    Description / Table of Contents: Summary Rats bearing metastases of theGuérin-tumour were submitted to a single whole-body irradiation with 1000 R from60Co, followed by an intravenous injection of isologous bone-marrow suspension. In the tumour-bearing animals, prior to treatment, there were anaemia, leucocytosis and thrombopenia proportional to the extent of the tumour growth. After irradiation and transfusion of marrow cells, anaemia and thrombocytopenia became more severe, probably due to bleeding from the necrotic tumour tissue. The increase leucopoesis in the tumour bearing animals was radio-resistant; the white cells in the blood fell to a slighter degree and for a shorter time than in tumour-free animals given the same treatment. No differences were noted between fresh bone-marrow and bone-marrow preserved at 70° in dimethylsulfoxide. The lethal result of a haematological radiation syndrome in the tumour-bearing animals was prevented by transfusion of bone-marrow. No definite effect on the course of the tumour growth could be observed from a single large dose of whole-body irradiation.
    Notes: Zusammenfassung Ratten mit metastasiertemGuérin-Tumor wurden einer einzeitigen60Co-Ganzkörperbestrahlung mit 1000 R unterworfen und anschließend mit isologen Knochenmarkzellsuspensionen intravenös behandelt. Bei den tumortragenden Tieren bestand vor der Bestrahlung eine Anämie, Leukozytose und Thrombozytopenie, deren Ausmaß Beziehungen zum Ausbreitungsgrad des Tumors zeigte. Nach Bestrahlung und Knochenmarktransfusion verstärkten sich die Anämie und Thrombozytopenie, was vorwiegend mit Blutungen in die nekrotisch zerfallenden Tumoren erklärt werden kann. Die gesteigerte Leukopoese der Tumortiere erwies sich als strahlenresistent. Die Leukozytenzahlen im Blut fielen für eine kürzere Zeit und nicht so stark ab wie bei gleichartig behandelten tumorfreien Kontrollen. Wirkungsunterschiede zwischen frischem und bei −70° in Dimethylsulfoxyd konserviertem Knochenmark fanden sich nicht. Die letalen Folgen eines hämatologischen Strahlensyndroms wurden bei den tumortragenden Tieren durch die Knochenmarktransfusion verhütet. Der Verlauf der Tumorprogression ließ sich jedoch durch die einzeitige hochdosierte Ganzkörperbestrahlung nicht sicher beeinflussen.

    Description / Table of Contents: Summary Relationships between blood lipids and clotting changes following phosphatide and triglyceride infusions. In an attempt to explain the clotting changes occurring after infusions of fat, we have examined both the action of pure phosphatide emulgator and complete fat emulsion, as well as the existence of correlations between clotting values and serum lipid fractions. Accordingly, the following clotting reactions proved to be mostly triglyceride-dependent: 1. an approximately 10% reduction of platelets, as compared to fasting values, 2 hours after the termination of the infusion, 2. a reduction of the stypven time for 4 hours following the end of infusion, 3. an elevation of the activity of the thrombin inhibitor for up to 2 hours post infusion. The antithrombin-III activity becomes reduced only after the administration of phosphatide. In addition to triglycerides, phosphatides are also involved in this platelet reduction, as was shown by a significant, slight fall in thrombocytes following infusion of phosphatide. A significant, slightly negative correlation between total fatty acid increase and reduction of stypven time indicates a similar behavior between the stypven-sensible cephalins and hyperlipemia. If the phosphatide emulgator is freed from cephalins extensibly, then the stypven time becomes significantly less reduced than in the case of cephalin-containing emulsions. Significant negative correlations between fasting values of thrombin inhibitor and total esterified fatty acids, as well as at 2 and 4 hours following infusion, point to the significance of endogenous heparin antithrombin for the outflow of neutral fats from the blood.
    Notes: Zusammenfassung Um die nach Fettinfusionen auftretenden Blutgerinnungsveränderungen zu deuten, haben wir vergleichend die Wirkungen des reinen Phosphatidemulgators und der kompletten Fettemulsion geprüft sowie Korrelationen zwischen Gerinnungswerten und Serumlipidfraktionen untersucht. Dabei ließen sich als vorwiegend triglyceridabhängig die folgenden Gerinnungsreaktionen nachweisen: 1. ein Plättchenabfall 2 Std nach dem Ende der Infusionen um etwa 10% gegenüber den Nüchternwerten, 2. eine über 4 Std nach Infusionsende andauernde Verkürzung der Stypvenzeit, 3. eine Aktivitätssteigerung des Thrombininhibitors bis 2 Std nach beendeter Infusion. Nur nach Phosphatidzufuhr verminderte sich die Antithrombin III-Aktivität. An der Plättchenverminderung sind neben Triglyceriden auch Phosphatide beteiligt, wie sich aus einem signifikanten, geringen Thrombocytenabfall nach Phosphatidinfusion ergab. Eine signifikante, schwach negative Korrelation zwischen Gesamtesterfettsäurenanstieg und Stypvenzeitverkürzung spricht für ein gleichsinniges Verhalten der stypvensensiblen Kephaline und der Hyperlipämie. Wenn der Phosphatidemulgator von Kephalinen weitgehend gereinigt wurde, so verkürzte sich die Stypvenzeit signifikant geringer als nach kephalinhaltigen Emulsionen. Signifikante, negative Korrelationen zwischen Thrombininhibitor und Gesamtesterfettsäuren bei den Nüchternwerten sowie 2 und 4 Std nach Infusionsende weisen auf die Bedeutung des endogenen Heparinantithrombins für den Neutralfettabstrom aus dem Blut hin.
