    Notes: The traditional and generally accepted mode of elimination of excess spermatid cytoplasm, known to take place at the time of sperm release, occurs as a result of the disengagement of the residual cytoplasmic mass. The present study suggests that cytoplasm is also eliminated from the head region of the spermatid in the period prior to sperm release. Data presented herein indicates that the amount of cytoplasm eliminated in this manner is substantial since the volume of spermatid cytoplasm diminishes by about 70% in the period of formation and resorption of tubulobulbar complexes. Reports from this laboratory (Russell and Clermont, '76; Russell, '79) indicated that characteristic evaginations of spermatid cytoplasm, termed tubulobulbar complexes, are resorbed by Sertoli cells in the form of numerous small pockets of cytoplasm. These pockets of cytoplasm are organelle-free and show a “watery” consistency. They are subsequently degraded by the Sertoli cell prior to sperm release.During the period of turnover of tubulobular complexes the organelles of the cytoplasmic lobe, which at first are loosely organized, become clustered and tightly packed. As a consequence, the cytoplasmic lobe appears electron dense and is stained intensely with toluidine blue. This feature is interpreted, although not proven, to be the result of elimination of the watery component of the cytoplasm via resorption of tubulobulbar complexes by the Sertoli cell. During the period in which tubulobulbar complexes form, there is an open channel in which cytoplasm may flow from the flagellum to the head and out the head into tubulobulbar complexes.

    Notes: The present study describes the development and maturation of the glomerular visceral epithelium and capillary endothelium in an attempt to clarify kidney function in the neonate. The puppy kidney undergoes nephrogenesis for at least two weeks after birth, and thus possesses nephrons of different ages and levels of maturation. Since nephron development varies considerably from inner to outer cortex, renal corpuscles were examined within three arbitrary zones (outer, middle and inner cortex). The visceral epithelium of the smallest glomeruli, i.e., those of the outer cortex, is composed of cuboidal cells which are densely clustered and rests directly upon the limited number of small capillaries. More deeply within the outer cortex, broad, flat epithelial processes extend around the capillary, but lack any visible pedicels. Mid cortical glomerular capillaries undergo an increase in diameter and length, and at the same time the podocytes flatten and separate from one another. Pedicels begin to appear in this zone as thickened, disorganized processes. Juxtamedullary glomeruli are the largest, due primarily to their extensive capillary branching (looping). Podocytes are nearly adult-like in appearance, with many long primary processes and pedicels.The endothelial morphology of glomerular capillaries varies from one loop to another, as well as within a single loop. The immature endothelium of outer glomeruli is initially characterized by extensive expanses lacking pores, multiple cellular layers and small, scattered pinholes. Greater capillary maturity is apparent in the mid cortex, due to an increase in pore number and diameter as well as greater attenuation of endothelial cells. However, evidence of remodeling and a lack of pores still exists in some capillaries. The glomerular capillaries of the inner cortex appear the most mature, but possess regions that are still immature when compared to the adult.

    Notes: To further characterize Sertoli cell-germ cell junctional specializations seminiferous tubules from sexually mature Sprague-Dawley rats were dissociated by enzymatic and mechanical methods. Ultrastructural analysis of cell suspensions prepared by incubation in collagenase alone or by mechanical methods revealed that spermatids remained attached to Sertoli cells or Sertoli cell fragments. Such cellular associations were found only between Sertoli cell fragments and spermatids in which the developing acrosome had made contact with the plasma membrane (step 8 and subsequent steps of spermiogenesis). Furthermore, the fragments were confined to that region of the plasma membrane over the acrosome. The Sertoli cell half of this adhesive site displayed the typical elements of Sertoli cell junctions, filamentous bundles and associated cisterna of endoplasmic reticulum, in apposition to the spermatids. The spermatids demonstrated no surface specializations at the attachment sites. In contrast, in cell suspensions prepared with trypsin, spermatids were free of attachments to Sertoli cells or their fragments. These results demonstrate that: (1) the junctions act to bind cells together, (2) adhesive type contact is established between Sertoli cells and spermatids at step 8 and subsequent steps of spermiogenesis, (3) contact is restricted to the spermatid plasma membrane over the acrosome, and (4) spermatids can be freed from the junctional specializations by treatment with trypsin.

    Notes: Horseradish peroxidase introduced into the lumen of the rat epididymis was taken up by the columnar cells of the epithelium by five minutes and more so after longer periods. The apical cells and particularly the clear cells in the caput and cauda epididymidis, respectively, showed significantly greater endocytotic activity than the principal cell in both locations. Within 14 days after castration, however, such differences in absorptive activity among the various cell types were essentially obscured because of increased endocytosis by the androgen-deficient principal cells. The results are discussed briefly in terms of the function of different epithelial cell types and secretory/absorptive activity in the epididymis.

    Notes: In the present study the early development of peripheral lymphoid organs (spleen, popliteal lymph node, mesenteric lymph node and Peyer's patches) is described in terms of homing patterns of T and B cells, demonstrated with immunohistoperoxidatic detection of characteristic membrane antigen in normal rats and with routine histology in neonatally thymectomized rats. In the first days after birth the peripheral lymphoid organs are almost exclusively populated by T cells. After neonatal thymectomy lymphocytes appear in the dome areas of Peyer's patches from four to six days after birth, in mesenteric and popliteal lymph nodes lymphocytes are found in the outer cortex from day 6 and day 8 respectively and in the marginal zone of the spleen from eight days onwards. These lymphocytes showed no membrane staining when reacted for T antigen with immunohistoperoxidatic techniques.The morphological evidence for considering Peyer's patches of rats as central inductive sites for the generation of B cells is poor. The discrepancy in the order of appearance of T and B cell (sub)populations in spleen compartments in normal ontogenetic development and lethally irradiated, stem cell reconstituted animals is discussed.

    Notes: Perisinusoidal (P.S.) cells occurring in the spaces of Disse in the livers of normal cats, dogs, miniature pigs, albino rats, human adults and children were examined by electron microscopy. The ultrastructural details of the P.S. cells and their topographic relationships with hepatocytes, sinusoidal lining cells and reticulum fibers are described. Species differences between P.S. cells were primarily a dissimilarity in lipid content: the main ultrastructural features were the same in all species studied. The P.S. cells of the rat liver displayed only low endocytotic activity, and no phagosome formation following intravenous administration of horseradish peroxidase. The close topographic relationship of the P.S. cells with the intralobular reticulum fibers was reminiscent of the intimate connection between fibroblasts and collagen fibers, or, in cat liver, of the reticulum cell - reticulum fiber association seen in lymphoid organs. Fibroblasts were not found inside the hepatic lobules. These findings support the conclusion that, the reticulum fibers of hepatic lobules are produced by perisinusoidal cells which, however, display also other functions.

    Notes: Normal swine testes, congenital cryptorchid swine testes, and normal human testes were exposed to HMM (heavy meromyosin) after either glycerination or saponin treatment in order to determine whether the fine filaments composing the crystalloids in the Sertoli cells are actin-like. The microfilaments of the crystalloids in the Sertoli cells of the cryptorchid swine testes bind HMM to form arrowhead complexes. Short bundles of microfilaments observed in the basal part of the Sertoli cells in both normal and cryptorchid testes also bind HMM. Similar bundles of HMM-bound filaments are observed in the vicinity of spermatocytes. The periodicity of the arrowhead complexes is about 35 nm, and all arrowheads on a given filament point in the same direction. In addition, the polarity of the HMM-bound filaments in a given crystalloid or bundle is uni-directional. A mechanism for the formation of the swine crystalloids has been suggested by Toyama (′75), and the results of this study strongly support this hypothesis.Fine filaments of Charcot-Boettcher's crystalloid in human Sertoli cells did not bind HMM. Therefore the fine filaments of the human crystalloid are not actin-like in nature.

    Notes: Two- to four- day- old beagle puppy kidneys were prepared fortransmission and scanning electron microscopy and compared to similarly prepared adult tissues. proximal tubules of puppy kidneys which contained nephrons in various stages of differentiation were examined andmaturational changes were described. Lateral surface contours of proximal tubular cells were initially smooth and relatively unfolded, but progressively acquired complex processes that may be recognized as lateral ridges and lateral-basal processes. Basal projections began as short, stubby processes and gradually took on either a narrow, plate-like or finger-like appearance. Mitochondria, lysosomes and apical vacuoles increased in number as the tubules matured. Mitochondria lacked orientation in outer cortical tubules, but exhibited some vertical arrangement within basal processes in inner cortical tubules. Despite features indicating advanced maturation of tubules in the inner cortex, puppy kidneys lacked the lipid droplets characteristic of the adult. Thus, differentiation of this portion of the developing nephron into S, S2 and S, segments was notpossible at day 4.Morphometric analyses of the lateral and basal membrane surface concentrationof proximal convoluted tubules from the puppy revealed all cells to have significantly smaller membrane area than that of the adult. However, the inner cortical cells of the puppy had a greater surface concentration than those of the outer cortex. The reduced transport capacity of the puppy proximal tubule may be related to the lack of segmentation and/or reduced lateral-basal surface area.

    Notes: The ability of the striated ducts of rat salivary glands to incorporate 3H-fucose into glycoprotein was studied by light and electron microscope radioautography. At 3.5 to 20 minutes after intravenous injection, the majority of the radioautographic grains in the ducts of the parotid gland were localized to the Golgi apparatus. By 40 minutes, the percentage of grains over the Golgi apparatus had decreased; a corresponding increase in grains occurred over small (0.1-0.4 μm) apical granules and the highly infolded basal and lateral plasma membranes. By two hours, less than 10% of the label was associated with the Golgi apparatus, while 26% and 28% were attributed to the apical granules and plasma membrane, respectively. By 8 to 12 hours after injection, the number of grains over the apical cytoplasm had decreased, suggesting luminal discharge of the apical granules. In contrast, the basal and lateral plasma membranes remained labeled up to 30 hours after injection as judged by the distribution of grains in light microscope radioautographs. Mitochondria appeared capable of independent incorporation of fucose, accounting for about 20% of the grains from ten minutes to two hours after injection. Comparable results were obtained in the striated ducts of the submandibular and sublingual glands.These results indicate that the striated duct cells readily incorporate 3H-fucose into newly-synthesized glycoproteins. A portion of these are secretory glycoproteins which are packaged and stored in the apical granules, and a portion are membrane glycoproteins which are incorporated into the extensive plasma membrane of these cells.

    Notes: Regeneration of submandibular gland (SMG) secretory parenchyma is remarkably impaired in salivary gland diseases and under experimental conditions such as in tissue culture and after isografting. In our study acinar regeneration was found to depend on the site where the SMG tissue was implanted. Implantation of several 2-3 mm3 fragments of SMG subcutaneously in the back of the same donor adult male rat resulted in initial necrosis and mononuclear cell infiltration of the autograft. Then there was epithelial proliferation with the appearance within 28 days of lobules which contained numerous duct-like structures and only a few or no acini. In contrast, implanting SMG fragments in the anatomical bed of the donor gland resulted in the appearance of a more differentiated autograft. Although the initial tissue changes were similar to those seen in the autografts in the subcutaneous tissues of the back, the SMG autograft in the neck also contained numerous acini by 42 and 56 days after implantation. These data support the view that the implantation site influences the course of cytodifferentiation in SMG autografts.

    Notes: The distribution of taste buds on the fungiform and circumvallate papillae of the cow tongue has been determined. The two tongues studied were from Holstein-Friesian cows four to six years of age; they contained 14,765 and 21,691 taste buds, respectively. The tip of the tongue is well supplied with fungiform papillae, and the posterior portion contains the circumval-late papillae. The midportion of the tongue contains relatively few taste papillae.The fungiform papillae contained 1,580 and 1,838 taste buds on the two tongues, respectively, and the circumvallate papillae were estimated to contain 13,185 and 19,853 taste buds. The highest concentration of taste buds therefore occurs in the circumvallate papillae; these relatively few papillae contain approximately 90% of the taste buds. On a circumvallate papilla, taste buds are found only on the papillary sidewall, with none either on the apical surface of the papilla or on the outer wall of the moat.

    Notes: The size and distribution of lipoprotein particles in the intestinal epithelium were determined before and after the start of suckling by infant rats. Prior to suckling very low density lipoprotein particles were within the smooth endoplasmic reticulum (SER), rough endoplasmic reticulum (RER), Golgi apparatus and interepithelial cell spaces of duodenal villus tip cells. Chylomicrons were produced in the SER, RER and Golgi apparatus of duodenal epithelial cells after the commencement of suckling.Ileal epithelial cells did not contain particles of VLDL dimensions prior to suckling. Ileal cells of rats suckled for 24 hours after birth had VLDL particles in the SER, RER and Golgi apparatus above and at the sides of the large supranuclear vacuole in each cell. Ileal cells at 10 and 15 days of age had fewer and smaller particles. These particles were also present in the interepithelial cell spaces and lacteals.It is suggested that VLDL particles are produced by duodenal cells prior to suckling when only endogenous lipid is available. The ingestion of exogenous lipids in milk is associated with the production of chylomicrons by duodenal epithelial cells and VLDL particles by ileal epithelial cells. VLDL particles within ileal cells become smaller and less numerous as suckling continues, possibly due to increased pancreatic lipase. This is interpreted as evidence that chylomicrons and intestinal VLDL particles are related.

    Notes: The course, relations and distribution of the inferior alveolar nerve and its branches in the cat are described. The nerves have been studied by dissection, histologically and by using electrophysiological techniques. Dissection revealed a basic pattern on which some individual variation was Superimposed. The inferior alveolar nerve has three branches Supplying thealveolar process (alveolar branches), one branch SUPPLying the canine and incisor region (caninelincisor branch) and four mental branches (posterior, mainand 2 anterior). Fibres Supplying the teeth were found in all except the mental branches. pulpal, periodontal and buccal gingival margin fibres from an individual tooth generally travelled together, but often in more than one branch. Branched axons Supplying both tooth pulp and an area of mental skin were found. The axons branched a t the point of separation of the appropriate mental nerve from the main trunk. A cutaneous midline overlap of 1-2 mm was found, but there was no transmedian innervation of tooth pulps.

    Notes: An investigation was performed on a new strain of genetically hypertensive mice to study those aspects of the renal glomerulus which have in the past been implicated in the etiology of renal parenchymal hypertension. Morphometric analyses were carried out utilizing a computerized graphic data analysing system on information obtained through transmission electron microscopy. Chronically hypertensive animals exhibited thinner basement membranes with numerous sub-epithelial focal thickenings, which were largely absent from the normotensive controls. No difference was noted in the width of the epithelial slit pores (interpedicelar spaces). The glomerular capillary loops of the hypertensive animals appeared otherwise unremarkable, as did the urinary space and parietal epithelium of Bowman's capsule. No evidence of renal parenchymal pathologies implicated in the etiology of systemic hypertension was observed, therefore, these animals would seem to be suitable models for human essential hypertension.

    Notes: In order to investigate whether fibroblasts in rodent periodontal ligament have a structural polarity, the position of the Golgi apparatus or the centriolar region in the cells was studied using light and electron microscopy. It appeared that, in the periodontal ligament of continuously erupting mouse and rat incisors, centrioles in fibroblasts on the tooth side of the ligament are preferably located in the anterior (occlusally directed) part of the cytoplasm. Polarity of fibroblasts in a single direction was less pronounced or absent on the bone side of the tissue. In the mouse, fibroblasts in the connective tissue adjacent to the incisor also contained an extensive system of cytoplasmic microtubules, whereas in the fibroblasts on the bone side of the ligament microtubules were less frequent.Unipolarity of fibroblasts was also observed in the periodontal ligament of the rat maxillary first molar, which is characterized by a limited eruption. Here, the Golgi region was usually situated in that pole of the cells that was directed towards the alveolar wall and the occlusal plane.It is suggested that structural polarity of fibroblasts in the periodontal ligament of rodent teeth is associated with orientation of functional activities of the cells, such as unidirectional movement or unidirectional deposition or phagocytosis of collagen.

    Notes: The innervation of the glomerular arterioles was investigated by light and electron microscopy autoradiography for localization of exogenous tritiated norepinephrine. By light microscopy accumulations of grains were seen associated with afferent arterioles and in lesser numbers with efferent arterioles and neighboring tubules. Accumulations of grains were noted to be in contact with juxtaglomerular granular cells. Electron microscopy autoradiography revealed that nearly two-thirds of the silver grains were on axons. Most of the label was on varicosities packed with small, clear and dense-cored, vesicles. Most varicosities, including those in contact with smooth muscle, juxtaglomerular granular or tubular cells, were labeled. Some varicosities which appeared unlabeled in a given section were labeled in subsequent sections. These findings are consistent with the notion that the glomerular arterioles are innervated mainly by adrenergic nerves. This view is supported by the previously reported observations of the concomitant virtual disappearance of fluorescent and acetylcholinesterase-positive nerves from the region of the glomerular arterioles after two injections of six-hydroxydopamine (a drug which selectively destroys adrenergic nerves) and the presence of small dense-cored vesicles in all axons of the juxtaglomerular region when examined by serial section electron microscopy.

    Notes: The variation in the origin of the (posterior) intercostal arteries was examined in 79 aortae of children who ranged in age from 1 day to 15 years and died of various causes. The distribution, size and number of the origins of the (usually present: 9) pairs of these arteries varied widely. The most commonly observed patterns were as follows: variation in distance between arteries of a given pair; variation in spacing between adjacent pairs; presence of a supernumerary single artery or of a pair; absence of one or both arteries of a given pair; unequal size of the orifices of a pair; complete division of a single artery into two branches beyond its origin; and an incomplete division of a single artery by a partial septum.Of interest appears to be the absence of one (in 59 instances) or both (in 17 instances) arteries of the same pair; the tenth intercostals were most commonly so affected. It would require a prospectively designed study to ascertain whether the absence of any of the intercostal arteries may have a clinical effect in man. Since these arteries supply directly or indirectly not only the intercostal and pectoral muscles, serratus anterior and the muscles of the back, but also the spinal cord, and in addition, provide cutaneous and a few mammary branches, functional correlations in the absence of given arteries may prove to havepractical implications.

    Notes: The histochemical characteristics, cross-sectional area and capillary of the skeletal muscle fibers of the anterior and posterior regions of the superficial masseter and the temporalis muscles are described for juvnile and adult rhesus monkeys of both sexes. Slow twitch fatigue resistant (S), fast twitch fatigue resistant (FR) and fast twitch fatigable (FF) fibers were found in varying proportions throughout the muscles; however some fibers with an intermediate myofibrillar ATPase activity were observed in the anterior masseter. No significant differences for any of the variables were found between male and female juveniles for a specific muscle sample site. However, consideable variation was found between juvenile and adult and between adult male and female monkeys in the percentages of different fiber types and the cross-sectional area of fibers in specific regions of the superficial masseter and temporalis muscles. We conclude from these observations that significant differences in funtion exist both within and between the different masticatory muscles of rhesus monkeys. Functional differences may result from the pronounced sexual dimorphism evident in the dentofacial complex of rhesusmonkey.

    Notes: The distribution of calcium in the enamel organ of the rat incisor was investigated using potassium pyroantimonate for ultrastructural localization of calcium. Substantial amounts of precipitate occurred in the intercellular compartment of the enamel organ and modest deposits were observed in specific organelles of the secretory ameloblast. Mitochondria, nuclei, granular endoplasmic reticulum, Golgi vesicles and secretory granules consistently contained small deposits of pyroantimonate. Complexing of calcium by the pyroantimonate was confirmed by EGTA decalcification and scanning electron microscope energy dispersive X-ray analysis. The observed distribution is discussed in light of potential for an intercellular pathway of calcium transport as well as controlled movement of the ion along the synthetic and secretory route followed by organic components of enamel.

    Notes: Photoreceptor morphogenesis in the sensory retina of chicks of 2 to 20 days incubation age was studied by scanning electron microscopy (SEM) and correlative transmission electron microscopy (TEM). At 9.5 days spherical inner segments extend into the subretinal space (optic ventricle). They are randomly arranged with chiefly smooth surfaces and contain centrioles, polyribo-somes and microtubules. Microvilli project primarily from Muller cells. By the twelfth day immature ellipsoid and myoid regions have formed. Microvilli are abundant on the lateral surfaces of inner segments and extend over the entire spherical surface by the fifteenth day. Occasional cilia with surrounding depressions at their bases were also observed. Inner segments are more symmetrically arranged due to close proximity of photoreceptor cells. Inner segments elongate during the sixteenth day; many display a transitional ovoid form. Microvilli become less numerous but some persist as calycal processes. By the eighteenth day, conical shaped outer segments appear. Thereafter, all photoreceptor cells are comparable to those in the mature retina. Abundant microvilli on the external surface of the sensory retina suggest a supportive role in supplying adequate nutrition to the sensory retina during morphogenesis. The establishment and continual development of the ellipsoid and myoid appearto be primarily responsible for the elongation of photoreceptor cells.

    Notes: Dichloromethylene diphosphonate (Cl2MDP) is a synthetic compound related in structure to inorganic pyrophosphate but is resistant to enzymatic and chemical degradation and is known to be a potent inhibitor of bone resorption. The administration of 20 mg/kg/day of Cl2MDP for ten days to growing rats results in marked increases in metaphyseal mineralized tissue mass due to slowed bone resorption. There was an increase in resorption areas covering anorganic bone viewed by scanning electron microscopy (SEM), however, the resorption pits, or Howship's lacunae, in these resorption areas were smaller and less defined than those encountered in controls. The appearance of these large areas of poorly delineated resorption pits is likely due to an inhibition of bone resorption coupled with slowed bone formation. Administration of Cl2MDP to growing rats also results in an increase in the numbers and size of osteoclasts. Because this would appear to be a histological paradox, in view of the ability of Cl2MDP to slow bone resorption, the osteoclasts were examined by transmission electron microscopy (TEM). The ruffled borders and associated cytoplasmic vacuoles were generally less extensive in the Cl2MDP-treated osteoclasts than in controls, even though clear zones were frequently seen. Examination of undecalcified light microscope sections reveal that the area of bone being degraded by adjacent osteoclasts was generally much smaller in the Cl2MDP-treated animals than in controls. Thus the collaborating TEM observations of smaller ruffled borders, with the SEM observations of smaller, less-de-fined resorption pits, with the light microscope observations of smaller bone areas being degraded by individual osteoclasts provide a morphological basis for the observed decreases in bone resorption following Cl2MDP administration.

    Notes: Previous studies in chick embryos (Goldberg, '77) indicated that unidirectional guidance of retinal axons toward the optic nerve is restricted to the vitread portion of the ganglion cell fiber layer (GCFL) of the retina; random fiber growth was noted after deflection of the optic axons sclerad to the GCFL.The present study on mice confirms these observations. Silver-stained flat mounts of retinal colobomas were examined. Many optic axons in colobomas do not exit normally from the eye, but travel randomly when deflected sclerad to the GCFL. Newborn mouse axons grew around retinal lesions in a highly directed manner. Such axons were always situated in the vitread portion of the GCFL.The unidirectional guidance found in newborn mice was absent in adults. Deflected adult axons traveled randomly regardless of their level within the GCFL. We propose that defective guidance largely accounts for failure of axonal regeneration in the adult mouse retina. The inability of the adult axons to fasciculate (adhere to one another and form fiber bundles) suggests that impaired cellular adhesivity may be part of the mechanism of regenerative failure.

    Notes: After treatment with various chemical and physical agents, flattened or ring-like saccules may occur in the cytoplasm of parietal cells of the gastric glands of several species of mammals. In the current investigation, similar structures appeared after treatment with high dosages of diazo-oxo-nor-leucine (DON), a glutamine antagonist. A tentative sequence for their formation is suggested. Saccules formed of unit membrane became abundant in some parietal cells of the treated mice. Single saccules often had narrow lumens and peripheral distensions. The saccules, either singular or several stacked together, became progressively more curved, enclosing a region of cytoplasm that often contained glycogen-like particles and occasionally vesicles or other organelles. Many of the concentric saccules were close to an intracellular canaliculus. Membrane bound cytoplasm containing glycogen-like particles occasionally occurred in the canaliculi, suggesting that exocytosis had occurred. Cytochemistry revealed that glycoproteins were associated with the concentric saccules, probably located on the luminal surface. The glycogen-like particles in all locations stained in a manner characteristic of glycogen. It is suggested that the concentric saccules may form from, vesicles of the tubulovesicular system.

    Notes: The morphology of early postimplantation mouse egg cylinders was studied using light and electron microscopy. Implantation sites at seven, seven and one-half and eight days of gestation were dissected from the myometrium and whole implants, including both decidua and egg cylinders were processed for electron microscopy. Pre-primitive streak egg cylinders were composed of two germ layers, a tall columnar ectoderm and an outer visceral endodermal layer. Ectodermal cells demonstrated large oval nuclei and an organelle sparse cytoplasm except for many free polyribosomes. The visceral endodermal layer was composed of two cell populations. One visceral endodermal cell type observed was tall columnar in shape and appeared absorptive as demonstrated by many microvilli, pinocytotic profiles and lysosomal granules. This population was confined to extraembryonic regions of the egg cylinder. The second visceral endodermal cell type, squamous in shape, evidenced only a few microvilli, pinocytotic profiles and lysosomal granules. This population was confined to the embryonic region of the egg cylinder. Concurrent with the formation of the primitive streak an increased number of cellular junctions and nuclear pores became evident in the ectoderm. Mesodermal cells were large and stellate-shaped exhibiting many filapodia which made contact with adjacent mesodermal elements. Later the cephalic region of the primitive streak proliferated resulting in the migration of wedge-shaped mass of cells, the head process. At the most ventral extremity of the post-primitive streak egg cylinder the cells of the head process became intimately associated with the ectoderm by areas of focal contact and gap junctions.

    Notes: Perforating canals arise exclusively from junctional canals just above the reserve zone and they do not branch after entering the proliferative zone. They are uniformly spaced and arranged in parallel array. The cartilage canals terminate near the beginning of the zone of hypertrophic cartilage cells. Vascular components within the perforating canals consist of a central arteriole surrounded by enlarged, interconnected capillaries which are individually in contact with the adjacent cartilage matrix. TEM shows that the capillary endothelium is extremely attenuated, possesses numerous fenestrations and lacks a continuous basement membrane. The central arteriole is enlarged through the midpart of the canal and then narrows to communicate with the capillaries near the bottom of the canal. The large capillaries ascend from their point of origin and recombine near the top of the growth plate to exit as a single venule. The vascular arrangement therefore describes a system in which the outgoing blood runs in close proximity, but counter to, the incoming blood. This vascular arrangement within the perforating cartilage canal would most likely allow the zone of maturing cartilage cells to receive the highest concentration of nutrients.

    Notes: The caudal neurosecretory system of the blueback herring, Pomolobus aestivalis, captured in freshwater, was examined. Neurosecretory cells were identified readily in terminal spinal cord segments. The axonal processes of these neurosecretory cells formed a discrete, ventrally placed bundle which terminated in a conspicuous ventral enlargement from the filum terminale. This enlargement comprises the neurohaemal contact zone or urophysis. The neurosecretory terminals were engorged with secretory granules in these fishes from a freshwater environment.The non-neuronal component of the urophysis also was examined in this study. These glial cells were dispersed throughout the urophysis. Most often they were found ensheathing the terminal neurosecretory processes. These cells showed a resemblance to neurohypophysial pituicytes. Although mentioned in the literature, these glial cells have not been examined with the electron microscope. The ultrastructural features of the glial cells and their relation to the neurosecretory constituents are described in this report.

    Notes: Enamel secretion by ameloblasts was investigated in the incisors of 100 gm normal and colcemid-injected male rats. Morphological studies were done on rats given a single intraperitoneal injection of 0.1 mg (1.25 mM) of colcemid and sacrificed 1 to 4 hours after injection. Protein synthesis and secretion were investigated with radioautography in normal and colcemid-treated rats injected with 3H-proline and sacrificed at intervals between 0.5 and 3.5 hours after injection. Colcemid was injected 0.5 hours prior to 3H-proline in each experimental rat. Electron microscopic examination revealed several morphological alterations between 1 and 4 hours after injection of colcemid. These changes included fragmentation of the normally elongated rough endoplasmic reticulum into shorter profiles; a disorganization of thenormally tubular configuration of the Golgi apparatus into a number of separate but intact stacks of Golgi saccules; the disappearance of secretion granules and profiles of smooth endoplasmic reticulum from Tomes' processes; and the accumulation of secretion granules at the mature face ofthe Golgi stacks, as well as in the infranuclear cytoplasm where they are normally not found. Radioautography revealed that protein synthesis by the rough endoplasmic reticulum had continued in colcemid-altered ameloblasts. Labeled secretion granules were found at the mature surface of the Golgi stacks and in the infranuclear cytoplasm, however they did not migrate into Tomes' processes. Consequently, labeled enamel matrix did not appear extracellularly at the same time as in normal controls. Quantitative radioautography in the light microscope revealed that the effect of colcemid, although reversed within 4 hours, had temporarily inhibited normal migration and exocytosis of secretion granules.

    Notes: An ultrastructural study of the intestinal absorptive epithelium in perch (Perca fluviatilis) has shown that the perch intestine can be divided into three segments: the proximal segment, the middle segment and the distal segment. The enterocytes of the proximal segment are found to be concerned with lipid absorption. The absorbed fat gives rise to the presence of two forms of inclusions: lipid particles and lipid droplets. Enterocytes of the middle segment exhibit the typical ultrastructural features of pinocytosis; these consist of extensive invaginations of the luminal surface membrane and accumulation of vacuoles in the apical cytoplasm. Exogenous proteins are ingested by absorptive cells from the intestinal lumen by a process similar to that described in neonatal mammals. In the distal segment the absorptive cells have few, short microvilli.Besides the absorptive epithelial cells, goblet cells, endocrine cells, pearshaped cells, and plasma cells are occasionally found.

    Notes: Differences in the exposure of spermatozoa surface components during epididymal passage have been examined using lactoperoxidase-catalyzed 125I-iodination or labeling with 125I-diazodiiodosulfanilic acid. Labeled surface proteins obtained from caput and cauda epididymides were solubilized in detergent, separated by sodium dodecylsulfate polyacrylamide slab gel electrophoresis, and identified by radiography. Densitometer scans of autoradiograms revealed increased amounts or exposures of surface proteins of ∼35,000, ∼39,000, ∼50,000, and ∼78,000 molecular weight on the cauda epididymal spermatozoa.

    Notes: Acid nitrophenyl phosphatases from sea urchin eggs and embryos were investigated by gel filtration. Four different forms were found in Hemicentrotus pulcherrimus, and three forms in Anthocidaris crassispina and Pseudocentrotus depressus.The first and second forms (designated AcP-1 and AcP-2) had the highest activity in the range of pH 5.6-6.0. The third (designated AcP-3) had an apparent optimum pH between pH 4.3 and 4.8, and the fourth (designated AcP-4) showed the maximum activity at pH 3.0. AcP-1 was much more thermolabile than AcP-2 and AcP-3 at 56°C. NaF inhibited AcP-2, AcP-3, and AcP-4 but not AcP-1. AcP-1, AcP-2, and AcP-3 were observed in the three species, whereas AcP-4 was not detected in A. crassispina and P. depressus.     Notes: Suspensions of nearly 100% viable golden hamster sperm were prepared by passing washed cauda epididymal sperm through a column of 0.25-0.3 mm glass beads. Incubations of these viable sperm under in vitro capacitation conditions in volumes of 0.1 or 1 ml (2-2.5 × 106/ml) resulted in 85-92% viable sperm after four hours and 45 minutes of incubation. More than 70% of these sperm were judged to have been capacitated after four hours and 45 minutes of incubation on the basis of their having undergone acrosome reactions and the presence of high numbers of sperm exhibiting the activated motility characteristic of capacitated hamster sperm.Thus, for the first time, procedures are available that will yield large numbers of viable capacitated sperm for biochemical analysis and that will also allow other studies of hamster sperm capacitation with minimum interference from molecules released from dead sperm.

    Notes: The external structure of the male gamete of Mytilicola intestinalis is studied under a scanning electron microscope and compared with transmission electron micrographs of thin sections corresponding to the different parts of same. The cell in question is long and filiform, showing, along a significant part of its length, two ridges or expansions helicoidally arranged and diametrically opposed. Four different parts or segments can be identified in this spermatozoon: segment A, characterized by its screw-like aspect; segment B, the longest, provided with well-developed helicoidal expansions; segment C, showing an uneven surface and lacking expansions; and segment D, with a smooth surface and decreasing diameter. The significance of this original structure in a spermatozoon, considered immobile until now, is discussed, stating different hypotheses with regard to the possible mobility of the cell just before fertilization takes place.

    Notes: The second meiotic division and polar body formation in mouse eggs fertilized in vitro were observed by phase-contrast and polarizing microscopy, and recorded by time-lapse cinematography. Eggs were collected from oviducts of mice that had been superovulated by injections of PMS and HCG. Some eggs, inseminated with spermatozoa that had been collected from caudae epididymides of mature male mice and cultured for two to three hours before insemination, were observed continuously on a glass slide under a phase microscope. Other eggs were inseminated in Petri dishes in a 5% CO2 incubator and examined every 20 minutes for 180 minutes. Compatible results in both sets of eggs showed that formation of the second polar body began 25-40 minutes after fusion of spermatozoon with the vitellus; it was completed 40-60 minutes later; anaphase II lasted approximately five minutes before the appearance of the furrow abstricting the second polar body. It is suggested that the furrowing associated with second polar body formation is guided by the same kind of forces that divide a cell mitotically.

    Notes: The effect of urethan on artificial induction of cleavage in eggs of the sea urchin, Hemicentrotus pulcherrimus, was studied. When the eggs were exposed for 20 minutes to seawater containing urethan (final concentration, 0.08 M) after butyric acid-activation and then treated with hypertonic seawater, the cleavage rate was enchanced by about 1.5 times as compared with the nontreated eggs. In the eggs exposed to urethan-seawater for over 70 minutes many clear spots appeared throughout the cytoplasm. Simultaneously, the pigment granules, which had been embedded within the cortex, migrated to the inner cytoplasm and encircled a monastral centrosphere and clear spots. The clear spots were composed of microtubules much like cytasters, and in the central region of them centrioles were not yet found. The eggs in which the pigment granules disappeared from the cortex may be more susceptible to cleavage induction by succeeding hypertonic treatment.

    Notes: The motility of rabbit spermatozoa recovered from the vagina, endocervix, uterus, and four regions of the oviduct was assessed visually by phase-contrast microscopy at intervals from one minute to 16 hours after a single mating. The percentage of motile cells in each sample was dependent on the temperature of recovery, ie, 23° vs 37°C, but was not influenced by the temperature of observation. Spermatozoa in the lower isthmus of the oviduct were the most temperature sensitive population to recovery at 23°C. When all manipulations and observations were performed at 37°C, the percentage of spermatozoa with progressive motility varied according to the region sampled and interval after mating. Populations from the vagina, uterus and upper regions of the oviduct usually had a high proportion of progressively motile cells with vigorous flagellar activity. Fewer spermatozoa showed progressive movement on recovery from the endocervix and lower 2 cm of the tubal isthmus and their flagellar activity was generally depressed. The decrease in flagellar beat frequency noted in the latter regions may be a major factor limiting sperm ascent in the female tract. A unique pattern of “activated” motility was seen exclusively in populations taken from the oviducts at 6 to 16 hours after mating. This motility pattern, consisting of alternating episodes of linear progressive and vigorous nonprogressive movement, may be analogous to the activated motility described for capacitated rodent spermatozoa.

    Notes: The sperm-specific proteinase acrosin (EC 3.4.21.10) is found in spermatozoa as a zymogen. We have looked for different forms of this zymogen in testicular, epididymal, and ejaculated spermatozoa from ram and have compared total sperm extracts made immediately after cell disruption with extracts made later from isolated sperm heads. We have concluded that the autoactivatable zymogen form, known generally as proacrosin, is the only form of acrosin within intact mature ram spermatozoa; no other zymogen form was detected, although lower levels of proacrosin were found in some samples of testicular spermatozoa. From studies of the activation process, it appears that ram proacrosin is truly autoactivatable; no evidence could be found for the involvement of any auxiliary enzyme. Estimations of the molecular weight of proacrosin using gel chromatography (60,000) and SDS-polyacrylamide gel electrophoresis (51,300) indicated that the zymogen is monomeric. Comparison with the molecular weight of ram acrosin (44,000 or 40,000, using the two respective methods) indicated that a single acrosin molecule is derived from each zymogen molecule. The sperm acrosin inhibitor (molecular weight 11,000 or 8,000) was present in testicular spermatozoa as well as in ejaculated spermatozoa; there was no evidence that it was produced as a result of zymogen activation.

    Notes: In various papers on the original spermatozoon of the Ostracoda, its helicoidal disposition has been indicated as the principle characteristic of this gamete, at cell structure level as well as in its external morphology. Through a combined study with scanning electron microscope (SEM) and transmission electron microscope (TEM), we have been able to establish the corresponding relationship between the cell architecture and the spermatozoon's external morphology. In the case of Heterocypris incongruens, the helicoidal relief of the gamete's external surface along the greatest part of its length, is the result of the twisting and undulating of a structure derived from the nucleus' external membrane, endoplasmic reticulum, and Golgi apparatus, called “feather-like organelle.” In keeping with the shape of this surface relief, the spermatozoon can be divided into three regions: An anterior one with a corkscrew form, a middle one showing a relief in the form of a screw with four threads, and a posterior or tail one without helicoidal relief. Finally, we discuss the different criteria existing on the possible orientation of this spermatozoon when it moves, as well as the functional advantages that the possession of a filiform, helicoidal, and mobile gamete represents.

    Notes: Immature rats, aged 27 days, were stimulated to develop preovulatory follicles by subcutaneous injection of 15 IU of pregnant mare serum gonadotrophin (PMSG). Two days later their oocytes were collected. They were cultured under conditions that permitted continuous observation. Times of the initial stages of maturation were carefully noted, in the absence and the presence of 10 μg/ml of bovine luteinizing hormone (LH). LH did not accelerate germinal vesicle disappearance. It was concluded that the immature PMSG-treated rat was not an appropriate model for the study of LH action; it was speculated that persistence of PMSG mimicked LH in all the oocytes from such donors.

Among them, two chymotrypsin-specific inhibitors, chymostatin and N-tosyl-L-phenylalanyl-chloro-methane, as well as p-nitrophenyl p′-guanidinobenzoate, inhibit fertilization of the sea urchins, Hemicentrotus pulcherrimus and Strongylocentrotus intermedius.A chymotrypsin-like protease is presumed to be a lysin of the sea urchins, since the inhibition of fertilization by chymostatin is remarkably diminished if the eggs are pretreated with trypsin or chymotrypsin to break the vitelline coat before insemination, and since N-tosyl-L-phenylalanyl-chloromethane, and p-nitrophenyl p′-guanidinobenzoate, as well as chymostatin, inhibit the fertilization.In all the sea urchins so far studied, elevation of fertilization envelopes is inhibited by leupeptin, antipain, soybean trypsin inhibitor, and p-nitrophenyl p′-guanidinobenzoate, all of which are potent trypsin inhibitors.Synthetic inhibitors have cytotoxic side effects on the eggs, but the microbial and plant inhibitors have no such effects.

    Notes: Ejaculated rabbit spermatozoa washed with buffer prior to decondensation by Triton X-100 and dithiothreitol were good templates for DNA synthesis by Escherichia coli DNA polymerase. This result agrees with the observations of Zirkin and Chang [1977], and implies that the sperm DNA is nicked. Template activity, however, was reduced if spermatozoa were extensively washed before decondensation, and if DNase inhibitors EDTA or Na2SO4 were present during decondensation. Template activity was also low if decondensation was induced with DNase inhibitors thioglycollic acid, Na2SO3 or sodium dodecylsulphate and dithiothreitol instead of with Triton X-100 and dithiothreitol. Calf thymus DNA was completely degraded when incubated with rabbit seminal plasma or buffer-washed spermatozoa, but much less degradation was observed if EDTA, Na2SO4, thioglycollic acid, Na2SO3 or sodium dodecylsulphate were also present, or if spermatozoa were extensively washed with buffer. Centrifugation of spermatozoa through 2.05 M sucrose completely removed contaminating DNase, and such spermatozoa were inactive as DNA templates after decondensation. The DNA template activity of swollen rabbit sperm nuclei thus parallels the activity of a contaminating seminal plasma DNase. This suggest that the nicks in sperm DNA enabling it to act as a template for DNA synthesis were generated by the DNase during decondensation and thus are not a natural structural feature of the DNA. The presence of breaks in the DNA of decondensed buffer-washed spermatozoa (DNase contaminated) was confirmed by their incorporation of phosphate from [γ-32 P] ATP in the presence of the enzyme polynucleotide kinase. These spermatozoa were found to contain as few as two breaks/mole of DNA, but sucrose-washed spermatozoa (DNase free) were free of breaks. The possible use of this enzymic procedure for the assessment of sperm genome damage and the evaluation of the quality of a sperm population are discussed.

    Notes: The structure of rabbit, fowl, and Xenopus laevis sperm chromatin was explored by study of the reaction of their decondensed nuclei with DNase 1 and micrococcal nuclease. Those of rabbit and fowl were readily digested by DNase 1, and the polyacrylamide gel electrophoresis profiles of DNAs extracted from the digests were similar, each being polydisperse with a single discrete band of DNA smaller than 72 base pairs. There were differences, however, between the sperm chromatins in the course of their digestion by micrococcal nuclease. A limit digest at about 45% acid solubility was obtained with Xenopus sperm chromatin, while 90% of fowl sperm DNA was rendered acidsoluble by the enzyme. The gel profiles of the limit digests were polydisperse, but only those of rabbit and fowl sperm chromatins possessed a discrete band of DNA smaller than 72 base pairs. Bleomycin did not react with DNA of rabbit, fowl, or Xenopus spermatozoa. Since bleomycin reacts with somatic cell chromatin, and the course of DNase 1 or micrococcal nuclease digestion of sperm chromatin was different from that found for somatic cell chromatin, it would appear that sperm chromatin does not have the repeating nucleosometype structure of somatic cell chromatin. The nuclease digestion studies further suggest that the organization of rabbit and fowl sperm chromatins is similar, and is different from that of Xenopus sperm chromatin. The dependence of the structure of sperm chromatin on the composition of its basic proteins, and a possible structure for a protamine-type sperm chromatin, are discussed.

    Notes: Sperm penetration through the zona pellucida and fusion of the sperm head with the vitellus were observed continuously and filmed under phase optics in cumulus-free living mouse eggs inseminated in vitro with capacitated epididymal sperm. Most spermatozoa penetrated the zona pellucida, traversed the perivitelline space, and fused with the vitellus at an angle nearly perpendicular to the surface. The mean duration required for sperm to penetrate the zona pellucida was 20 minutes with a range of 15-26 minutes. Sperm traversed the perivitelline space in less than one second. The initial contact of sperm with the vitellus generally took place at the tip of the sperm head. When the tip of the sperm head contacted the vitellus there was an immediate reduction in the rate of flagellation, followed by the gradual sinking of the sperm head into the vitellus.

    Notes: Mature unfertilized ova from superovulated hamsters were freed from all investments and frozen at -50°C. They were cooled at about 1°C/min to 0°C then at 0.8° to 0.6°C/min to -50°C. At 0°C, dimethyl sulfoxide was added to a final concentration of 1.25 M. The ova were stored at -50°C for up to four months. Thawing was performed at 2-4°C/min and followed by several washes with insemination medium. Approximately 90% of the ova were normal in appearance after thawing. The frozen and thawed ova with normal appearance could be penetrated by hamster or human spermatozoa at a rate comparable to unfrozen controls. The ability of hamster ova to tolerate storage at a relatively convenient temperature (-50°C) for long periods (tested for up to four months) makes possible their shipment at low cost to institutions lacking this resource. There they can be used for basic biological studies of sperm-egg interaction or in the clinical assessment of human sperm quality.

    Notes: Significant release of the acrosomal enzymes arylsulfatase, β-N-acetylhexosaminidase and hyaluronidase was observed following the treatment of ejaculated rabbit spermatozoa for 12 hours in 20% rabbit serum for inducing in vitro capacitation, and these sperm were capable of in vivo fertilization; however, the treatment of sperm for 15 minutes in high ionic strength (380 mOsm/kg) or low ionic strength medium (305 mOsm/kg) for in vitro capacitation did not result in any significant release of the above enzymes nor were the sperm capable of in vivo fertilization. Serum-treated spermatozoa remained significantly motile following the 12 hour treatment, 51% underwent the acrosome reaction and were capable of fertilizing 66% of the ova in vivo. Identical serum treatment of lysosomes from rabbit liver resulted in a comparable release of the lysosomal enzymes. Serum treatment for in vitro capacitation resulted in vesiculation of the anterior margin of half the spermatozoa, but left their inner acrosomal membranes and equatorial segments intact. A biochemical relationship between the release of acrosomal enzymes and capacitation is suggested.

    Notes: Sperm of Hemicentrotus pulcherrimus undergo the acrosome reaction in the jelly coat or on the surface of the vitelline layer of Pseudocentrotus depressus eggs and can fertilize the latter, whereas those of P depressus do not undergo the acrosome reaction even after they strike the vitelline layer of H pulcherrimus eggs and cannot cross-fertilize.In the latter combination, however, if cross-insemination is performed in the presence of homologous egg water, cross-fertilization becomes less difficult than in normal seawater, and percentage cross-fertilization rises as does percentage acrosome reaction. It is suggested that the cross-fertilizability of this combination depends highly on the inductivity of the acrosome reaction. The reason why such a causal relation is observed is discussed.

    Notes: In the course of the reorganization and degeneration of the proximal centriole in the mature acentriolate spermatozoon of the Mongolian gerbil, both the proximal and distal centrioles appear in the early cap phase of spermatid development. During the acrosome phase, both distal and proximal centrioles become highly active in the formation of a segmented column. The proximal centriole becomes actively involved in the formation of the capitulum, while the distal centriole forms the axonemal complex and dense fibers. During the maturation phase of spermatid development, the “pinwheel” arrangement of the proximal centriole becomes an “S”-shaped structure, turned 90° on its vertical axis. The few “doublet” microtubules that can be detected later in that stage completely disappear during spermiation. The distal centriolar area develops a single central pair of microtubules and membranous elements. Another prominent feature in the neck region of the gerbil spermatozoa is the presence of two dense rudimentary columns in association with the mitochondria. Although their density is similar to that of the other columns, these two columns have no connection with the dense fibers; in fact, they are closely associated with the mitochondria.

    Notes: Ejaculated spermatozoa of boars, bulls, rabbits, and rams were embedded in glycolmethacrylate and thin sections stained with phosphotungstic acid at low pH in order to observe the distribution of glycoproteins of the plasma membrane. Colloidal iron hydroxide was also used to detect the free acidic groups present on the sperm surface. Species-specific patterns of localizations of glycoproteins and linked negative charges were observed. The distribution was sometimes homogeneous as in bull, but generally heterogeneous in the other species. The significance of the results on sperm surface components and the practical interest to know their normal distribution are discussed.

    Notes: Ejaculated spermatozoa of boars, bulls, rabbits, and rams were fixed with glutaraldehyde and embedded in glycolmethacrylate. Thin sections were treated with phosphotungstic acid at low pH in order to localize cellular glycoproteins stained by the PAS technique at the light microscope level. At the ultrastructural level the glycoproteins were segregated in the anterior segment of mature spermatozoa. The distribution of these glycoproteins in the anterior segment was not homogeneous; it appeared species-specific in detail, but as a rule, the maximum concentration was observed in a superficial layer, especially in the marginal thickening. The localization of other acrosomal components (eg, crystalline and basic proteins) is also reviewed. The origin and significance of the segregation of proteins and glycoproteins in the acrosome are discussed in relation to the fact that the acrosomal enzymes analyzed so far are glycoproteins.

    Notes: Embryo transfer experiments were carried out to study the developmental capacity of cultured rabbit embryos when transferred to recipients of variable postovulatory maturity. Rabbit embryos were flushed from the oviduct at 26 hours postcoitum (pc) and cultured in a modified Ham's F-10 medium supplemented with bovine serum albumin (BSA) for a period of 70 hours. At 96 hours pc the cultured embryos, which ranged from the early morula to the expanding blastocyst stage, were transferred to pseudopregnant recipients mated to vasectomized males 36 to 96 hours prior to the transfer procedure. Greatest embryo survival occurred when transfers were made to either the oviducts or uterine horns of recipients at 48 hours pc. Intermediate results for both implantation rates and number of young born were obtained with recipients at 36, 60, 72, and 84 hours pc. Transferred embryos consistently failed to survive the uterine environment of recipients 96 hours pc at transfer although this group was synchronous with embryonic chronological age. Oviductal transfers were generally more successful than uterine transfers. Markedly higher rates of embryo survival resulted from embryos that were collected 60 and 72 hours pc and transferred directly to synchronous recipients without an interim period of culture. Dissimilarity of development for in vivo grown rabbit embryos and those cultured in synthetic medium is demonstrated.

    Notes: The method of in vitro fertilization was applied to test a previous suggestion that the lowered fertilizability of the tubal oocytes of female KE strain mice and the high resistance of their zona pellucida to proteolytic enzymes, are due to the premature cortical reaction taking place near the time of ovulation. Therefore higher fertilizability of ovarian oocytes is expected.The effectiveness of F1 hybrid sperm penetration into ovarian and tubal KE oocytes confirmed these assumptions. The ovarian KE oocytes recovered 9-10 hours after administration of human chorionic gonadotropin (HCG) showed significantly higher penetrability (70-83%) than did the tubal oocytes recovered 12 hours after HCG (about 50%) and 14-16 hours after HCG (20%).Similar results were obtained with C57 oocytes. Sperm penetration into ovarian oocytes (10 hours after HCG) was much more effective (67%) than into tubal oocytes (18%); this finding correlated with more rapid zona dissolution by chymotrypsin. On the basis of these results one might speculate that premature cortical reaction takes place also in the C57 strain.

    Notes: When female Xenopus laevis are injected with [3H]-vitellogenin or [14C] N-acetyl glucosamine, most of the labeled material becomes associated three days later with oocytes having a diameter of 0.9-1.1 mm; smaller and larger oocytes are less labeled. With time, the pattern of labeling shifts to larger oocytes, indicating that those oocytes initially labeled continue to grow. We have measured such shifts as a function of time to provide estimates for oocyte growth rates from the end of stage III (diameter = 0.6 mm) to stage VI (diameter = 1.2 mm). The total time required for oocytes to progress through this size increase is 16-24 weeks in unstimulated females and 9-12 weeks in human chorionic gonadotropin (hCG)-stimulated females. The fastest rate of growth occurs from mid-stage IV (approximately 0.8 mm diameter) until midstage V (1.2 mm diameter), which corresponds to the period of most pronounced vitellogenin uptake. The relative proportion of oocytes within this size range is also reduced, as predicted under steady-stage conditions. Evidence is also presented which indicates that the steady-state level of full-grown oocytes is maintained by a combination of replenishment and atresia. These results provide the first description of the kinetics of oocyte growth in X laevis females maintained under normal laboratory conditions and should be useful for any considerations of macromolecular events occurring during oogenesis.

    Notes: Finite element equivalents of the equations governing shearing and buoyancy driven flows are derived, and reduced to upwind forms suitable for the solution of problems in which the Reynolds and Rayleigh numbers are large. A modification to the central difference iterative method is studied which increases the Reynolds and Rayleigh numbers for which a central difference form may be used. A comparison is made between the results obtained using the central and upwind forms of the finite element method and those predicted by finite difference methods in the case of flow in a cavity. A mesh refinement study is made. The upwind forms of the finite element equations are applied to the solution of a complex flow problem involving the flow of glass in a throated furnace in the case of constant- and temperature- dependent viscosity and conductivity.

    Notes: In Reference 1 a class of first-order factorization methods for the solution of large, sparse systems of equations, of which the coefficient matrix is a symmetric M-matrix, was introduced. Asymptotic results for the computational complexity was proved in the case of systems arising from finite difference approximation of second-order self-adjoint elliptic partial differential equation problems in two dimensions with Dirichlet boundary conditions. In this paper the result is extended to cover even mixed boundary conditions and problems with discontinuous material coefficients. Results from numerical experiments with various finite difference and finite element approximations are presented and comparisons with direct and other interative methods are made.

    Notes: This paper describes a numerical procedure for the solution of metalworking problems of a strain-hardening material. The overall metalworking process is split up into a number of stages and the governing equations for a typical stage are set up in terms of incremental displacements. The incremental deformation occurring in each successive stage is found by an interative numerical procedure based on the concept of piecewise linearization. The ‘composite’ solution yields the distribution of stress and strain in the work material at various stages of the process. Finally, application of the method to finite axisymmetric compression is described.

    Notes: A series of open-type elements which are compatible with axisymmetric thin shell elements have been dervied. These elements allow the effects of intermediate openings and discrete support systems for rotational shells to be realistically modelled in the dynamic regime. Also, consistent load vectors for several common cases are derived.The availablility of these elements in a public domain computer program enhances the possibility for modelling systems which are basically axisymmetric but which include intermediate or end supports using a rotational shell code.

    Notes: The problem of non-radial loading (in the stress space) of structures of elastic-plastic metals is discussed. A concept of combined isotropic and kinematic hardening is proposed. For a material displaying such mixed hardening, constitutive equations are derived and incorporated in a finite element program for the analysis of elastic-plastic 2D-structures. When determining the numerical solution of the non-linear equations, the program enables the investigator to select an optimal combination of step by step and iteration procedures. The computed results are compared with experimental results and with results from calculations based on the overlay model, Reference 25.

    Notes: Orthogonal base functions that are complete first-order polynomials but contain higher order terms are developed for normalized rectangular and triangular elements. To obtain exact expressions for kinetic energy under rigid body rotation, an internal node is introduced at the centroid of each element. Then the corresponding mass matrix is both diagonal and consistent. A mixed variational formulation is used in the derivation of spatially discretized field equations. Central Difference time integration is utilized to provide sample results which are compared with other numerical or analytical solutions.

    Notes: It is shown here that certain interpolating polynomials of degrees four and five may not always be uniquely defined on triangular-shaped elements which have one curved side. Conditions which indicate non-uniqueness are given, together with some geometrical interpretations concerning the location of the node on the curved side. A numerical example is given to demonstrate that there ar curves for which every point is unsuitable to be chosen as a node.

    Notes: A numerical model is proposed to simulate the space-time behaviour of a low-density plasma in a linear multipole. Semi-discrete Galerkin techniques are used in conjunction with triangular finite elements. For both space and time discretizations, special techniques are developed to numerically separate the effect of rapid diffusion along the field lines from the slow perpendicular diffusion. Numerical results are exhibited to show how these techniques succeed in eliminating the problems caused by the strong anisotropy of the diffusion coefficient in the strongly curved and very inhomogeneous multipole configuration.

    Notes: An isoparametric Hermite cubic element is developed for calculating compressible flows past complex aerofoils in two dimensions. A simple intermediate transformation is used to derive an isoparametric transform whose Jacobian is always non-zero and which attains very high accuracy on the boundary. Application of the method and assessment of the error is illustrated by flow past a circular cylinder.
