    Notes: Sorbitol dehydrogenase was bound to the surface of acyl-azide-activated collagen membranes and its kinetics was investigated as a model of two-substrate or cofactor-requiring enzyme reactions. The study was performed with the “rotating membrane reactor” especially designed to obtain a precise variation of the external mass-transfer coefficient, and thus the direct visualization of diffusional effects on the bound enzyme behavior. Diffusional limitations for NADH were found to decrease the apparent affinity for NADH, but to increase the apparent affinity for fructose. Such opposite effects of diffusional limitations on apparent affinities are generally applicable to reactions involving two substrates or a substrate and a cofactor of widely different affinities.

    Notes: A model that continuously predicts the concentration of microorganisms in complex medium fermentations is suggested. The model uses carbon dioxide evolution as its primary input and assumes that respiration activity can be differentiated into growth-related and maintenance-related functions. This model can be programmed on computer-coupled vessels and used to standardize on a physiological fermentation inoculum transfer time. The cell concentration estimate can also be used to calculate specific growth rate and can be combined with additional monitored information to calculate other important fermentation parameters such as specific oxygen uptake.

    Notes: Conditions for the laboratory-scale production of acetoin plus diacetyl by Enterobacter Cloacae ATCC 27613 were studied. Thirty-five g acetoin plus diacetyl/50 g sucrose were obtained when fermentation was carried out in 2. 5 liter medium containing 12.5 g peptone and 12. 5 g yeast extract, at pH 7.0, in a 5 liter conical flask on a shaker (240rpm) at 28-30°C for 48 hr. Recovery of pure diacetyl was 85% of the total plus diacetyl.

    Notes: Chitosan samples manufactured under different conditions were compared for effectiveness of coagulating an activated sludge suspension grown on vegetable canning wastes. Computer analysis of data from Buchner funnel filterability tests resulted in quadratic polynomial equations describing the response curves for volume of filtrate versus dosage, expressed as g/liter chitosan/100 g sludge suspended solids (SSS). The quotient of the filtrate volume and dosage at the inflection points of the equations obtained for 10 test samples and 1 commercial chitosan sample were compared to evaluate the response (effectiveness) per unit amount for each chitosan product. The product made by a standard procedure (deproteinated with 3% NaOH at 100°C for 1 hr, demineralized with 1N HCL at ambient temperature for 30 min, and deacetylated with 50% NaOH at 145-150°C under N2 for 5 or 15 min) gave the best performance as a coagulating agent for this activated sludge system. Other products, including the commercial preparation, required higher dosages to achieve the same effectiveness. Products deacetylated in the presence of sir rather than nitrogen decreased waste treatment effectiveness, which approximated the trends of reduced viscosity and molecular-weight distribution. The products containing minerals were less effective than products from which minerals had been removed prior to deacetylation, but they were more effective than the enzyme treated sample and the commercial product. In general, although chitosan products obtained after 15 min deacetylation were more effective than those receiving 5 min deacetylation, effectiveness did not correlate linearly with viscosity and molecular-weight distribution trends. However, chitosan products deacetylated for 15 min did show that the higher-molecular-weight products (0.65-1.1 × 106) were more effective coagulating agents for activated sludge than the manufactured product having the lowest molecular weight (0.47 × 106) and the commercial reference sample (0.56 × 106). Thus, higher values for molecular weight were predictive of greater effectiveness for coagulation of activated sludge suspensions.

    Notes: A novel fermentation device, the rotorfermentor, is described and some experimental results are presented on power requirements and oxygen mass transfer characteristics of the rotorfermentor. This fermentation device is designed to achieve high cell concentrations in batch and continuous cultures. Basically, the rotorfermentor consists of a rotating microporous membrane which is enclosed within a stationary fermentor vessel. The metabolic products in the broth are continuously removed by filtration through the rotating microporous membrane while the growing cells can be retained inside the fermentor. This dual function of cell growth and concentration with the simultaneous removal of metabolic products is the essential characteristic of the rotorfermentor.

    Notes: A two-parameter theoretical model is developed to evaluate the effect of immobilized enzyme deactivation on substrate conversion in fixed- and fluid-bed reactors under diffusion-free conditions. The method describes a simple reaction in which three different immobilized enzyme deactivation forms are considered, and an expression is developed to evaluate the effect of immobilized enzyme deactivation on yield in a consecutive reaction. Comparison of reactor performances for the two reactor types reduces to a comparison of the appropriate dimensionless parameters. The practical implications of the development are illustrated through an example.

    Notes: Porcine trypsin (EC 3.4.4.4) converted, within approximately 2 hr at 50°C, its 1000-fold weight of water-insoluble, heat-denaturated cheese whey protein into a water-soluble product. In the course of this digestion, the enzyme increased the α-amino nitrogen of the protein by a factor of 〉20, from 0.40 to 9.40%. After digesting the water-insoluble whey protein, fully active trypsin could be recovered from the soluble digest with the aid of a cellulose-based affinity adsorbent. The enzyme which was eluted from a column of p-aminobenzamidine, bound to succinylated aminododecylcellulose, was fully active and showed essentially unchanged kinetic properties with a synthetic substrate, L-benzoyl-arginine p-nitroanilide. It was possible to perform, with the same amount of trypsin, three subsequent and equally effective solubilizations of whey protein, followed by a fourth digestion which still yielded a soluble product, but was considerably slower and incomplete. During each digestion, an estimated 30% of the trypsin was lost. The was not due to a decreased efficiency of the affinity adsorbent, as its trypsin-binding capacity was essentially unaffected after over 10 cycles of use.

    Notes: A mathematical model has been employed to examine the interplay of reaction and mass transfer in immobilized enzyme systems involving reaction-generated enzyme poisons. Deactivation rates can be significantly reduced in some cases by catalyzing a purification reaction in which the poison is transformed into an innocuous substance. This conclusion is illustrated experimentally for reaction-generated H2O2 in a continuous-flow stirred slurry reactor containing glucose oxidase immobilized on activated carbon.

    Notes: Two analytically solved mathematical models are presented for a reactor ystem employing immobilized whole cells as a biocatalyst. The whole cells are entrapped or pumped through the shell side of the dialyzer reactor unit. The reactant mixture is circulated through the cialyzer tube side. Nutrient diffuses across the hollow fiber membrane from the tube side to the shell side, where it reacts to form product, which then back diffuses into the reactant mixture stream. The use of a high recirculation ratio of nutrient through the dialyzer tubes to nutrient feed rate to the entire system, allows the system to be modeled as a continuous-flow stirred-tank reactor. The first analysis details the development of an effectiveness-factor correlation for first- and zero-order kinetics. The second analysis presents the solution to an unsteady-state-system mass balance with Michaelis-menten kinetics.

    Notes: The stability of the model of a completely mixed activated-sludge process holding the recycle sludge concentration, XR, as a system constant subjected to pH, temperature, potassium cyanide, and phenol shock loading was investigated. Soft-drink bottling wastewater was used and maintained at 1000 mg/liter chemical oxygen demand (COD). The hydraulic ratio and recycle sludge concentration were maintained at 0.3 and 7000 mg/liter, respectively. An initial dilution rate of ¼ hr-1 was maintained for pH and temperature shock loading, with ¼ and ⅛ hr-1 for KCN shock loading and ¼, ⅛, and \documentclass{article}\pagestyle{empty}\begin{document}$\frac{1}{16}$\end{document} hr-1 for phenol shock loading. It was found that the present system could handle pH shock loading as low as 4.0 and as high as 10.4 without any serious disruption of biological solid concentration and filtrate COD. At pH 4.0 shock loading, filamentous organisms were predominant. Temperature shock loading could be handled from 23 to 36°C without any leakage of effluent filtrate COD. At 46°C temperature shock, a 14 hr period was required to recuperate to the new steady state and provided only 85% of COD removal efficiency. For KCN (50 mg/liger) and phenol (85 mg/liter) shock loading, the dilution rates should be lower than \documentclass{article}\pagestyle{empty}\begin{document}$\frac{1}{16}$\end{document} hr-1 in order to shorten the transient period and improve the effluent quality. Biological kinetic constants included cell yield value, maximum growth rate, and the saturation constant, which was varied with the qualitative shock applied.

    Notes: Phanerochaete chrysosporium was grown in fermentors on NaOH-extracted maple, pine, and cedar barks at the optimum substrate concentration of 1% (w/v). The yields (mg protein/liter) on maple, pine, and cedar were 1500, 1200, and 880, respectively, which are probably due to the different lignin contents of the barks. Lignin is not utilized. The productivities at 30°C obtained for pine (4.07 × 10-2 g protein/liter hr) and cedar (2.63 × 10-2 g protein/liter hr) barks were greater than for maple (2.63 × 10-2 g protein/liter hr). The substrate (bark) was the limiting component of the fermentation. Over the 26-38°C temperature range protein productivity increased by a factor of three (1.55 × 10-2 vs. 4.61 × 10-2 g protein/liter hr) for maple bark. Low agitation rates resulted in an overproduction of cellulase and reduced levels of microbial protein.

    Notes: The effect of oxygen supply on L-leucine fermentation was investigated employing a leucine-producing mutant of Brevibacterium lactofermentum. Since it was not possible to measure oxygen tension below 0.01 atm by a Teflon-coated oxygen electrode, the degree of satisfaction of the cells' oxygen demand (cells' respiration rate/maximum oxygen demand of cells, rab/KrM) and the redox potential of the culture medium (E. mV) were used as indices to oxygen supply in cultures under low oxygen tension. When the oxygen demand of the cells was satisfied (rab/KrM = 1.0) and the E value was between -90 and -110 mV, L-leucine formation was 26.5 mg-ml. When the oxygen demand of the cells was not satisfied (rab/KrM = 0.85) and the E value was between -200 and -220mV, L-leucine accumulation was 29.7 mg/ml. When the oxygen supply was extremely limited (rab/KrM = 0.27) and the E value was -280 mV. L-leucine formation was 12.9 mg/ml. A new method which simultaneously measures the redox potential and dissolved oxygen was applied to the determination of the critical dissolved oxygen level for cell respiration (PL crit), which was too small to be detected by conventional oxygen electrodes. The value of PL crit of the leucine producer was estimated as 0.0002 atm.

    Notes: Methanol oxidase produced by the yeast Hansenula polymorpha DL-1 was used for the enzymatic oxidation of methanol to formaldehyde. The kinetics of enzyme and protein release during cell desruption were studied at the laboratory scale with a Braun homogenizer and the pilot plant scale with a Manton-Gaulin homogenizer. Conditions were defined for maximum release and retention of high activity in cell-free extracts. Methanol oxidase was immobilized by adsorption on DEAE-cellulose from enzymes in cell-free extracts or from ammonium sulfate purified purified fractions. The kinetics of formaldehyde formation with both soluble and immobilized enzyme was studied in batch and continuous reactors.

    Notes: Immobilized soybean β-amylase was prepared by using porous cellulose beads. The expressed activity of the β-amylase-cellulose beads conjugated below 35 mesh was 59-69% of the initial activity and the protein content was 10-13%. General properties of the conjugate were almost identical with those of the native enzyme except for the Km value. The Km value of the conjugate was 40mM and the Km value of the native enzyme was 0.6mM. This large difference was probably caused by pore structure, i.e., a pore diffusion problem. The film diffusion problem occurred at the flow rate below a linear velocity of 3 cm/min. Maximum maltose contents of the hydrolyzates prepared by the conjugate and the native enzyme were 69 and 71%, respectively. After a continuous column operation at 50°C for 17 days, the activity of the column was 60% of the activity. The half-life of the column at 40°C was 40 days.

    Notes: This article presents a review of the enzyme mechanisms involved in degradation of cellulose by the white-rot fungus Sporotrichum poulverulentum. The hydrolytic enzymes involved include: (1) five endo-1,4-β-glucanases; (2) one exo-1,4-β-glucanase, and (3) one or several 1,4-β-glucosidases. A recently discovered oxidative enzyme of importance in in vitro cellulose degradation seems to be a cellobiose oxidase. An oxidoreductase, cellobiose:quinone oxidoreductase, is of importance both in cellulose and in lignin degradation. Regulatory mechanisms of the extracellular enzyme activities, such as monosugar levels causing catabolite repression of the endoglucanases, have also been investigated. The enzymes used by S. pulverulentum in cellulose hydrolysis are compared to those used by Trichoderma viride. Very similar types of enzymes are used in both cases. However, no oxidative enzyme has so far been found to be involved in extracellular cellulose degradation in the case of T. viride. Recommendations for further research are given.

    Notes: Graphites of different manufacture and origin exert positive but different accelerations on the rate of oxidation of NADH to NAD+ in aqueous solution; different proportions of the oxidized form of NAD retain enzymatic activity depending on the nature of the graphite. Oxidative pretreatment of the graphite surfaces increases the rates of NADH oxidation, but subsequent silanization of the surfaces to attach alkylamine groups causes the rates to decrease. The experimental results suggest the presence of at least two types of sites on graphite surfaces: One very reactive site which produces a high percentage of an enzymatically inactive reaction product of NADH and is itself deactivated during the course of reaction, and another type of site which promotes the oxidation of NADH to enzymatically active NAD+ in high yields.

    Notes: Formaldehyde, an intermediate in methanol oxidation, plays an important role in controlling methanol utilization by microorganisms which fix carbon via the ribulose monophosphate (RMP) cycle. The extracellular formaldehyde concentration profiles in batch cultures at low and high starting methanol concentrations with and without the presence of semicarbazide in the media, suggest the role played by formaldehyde. The impact of formaldehyde on growth is demonstrated by the time dependent and the initial methanol concentration-dependent cell-mass-yield coefficient. Kinetic studies of the enzymes involved in the oxidation and incorporation of C1 units suggest that enzyme inhibition cannot account for the observed growth inhibition. A mechanism is proposed to explain methanol and formaldehyde utilization that stresses the transcriptional regulation of mass flow around formaldehyde in assimilation and oxidation pathways through repression and induction.

    Notes: A penicillin sensitive enzyme electrode has been used to analyze the concentration of benzylpenicillin in fermentation broth. The electrode response time was in the region of 2 min and the response to penicillin concentration was linear within the range of 1 to 10mM. The buffering capacity of the medium influenced the sensitivity of the electrode. At low buffer capacity the sensitivity of the enzyme electrode to penicillin was very high, but then the sensitivity to small changes in buffer capacity was relatively large. At high buffer capacity the sensitivity to penicillin was reduced and the electrode became less dependent on changes to buffer capacity. Constant calibration curves were repeatedly obtained with the electrode when used for 2 hr daily in a fermentation medium over a six day period. Three methods devised to calibrate the electrode for use in fermentation media were investigated. Methods one and two, based on the relationship between electrode sensitivity and buffer capacity in phosphate buffer and in sterile media, gave rather high penicillin concentration values. The third method based on an internal standard was the most satisfactory.

    Notes: A new sensor for the continuous determination of ethanol during the production of yeasts, growing aerobically on fermentable sugars, is described. The operating principle is based on the detection of ethanol vapor in the exit air. A description of the sensor with a diagram of the measuring electronics is supplied. It was designed for the determination of ethanol contents up to 3 g/liter in aqueous solutions at 30°C. The sensitivity is very high  -  1 ppm ethanol being detectable under these conditions. The aeration rate dose not affect the output signal in a wide range (0.5-2 v/v/m). Besides the unspecific sensitivity of the sensor to other easily oxidizable substances, the influence of the pO2 in the exit air, however, must be taken into account. The application of the sensor is shown in a fed-batch culture of bakers' yeast where aerobic fermentation is caused by increasing the glucose feed rate.

    Notes: Hydrolysis of cellulose by Trichoderma viride cellulase reached a plateau after some 25 hr. If the initial enzyme-to-substrate ratio was low, resuspension of substrate in fresh enzyme or addition of enzyme resulted in further high rate hydrolysis. This did not occur if the initial ratio was high. Over 75% hydrolysis might be achieved in the former case, while less than 60% in the latter. A model postulating inactivation of adsorbed enzyme-substrate complex which blocked further hydrolysis was proposed, and it was found to fit the data well. The proposed model had five parameters, four of which could be checked by graphical methods, and all of which had physical meanings. The parameters were estimated by a nonlinear least-squares minimization FORTRAN computer program, using numerical integration and optimization of the parameters. The model was used to predict the resuspension data, powdered enzyme addition data, cellobiose addition data, and cellulose addition data; the deviations from the model are discussed. It was found that average values could be used for four out of the five parameters, while the fifth (initial enzyme concentration) did not correlate with independent measurements such as the filter paper activity or protein concentration.

    Notes: The effect of waste characteristics on the removal of suspended solids and soluble phosphorus by alum, Al2(SO4)3·18H2O, was investigated in an activated sludge system with no cellular recycle. Under the conditions presently studied, it was found that sludge organisms grown in nitrogen- and phosphorus-restricted media possess exceptionally large capsules and produce a higher surface electric charge per unit of fry weight. Chemical demand for separation of these capsulated cells is obviously higher than those solids cultivated under a well-balanced nutrient condition. For phosphorus removal, the molar ratio of Al+3:P is a function of the initial concentrations of soluble phosphorus and suspended solids in solution. The relationship between percent phosphorus removal and the quantity of chemical coagulent used shows nonstoichiometric relationship; that is, the required molar ratio of Al+3:P is always greater than one and directly depends upon the content of suspended solids in the flocculated media. Moreover, the present study indicated that chemical separation of dispersed microorganisms occurred following phosphorus removal.

    Notes: The kinetics of microbial growth and product formation are described as applied to the high cell concentration scheme of the rotorfermentor. A bench scale pilot plant was designed and built in order to demonstrate the operational feasibility of the rotorfermentor. The fermentation of glucose to ethanol by Saccharomyces cerevisiae ATCC 4126 was used. When the rotorfermentor was used with a glucose feed concentration of 104 g/liter almost 100% glucose utilization was obtained and the ethanol productivity rate was 27.3 g ethanol/liter hr which was found to be about 10 times greater than the ethanol productivity obtained from an ordinary continuous stirred tank (CST) Fermentor. The ethanol experimental results obtained from the rotorfermentor and an ordinary CST fermentor were used as a basis to assess the economic feasibility of the rotorfermentor. The economics of an industrial scale ordinary CST fermentor with and without cell recycle is compared with a rotorfermentor unit for the same ethanol production throughput. For the process conditions considered on this case, calculations showed that the rotorfermentor may replace both a CST fermentor and cell centrifuge resulting in lower capital equipment costs and lower power consumption requirements.

    Notes: The maintenance coefficient, ms (mmol substrate/g cell dry wt hr), of two distinct groups of C1-utilizing bacteria has been determined by growing the organisms in an aerobic continuous culture limited by different C1 growth substrates. For growth on methanol, ms = 2.5 ± 0.3 for Pseudomonas C; 3.9 ± 0.7 for Ps. methylotropha (these bacteria utilize methanol via the ribulose monophosphate pathway of formaldehyde fixation) 1.5 ± 0.2 for Pseudomonas 1, and 2.3 ± 0.4 for Pseudomonas 135 (the latter bacteria utilize C1-compounds via the serine pathway). For growth on formaldehyde, ms = 1.5 ± 0.3 for Pseudomonas 1 and 2.7 ± 0.7 for Pseudomonas 135, whereas on formate the values for ms are 1.0 ± 0.2 and 4.4 ± 1.3; respectively. Although the maintenance coefficients did not differ systematically between the two groups of bacteria, the maintenance requirements per generation of the serine pathway bacteria were considerably higher (8.7 vs. 3.9) owing to their slower growth rate. The maximum molar yield values, YMmax (g cell dry wt/mol substrate utilized), corrected for the maintenance energy of bacteria which utilize C1-compounds via the ribulose monophosphate pathway averged 10.1 when grown on methanol, while the values for bacteria which use the serine pathway averaged 13.5. On formaldehyde an average value of 11.5 is obtained and on formate the average value was 7.4 in the serine pathway bacteria.

    Notes: Material and energy balances for fermentation processes with product formation are developed using regularities which are frequently useful in analyzing experimental results. Available electron balances are presented and used to analyze the results of experiments in which biomass and lysine are produced in batch culture using molasses and corn extract as organic substrates. Measured values of biomass productivity, lysine productivity, O2 consumption rate, heat evolution rate, CO2 evolution rate, and rate of utilization of reducing sugars (RS) are used to obtain estimates of biomass energetic yield, lysine energetic yield, and fraction of substrate energy evolved as heat. For two of the three experiments, based on the entire fermentation, the measured mass rate variables are very consistent and energy measurement errors are less than 10% when proper consideration is given to both the RS and the amino acids present in the organic substrate. The average measured value of 26.5 kcal/equivalent (equiv) of available electrons transferred to oxygen compares favorably with the known average value of 27 kcal/equiv.

    Notes: Modeling of microbial growth using nonmiscible substrate is studied when kinetics of substrate dissolution is rate limiting. When the substrate concentration is low, the growth rate is described by an analytical relation that can be identified as a Contois relationship. If the substrate concentration is greater than a critical value Scrit, the potentially useful hydrocarbon S* concentration is described by S* = Scrit/(1 + Scrit/S). A relationship was found between Scrit and the biomass concentration X. When X increased, Scrit decreased. The cell growth rate is related to a relation μ = μm[A(X/Scrit)(1 + Scrit/S) + 1]-1. This model describes the evolution of the growth rate when exponential or linear growth occurs, which is related to physico-chemical properties and hydrodynamic fermentation conditions. Experimental data to support the model are presented.

    Notes: The production of single-cell protein (SCP) from ethanol by different imcroorganisms is briefly reviewed. The trends in modeling the SCP production are outlined with a stress on the need to incorporate metabolic and engineering considerations into the pertinent models. Data on batch yeast growth on ethanol are analyzed, a metabolic model involving design parameters is suggested, parameters values are estimated, and their significance is discussed. The model essentially describes the response of a system to disturbances (substrate, metabolites, oxygen, etc.). An effective numerical integration procedure is necessary in the solution of the model which has “stiff” character.

    Notes: By means of stimulus-response experiments with a Li+ tracer, a description of the fluid flow in a 30 m3 reactor, used for anaerobic treatment of wastewater, was derived. It appeared that of the three parts that could be distinguished in the reactor (the sludge bed. The sludge blanket, and the settler), the sludge bed could be described as a combination of two perfectly mixed regions with bypassing and return flows; the sludge blanket was perfectly mixed, while the fluid flow in the settler could be described as plug flow. From experiments performed under different conditions it was concluded that 60 m3 gas/day were sufficient to provide good mixing and also that the height of the sludge bed should be 2-3 m. From this last conclusion it follows, e.g., that scale-up of the 30 m3 reactor should be done by increasing the cross section of the reactor while homogeneously distributing the influent over the bottom.

    Notes: The shake flask is a very common and useful tool for the study of submerged fermentations on a small scale. However, the oxygen supply may easily become a limiting factor. A model for the aeration in shake flasks is presented that enables one to predict whether in the course of an experiment the oxygen supply is becoming a growth-limiting factor or not. The results of measurements of the transfer coefficient of a cotton plug and the oxygen mass-transfer coefficient kla are also given.

    Notes: Work has been carried out on the production of citric acid in batch culture using Aspergillus foetidus. It is shown that the dry weight increases throughout the fermentation. The supply of nitrogen is exhausted early and the subsequent increase in dry weight is due to accumulation of carbon by the cells. It appears that the exhaustion of nitrogen is a necessary prerequisite for the production of citric acid, which is produced by carbon-storing cells. The pH history in the early part of the fermentation did not affect the final yields.

    Notes: Immobilization of an enzyme within an insoluble material permeable to substrate can change the apparent behavior of the enzyme. In particular, external mass transfer and intraparticle diffusion effects can significantly influence the dependence of observed reaction rate on operating parameters such as temperature and pH. This analysis shows that, under very general conditions, the influence of diffusion alone is to diminish the sensitivity of the observed rate to any parameter that is uniform throughout the catalyst particle. However, the optimal value of the parameter is not changed because of intrapellet diffusional effects.

    Notes: Comparison of the equations that describe the relationship between the maximum cell yield coefficient, the maintenance coefficient, and the specific growth rate at steady-state conditions revealed that the equations used for axenic cultures are congruent with those commonly used for mixed-culture system such as activated sludge. A unified basis was proposed. The expression of the yield and maintenance coefficients in carbon units according to the unified basis permitted one to evaluate literature data on both axenic and mixed-culture systems. From this it appears that the maximum cell yield ranges from 0.50-0.80 (mg biomass carbon formed/mg substrate carbon used) for both axenic and mixed systems. However, the maintenance coefficient (mg substrate C/mg biomass C·hr) for the axenic cultures was between 0.010 and 0.100, but for activated sludge communities it was between 0.001 and 0.010. Microorganisms were isolated from sludge communities with these apparently low maintenance requirements and grown axenilly. Their maintenance coefficients but not their maximum yield coefficients decreased with decreasing specific growth rates. The consequences of this finding with regard to species selection in mixed-culture systems and the concept of cellular maintenance requirement are discussed.

    Notes: Fusarium moniliforme was grown on a carob aqueous extract in a chemostat for fungal protein production. The substrate was adjusted to provide 0.5% carob sugars supplemented with inorganic salts. The dilution rate varied from 0.086 to 0.227 hr-1 under constant conditions of temperature (30°C), pH (4.5), and oxygen saturation (60-80%). A yield of 0.709 g dry mycelium/g consumed carob sugar and a productivity value of 0.687 g dry mycelium/liter hr-1 were obtained at μ = 0.205 hr-1. The maintenance coefficient was 0.077 g carob sugar/g dry mycelium hr-1. While the carbohydrate and purine content of dry mycelium increased at μ values from 0.114 to 0.205 hr-1 both true (Lowry) and crude (N × 6.25) protein contents decreased at the same μ range. Maximum values of 36.3% true and 47.9% crude protein of dry mycelium were obtained at μ = 0.114 hr-1, whereas a minimum purine content of 99.8 μmol/g corresponding to 6.42% nucleic acids was recorded at μ = 0.086 hr-1. It was concluded that a continuous fermentation of carob aqueous extract using F. moniliforme should be operated at growth rates of approximately 0.205 hr-1 in order to maximize protein production.

Some factor affecting the extent of succinylation are described, e.g., heating of protein (80°C for 5 min) prior to dervatization reduced maximum succinylation of available ε-amino groups from 88 to 54%. Succinylation of amino groups progressively increased as the concentration of succinic anhydride was increased and there was a concomitant decrease in nucleic acid content of the precipitated protein concentrate from 12.5 to 5.3 for 0 and 88% succinylation, respectively, succinylation enhanced the solubility of the protein concentrates particularly below pH 7 and it decreased the isoelectric point pH. 4.5 to 4.0.

    Notes: Cell recycle and vacuum fermentation processes are described for the continuous production of ethanol. Preliminary process design studies are employed to make an economic comparison of these alternative fermentation schemes with continuous and batch fermentation technologies. Designs are based on a production capacity of 78,000 gal 95% ethanol (EtOH)/day employing molasses as the fermentation substrate. The studies indicate that a 57% reduction in fixed capital investment is realized by continuous rather than batch operation. Further decreases in required capital investment of 68 and 71% over batch fermentation were obtained for cell recycle and vacuum operation, respectively. However, ethanol production costs were dominated by the cost of molasses, representing over 75% of the total manufacturing cost. But, when a reasonable yeast by-product credit was assumed, the net production cost for 95% ethanol was estimated at 82.3 and 80.6 cent/gal, for the cell recycle and vacuum processes, respectively.

    Notes: Solid-state fermentations (78% initial moisture content) of alkali-pretreated Eastern Hard Maple sawdust were conducted in tray and tumble fermentors using chaetomium cellulolyticum. Crude protein content of the solids rose from 0.9 to 11% in the tray fermentor and 8% in the tumble fermentor in 20 days. These levels were almost equal to those achieved in corresponding slurry-state fermentations (1-5% (w/v)) of the same substrate. Specific growth rates were two to four times lower in the solid-state fermentors but this was offset by their greater solids-handling capacity: the rate of protein production per unit volume of fermentation mixture was comparable to that of the 5% (w/v) slurry and two to three times higher than that of the 1% (w/v) slurry.

A transient continuous-culture experiment was also conducted to investigate hydrocarbon-limited growth; the hydrocarbon feed flow rate was stopped for several hours and then resumed at a reduced steady-state flow rate. Interfacial tension, Sauter mean diameter, pseudosolubility, fraction of cells in the aqueous phase, oil-phase volume fraction, and cell concentration were measured to characterize the system. The microorganisms appear to utilize both the submicron drops and the microscopic drops. The effects of interfacial tension, pseudosolubility, and unoccupied interfacial area on the kinetics of hydrocarbon fermentation are discussed here. A conceptual model for hydrocarbon uptake is presented and discussed.

    Notes: Whole cells of Pseudomonas testosteroni, induced to synthesize steroid-transforming enzymes beforehand, have been immobilized by entrapment in polyacrylamide gel. The immobilized cells have then been used to catalyze the continuous Δ1-dehydrogenation of Reichstein's substance S under various conditions in the presence of phenazine methosulfate (PMS), an electron acceptor for the cell-free Δ1-dehydrogenase. The presence of PMS substantially increases the rate of reaction when fed with the steroid substrate to a continuous stirred tank reactor containing the immobilized cells. The operational half-life of the Δ1-dehydrogenase activity of the cells, about 103 hr under the best operating conditions, is essentially unaffected by the presence of PMS. Though the acceleration of the reaction may be due to PMS-mediated passage of electrons from some component in the electron transport chain to molecular oxygen, the lack of a similar effect with methylene blue is consistent with the conclusion that PMS functions directly as the electron acceptor for the Δ1-dehydrogenase.

    Notes: Nitrification and denitrification are important microbiological reactions of nitrogen. In this work, the kinetics of these reactions have been investigated based on a Monod-type expression involving two growth limiting substrates: ammonium nitrogen and dissolved oxygen for nitrification and nitrate nitrogen and dissolved organic carbon for denitrification. The kinetic constants and yield coefficients were evaluated for both these reactions. Past experimental work was used to determine the constants for the nitrification reaction. For the denitrification reaction, experiments were performed in a stirred tank reactor under conditions such that only one substrate was growth limiting. Steady-state values of the substrate concentrations in the reactor were determined at various dilution rates. These data were analyzed to obtain the kinetic and stoichiometric constants. From these constants it was concluded that in the range of nitrate nitrogen concentrations encountered in waste water, the denitrification reaction can be considered a first-order reaction. It was also found that three times as much organic carbon is required as nitrate nitrogen for complete nitrogen removal.

    Notes: The growth behavior of Chaetomium cellulolyticum, a new cellulolytic fungus, has been examined in slurry fermentation systems using various chemically pretreated sawdusts from hardwoods as substrates. Both acid- and alkali-pretreatment methods were used and the fermentation media included the spent pretreatment liquor in an attempt to concurrently maximize substrate utilization and minimize the biological oxygen demand (BOD) level in the process effluent. Diauxic growth patterns were found in the three cases studied, suggesting an initial utilization of soluble hemicellulose sugars followed by utilization of the insoluble cellulose. This behavior patterns was supported by separate growth experiments using the major sugars of hemicellulose as carbon sources. The organism was found to be a good convertor of both cellulose and hemicelluloses into single cell protein (SCP). In terms of rate and extent of protein production in the insoluble biomass product, acid pretreatment appears to be better than alkali pretreatment if the product is intended as ruminant feed.

    Notes: The feasibility of using hollow fiber membrane dialyzers (C-DAK) for immobilization of microbial whole cells was investigated. The cells are located on the shell side of the dialyzer, while substrates and products are free to diffuse across the hollow fiber membranes. The biochemical reaction studied was the conversion of L-histidine to urocanic acid and catalyzed by L-histidine ammonia-lyase. C-DAK dialyzers containing a heat-treated suspension of Pseudomonas fluorescens ATCC 11299b (with L-histidine ammonia-lyase activity) were incorporated into constant volume recycle reactor systems for continuous product formation. A simple model successfully correlated the data and predicted performance. It was found that the reaction was not likely to be diffusion limited, and such a cell immobilization scheme is convenient and workable for continuous production of biochemicals.

    Notes: Several microorganisms having higher nicotinamide adenine dinucleotide kinase (NAD kinase, EC 2.7.1.23) activity were immobilized into polyacrylamide gel lattices. The enzyme activity yield by immobilization was highest in Achromobacter aceris AKU 0120. By the incubation of the immobilized A. aceris cells at pH 4.0, the NAD kinase activity increased and the adenosine triphosphate(ATP)-degradation activity disappeared completely. Enzymic properties of the immobilized A. aceris cells were investigated and compared with those of intact cells. The optimal pH and the optimal temperature of immobilized cells were the same as those of intact cells. Immobilized cell NAD kinase was more stable than that of intact cells. The operational half-life of immobilized cells was 20 days when the substrate solution was passed through a column packed with immobilized cells at a flow rate which gives a space velocity (SV) of 0.1 hr-1 at 37°C. On the other hand, the half-life of the intact cells was only 6 hr.

    Notes: Pullulanase was immobilized successfully by simple, inexpensive methods that may be useful for industrial application of this enzyme. A tannin-pullulanase(TP) complex was obtained by addition of tannic acid to the culture filtrate of thermophilic Streptomyces flavochromogenes. TP could be bound to TEAE-cellulose (TTCP). Immobilization in this manner took place with quantitative retention of activity. The immobilized enzymes were stable for more than six months. The optimum temperatures of the native enzyme and TP were both 50°C; that of TTCP was 45°C. In the presence of 5mM Ca2+, the activity of TTCP was increased approximately twofold and the optimum temperature was raised to 50-60°C. Pullulanase was not significantly eluted from TP or TTCP by NaCl Solution (0.1-0.5M).

    Notes: The effect of soluble thiol reagents on the extractability of protein from yeast cells was studied. The incubation of yeast cells with dithiothreitol, 2-mercaptoethanol, or monothioglycerol markedly stimulated the release of soluble carbohydrates into the medium. There was a concomitant improvement (over twofold) in the extractability of protein from the yeast cells. The thiol reagents activated the proteolytic enzymes of the yeast cells. Unless inactivated, these enzymes hydrolyze the extracted protein.

    Notes: When a steady-state oxygen concentration is measured with a membrane-covered probe in an open system, the oxygen consumption in the unstirred layer gives rise to an error of measurement whose seriousness depends on the kinetics of the oxygen-consuming process. First-order oxygen consumption in the sample causes a proportional reduction in the signal so that the calibration in curve remains linear. A zeroth-order process causes the calibration curve to be offset from the origin, but it remains linear. Oxygen consumption according to the Michaelis-Menten equation causes the calibration curve to become nonlinear with the maximum deviation at the lower end of the scale. The error determines a lower limit for usefulness of membrane-covered probes. Steady-state kinetics at oxygen concentrations in the order of KM cannot be determined with a membrane-covered probe for enzyes with KM for oxygen lower than 0.01μM. In a dense culture of respiring microorganisms, no oxygen will reach the probe when the bulk concentration of oxygen is in the order of KM.

Determinations of stability of bound enzyme showed that the most active and stable preparations were obtained by application of the carrier with amino groups activated with glutaraldehyde (activity half-life 96.6 days).     Notes: A new type of continuous fermentor has been developed which features an agitation-aeration system based on pulsed flow across perforated plates the use of plug flow to achieve a concentration gradient. The influence of the agitation-aeration parameters (in this case the pulsed speed) has been measured, and mathematical models have been produced for the gas hold-up, the power dissipated in agitation and aeration, the oxygen-transfer rate, and efficiency. The oxygen transfer is high, up to 600 mmol/liter hr with a transfer efficiency much higher than that published for any other technique.

    Notes: Glucoamylase from four different companies was studied: three had similar stability (half-life at 50°C about 140 hr); the fourth was less stable (half-life at 50°C about 20 hr). The immobilized enzymes were all less stable than their soluble counterparts: immobilized enzyme stability depended on the soluble enzyme used, the support, and method of immobilization. Thus enzyme bound to Enzacryl-TIO was less stable than enzyme bound to hornblende (metal-link method); this, in turn, was less stable than enzyme bound to hornblende by a silane-glutaraldehyde process. Bound enzyme stability was also improved by the presence of substrate or product (starch maltose or glucose). After 110 hr at 50°C in the presence of maltose (10% (w/v)) one preparation (a more stable soluble enzyme boul1d to hornblende by a silane-glutaraldehyde process) retained over 95% of its activity: activity loss was too low to permit the estimation of a half-life.

Asparaginase was bound on the AHA-spiropyran gel under visible light and was released in the dark. The optimum conditions for photocontrolled binding and releasing of asparaginase were a 0.05M phosphate buffer concentration and pH 7.0. Seventy-five percent of the bound asparaginase was released from the AHA-spiropyran gel column in the dark. Ninetyfold purification of asparaginase was performed with the AHA-spiropyran gel Column.

    Notes: Mathematical models presented in a previous paper (Biotechnol. Bioeng., 20, 865 (1978)) describing unsteady-state bacterial growth on organic polymers are modified to predict the holding time necessary to attain a given treatment level in a steady-state activated sludge process. Various hydraulic regimes for the activated sludge aeration basin are assumed, and it shown that the hydraulic regime strongly influences the total holding time necessary to attain a given treatment objective. Maintaining the highest possible concentrations of bacteria, extracelluar enzyme, cleavable polymer bond, and utilizable oligomer in the aeration basin during the course of treatment yields the highest possible organic nutrient uptake. However, it is shown that aeration-basin hydraulic regimes that maintain relatively high levels of bacteria and extracellular enzyme also maintain relatively low levels of polymer bond and utilizable oligomer and vice versa. Thus the efficacy of a particular aeration-basin hydraulic regime depends on its ability to maintain at high levels those concentrations that most strongly influence the kinetics of enzymatic hydrolysis or utilizable oligomer uptake.

    Notes: The fast enzymatic hydrolysis of D,L-phenylalanine methylester (DLE) to L-phenylalanine (LA) and D-phenylalanine methylester (DE) with immobilized α-chymotrypsin was chosen as a model reaction. Under the experimental conditions applied in the present investigations the pore diffusion is the rate-limiting step of this reaction owing to the pH gradient in the particles. The effectiveness of the catalyst is experimentally determined as a function of the substrate concentration based on measurements of the enzyme protein content of native and immobilized enzyme. The proteolytic reaction is theoretically treated by also using a pore diffusion model which takes into account the concentration gradients of substrate and product, pH- and enzyme activity profiles, as well as the change of buffer capacity of the solute in the catalyst particles. The model parameters were experimentally determined for the investigated system. It can be shown that conditions are possible for which the effectiveness of the catalyst exceeds unity.

    Notes: Combining acid cottage cheese whey and the lime-sulfide effluent from tannery unhairing processes spontaneously coprecipitates the whey proteins with the large peptides and proteins of the tannery waste. The floculation of the denatured protein material also carries down the hide pigments, excess lime, and the casein fines from the whey. The clear supernatant contains lactose, sulfur in various states of oxidation, free amino acids, peptides, and ammonium salts, but no detectable macromolecular proteins. The recovered solid products, which contain more than 20% of the original nitrogen, appear to have a good balance of essential amino acids although actual composition varies with the composition of the raw wastes. Feed supplements may possibly by obtained by this method from two presently wasted industrial effluents.
