    Notes: Summary In a seven year old boy with acute abdominal pain and intestinal bleeding, laparotomy revealed an invaginated carcinoma adenomatosum cylindrocellulare in the distal part of the jejunum. Histological and pulse cytophotometric investigations support the diagnosis of a highly differentiated but locally destructive and malignant tumor.

    Notes: Summary We have correlated the light-microscopic features in the un-myelinated hypothalamus with gonadal atrophy in 15 women of 30-111 years of age and in 7 men between 29 and 82 years. In the postmenstrual cases there is a distinct concordance of gonadal atrophy and the manifestation of nucleolar changes (augmentation, multiplication and vacuolization) in many nerve ceils of the arcuate and subventricular nuclei. In younger, still fertile women this nucleolar finding was seen only rarely and sporadically, was limited to the arcuate nucleus and was absent in the subventricular nucleus. We interpret this nucleolar finding as a feedback effect. In man, too, this coincidence is obvious with age and in gonadal atrophy, with fewer nucleolar changes in old age than are seen in women. This difference is probably caused by a more rapid drop of the estradiol than of the testosterone level.-The hypertrophy of the subventricular nucleus (Sheehan and Kovacs) was also observed in our postmenstrual cases.

    Description / Table of Contents: Résumé Trente et un cas de tumeurs malignes de la thyroïde, classifiés à l'origine comme des carcinomes anaplasiques, ont été réexaminés en immunohistochimie pour la présence d'anticorps IgM, IgG, IgA, de cytocératine, calcitonine, lysocyme, et alpha1-antitrypsine en utilisant la méthode péroxydase/antipéroxydase. Les résultats de cette reclassification ont été comparés avec des données cliniques: symptômes cliniques, modalités thérapeutiques, et évolution clinique. On a employé la radiothérapie postopératoire dans 80% des cas et la chimiothérapie dans aucun cas. Sept des 31 tumeurs étaient positives pour les anticorps IgM (n=4), IgG (n=2), et IgA (n=1). Dans tous ces cas, les marqueurs épithéliaux étaient négatifs. De cette manière, on a pu reclasser comme lymphomes malins primitifs non seulement 3 tumeurs à petites cellules mais aussi 4 tumeurs avec des zones de cellules géantes.
    Abstract: Resumen Treinta y un casos de neoplasias malignas de la glándula tiroides originalmente clasificadas como carcinoma anaplásico fueron reexaminados por inmunohistoquímica utilizando métodos de peroxidosa:antiperoxidasa (PAP) para IgM, IgG, IgA, citoceratina, calcitonina, lisocina, y alfa-1-antitripsina. Los resultados de la reclasificación fueron comparados con la información clínica, incluyendo sintomatología, modalidades de tratamiento, y evolución final. Se realizó radioterapia postoperatoria en más de 80% de los casos, quimioterapia en ninguno. Siete de 31 tumores exhibieron tinción positiva para anticuerpos IgM (n=4), IgG (n=2), e IgA (n=1). Todos estos casos fueron negativos para marcadores epiteliales. Sorprendentemente, no sólo la totalidad de los tumores de células pequeñas (n=3) sino también 4 tejidos con áreas predominantes de células gigantes, estuvieron en el grupo de aquellos reclasificados como linfoma maligno.
    Notes: Abstract Thirty-one cases of thyroid malignancies which were originally classified as anaplastic carcinoma were reexamined immunohistochemically using PAP methods (peroxidase:antiperoxidase) for IgM, IgG, IgA, cytoceratin, calcitonin, lysozyme and alpha-1-antitrypsin. The reclassification results were compared with patient data such as clinical symptoms, treatment modalities, and clinical outcome. Postoperative radiotherapy was carried out in more than 80% of cases, chemotherapy in none. Seven of 31 tumors showed a positive staining for IgM (n=4), IgG (n=2), and IgA (n=1) antibodies. All of these cases were negative for epithelial markers. Surprisingly, not only all small cell tumors (n=3) but also 4 tissues with predominantly giant cell areas were among those reclassified as primary malignant lymphoma.

    Notes: Abstract On a rat, which has spontaneous died 12 months after 4 times i.p. application of aflatoxin during pregnancy, there has been found a frontal tumor barely of the size of a pepper-corn which came out from the leptomeninx. A comparison to the Abrikossoff tumor concerning the light- and electron-microscope as well as the nucleic-acid concentration is justified. The protein content of the granula of the tumor cells is higher than the values known in the granular-cell myoblastoma of man.

    Description / Table of Contents: Zusammenfassung Mit dem in der Impulscytophotometrie zu DNS-Messungen benutzten Fluorochrom Ethidiumbromid1 wurden Schnittpräparate menschlicher Epidermis und Tupfpräparate menschlicher Leberzellen behandelt. Die an Fluoreszenz- und Durchlichtphotographien demonstrierten Färberesultate zeigen, daß keine nukleinsäurespezifische Bindung des Farbstoffes erfolgt. Aufgrund der spektralen Eigenschaften der mit dem Fluorochrom tingierten Zellelemente erscheint eine quantitative mikrofluorometrische Substanzmengen-bestimmung nur bedingt möglich.
    Notes: Summary In order to test Ethidiumbromide1 as a marker for DNA in cells, slides with sections of human epidermis and smears of liver cells were stained in a buffered solution of this substance. The staining results demonstrated by photographs indicate, that Ethidiumbromide does not bind selectively to nucleic acids. As a consequence of the spectral properties of the stained tissue elements this fluorochrome does not seem to be suited for quantitative determinations of nucleic acids by measurements of the fluorescence intensity.

    Notes: Summary This is a report of the second known case of a primary malignant granular-cell tumor of the cerebral hemispherical white matter. Two cell types may be distinguished apparently representing different developmental stages of otherwise identical tumor cells. Quantitative histochemical and biochemical studies have shown that tumor cells were containing markedly elevated levels of DNA and RNA. Only few ribosomes and polysomes could be detected, however, by electron microscopy. While cytophotometry disclosed only slightly elevated cytoplasmic proteins, there was a distinct increase in total protein concentration of tumor tissue per unit weight, as measured by conventional techniques. It is suggested that this increase is mainly attributable to structural proteins of cellular and subcellular elements of multiplicating malignant cells. A ratio of RNA to DNA in excess of 1 was found for white matter derived from the central tumor, while the ratios of control tissue were lowered to values far below one due to postmortem changes greatly reducing tissue concentrations of RNA. No detailed characterization of RNA was attempted. The nuclear DNA content of tumor cells reached values equivalent to chromosomal hexadekaploidy. This was in sharp contrast to control data from an Abrikossoff tumor of the oral cavity and from a neurohypophyseal tumorette (“choristoma”), respectively, displaying diploidy only associated with a much lesser increase of cytoplasmic RNA and proteins. Qualitative lipid studies were consistent with a marked active demyelination of the tumor centre as indicated by a severe reduction of cerebrosides and sulfatides and the presence of cholesterol esters. In addition there was a striking loss of phosphatidylethanolamine and a lesser one of sphingomyelin of white matter of both the tumor-stricken and the contralateral unaffected hemispherical regions, possibly suggesting a generally disturbed metabolism of myelin. It is not clear whether these general changes were resulting from the presence of the unilateral tumor or from precocious cerebral involution.

    Notes: Summary Neuron-specific enolase (NSE) is an enzyme detectable in nervous and neuroendocrine tissue. Increased serum levels of NSE are found in small cell lung cancer and in patients with neuroblastoma, in whom NSE is used as a serum tumor marker. We have investigated 32 patients with histologically proven medullary thyroid carcinoma, a tumor of neuroendocrine origin, in which the classical tumor marker calcitonin (CT) was pathologically elevated. Positive immunocytochemistry for NSE and CT in C-cells was obtained in all cases. Increased serum NSE levels were found in only 5 of 32 patients, there was no correlation between NSE and CT concentrations. We also compared NSE and CT serum levels during long-term follow-up and again found no correlation between NSE and CT. After i.v. stimulation tests with pentagastrin and calcium, no correlation was found between NSE and CT serum levels. We conclude, therefore, that in medullary thyroid carcinoma NSE is useful for immunocytochemistry but not a reliable serum tumor marker.

In einer Patientenumfrage (n = 77) 0,5–35 Jahre nach Splenektom wegen Traumas, konnte keine Erhöhung der Sepsis und Pneumonierate beobachtet werden. Unter 38 632 Obduktionen wurden die Todesursachen der Obduktionen nach Splenektomie (n = 331) analysiert. Eine Todesursachenhäufung nach Splenektomie, wie Sepsis, Thrombose und Infarkt, kann nicht festgestellt werden.
    Notes: Summary The long-term complication rate following splenectomy was investigated in two retrospective studies analysing the incidence of sepsis, pneumonia, thrombosis and myocardial infarction. The follow-up included 77 patients who had untergone splenectomy 0.5–35 years previously due to traumatic rupture. No significant increase in the incidence of sepsis or pneumonia could be detected. Postmortem examinations of 38,632 patients included 331 cases with splenectomy. The incidence of sepsis, thrombosis or myocardial infarction as the cause of death was no higher among patients who had undergone splenectomy.
