    Notes: Correlation between chemical structure and Rf-values was studied in the case of aniline and its derivatives using high performance TLC and a U-chamber. The characteristic group constants referring to each substituent were determined by chromatographing derivatives with one substituent and aniline in toluene and dibutyl ether respectively. On the basis of these constants the RM and Rf values of derivatives with more substituents were calculated. Comparing the calculated and measured values good agreement was found.

    Notes: The principal analytical details for the third of three possible modes in high performance thin-layer chromatography are given, namely the anticircular mode. Separation is achieved by allowing the mobile phase to enter the plate layer on a precise outer circle line, from where it flows towards the centre with nearly constant speed. This technique is theoretically and practically the fastest of all three possible in HPTLC. It permits maximum sample capacity with a minimum of time, layer and mobile phase consumption. It is therefore the most economical HPTLC technique.A new carrier-free mobile phase transfer principle is used. The conditions for qualitative and quantitative analysis are good: repeatability, reproducibility and accuracy of routine TLC analyses are superior to those achieved by the classical trough technique. The specially narrow spot-path in anticircular HPTLC facilitates automated quantitation. Compared with the linear and circular modes, the anticircular mode shows better separation and significantly increased sensitivity at higher Rf-values. The drawback, however, is that the separation power (expressed by the separation number) is lower compared with the other two modes.

    Notes: Practical details are given to produce low dead volume Pt/Ir to glass connections and a variety of components for effluent splitting and pneumatic solute switching. It is shown that the column effluents from a glass capillary column can be split in any desired ratio and maintained constant, regardless of column flow rate. Bandbroadening in the splitter remains negligible even for very low flow rates. A complex pneumatic system to be used for heartcutting with two glass capillary columns is shown.

    Notes: Micro capillary columns were successfully applied to liquid chromatography by employing the principles used in micro high performance liquid chromatography. Fundamental investigations on the use of capillary columns in LC were performed for the various column parameters. Good separations of five aromatic hydrocarbons and four kinds of phthalic esters were obtained on a 62 μm I.D. capillary column, coated with SE-30.

    Notes: The real plate number concept is interpreted in terms of the fundamental properties of a chromatograph. It is shown that the linear relationship between the total peak width at half height, (b0.5)T, t, and k is only an approximation of the more general linear relationship between (b0.5)2T, t and (1 + k)2. In the case where the relationship between (b0.5)T, t and k approaches linearity, it is concluded, firstly, that b0 = a, i. e., b0 is a column property and independent of extra-column phenomena and, secondly, that nreal=ntheor. When the relationship becomes non-linear, b0 is determined partly by extra-column phenomena, but not in terms of a useful relationship. It is concluded that it is unnecessary to introduce the parameters a, b0 and nreal, since they all have counterparts in the established theory of chromatography. These counterparts prove to be equally useful in the practical analysis of chromatographic systems.

    Notes: Volatile fatty acids in aqueous media were esterfied in situ with phenyldiazomethane (PDM). Complete esterification was achieved in 3 h at 40°C under continuous stirring of an ether/water system. The process involved immediate transfer of benzyl esters into the organic phase. The separation of benzyl esters mixtures was carried out in glass capillary GC columns. The retention data and FID molar responses were determined for C1 to C6 acid benzyl esters, including 2-methylpropionic, 3-methylbutyric, 2-methylvaleric, and 2-ethylbutyric acid. An unusual relationship was observed between benzyl ester relative molar responses and carbon atom number.

    Notes: The error propagation is given for routine analysis by in situ evaluation in TLC using the methods with internal or external standard. The internal standard method will need four different measurements of peak height or area, while the external standard method uses only two measurements. Therefore in the latter case the error of the spotting volume will cause errors in the determined concentration. By error propagation it can be shown, that the internal standard method gives better results, if the error in measuring peak height or area is less than the error of the spotting volume.

    Notes: A method is described for the simultaneous quantitative determination of monoamines and related compounds from urine and brain tissue samples in the pg-range, using a GC/MF instrument LKB-2091 provided with a multiple ion detection (MID) system and capillary columns. The simultaneous detection of “fingerprints” with fourteen and more compounds during one single GC run was achieved after the intoduction of “time gates” for the detection of the different characteristic masses. Several modifications to the LKB equipment permitted exact reproducibility of retention times, which is essential for the use of “time gates”. The detection of “fingerprints” with these acidic, basic and alcoholic substances was achieved by the extraction of the dried biological samples with silylating agents.

    Notes: A rapid and sensitive method to determine volatile organohalides in different types of waters has been developed. The chromatographic system is based on the resolving power of the glass capillary column and the high sensitivity of the electron capture detector. The capillary columns were connected to the detector so that the eluate from the column is mixed with thermostatted purge gas to minimize band broadening due to dead volumes. The low column bleeding and decreased band spreading from capillary columns enhance the sensitivity to very low levels: 1 fg (10-15g) for carbon tetrachloride, despite dilution of the column eluate by the purge gas.The water samples of 100 ml were extracted with 1 to 10 ml n-pentane which is the most practical alkane with respect to purity. When using high water to pentane ratios corrections had to be made, due to low extraction yield, to get accurate quantifications.A column with Carbowax 400 stationary phase permits direct tap water injection. The water peak concealed the substances eluated after CHCl3 and CHBrCl2.The gas chromatographic separation of haloforms in tap water could be shortened to 30 seconds using a 10 m SE-52 column with an increased film thickness.Besides tap water and seawater, industrial waste water from a pulp mill was analyzed. Ten different halogenated organic compounds, ranging from 1 ng/I to 760 μg/I, were quantified in the different waters. Bromoform, which had not previously been found in seawater, was detected.Chromatogram of a standard mixture containing 11 organohalides. Stationary phase, SE-52; 33 m × 0.3 mm i.d. inj. temp. 200°, column temp. 50°, interface 250°, detector temp. 250°.     Notes: Splitless injection is based on the solvent effect as a mechanism condensing large vapour clouds down to infinitely shortened bands. The effect is controlled by the parameters column temperature, volatility and amount of solvent, and rate of injection. By properly selecting the variables the effect can easily be optimized for any combination of sample and column. It is the purpose of this paper to provide the mechanistic understanding as required for this optimization, as well as some rules for the experimental realization. Potentialities and limitations of splitless injection are discussed, and the role of the solvent effect in on-column injection is emphasized.

    Notes: A roast coffee infusion (nicaragua arabica) was acidified (pH 1,5) with hydrochloric acid and extracted with diethyl ether in a perforator. After evaporation of the solvent the residue was treated with N-methyl-N-trimethylsilyl trifluoroacetamide and analyzed by glass capillary GC/MS. The main components were identified by their electron impact and isobutane chemical ionization mass spectra. 1,2,4-trihydroxy benzene, 1-ethyl-3, 4-dihydroxy benzene and 2,3-dihydroxy toluene were identified as new constituents of roast coffee with possible emetic properties.

    Notes: Basic methodology to effect the separation of complex samples by gas chromatography is discussed from an experimental point of view. Several possibilities are presented which are commonly used to enhance the information content of a gas chromatographic run. These include incorporation of more than one set of retention data, multiple detectors, and special column switching techniques. These and other techniques have sometimes been labeled “multidimensional” in the literature. An attempt is made to define the term “two-dimensional chromatography”.The review is divided into sections which point out the different directions which have been taken to deconvolute chromatographically unresolved components. Particular emphasis is placed on technology which is capable of dealing with samples of extreme complexity, and instrumentation for trace- and ultratrace analysis is dicussed with specific examples. The future of two-dimensional techniques in gas chromatography is looked into.

    Notes: New methods for peptide separation are being developed. Resolution of peptide mixtures on HPLC with good reproducibility indicates the progress that has already been made in development of supports with useful separation capabilities. At present adsorptive characteristics of the stationary phases may place limitations on the applicability of the technique to underivatized peptides. Hopefully, the application of ionpairing techniques and the development of new support materials will expand the range of application for protein and peptide purification.

    Notes: Basic methodology to effect the separation of complex samples by gas chromatography is discussed from an experimental point of view. Several possibilities are presented which are commonly used to enhance the information content of a gas chromatographic run. These include incorporation of more than one set of retention data, multiple detectors, and special column switching techniques. These and other techniques have sometimes been labeled “multidimensional” in the literature. An attempt is made to define the term “two-dimensional chromatography”.The review is divided into sections which point out the different directions which have been taken to deconvolute chromatographically unresolved components. Particular emphasis is placed on technology which is capable of dealing with samples of extreme complexity, and instrumentation for trace- and ultratrace analysis is dicussed with specific examples. The future of two-dimensional techniques in gas chromatography is looked into.

    Notes: Two new acyltetramic acids related to streptolydigin have been isolated from fermentations of Streptomyces lydicus. The principal members of this complex were resolved by TLC on silica gel. However, the methods of detection, permanganate spray or bioautography, were not suitable for both crude fermentation broths and purified extracts. Gas chromatography is unsuitable for the detection of either underivatized or silylated streptolydigins. High performance liquid chromatography (HPLC) particularly on triethylaminoethyl cellulose is rapid and sensitive and is the method of choice for the analysis of both crude and purified samples. Using high performance liquid chromatography, two components were detected in the complex, which are not observed using any of the other chromatographic procedures.

    Notes: A thorough survey of the past and current available literature shows that the diffusion of the solvents toluene, ethylbenzene, ortho-, meta-, and para-xylenes in polypropylene has not been studied so far, although such studies find practical applications in various industrial problems. Diffusitives and the activation energies of the above mentioned solvents in polypropylene were determined in the temperature range of 45-120°C by gas chromatography. The values obtained in this communication compare fairly with those determined by conventional techniques for analogous systems, confirming the reliability of the gas chromatography as a powerful tool for diffusion measurement and the authenticity of the data presented herein.

    Notes: A definition for an absolute retention index is given. This index is independent of whether the process is isothermal or temperature programmed. The relation between this index and Kovats's index is discussed. General equations are derived for the retention times of homologous series, from which the position of the air peak or missing members can be determined. Methods used in the literature for air peak determination are improved and extended to other cases of interest. The retention index in temperature programmed chromatography is reexamined in the light of recent publications on temperature programming.

    Notes: Filamentary crystal growth on the inner surface of a glass analytical capillary column using a modified ammonium hydrogen difiuoride procedure, produced highly efficient wall coated open tubular columns.Baseline separation of the acetate derivatives of 2,3,4,6- and 2,3,5,6-tetrachlorophenol was achieved and was instrumental in determining pentachlorophenol loadings along a watershed. The application of capillary columns, related to the analyses of organochlorine pesticide residues is also demonstrated. Parent and degradation products of twenty-five commonly monitored residues were effectively resolved on a 20 MOV-101 column.

    Notes: Equations describing gradient elution in TLC, with a mobile phase of a constant composition in each step, have been derived. These equations are useful for calculating retention volumes from isocratic TLC data. Equations originally derived for gradient HPLC were adapted for TLC. The validity of the equations has been experimentally verified. The satisfactory agreement between experiment and theory for binary mobile phase composed of methyl ethyl ketone and n-heptane suggests that TLC can be used as a pilot technique for gradient HPLC.

    Notes: During one year continuous use of on-column injection, the typical advantages described in our first report have fully been confirmed. In addition the analysis of large sample volumes has proved promising. Only minor modifications have been applied to the on-column injector device. Broad evidence has been gathered showing that full separation efficiency of the capillary columns after on-column injection is attained only when cold trapping or the solvent effect, as band shortening mechanisms, are working- While, under the conditions of on-column injection, cold trapping is less efficient than with other injection techniques, the opposite holds true for the solvent effect. Compared with splitless injection, the danger of excessive solvent condensation on the column is increased. A working rule is presented for establishing the optimal chromatographic conditions for handling large sample volumes while ensuring full separation efficiency yet avoiding harm to the column.

    Notes: The simultaneous demonstration of lysozyme using the unlabelled antibody enzyme method and mucosubstances by staining with the periodic acid-Schiff (PAS) and Alcian Blue pH 2.5 techniques has led to the identification of a new subpopulation of mucus-producing cells containing lysozyme in small intestinal specimens from normal rats and from patients with inflammatory bowel diseases. These cells, containing both mucosubstances and lysozyme, are located in the lateral walls of the crypts of Lieberkuhn, but can occasionally be found also in or near the tips of the villi. The specific staining for lysozyme was observed in the apical and/or basal cytoplasm of these mucus-producing cells and was readily detected in sections counterstained for mucosub-stances with Alcian Blue. The localization of these mucus-producing cells was similar both in the normal rat and in the pathological human specimens. Absorption controls and controls where a non-immune serum was substituted for the specific antilysozyme serum confirmed the specificity of the lysozyme localization.

    Notes: Development of porcine conceptuses included transitions of five stages: blastocysts of spherical, ovoid, and tubular forms containing an embryonic disc and trophoblast, extremely elongated filamentous blastocysts, and stages of embryogenesis between days 9 and 18 after mating. Embryonic survival was reduced by 17% during this period. In this litter-bearing species, intense alteration in distribution patterns occurred at days 11 and 12, when blastocysts rapidly elongated to filamentous forms. Increased embryo mortality did not result from rapid changes in distribution patterns of conceptuses within the same uterine horn at this time. Filamentous blastocysts quickly reached lengths often exceeding 60 cm, and these conceptuses became regularly spaced with no overlap of tubular membranes from other embryos in that horn. The number of concep-tuses within a uterine horn ranged from 1 to 13. Protein in individual concep-tuses was used as an indicator of growth and denoted exponential increase, but at a lower rate for blastocysts of spherical, ovoid, and tubular forms as compared with that found in filamentous blastocysts and embryogenesis stage conceptuses. Growth of a conceptus, based upon its protein content, was independent of the developmental stage or potential loss of those neighbors nearest that conceptus.

    Notes: The development of the middle ear and tympanum of Gallus gallus has been studied in embryos Hamilton-Hamburger stages 20-46. Particular attention was paid to the pattern of expansion of the pharyngeal pouch forming the tympanic cavity, and the histogenesis of the tissues of the region of the vestibular window. It is concluded that pouch expansion is brought about by simple epithelial growth into regions devoid of mesenchyme. The mesenchyme does not show significant cell death, but differentiates as connective tissue, macro-phages, and sinus vascular spaces. The epithelium of the mature cavity is of endo-dermal origin, and there is no indication of celomic contributions. We provide a detailed morphological analysis of the development of the footplate from cells of both the second visceral arch and the otic capsule. These detailed observations on the patterns of chondrification of columella and capsular tissues permit the presentation of a model for cellular interactions leading to the differentiation of the annular ligament.

    Notes: The prefusion epithelium of human palatal processes was examined for evidence of specialization which might facilitate epithelial adherence with the opposing palatal process. A surface coat stained with ruthenium red (RR) was found on all apical aspects of the palatal epithelium. In the prefusion regions, RR staining was also observed in the spaces between the superficial cells of the epithelium and in necrotic cells. Adjacent oral and nasal epithelium excluded the RR below the level of the apical junctional complex. In the absence of RR, a dense material was observed in the most superficial intercellular spaces of the prefusion region. Many superficial cells in the area were in various stages of necrosis. The combination of degenerating surface cells and an accumulation of a poly-anionic substance such as glycoprotein may facilitate epithelial adherence between opposing human palatal processes.

    Notes: Normal, rachitic, and vitamin D3-replete chicken growth plates were studied utilizing the potassium pyroantimonate-osmium tetroxide procedure. A marked membrane and mitochondrial calcium was revealed in all specimens in the maturing and early hypertrophic zones which disappeared as heavy matrix mineralization began.The most significant difference shown in the specimens was in the marked intracellular lipid content of chondrocytes in all zones of the rachitic and D3-replete growth plates. There was negligible lipid present in normal specimens.It is suggested that as most of the mechanisms postulated as necessary for calcification are present in rachitic chicks, perhaps the increased intracellular lipid pool results from the formation of abnormal lipids for insertion into the plasma membrane and thus prevents normal calcium transport.Chains of intracellular vesicles were also visualized in maturing and hypertrophic chondrocytes. These were more often seen in rachitic growth plates and in increased numbers in the early D3-replete specimens. The etiology is unknown at the present time.

    Notes: At least six different types of nuage, as judged by morphological appearance, were found in spermatocytes of the adult rat testis. These were characterized by form, distribution, and association with other organelles. The appearance (with time) and the dissociation of some nuage types were recorded in conjunction with other events of the spermatogenic cycle. The data indicate that all types of nuage behave in a dynamic way. Especially interesting were the findings concerning the chromatoid body. This structure was first observed in mid-pachytene spermatocytes. By late diplotene and during the first meiotic division, it had lost its prominence such that its eventual fate could not be determined. Large dense (0.5 μm) bodies appeared de novo in the cytoplasm of newly formed secondary spermatocytes. They were seen scattered throughout the cell and in association with mitochondria. With time these bodies appeared to coalesce to form the large definitive chromatoid of a type similar to that known to be present throughout most of the remainder of spermiogenesis.

    Notes: Dorsal roots and spinal ganglion cells of adult cats were studied for acetylcholinesterase (AChE) content. An inverse correlation exists between cell size and AChE activity; large neurons are AChE-negative whereas small ones are intensely AChE-positive. A few AChE-positive fibers were demonstrated in the dorsal roots. A possible correlation between AChE activity and substance P content of primary sensory neurons is discussed.

    Notes: The actions of the deep and superficial portions of the ptery-goideus muscle and the depressor mandibulae were studied in the agamid lizard, Uromastix aegyptius, using fine wire electrodes. The EMG signals were recorded, while simultaneously recording the jaw positions during feeding on movie film at 32 fps. Muscle activity varies with different types of food. The deep and superficial portions of the pterygoideus performed different functions. The deep ptery-goideus functioned during opening of the jaws, presumably to protract the jaw. The superficial pterygoideus was strictly a jaw elevator. The depressor mandibu-lae is not the initiator of jaw opening. When biting on tough food items, the deep pterygoideus and depressor mandibular were active.

    Notes: The L-proline analog, L-azetidine-2-carboxylic acid, (LACA) was injected into embryonated eggs of the common fowl, Gallus domesticus at daily doses of 350 μg/egg on one or several days between 8 and 12 days of incubation. Treatment at nine-days of incubation preferentially retarded embryonic growth to the twelfth day but recovery of growth rate occurred by 15 days of incubation. Relationships between growth and LACA-inhibited aspects of collagenogenesis are discussed.The earliest aged embryos from which isolated stem cells from membrane bones will form secondary cartilage is ten days of incubation. Secondary chondro-genesis on the quadratojugal, a membrane bone of the skull, was inhibited by treatment of whole embryos with LACA at nine days of incubation but not by treatment at eight days. We concluded that an event involving collagen began at nine days of incubation, was blocked by LACA, and was part of the process of chondrogenic determination of these stem cells. Addition of LACA to the medium in which already determined stem cells from the quadratojugal were cultured prevented expression of the chondrogenic phenotype. This proline analog is then a useful probe for events relating both to determination and to expression of the differentiated state, and allows conclusions to be drawn regarding the role of col-lagenogenesis in these events.

    Notes: Mouse uterine glandular epithelium during the lactationally delayed implantation period and after estradiol induction of implantation was investigated using light and electron microscopy. During the delayed implantation period the lumen of this simple tubular gland is narrowed. The glandular epithelial cells have a well developed Golgi complex lateral to the nucleus, and numerous cisternae of smooth and rough endoplasmic reticula (RER) and many electron lucent apical vesicles of sizes up to 0.2 μm in diameter near the luminal border. The basal region contains lipid droplets and dispersed, irregular cisternae of RER. Twenty-four hours after the administration of 17β-estradiol the glandular lumina become dilated but the luminal content does not stain with azure B. Ultrastructurally the glandular cells are not remarkably different from those seen during the delay period. However, by 48 hours after estradiol administration the glandular lumina are not only dilated but filled with material which stains intensely with azure B and is ultrastructurally dense and homogeneous. The apical region of the glandular cells contains granules up to 0.4 μm in diameter composed of electron dense material similar in density to that seen in the glandular lumen. In addition, the Golgi complex has assumed a position apical to the nucleus, and the basal RER has an increased number and more orderly arrangement of cisternae.The changes seen in the uterine glands after the induction of implantation during the delay period are apparently indicative of increased secretory activity of the glandular epithelia. However, the contribution of the glands to the changes in uterine fluid composition has yet to be established.
