    Notes: The effect of a Ti seed layer on the magnetic properties and microstructure of Co77Cr19Ta4 was studied in Co77Cr19Ta4/Ti/M (M=Co91Zr3Nb6, Fe, Co) double-layered perpendicular recording media. A thin Ti film of ∼5 nm enhanced the c-axis alignment perpendicular to the film plane, resulting in a large perpendicular anisotropy Ku and the best magnetic properties under the present experimental conditions. The coercivity at this Ti thickness (3–3.7 kOe) was approximately half of the magnetocrystalline anisotropy field Hk estimated from the Ku value, even with the present thick 50 nm CoCrTa. The recording performance using a single-pole-type writing head showed that media with a 5 nm Ti layer had higher recording sensitivity compared to media without Ti layers, and a higher SN value at densities greater than 200 kFRPI. No significant loss of recording resolution due to the nonmagnetic Ti space between the CoCrTa and the back layers was observed by the use of the 5 nm Ti film. Moreover, the results for media with Fe back layers indicated that a thinner back layer with a high flux density can be used without any reduction of recording sensitivity and resolution. © 2000 American Institute of Physics.

    Notes: Simulations of perpendicular recording media were carried out to determine the effect of switching speed limitations upon recording performance. Simulations of hysteresis loops at various applied field sweep rates show that the onset of a switching speed limited increase in coercivity occurs for sweep rates in excess of 2×1013 Oe/s. Switching speeds of individual grains were found to be around 18 to 38 ps, depending on the medium thickness and the magnetization state of surrounding grains. Recording simulations show that for media thicknesses of up to 180 Å, writing frequencies of 5 Gbit/s are feasible before loss of output occurs. © 2000 American Institute of Physics.

    Notes: We present a lithographically made Al single-electron transistor that shows gate modulation at room temperature. The temperature dependence of the modulation agrees with the orthodox theory, however, energy-level quantization in a tiny metallic island affects the device characteristics below 30 K. The charge-equivalent noise of the device at 300 K was measured to be ∼4×10−2 e/Hz1/2 at 1 Hz and is expected to be 1000 times lower in the white-noise regime at higher frequencies. © 2000 American Institute of Physics.

    Notes: Polymerization of C60 clusters epitaxially grown on Si(111)-(7×7) substrates was found to be induced by electron injection from the probe tips of scanning tunneling microscopes (STM) as the sample bias was increased from +4.0 to +5.5 V, exhibiting an evolution behavior characterized by an incubation, a linear growth, and a saturation. The incubation time and the growth rate are dependent greatly on the sample site, which is explained by a model taking into account the pre-existing stress as the driving force of the polymerization and the internal stress built up as a consequence of polymerization producing a stress for backward reactions. © 2000 American Institute of Physics.

    Notes: The purpose of the present study was to elucidate the effects of unilateral masseter muscle pain on the jaw-jerk reflex. The latency and peak-to-peak amplitude of bilateral electromyographic activity recorded at the masseter muscles during the jaw-jerk reflex were measured in 18 patients with craniomandibular dysfunction (CMD) with strictly unilateral masseter pain or tenderness and 10 control subjects using a computerized recording and analysis system. The reflex was elicited, at the mandibular rest position, by tapping the centre of the chin downwards with a reflex hammer incorporating a microswitch that triggered the sweep of the recording apparatus upon contact with the chin. In the CMD group, the jaw-jerk latency on the affected side (6·89±0·98 ms) was significantly shorter (P〈0·01) than that on the unaffected side (7·59±0·92 ms). In the control group, there was no difference between the jaw-jerk latencies on the right (7·06±0·64 ms) and the left (7·08±0·65 ms) sides. The range of side asymmetry for jaw-jerk latencies in the CMD group was greater than that in the control group. In six patients, the latency difference exceeded 1 ms. The asymmetry of latency of the jaw-jerk reflex was thought to be due to facilitation on the side with masseter pain or tenderness. This facilitation on the ipsilateral side might be produced by enhanced gamma drive induced by sustained nociceptive stimulation. Such effects may be related with clinically derived concepts regarding such muscle dysfunction as myospastic activity or trigger points.

    Notes: Summary  We report 2 cases with haemorrhagic complications following percutaneous transluminal angioplasty (PTA) for carotid stenosis. Computed tomography (CT) scanning of these cases demonstrated diffuse subarachnoid haemorrhage in 1 case, and intracerebral haemorrhage in the other case on the next day after PTA. In the latter case, we measured cerebral blood flow velocity and mean transit time with transcranial doppler (TCD) and dynamic CT scan, which demonstrated remarkable increases in the blood flow velocity and peak height, respectively. From these results, postoperative hyperperfusion was suggested to have caused haemorrhagic complications.

To investigate whether common variants in the human genetic background are associated with pathogenesis of ischemic heart diseases, we systematically surveyed 41 possible candidate genes for single-nucleotide polymorphisms (SNPs) by directly sequencing 96 independent alleles at each locus, derived from 48 unrelated Japanese patients with myocardial infarction, including 25.8 kb 5' flanking regions, 56.8 kb exonic and 35.4 kb intronic sequences, and 1.8 kb 3' flanking regions. In this genomic DNA of nearly 120 kb, we identified 187 SNPs: 55 in 5' flanking regions, seven in 5' untranslated regions (UTRs), 52 in coding elements, 64 in introns, eight in 3' UTRs, and one in a 3' flanking region. Among the 52 coding SNPs, 26 were non-synonymous changes. Allelic frequencies of some of the polymorphisms were significantly different from those reported in European populations. For example, the Q506R substitution in the coagulation factor V gene, the so-called "Leiden mutation", has a reported frequency of 2.3% in Europeans, but we detected the Leiden mutation in none of the Japanese genomes that we investigated. The allelic frequencies of the –33A〉G SNP in the thrombomodulin gene were also very different; this allele occurred at a 12% frequency in the Japanese patients that we examined, although it had been detected in none of 82 Caucasians reported previously. These data support the hypothesis that some SNPs are specific to particular ethnic groups.

    Notes: Abstract. An inv(11)(q13.5;q25) inversion was previously identified in a 9-month-old male patient with complex cyanotic heart defects, altered lung lobation, symmetric liver, and abnormally lobulated spleen (polysplenia). This chromosomal rearrangement was inherited from the phenotypically normal father. We termed these regions DHTX-A (disrupted in heterotaxy) –A at 11q13.5 and DHTX-B at 11q25. Here, we report the isolation and characterization of the inversion breakpoints and the gene that is disrupted by the DHTX-A breakpoint. The putative DHTX is identical to the UVRAG gene, which was originally identified as a gene that complements the UV sensitivity of xeroderma pigmentosum complementation group C. The 4-kb mRNA was found to be encoded by a large gene, at least 300 kb long, composed of 15 exons. The function of the gene product remains largely unknown. However, the near central portion of the UVRAG protein is predicted to contain a coiled-coil domain, which has been implicated in mediating protein-protein interactions. Southern analyses and fluorescence in situ hybridization (FISH) revealed that the DHTX-A breakpoint in the patient and his father lies within the intron between exons 6 and 7 of UVRAG. Northern blot analysis indicated strong expression in human fetal and adult tissues and in mouse embryonic day-7 and adult tissues, respectively. Whole mount in situ hybridization also showed that the Uvrag gene is expressed in the presomite-stage embryo. Several hypotheses are discussed to explain the relationship between the chromosomal inversion and the accompanying phenotypes.

    Notes: Abstract. Single-nucleotide polymorphisms (SNPs) can make an important contribution to our understanding of genetic backgrounds that may influence medical conditions and ethnic diversity. We undertook a systematic survey of genomic DNA for SNPs located not only in coding sequences but also in non-coding regions (e.g., introns and 5' flanking regions) of selected genes. Using DNA samples from 48 Japanese patients with rheumatoid arthritis (RA) as templates, we surveyed 41 genes that represent candidates for RA, screening a total of 104 kb of DNA (30 kb of coding sequences and 74 kb of non-coding DNA). Within this 104 kb of genomic sequences we identified 163 polymorphisms (1 per 638 bases on average), of which 142 were single-nucleotide substitutions and the remainder, insertions or deletions. Of the coding SNPs, 52% were non-synonymous substitutions, and non-conservative amino acid changes were observed in a quarter of those. Sixty-nine polymorphisms showed high frequencies for minor alleles (more than 15%) and 20 revealed low frequencies (〈5%). Our results indicated a greater average distance between SNPs than others have reported, but this disparity may reflect the type of genes surveyed and/or the relative ethnic homogeneity of our test population.

    Notes: Abstract A stilbene synthase gene was isolated from three Vitis spp. (V. vinifera, V. labrusca and V. riparia). These genes were placed under the control of the cauliflower mosaic virus 35S promoter and introduced into kiwifruit (Actinidia deliciosa) plants by Agrobacterium-mediated gene transfer. The introduced gene(s) were expressed and piceid (resveratrol-glucoside) rather than resveratrol was produced in the leaves of the transformants. Resveratrol produced by the action of the integrated gene(s) seems to be metabolized into piceid by an endogenous glycosyltransferase. Among the transformants obtained, the highest piceid content in the young leaves was 182 μg/g fresh weight. Although these transformants did not show resistance against Botrytis cinerea, which causes gray-mould disease, the fruits may have some beneficial effects on human health.
