    Notes: Abstract Intragenic restriction site polymorphisms in amino acid residue 72 in exon 4 and a Mspl polymorphism in intron 6 of the p53 tumour suppressor gene can both serve as polymorphic markers. Probe YNZ22 (D17S5) is a highly polymorphic, variable number of tandem repeat (VNTR) marker which maps to chromosome 17p13.1 where the p53 gene is located. Locus specific amplification by polymerase chain reaction (PCR) technique and subsequent non-isotopic single-strand conformation polymorphism analysis of the PCR fragments was used for the detection of loss of heterozygosity (LOH) of 17p including the p53 gene locus. In combination with a PCR-based method for the analysis of the VNTR locus D17S5 using unique sequences flanking the polymorphic region of YNZ22 we investigated tumour DNA and corresponding constitutional DNA from 69 patients, including 39 patients with gastric cancer, 21 patients with osteosarcomas and 9 patients with Ewing's sarcomas. Using all three methods, 49/69 (71%) patients were informative for LOH, which revealed allelic loss in 5/39 (12.8%) gastric cancers, 1/9 (11.1%) Ewing's sarcoma, and 4/20 (20%) osteosarcomas.

    Notes: Abstract We have examined the correlation between matrix metalloproteinase (MMP) expression and metastatic properties of a low metastatic osteosarcoma cell line, osteosarcoma takase (OST), under stimulation by tumour necrosis factor α (TNFα). In vivo, OST cells exhibited significantly increased colonization in the lungs of nude mice in a dose-dependent manner when they were treated by TNFα prior to injection. In vitro, TNFα enhanced tumour cell invasion through the reconstituted basement membrane in a transwell chamber up to 2.5-fold. Gelatin zymography and sandwich enzyme immunoassays demonstrated marked production of MMP-9 [92-kDa gelatinase/type IV collagenase (gelatinase B)] but not MMP-2 [72-kDa gelatinase/type IV collagenase (gelatinase A)], MMP-3 (stromelysin-1) or MMP-7 (matrilysin). Motility of the tumour cells and adhesion to cultured endothelial cells were slightly increased by the TNFα treatment up to 1.6-fold and 1.4-fold, respectively, while the growth rate was decreased. These results suggest that upregulation of MMP-9 together with enhanced motility and endothelial adhesion contribute to the increased metastatic ability of OST cells induced by TNFα treatment.

    Description / Table of Contents: Zusammenfassung Es wurde bei 11 Schweinen eine orthotope Xenotransplantation der Hundeleber durchgeführt. Die Überlebensdauer war durchschnittlich 4,3 ± 1,5 Std. Die Todesursachen waren generalisierte Blutungen. Um die hyperakute Abstoßungsreaktion zu bremsen, wurden vor der Verpflanzung der Hundeleber bei 5 Fällen Hundeniere und Hundemilz je 20 min und bei 5 anderen Fällen eine zweite Hundeleber am Femoralkreislauf eingeschaltet. Wegen der stark aufgetretenen Kreislaufreaktion (Tachykardie und Hypotonie) war die Überlebensdauer noch kürzer. Aus gleichen Gründen wurden hohe Dosen Prednisolon intraund postoperativ bei 5 Fällen verabreicht. Die Überlebensdauer von 4 Schweinen war 9,0 :L 2,2 Std. Ein Schwein überlebte 5 Tage. Bei diesem Schwein wurde Hundeproteinsynthese beobachtet.
    Notes: Summary Orthotopic xeno-transplantations of dog livers into 11 pigs were carried out. The survival time was on average 4.3 ± 1.5 h. The cause of death was generalized haemorrhage. To inhibit the hyperacute rejection, before trans-planting the dog liver we connected dog kidneys and dog spleens in 5 cases and in another 5 cases a second dog liver to the femoral circulation, 20 min, before the transplant, in each case. In view of the very strong circulatory reaction (tachycardia and hypotension) the survival time was even shorter. For the same reason high doses of prednisolone were administered intra- and postoperatively in 5 cases. The survival time of 4 pigs was 9.0 ± 2.2 h. One pig survived for 5 days, and in this animal we were able to observe dog-protein synthesis.

    Description / Table of Contents: Zusammenfassung In der vorliegenden Arbeit wurden durch MNNG (N-methyl-N′-nitro-N-nitrosoguanidin) oder ENNG (N-ethyl-N′-nitro-N-nitrosoguanidin) erzeugte Tumoren im Drüsenmagen der Ratte feinstrukturell untersucht. Ziel der Untersuchung war es, die Polymorphie der Tumorzellen in dem experimentellen Carcinom zu beschreiben und insbesondere die Beziehungen zwischen den verschiedenen metaplastischen Zelltypen und den Carcinomzellen zu untersuchen. Im Tumorgewebe konnten besonders im Invasionsbereich neben undifferenzierten Adenocarcinomzellen zahlreiche gut differenzierte Zellen nachgewiesen werden. Es fanden sich Becherzellen, endokrine Zellen, Zellen, die lamellierte Myelinfiguren in ihrem Cytoplasma enthielten und Plattenepithelcarcinomzellen. Vor allen Dingen wurden regelmäßig Zellen mit gut differenzierten Mikrovilli beobachtet, die Tonofilamente und den “membrane coating granules” ähnliche Granula enthielten und somit typische Zeichen sowohl des Plattenepithels als auch der Drüsenzellen aufwiesen. Daher wurden diese Zellen als Zwischenstufen zwischen Adenocarcinomzellen und Plattenepithelcarcinomzellen gewertet. Das Vorkommen derartiger Zwischenstufen scheint uns darauf hinzuweisen, daß Adenocarcinomzellen in dem durch MNNG oder ENNG erzeugten experimentellen Magencarcinom die Fähigkeit besitzen können, sich nach Zellteilung zu den genannten metaplastischen Zelltypen zu differenzieren.
    Notes: Summary In the present study electron microscopic investigations of tumors of the glandular stomach of rats induced by oral administration of MNNG or ENNG are dealt with. The aim of the study was to describe the different cell types found in the carcinoma and to elucidate the possible realtionship between the undifferentiated carcinoma cells and the more differentiated cell types found in the tumor. The results show that besides undifferentiaded carcinoma cells, several differentiated cell types such as goblet cells, endocrine cells, cells with lamellated inclusions in their cytoplasm, and squamous carcinoma cells can be observed. The most conspicuous findings were carcinomatous gland cells with well differentiated microvilli on their luminal surface and typical tonofilamentous structures in their cytoplasm. These cells exhibited signs of the squamous epithelium and of gland cells. Therefore they may be considered as intermediate variants between adenocarcinoma cells and squamous carcinoma cells. The occurrence of such intermediate steps points to the possibility of differentiation by cell division of adenocarcinoma cells into several metaplastic cell types in the experimental stomach carcinoma induced by MNNG or ENNG.

    Notes: Summary Recent investigations have shown that telangiectatic osteosarcoma has a poorer prognosis than other osteosarcomas. To elucidate the histogenesis of TOS two cases were investigated on the electron microscopic level. The results show that besides anaplastic, osteoblast-like, and fibroblast-like tumor cells angiosarcomatous components can be observed in this malignant bone tumor, which are characterized by endothelial cells with pinocytotic vesicles, tight intercellular junctions, fine fibrils, and so-called Weibel-Palade bodies in their cytoplasm. From these results, it is concluded that telangiectatic osteosarcoma is derived from multipotent mesenchymal cells with potential differentiation into various directions, such as osteoblast-like cells, endothelial cells, and fibroblast-like cells.

    Description / Table of Contents: Zusammenfassung Durch orale Applikation von MNNG (N-methyl-N′-nitro-N-nitrosoguanidin) über 35 Wochen oder ENNG (N-äthyl-N′-nitro-Nnitrosoguanidin) über 20 Wochen wurden experimentelle Carcinome im Drüsenmagen von Ratten erzeugt. Die Tiere wurden in regelmäßigen Abständen nach dem Fütterungsbeginn getötet und die Tumoren histologisch aufgearbeitet. Dadurch waren die Entwicklungsphasen der Carcinogenese analysierbar, die für die Frühdiagnose des Magencarcinoms von Bedeutung sein können. Die histologischen Veränderungen während der Carcinogenese wurden in 4 Stadien eingeteilt : 1. Die regenerative Hyperplasie findet sich 1–9 Wochen nach Fütterungsbeginn mit MNNG vorwiegend im Bereich von Erosionen und Geschwüren. Es handelt sich um unregelmäßige Drüsenproliferationen ohne Zellatypien. 2. Die glanduläre Hyperplasie, die am häufigsten 12–17 Wochen nach MNNG-Gabe auftritt, besteht aus einer verdickten Schleimhaut mit vermehrten Drüsenschläuchen, die ebenfalls gut ausdifferenziert sind und kaum Atypien zeigen. 3. Die Dysplasie zeigt sehr unregelmäßige Drüsen mit ausgeprägten Zellatypien, deren Ausdehnung jedoch auf die Schleimhaut beschränkt ist. 4. Die Neoplasien bestehen aus undifferenzierten Drüsenproliferationen, die bis in die Tunica serosa vordringen. Zum Teil handelt es sich noch um Adenome, meist jedoch um Adenocarcinome. In einigen Fällen waren Metastasen in Pankreas, Leber und Lymphknoten nachweisbar, in einem Fall in einer Rippe. Histochemisch zeigten die Tumoren im Vergleich zu den Kontrollen keine charakteristischen Veränderungen.
    Notes: Summary Experimental carcinomas in the glandular stomach of rats were induced by oral administration of MNNG (M-methyl-N′-nitro-N-nitrosoguanidin) for 35 weeks or ENNG (N-ethyl-N′-nitro-N-nitrosoguanidin) for 20 weeks. Rats were killed at different times after beginning of carcinogen treatment and tissue specimens were prepared for histologic investigation. Particular interest was placed on the development of tumors and on pathological findings possibly contributing to early diagnosis of stomach cancer. During the development of tumors, several dysplastic reactions were observed in the antral mucosa. They could be classified into 4 groups : One was regenerative hyperplasia (1) that meant irregular glandular proliferations without cell atypism at the margin of erosions and ulcers. This lesion was mainly found 1–9 weeks after administration of MNNG. In glandular hyperplasia (2) either crypts or glands were extended and mucosal layers were thickened. No signs of cell atypism were observed. This lesion was mainly found 12–17 weeks after administration of MNNG. Dysplasia (3) was combined with considerable structural modifications and cellular atypism. However, this lesion was limited to the mucosal layer. Neoplastic changes (4) were characterized by marked cellular atypism and extension to tunica submucosa and tunica serosa. Some tumors showed the histological patterns of benign tumors, but most of them were adenocarcinomas. In some cases metastases into pancreas, liver and lymph nodes and in one case into the 12th rib were observed. No particular enzyme patterns were found by histochemistry.

    Notes: Summary A total of 20 highly malignant osteosarcomas were studied by DNA flow cytometry after preoperative chemotherapy according to the COSS 80/82 protocol to assess their nuclear DNA content and the impact of chemotherapy on DNA ploidy and proliferation. Of the cases studied, 70% revealed aneuploid DNA stem lines with a median value of 1.67, and a median S-phase proportion of 16.1%. These data differed substantially from the results of a preceding study on untreated osteosarcomas revealing a higher frequency of DNA aneuploidy, higher DNA indices, and higher proportions of cells in S-phase. The pretreated tumors also showed a distinct relation between DNA content and the grade of tumor regression. No aneuploid DNA stem line was found in the responder group-I, whereas group-V (osteosarcomas with more than 50% viable tumor tissue) consisted almost completely of aneuploid DNA populations; all tumors with more than 2 DNA aneuploids were found in the latter group. All populations with a DNA index (DI) over 2.0 were found in the nonresponder groups IV and V, where all DNA aneuploids had an S-phase above 12%. No differences in DNA ploidy or proliferation were found in the various histological subtypes of pretreated osteosarcomas. These data indicate that flow cytometric DNA measurement in osteosarcoma may not only serve as a tumor marker, but also support the evaluation of morphologically established regression grades, thereby verifying the patient's prognosis: a high DI (over 2.0), a high number of DNA aneuploids (more than 2), and a high proliferation (S-phase above 12%) seemed to indicate a poor regression of the primary lesion, which may offer a pretherapeutic way of prognostic evaluation.

    Notes: Summary Seven giant cell tumors of bone and four malignant fibrous histiocytomas were studied immunohistochemically with different monoclonal antibodies to the mononuclear phagocyte system (MPS), to HLA-DR antigens, and to a proliferation-associated nuclear antigen (KI-67), in order to clarify the role of macrophages in these tumors. A part of the mononuclear cells stained positive with antibodies against the MPS. Antibody 25-F-9 against mature tissue macrophages showed the strongest reaction. The osteoclast-like giant cells also stained positive with this antibody. Fibroblast-like stromal cells, however, showed negative reactions to all antibodies against MPS cells. A double-labeling immunohistological technique was used to detect the proliferating cell population in these tumors. The fibroblast-like cells that were negative for MPS markers, were positively labeled with the monoclonal antibody Ki-67 against a proliferation-associated nuclear antigen, whereas a negative reaction to Ki-67 was seen in cells positive with antibodies to the MPS. These results support the concept that macrophages are a reactive population in these tumors, whereas the fibroblast-like mesenchymal cells are the proliferating tumor cells.

    Notes: Summary At clinical presentation, the majority of malignant tumors are composed of multiple clonal subpopulations of tumor cells with different phenotypic characteristics. Using the experimental tumor model ER 15-P, a methylcholanthrene-induced pleomorphic sarcoma of the C57 Bl6J mouse, we studied a system of long-term in vivo passages of this primary tumor for cell morphological changes, and alterations in the potential for spontaneous lung metastases. Transplants from the primary after the 4th, 20th, 40th and 80th i.m. passage (referred to as T4, T20, T40, and T80 respectively) together with their lung metastases were investigated by light microscopy, immunohistochemistry, and electron microscopy. In addition, the potential for metastasis to the lungs in each group was determined and compared with that of the parent T4 tumors. T4 tumors were mainly composed of spindle-shaped tumor cells with the ultrastructural features of fibroblasts and myofibroblasts, often arranged in a storiform or fasciculated growth pattern, and intermingled with tumor giant cells. Some small areas contained polygonal or rounded tumor cells, ultrastructurally undifferentiated, and sometimes arranged in a hemangiopericytoma-like growth pattern. Although electron-microscopical findings clearly demonstrated the mesenchymal origin of these tumor cells, immunostaining with a polyclonal antibody to vimentin was unspecific in all tumor cells and normal mouse tissue. Monoclonal antibodies to vimentin from different sources were completely negative in tumor cells and murine stromal components. In contrast, myofibroblast-like tumor cells showed immunohistochemically, a moderate to strong co-expression with monoclonal antibodies to desmin, muscle actin and α-smooth muscle actin. On the basis of these morphological findings, the primary ER 15-P was classified as a pleomorphic myofibrosarcoma. The lung metastases of T4 tumors were mainly composed of undifferentiated round to polygonal tumor cells, while the number of desmin-positive, muscle- and α-smooth muscle-actin-positive cells was reduced. The morphological features of T20 tumors and their lung metastases were the same as in T4, indicating a relative stability of the phenotype up to that stage. In contrast, T40 and T80 tumors and their lung metastases were found to contain almost exclusively undifferentiated tumor cells and many tumor giant cells. While fibroblast-like tumor cells were seen only occasionally, myofibroblast-like tumor cells had almost completely disappeared. The potential for lung metastases was nearly constant in all groups, suggesting metastatic stability. Obviously, the undifferentiated tumor cells of this model are associated with a higher metastatic potential.
