of the phenotype of hairy cells.

    Notes: Summary A 33-year-old woman presented with abdominal pain and distention, diarrhoea and marked eosinophilia in blood and ascites. As other causes could be excluded, the subserosal type of eosinophilic gastroenteropathy was diagnosed. The low plasma fibrinogen level (〈 100 mg/100 ml) found in this patient is an as yet undescribed feature. During prednisolone therapy it increased concurrently with the fall of blood eosinophils and the relief of clinical symptoms. Interest was further directed to the ascitic fluid where not only the presence of eosinophils (74%) enveloped in fibrin yarn and of basophils (2%) but also of 24% T lymphocytes (among them 75% CD4+, 24% CD8+, 4% CD25+, 〈1% CD19+, 〈1% natural killer cells) could be demonstrated. These lymphocytes are likely to be the source for lymphokine production chemoattracting eosinophils into the intestine. In addition they seem to be involved in IgE hyperproduction, which after adequate therapy and complete resolution of the clinical symptoms, tended to decrease slowly.

    Description / Table of Contents: Summary Supravital blood platelets stained with acridine orange or auramine and suspended in EDTA-or citraplasma lost their fluorochrome granules during blood clotting induced by recalcification. The same is true for platelets in clotting whole blood. The granules persisted if thrombin was used as clotting agent. In this case the granules faded after blue light stimulation. Granules formed by nile-blue sulfate in platelets under identical conditions exhibited the same behaviour as granules of the above mentioned fluorescent stains. Acridine orange in concentrations above 1:15,000 strongly inhibited the conversion of fibrinogen to fibrin but didn't diminish the activity of platelet factor 3 in platelets.     Notes: Zusammenfassung Mit Acridinorange (AO) oder Auramin gefärbte, in EDTA- oder Citratplasma suspendierte Blutplättchen verloren ihre Fluorochromgranula im Verlauf von Blutgerinnungsvorgängen, die durch Recalcifizierung ausgelöst worden waren. Entsprechendes galt für Thrombocyten in spontan gerinnendem Vollblut. Unter Thrombinzugabe blieben die Farbstoffgranula erhalten, erwiesen sich aber dann gegenüber Blaulichteinwirkung labil. Die bei Hellfeldbetrachtung sichtbaren Nilblausulfat-Granula in Blutplättchen verhielten sich unter den erwähnten Bedingungen wie die Fluorochromgranula. AO störte bei gerinnungsanalytischen Untersuchungen in Konzentrationen über 1:15000 die Fibrinogen/Fibrin-Umwandlung, beeinträchtigte aber in Form der Speichergranula in den Plättchen nicht die Thrombocytenfaktor-3-Aktivität.

    Notes: Abstract Seven thermophilic anaerobic bacteria which ferment xylan were isolated from natural geothermal features in the western United States. Typically, these strains were Gram-negative non-sporeforming rods with an unusual double-layered cell wall which resembled that observed in Thermobacteroides acetoethylicus. The strains differed from known thermophilic anaerobes in their ability to utilize a very wide variety of carbohydrates and in their ability to grow in a chemically-defined medium and/or at pH 3.5. Four of the strains contained cryptic plasmids of 1.2 or 1.5×106 daltons. The taxonomic characteristics of the strains are discussed in terms of their relatedness to those of Thermoanaerobium, Thermoanaerobacter, and Thermobacteroides species.
