    Notes: The enzyme β-galactosidase (lactose) obtained from several microbial sources was immobilized on zirconia-coated porous glass particles. The immobilized enzymes were characterized by determining pH profiles, kinetic constants, thermal profiles, and operationalhalf-lives in lactose and whey ultrafiltrate solutions. Studies were carried out on continuous reactor performance, and enzyme requirements for scale-up were estimated. Lactose or whey hydrolyzed by this technique could find use commercially as a sweetener in a number of dairy products.

    Notes: The kinetic behavior of water insoluble enzyme columns was studied using the hydrolysis of GPNA (N-glutaryl-L-phenylalanine-p-nitroanilide) by α-chymotrypsin as a model. Thermal denaturation of soluble and insoluble enzyme was compared in a batch reactor and in a tubular reactor. Convection of metabolites, enzyme reaction, diffusion of metabolites across porous particles and the kinetic behavior of a water insoluble enzyme column were tested.The respective and mutual effects of the first two phenomena were comprehensively analyzed for both stationary state and transient conditions. When the ingoing substrate concentration was varying with time, convection-reaction equations were solved by digital computation. The possible effects of diffusion limitations and unstirred layers are discussed.

    Notes: Changes in the catabolic pattern of Saccharomyces cerevisiae, growing in continuous culture, were effected by altering the glucose feed rate or the dissolved oxygen concentration. The cytochrome concentrations and the adenosine phosphate pool level of the yeast in a series of steady states and during three transitions were measured and compared with the glucose uptake rate (QG), the respiration rate (QO2), and the rate of ethanolic fermentation (QE).Respiration was decreased at high glucose feed rates only if oxygen was low but cytochromes were glucose repressible at both high and low oxygen concentrations. In the main, QE and the levels of ATP, ADP, and AMP were decreased and cytochrome concentration were elevated at low QG values. No consistent relationship between any of the adenosine phosphate parameters and QO2 was discernible.Evidence is presented for the concept that the QG directly controls the adenosine phosphate pool level and that a relationship between the concentration of adenosine phosphate anhydride bonds and the adenosine phosphate level is constantly maintained.

    Notes: The immobilization of glucose oxidase and catalase by adsorption within the pores of controlled-pore titania has yielded a remarkably stable enzyme system. Catalase apparently acts as both a stabilizer and an activator for glucose oxidase within the pores of this material. Hydrogen peroxide concentrations and flow rates have a marked effect upon the apparent activity of the immobilized enzyme system. The carrier parameters were varied to obtain optimum loading and stability information.

    Notes: β-galactosidase from E. coli (β-D-galactose galactohydrolase, EC 3.2.1.23) has been entrapped in a crosslinked 2-hydroxyethyl methacrylate gel with a 35% retention of activity. The kinetic behavior of the gel-entrapped enzyme has been studied in a recirculation reactor system, the substrate being o-nitrophenyl-βhyphen;D- galactopyranoside. Kinetic constants were determined for particle sizes ranging from 69 to 231 μm in diameter and compared to those of the free enzyme. External diffusion effects were eliminated by operating at high recirculation flow rates. A fourfold increase in Km(app) was observed for the 231 μm particles, consistent with existing theoretical treatments for internal diffusion effects.An Arrhenius plot of rate data showed significant curvature at higher temperatures, which was attributed to the effects of internal diffusion. The pH-activity profile of the gel-entrapped enzyme was bell-shaped at high substrate concentration and, in contrast to the free enzyme, could be fitted to the titration curve of two ionizable groups, a basic group having a pK of 8.6. The gel-entrapped enzyme had a higher pH optimum and retained a larger percentage of its maximal activity at alkaline pH than the free enzyme; its pH stability at high pH was also much better.The thermal stability of the gel-entrapped enzyme was studied and found to be 14 days at 22°C and 65 min at 45°C.

Maximal rates at which E. coli can grow on glucose as carbon source under various limiting oxygen-supply conditions were determined in a bench-scale fermentor.     Notes: Many enzyme-catalyzed reactions involve the liberation or consumption of hydrogen ions. In this paper a mathematical model is employed to investigate how such reactions behave when the enzyme is immobilized. Shifted pH optima, disappearance of an optimum pH, insensitivity to bulk pH, and very large effectiveness factors are some of the phenomena which appear as a result of pH coupling between the reaction and the enzyme's activity. Several of the qualitative features revealed by the model are consistent with earlier experimental observations. In addition, preliminary guidelines for optimal choice of enzyme support are suggested.

    Notes: Partially purified lactoses (β-D-galactoside galactohydrolase, EC 3.2.1.23) from Aspergillus niger, Ladobacillus helveticus, and Saccharomyces lactis were immobilized on diazotized porous glass particles (mean pore diameter, 86.5 nm: particle size diameters, 75-125 μm). In acid whey containing 4-4.5% lactose, A. niger lactase gave the highest activity (89 μmoles lactose hydrolyzed/g glass, min) at 55°C and pH 4.5. Glass-immobilized A. niger laclases (lactase-BG) retained much hydrolytic activity after storage and periodic use for 165 days at 55°C. For values of X greater than 30%, hydrolysis of 0.12M lactose in acid whey by a continuous flow column packed with 2 ml of lactase-BG particles could be correlated by X = 17.2(V/F) + 12.5 where X = lactose hydrolysis, percent of lactose originally present; V = volume of packed bed of lactase-BG, ml; F = flow rate of acid whey, ml/min.

    Notes: In an effort, to develop comprehensive mathematical models for use in optimizing fermentation processes, product rate data taken at constant pH's for the lactic acid fermentation (Lactobacillus delbrueckii) were analyzed. It was observed that the rate of acid synthesis-time trajectories exhibited a “shoulder” effect at pH's less than 5. That is, a nearly constant rate of synthesis for up to 10 hr in the late growth phase, out of a maximum total fermentation time of 70 hr. This effect was used as a clue from which to structure the proposed model with parallel pathways. Simulating shunting pathways does, in fact, demonstrate that, the notion of parallelism is consistent with the expression of a shoulder. It is postulated, therefore, that a differential pH effect between two parallel pathways can account for the presence of a shoulder when both routes prevail, and no shoulder when either pathway predominates.

    Notes: Maltase, phosphatase, malate dehydrogenase, glucose-6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase activities were determined in the supernatant of brewer's yeast cells from samples taken at various intervals during a 90 min treatment in a Dyno-Mill disintegrator.In an attempt to find optimum operating conditions, the following parameters were varied: speed of rotation of the agitator, flow rate, yeast concentration in the slurry and bead diameter, and the specific rate of extraction was determined for each such variation. Analysis of the data showed that, superimposed upon extraction, there was a progressive but slow loss of total activities. Enzyme release could be expressed as an approximately first-order chemical reaction. The results compared well with those of Follows et al. (Biotechnol. Bioeng.), 13, 549 (1971) and showed that the Dyno-Mill can be operated under proper conditions without significant loss of enzyme activity and with high yields.

    Notes: The complete mixing activated sludge (CMAS) system is gaining in popularity for treating both domestic and industrial wastewaters. Experience over the past 20 years has produced a simple mathematical model which can be used in both the design and the operational evaluation of CMAS systems. Laboratory pilot plants and full scale field units have furnished the basic data needed to confirm the validity of the mathematical model. The basic concepts of the model are discussed in light of field evaluations. Areas of conflict and confusion which have arisen in the past will be presented and discussed. Design examples and operational evaluations are presented for several different wastewater systems.

    Notes: Anaerobic fermentation of organic solid waste can provide a significant source of fuel gas (methane). Application of this process requires a better understanding of the kinetics of the biological system. The literature is replete with kinetic studies of this process as applied to waste solids from water pollution control systems. Much of this work has been conducted in the mesophilic temperature range. Increased temperatures yield higher reaction rates that will improve the economics of the process. The rate limiting step in the fermentation of refuse is the hydrolysis of the complex organic solids, in particular cellulose. Cellulose is a major component of the refuse. A laboratory study employing domestic refuse has shown the effect of temperature on the rate of methane fermentation. The optimum mesophilic temperature was found to be 42°C, while the optimum thermophilic temperature was at least 60°C. No data was obtained beyond the 60°C temperature. Reaction rate constants are presented for anaerobic fermentation of domestic refuse. Because of the characteristics of the substrate it-was not possible to obtain the necessary measurements for evaluation of constants in the Monod model. An overall system constant was developed.

    Notes: Conventional sources of protein cannot meet the present or projected needs for human consumption. Single cell proteins from fermentation of petroleum and cellulosic wastes are likely sources of additional protein. The volume of cellulosic wastes is sufficient to supply all additional protein needs on a continuing basis for cellulose is a renewable resource. Both mesophilic and thermophilic microorganisms utilize cellulose at reasonable rates. Biodegradation of lignin and lignin-cellulose complexes constitutes a major obstacle to commercial utilization of cellulosic wastes. Thermophilic actinomyces appear to be the most effective organisms for single cell protein production from cellulosic wastes.

    Notes: The yeast Candida tropicalis utilizes both glucose and hydrocarbons as sole carbon sources. When grown on hydrocarbons, the cells contain twice as much lipid as when grown on glucose. In transient continuous culture experiments, following a substrate change from glucose to hexadecane, an adaption phase occurred. During this phase the lipid concentration per cell increased greatly. It is proposed that a high cellular lipid concentration is necessary for hydrocarbon assimilation, and this is not just a reflection of the lipophilic nature of the substrate.

    Notes: Efforts were made to eliminate the influence of other factors as far as possible in order to obtain reliable results on the effects of oxygen on the growth of baker's yeast. A cultivation method is presented which permits the study of the effects of aeration intensity under conditions where the influence of catabolite repression is eliminated. A completely synthetic medium with glucose as the only carbon and energy source is also described.The capacity of yeast to perform aerobic metabolism varies when cultivated under different intensities of aeration. A clear maximum is observed for growth with 10% oxygen in the aerating gas mixture. Under conditions where catabolite repression does not function yeast has the potential for oxidative metabolism even under oxygen-limited growth. The main agent controlling the ability of yeast to support growth using only the oxidative metabolism is the available oxygen.At high oxygen tensions the metabolism is disturbed.

    Notes: An intracellular, thermostable, neutral α-galactosidase (α-D-galactoside galactohydrolase EC 3.2.1.32) was produced in pilot plant quantities from a strain of Bacillus stearothermophilus. The organism was cultured at 50°C in a soluble neutral medium containing water extract of soybean meal (3%) and 0.5% yeast extract. The enzyme biosynthesis was inducible and sensitive to catabolite repression. After autolysis of the cells, the α-galactosidase was selectively and quantitatively complexed from clarified beer directly onto DEAE Sephadex; and enzyme-rich fractions were batchwise eluted with an increasing gradient of NaCl solutions. The eluates were given two consecutive isopropyl alcohol precipitations, and the aqueous solutions of the second precipitate were dialyzed and lyophilized. Final product activity recovery was 72% based on the crude fermentation beer. Best specific activity was 5.2 u/mg protein. Further laboratory purification (DEAE Sephadex and Bio-Gel P200) yielded a product with 14.2 u/mg protein.

    Notes: The death kinetics of a strain of Saccharomyces cerevisiae were studied in an industrial scale spray drier. In solution studies, the death kinetics of yeast was found to be comparable to pathogen destruction. From the studies in drying of yeast a prediction of a 4 log cycle decrease in viable cells of pathogens could be made for normal processing conditions. This should insure the safety of spray-dried foods unless after contamination occurs. It was found that during drying, although the rate of death is high, the activation energy is greatly decreased over that of death in aqueous solution (reduction from 130 kcal/mole to 5 kcal/mole). The reduction in Ea may be attributed to the thermodynamic compensation phenomenon in which the resulting negative entropy of reaction acts to protect the cells through a water-protein interaction. However, the possibility of a change in death mechanism cannot be precluded. Overall, these results suggest the danger in extrapolating death kinetics to high temperature.

It has been found that the Enzyme Rate Equation (Atkinson and Lester), Biotechnol. Bioeng., 16, 1299 (1974), together with an analytical solution which describes the asymptotic conditions associated with a large particle size, provides an adequate estimation of the values resulting from the numerical solution outside the region of the parameter space defined by 0.4 〈 Mg′, M0′ 〈 10.When the dimensionless parameter (B0′/Bg′)(Mg′2/M0′2) is greater than unity the overall rate of reaction is limited principally by the external concentration and when the parameter has a value less than unity, by the external oxygen concentration.The results are generally applicable to enzymes whose kinetics are similar to those of glucose oxidase or for which the equation describing glucose oxidase kinetics provides an adequate curve-fit of experimental data.

    Notes: An apparatus is described for the rapid measurement and recording of methanogenic activity in anaerobic fermentations, and its application is demonstrated in the evaluation of the anaerobic contact process, using pear waste. The method is based on recording the rate of manometer liquid displacement in a Warburgtype vessel by means of optical sensors, appropriate electronic circuitry, and an event marking recorder or time-interval printer. Optimum conditions for measuring methanogenic activity included a pH of 6.7-6.9, a final phosphate buffer concentration of 0.07-015M, and formic and acetic acid contents of over 500 and 200 mg/liter, respectively. In comparisons of fermenter liquid and settled effluent, methanogenic activity can be assumed to be proportional to the number of methane formers present. The apparatus should be generally useful in recording rates of gas production or consumption.

    Notes: Immobilized glucoamylase, invertase, and β-galactosidase were prepared by using N-vinylpyrrolidone monomer (VP) under γ-ray irradiation. The enzyme-VP solutions were gelled by irradiation with 2.9 Mrad and the added enzymes were almost completely entrapped. Activity losses on entrapping were 55% for the VP-glucoamylase gel, and more than 90% in the case of VP-invertase and VP-β-galactosidase gels. No leakage of enzyme from these gels could be detected within 1 hr. The VP-glucoamylase gel was capable of hydrolyzing dextrin (mol wt 10,400) to glucose and the glucose equivalent was equal to that obtain able with native enzyme. The optimum temperature, heat stability, pH activity curve, and pH stability of VP-glucoamylase gel were slightly inferior to those of native enzyme, while Km was a little larger than that of native enzyme.

It was found to be comparable to galactosidases from other E. coli strains in stability, pH and temperature maxima, and activity requirements, but it had a more favorable ratio of activity toward lactose versus synthetic substrates. The galactosidase was immobilized on porous glass beads by three covalent bonding methods. Kinetic data for the free and bound enzymes were determined using natural and synthetic substrates. Activity characteristics of the free and immobilized enzymes were comparable, however, the bound forms were less stable to heat.

    Notes: Polysaccharide was synthesized by Aureobasidium pullulans (or Pullularia pullulans) 2552 in a sucrose medium. The field apparent viscosity of the culture medium from shake flask experiments rose to 24,500 cP and then dropped toward its initial value as the fermentation progressed. The magnitude of the maximum apparent viscosity depended on the initial pH of the fermentation broth. The inoculum age influenced the cultivation period before which the maximum viscosity was reached. Rheograms of the fermentation broths showed a change in viscosity behavior from Newtonian to pseudoplastic, and then toward Newtonian characteristics during the fermentation. The calculated non-Newtonian index was found to be a sensitive factor for the indication of the non-Newtonian behavior. Such behavior could not be detected from rheograms. Viscosity profiles of polysaccharide isolated from various stages of the fermentation showed a change from Newtonian to pseudoplastic behavior depending on the concentration (0-2%) of polysaccharide.

    Notes: An experimental investigation on the rheology of penicillin broths was undertaken in order to obtain more understanding of this important aspect of the fermentation process. The measuring technique consisted in observation of the torque exerted upon a rotating turbine impeller. The experimental data were interpreted in terms of a model which basically is a synthesis of a known relationship for the rheological behavior of printing ink (Casson equation) and some considerations analogous to the rheological description of polymer solutions (excluded volume concept). One of the key variables in the model is a morphology factor, which can be used for a quantitative description of mycelial morphology. The value of the morphology factor can be determined experimentally by a simple viscosity measurement in combination with a mycelial dry weight determination. There are strong indications that the model may be applicable to mycelial broths other than those of penicillin.

    Notes: The utility of an immobilized enzyme depends not only on initial loading but also on operational half-life. The loss of activity of an immobilized enzyme in a column reactor may occur in several ways. It is therefore of interest to determine whether the decay in activity is due to the rupture of any of the enzyme-carrier bonds in the composite. In order to do this, it is first necessary to establish whether the enzyme on the composite is in fact covalently bound or adsorbed. In this report we have shown that there are a number of covalent links per protein molecule. We have also shown that the bond energies are sufficient to prevent shearing of the enzyme itself from the composite under any stresses which may occur in a chromatography column or packed-bed reactor during continuous operation. The effect of the siloxane linkage on composite stability is also discussed.

    Notes: A fiber-optic retroreflective turbidimeter has been developed to automatically and continuously assay the cell concentration in a fermentor by measuring the turbidity of the solution as a function of the light scattered at 180° to the incident light. The output signal is nearly directly proportional to the cell concentration in a fermentor when the sample stream contains from 0 to more than 50 g of cells per liter (wet weight). The device consists of a bifurcated fiber-optics light pipe with its distal end inserted into a flow cell through which the material to be analyzed passes. A light source on one proximal branch of the light pipe illuminates the sample stream; light that is back-scattered from participates in the stream re-enters the light pipe and is returned to a photodetector on the other proximal branch of the light pipe. A signal conditioning system connected to the optical head by a cable provides gain and zero adjustment.

    Notes: Purified enzymes encapsulated in liquid surfactant membranes have been shown to retain their catalytic activity. In general, previous work on encapsulation has been confined to single enzymes. The system has now been extended to encapsulate a bacterial cell-free homogenate. Liquid membrane-encapsulated bacterial cell-free homogenate reduces effectively NO3- to NO2- and other nitrogen compounds of lower oxidation state. This technique of removing nitrates and nitrites may have application in waste-water treatment. Also, it has been shown that encapsulated cell-free homogenate does not leak and there is no absorption of the substrate onto the liquid surfactant membrane surfaces. The reduction in the reaction rates is discussed in terms of solubility of the substrate and the rate of permeation of the substrates through the liquid surfactant membrane.

    Notes: Glucose-limited chemostat cultures of Candida utilis were cultivated at various pH levels (3.0-7.5), temperatures (15-37.5°C), dilution rates (0.06-0.42 hr-1), and with different nitrogen sources (NH4+ and NO3-). The ratio of total nucleic acid to protein increased with increase in dilution rate at constant temperature and decreased with increase in temperature at constant dilution rate. The pattern of these variations is consistent with the hypothesis that the nucleic acid to protein ratio is a function of the ratio of the actual dilution rate to the critical dilution rate corresponding to each one of the cultivation temperatures. This ratio is called “reduced dilution rate.” A basis is proposed on which various microorganisms may be compared with respect to the ratios of cell protein to nucleic acid, RNA, ribosomal RNA, and polysomes.

    Notes: Enzymes can be encapsulated within a semipermeable membrane which allows reactants to enter and the products to diffuse out. The mass transport from the external fluid to the membrane and the combined mass transport and biochemical reaction from the membrane inwards can be modeled with recognized formulations; measurements of the overall reaction rate lead then to estimates of the permeability of the membrane itself.With capsules enclosing catalase, the permeability of collodion membranes to H2O2 is found to be large (〈2 × 10-2cm/sec) in comparison to rates in the other two diffusion zones. For this first-order reaction system, an analytical solution to the transient case of the well-stirred finite bath is found using the Laplace transform. With capsules enclosing urease, the nonlinear Michaelis-Menten kinetics apply to the enzymatic step. The steady-state operation of a column packed with urease microcapsules is analyzed with the aid of numerical computation and the membrane permeability for urea is found to be 10-3cm/sec.

    Notes: The predation of bacteria by protozoan in both continuous and batch cultures was studied using experimental and modeling techniques. The predator organism was the ciliate, Tetrahymena pyriformis. The bacterium, Aerobacter aerogenes, served as the prey. Several batch growth responses were observed each initiated at a different nutrient level. Continuous cultures were conducted over a range of dilution rates. The models studied were partially successful in simulating the empirical data. Deviations between the model and the actual population behavior are discussed and possible explanations for the differences proposed.

    Notes: The kinetics of C. tropicalis growth were investigated with pure n-hexadecane as dispersed phase substrate. Two distinct growth phases were found: In the first phase, exponential growth was independent of stirrer speed. The onset of the second phase, one of linear growth, was determined by stirrer speed. By the use of two different fermenter types, it was shown that the drop size of the dispersed phase was not primarily responsible for the observed kinetics. It was considered that the formation of biological flocs determined the observed growth pattern. This was substantiated by the results of continuous cultures in the different fermenter types, with various substrate concentrations.

    Notes: A new correlation is given for the prediction of the volumetric coefficient for mass transfer (KLa) in stirred tanks from dispersed gas bubbles to basal salt solutions of ionic strengths representative of fermentation media. The correlation includes the effects of both the operating parameters (agitation power per unit volume and gas superficial velocity) and the physicochemical properties of the system: interfacial tension, viscosity, density, diffusion, coefficient and, in particular, ionic strength. The effect of the latter was found to be most significant in the Newtonian systems of water-like viscosity investigated; no previous correlations have included the effect of ionic strength. KLa values were determined by using a dissolved oxygen probe to monitor the steady-state oxygen tension in continuous flow experiments, and/or the rate of change of oxygen tension in unsteady-state semibatch experiments. In the latter cases, results were computed by a nonlinear, least squares computer program which fitted the experimental data to a model of probe transient response characteristics. The general applicability of the model and the computational procedure was verified by comparing the results to those obtained with the same electrolyte solution in the steady-state mode. The experiments were run over a wide range of agitation power inputs, including those typical of both soluble- and insoluble-substrate fermentations. The correlation appears to be valid for both oxygen mass transfer with and without homogeneous chemical reaction in the liquid phase; in the former case, for example, sulfite oxidation, knowledge of the chemical reaction enhancement factor is required. In addition to predicting oxygen transfer capabilities, the correlation may be used for other sparingly soluble gases of interest in fermentation systems, such as methane, hydrogen, and carbon dioxide.

    Notes: Barely β-amylase (α-1,4-glucan maltohydrolase, EC 3.2.1.2) has been immobilized by covalent fixation to amino derivatives of epichlorohydrin crosslinked Sepharose mediated by cyclohexyl isocyanide and acetaldehyde. The enzyme conjugates contain up to 35% of the total activity of the β-amylase added to the coupling mixture. The profiles of activity versus pH and ionic strength are essentially the same for free and immobilized β-amylase, whereas the resistance to inactivation during storage and use is considerably enhanced by immobilization. Columns with immobilized β-amylase have been used for continuous degradation of starch. At 45°C, half of the initial activity remains after seven weeks, and the corresponding figure at 23°C is 85 percent.

    Notes: 5′Xanthylic acid was efficiently converted to 5′guanine nucleotides (5′GMP, 5′GDP, and 5′GTP) without being degraded to guanine via 5′GMP by decoyinine resistant mutants of strain KY 13315 which had been isolated from Brevibacterium ammoniagenes and was practically devoid of 5′nucleotide degrading activity. The concentration of phosphate in the medium showed a profound effect on the ratio of the accumulated 5′guanine nucleotides, making it possible to direct the fermentation towards 5′GMP or 5′GTP. A direct accumulation of 5′guanine nucleotides from carbohydrate was possible by mixed cultivation of a 5′XMP accumulating strain and a 5′XMP converting mutant. A maximum concentration of 9.67 mg of 5′guanine nucleotides per ml was obtained directly from glucose in such a mixed culture.

    Notes: Of 24 different polymer and surfactant materials examined, a carboxypolymethylene (“Carbopol”) was found to cause enhancement of respiration rates in an Aspergillus niger culture by as much as 200%. Enhancement of other fermentation parameters, such as cellular growth and amylase production, was also observed.The enhancement effects of Carbopol were examined with clusters of spores and mold pellets. In the first case, it appears that the ionized carboxyl groups of Carbopol induced electrostatic repulsion among the spores thus initiating pulp growth with increased interfacial area of contact between the mold and the nutrient medium. In the second case, the Carbopol additive formed a thin film attached to the surface of the pellets which seemed to be responsible for an increased rate of potassium transport and, hence, fermentation yields. Additive utilization as substrate and physiological changes in the culture were not observed in these cases.It was also found that the probability of pellet formation, the size of pellets formed, and the number of spores per pellet can be correlated to the energy input to the fermentation system.

    Notes: The steady state, nonlinear diffusion equations which describe reactions in constrained enzyme solutions are of great interest in many biological and engineering applications. As in other types of nonlinear differential equations, exact analytical solutions do not exist except in some simplified cases. In this paper, a general procedure is presented for solving numerically for the substrate concentration profile and effectiveness factor utilizing the transformation method suggested by Na and Na. Design correlations for enzyme solutions constrained within spherical membranes are included. The use of a unique definition of the Thiele Modulus in these charts permits the clear illustration of the effects of substrate concentration and external mass transfer resistances on the overall effectiveness factor for the catalyst particle.

    Notes: Studies are reviewed on the composition of hemolymph from Japanese beetle larvae, Popillia japonica. This research was conducted to determine the chemical environment required for sporulation of the milky disease organism, Bacillus popilliae. Amino acids, proteins, organic acids, lipids, carbohydrates, and oxygen content were determined in hemolymph from healthy and diseased larvae. Oxygen content of hemolymph was measured by microelectrodes implanted in the hemocoel of living larvae. Vegetative growth of the pathogen reduces the dissolved oxygen; however, as sporulation occurs, oxygen concentration approaches normal values. Trehalose, the major hemolymph sugar, is reduced by ca. 50% during the course of milky disease. Neutral lipids, phospholipids, and hydrocarbons decrease markedly with infection. Both branched-chain and normal alkanes occur in hemolymph. Milky disease causes an increase in concentration of pyruvic, malic, glycolic, tartaric, and glyoxylic acids. The major hemolymph protein, a lipoglycoprotein, has a mol. wt. of ca. 500,000. Reduced concentration of this protein is accompanied by the appearance of a smaller protein. Amino acids that increase significantly as a result of infection are: Glutamic acid, aspartic acid, β-alanine, phenylalanine, threonine, lysine, and serine. Hemolymph contains few peptides of low molecular weight.

    Notes: Cellulase production by strains of Myrothecium verrucaria, Stachybotrys atra and Trichoderma viride was examined. Myrothecium verrucaria was found to give the greatest yields. A variety of media were examined as potential substrates for the industrial production of cellulase. The salts content of the medium was varied and was found to affect cellulase production. Glucose, carboxymethylcellulose (CMC), filter paper and three industrial wastes were examined as possible cellulase inducers. Filter paper was found to be the most effective, followed by sugar cane bagasse and CMC.

    Notes: Quantitative studies are reported of external film and internal pore diffusion effects for glucose oxidase immobilized on porous glass in a continuous, tubular, packed bed reactor. The McCune and Wilhelm model for external film diffusion is shown to describe these data over a particle Reynolds number range from 0.2 to 25. The Thiele analysis provides a good description of internal pore diffusion resistances. It appears that immobilized enzymes free of internal diffusional restrictions generally require the use of supports with pore sizes larger than those currently available in porous glass.

    Notes: A double polarographic device is described which permits regulation of dissolved oxygen concentration at any preset pO2 in suspensions or cultures of photosynthetic algae. It also simultaneously permits measurements of the rate of oxygen evolution during photosynthesis. The apparatus is designed to operate in closed vessels in the absence of a gas phase, and may be used at increased pressures tested up to 500 atmospheres. Regulation of oxygen levels was maintained constant at various preset concentrations equivalent to atmospheric pO2 values ranging between 0.02 and 0.21 at 25°C.

    Notes: A substrain of Bacillus cereus 569/H produced under controlled fermentation conditions in a pilot plant fermentor phospholipase-C. A partially purified preparation showed good storage stability as a lyophylized powder and in frozen solutions. The preparation contained very small amounts of phosphomonoesterase and proteolytic activities and essentially no ribonuclease activity. The level of hemolytic activity of the preparation was much lower than that of a commercial preparation of phospholipase-C from Clostridium. Treatment of sarcoplasmic reticulum membrane with phospholipase-C from B. cereus and from Clostridium showed that the B. cereus enzyme caused hydrolysis of 96% of the membrane phospholipids whereas the enzyme from Clostridium could hydrolyze only 80% of the phospholipids.

    Notes: A mixed culture derived from soil and activated sludge organisms was used to degrade phenol which was inhibitory to microorganisms at higher concentrations. The purpose of the experiments was to determine the kinetic parameters governing growth of the organisms by measuring growth rates in batch culture. To maintain a constant inoculum for the experiments inoculum was taken from a continuously operating continuous culture. Two populations were studied corresponding to two separate residence times in the continuous culture apparatus. One contained predominantly filamentous organisms, the other nonfilamentous. Five kinetic models were applied to the data and the best kinetic parameters for each model were determined by nonlinear least squares techniques. The models were then evaluated for best relative fit to the data. No significant differences were found between the models on the basis of fit and so a choice was made on the grounds of simplicity. A model proposed by Haldane was chosen as the best. No function however gave a satisfactory fit at the highest growth rates obtained. This experimental maximum in the plot of growth rate against substrate concentration was very sharp.

    Notes: It is shown that two steady states exist in certain regions of operation of a 2-liter continuous stirred tank biological reactor. Transition was made from one steady state to another by applying shock loads of either phenol substrate which is inhibitory to the culture at high concentrations or by adding large additional amounts of concentrated organisms. The existence of the multiple steady states is ascribed to the existence of wall growth, and their position is determined by the amount of wall growth. Transient behavior of the system did not follow the predictions of the simple wall growth model but the culture appeared to undergo a lag period immediately after applying the shock load to the system. It is concluded that the stability of a continuous culture utilizing an inhibitory substrate is improved by increasing the degree of wall growth and decreasing the substrate feed concentration. It is also concluded that small scale experiments can usually not be interpreted correctly unless the effect of wall growth is taken into account.

    Notes: The response of aerobically growing heterogeneous microbial populations of sewage origin to step increases and decreases in pH were studied in both once-through and cell recycle systems. The pH range studied was 2.7 to 8.0. All studies were conducted at a dilution rate of 0.125 hr-1, and all shocks were administered from a base or preshock pH level of 6.4 to 6.7. In each experiment, the preshock or initial “steady state” was assessed, the pH of the feed changed, and the resulting transient behavior of the system examined until attainment of the new or final “steady state” was approached. The major objectives of the work were to characterize the nature of the response with respect to biomass and effluent substrate concentrations, types of microbial populations present and chemical composition of the biomass, and to obtain guidelines as to allowable change in pH in waste streams. It was found in once-through systems that substrate removal efficiency recovered from pH levels as low as 3.0 after rather long periods of transient leakage of substrate. Cell recycle attenuated the severity of substrate leakage. In all cases of severe acid shock, the microbial population changed from predominantly bacterial-protozoan to one consisting predominantly of filamentous fungi. Changes in chemical composition of the sludge (protein and carbohydrate content) were consistent with the population changes. Based upon the results, it can be conservatively estimated that changes in pH of no more than one unit from the neutral preshock range can be tolerated without possible disruption of biochemical efficiency of substrate removal.

    Notes: Wheat germ agglutinin protein, which is able to agglutinate tumor cells better than normal cells, was covalently bound to polyacrylamide gel beads. The specific binding activity of the protein was preserved on these beads and was expressed heterogeneously by the binding of mouse leukemia cells (L1210) to the protein coupled gels. The selective activity of the immobilized protein was maximal when the number of sites available to covalently couple the protein was lowest. The application of this observation to the general field of covalent immobilization of proteins and enzymes may be of considerable utility.

    Notes: Collagen was used as carrier for the immobilization of invertase, lysozyme, urease, glucose oxidase, penicillin amidase, and glucose isomerase. Immobilization was accomplished by either impregnation of a preswollen collagen membrane with enzyme solution or electrocodeposition of collagen and enzyme from a collagen dispersion containing dissolved enzyme. The collagen-enzyme complexes prepared are in membrane form.Membranous collagen-enzyme complexes were used to construct biocatalytic reactors such as the capillaric coil modular reactor. Such biocatalytic reactors were used in a recirculation system for the conversion of substrates. The biocatalytic reactors showed initial decreases of activity to stable limits which are maintained over a large number of reactor volume replacements. The stable limits correspond to approximately 35% of the initial activities for lysozyme and invertase, 25% for urease, 15% for glucose oxidase.The mechanism of complex formation between collagen and enzyme involves multiple salt linkages, hydrogen bonds, and van der Waals interactions. This protein-protein interaction which leads to stable complexes by both impregnation and electrocodeposition processes is unique among the enzyme immobilization methods currently available.

    Notes: Kinetic studies on the growth of Aspergillus strains as well as constitutive and inductive polygalacturonase formation have been made in bath fermentations, without pH control. Equations describing growth and product formation of the four-stage growth of microorganisms were applied for the first time to diauxic growth and enzyme formation of Aspergillus strains. Diauxic growth of the cultures has been found in both sucrose- and peetin-containing media. Enzyme concentration in the second transient phase, calculated by the use of new equations, proved to be negligible. In the exponential and declining phases, the calculated values of mycelial and enzyme concentrations were in good agreement with the values observed. Types of classification of the product formation of Luedeking and Piret refer to the second cycle of diauxie. The types of product formation described by Kono and Asai and Luedeking and Piret are mostly in good agreement with each other. The greatest difference has been found in the case of inductive endo-polygalacturonase formation, where the final enzyme formation could not be plotted because of the decrease (autolysis) in mycelial weight.

    Notes: DL-Norleucine, a nonsulfur analogue of methionine was found to markedly stimulate synthesis of cephalosporin C by Cephalosporium acremonium strain CW19 in three different chemically defined media. Methionine, but not norleucine, stimulated cephalosporin C biosynthesis in a crude medium. The lack of stimulation by norleucine in complex medium was shown to be due to lack of uptake of this amino acid by mycelia growing in such a medium. In defined media containing a suboptimal methionine concentration, norleucine stimulated antibiotic production up to the level reached by optimal methionine. At an optimal dose of methionine, norleucine elicited no further increase in cephalosporin C production, indicating that these two amino acids act by the same mechanism. The data strongly indicate that stimulation by methionine is not a function of its ability to donate sulfur for antibiotic formation. Methionine was found to neither repress nor inhibit cysteine metabolism.

    Notes: It is sometimes easier and cheaper to extract a drug's target sites from cells than it is to purify, concentrate or manufacture the drug itself. This is especially true in the case of DNA and some of the exotic antibiotics that react with it. Phleomycin, an inhibitor of replication, was considered as a specific case in point. Biologically active components of this antibiotic mixture bind nondestructively to DNA, in vitro and in vivo, and thereafter may be processed by simple, economical methods usually reserved to macromolecules of much greater size.

    Notes: A simplified model of cell metabolism, consisting of a series of linked reversible enzymatic reactions dependent on the concentration of a single external substrate has been developed. The general mathematical solution for this system of reactions is presented. This general solution confirms the concept of a rate-limiting step, or “master reaction”, in biological systems as first proposed by Blackman. The maximum rate of such a process is determined by, and equal to, the maximum rate of the slowest forward reaction in the series.Of practical interest in modeling the growth rate of cells are three cases developed from the general model. The simplest special case results in the Monod equation when the maximum forward rate of one enzymatic reaction in the cell is much less than the maximum forward rate of any other enzymatic reactions.More realistic is the case where the maximum forward rates of more than one enzymatic reaction are slow.     Notes: The known species of yeasts produce at lest 180 different polysaccharides having mannose as a major constituent. The type of mannan formed is characteristic of the yeast species, and can be used as an aid in identification and classification. The immunological characteristics of yeasts depend largely upon the nature of the mannans. Some of the yeast mannans can be produced in relatively high yields, and have been suggested as replacements for plant and bacterial gums as thickeners, dispersing agents, and similar applications. However, more significant uses, based on the finer differences in their chemical structures and the specific reactions they undergo, may result from a closer study of these polymers.

    Notes: The thermal stability of glucose oxidase in solution was studied as a function of time and temperature between 37-60°C. As expected, the rate of thermal inactivation increased with temperature and at 60°C more than 80% of the enzyme's activity was lost after 0.5 hr incubation. Similar stability measurements on enzyme solutions containing water soluble synthetic polymers showed that several of the polymers significantly enhanced the thermal stability of glucose oxidase. Copolymers of vinyl acetate with either vinyl pyrrolidone or vinyl alcohol were found to be particularly effective. The molecular weight of the added polymers was found to be unimportant in the stabilization process but both polymer concentration and compositions were shown to be critical parameters.

    Notes: Carrier-bound immobilized α-amylases and glucoamylase have been obtained by photopolymerizing aqueous solutions containing acrylamide, a crosslinking monomer, an enzyme-reactive monomer, an oxygen scavenger, and amylase, and allowing the amylase to react with the entrapping enzyme-reactive gel matrix evolved. Partial starch hydrolyzates (28 DE corn starch syrup) have been continuously converted to 90-94% dextrose syrups in columns packed with immobilized amylases. The enzymatic heat stability of immobilized glucoamylase gels is discussed.

    Notes: Lysozyme immobilized on polyacrylamide beads or cellulose fibers is found to retain activity for hydrolysis of the cell walls of Micrococcus lysodeikticus. The immobilization on cellulose is somewhat reversible; the polyacrylamide immobilized lysozyme does not release any enzyme upon washing as evidenced by UV and lytic activity tests. The specific catalytic activity of the lysozyme-polyacrylamide system is found to decline as the density of derivatized surface groups is increased; a model of protein deactivation due to excess surface coupling is presented as a possible rationale for such specific activity variations.

    Notes: Experimental evidence is presented which indicates that both chemical and autoflocculation of microorganisms occur by the same mechanism. Basically, long chain polymeric species attach themselves between the microbial particles and agglomerate them into flocculant particles of sufficient magnitude to subside from suspension under quiescent conditions. Properties of the polymers which are produced during autoflocculation are investigated and these polymers are shown to be capable of causing the agglutination of inorganic colloids such as alumina. It is hypothesized that these biologically generated flocculating polymers could belong to a class of compounds known as transport enzymes. The experimental data qualitatively agrees with this hypothesis.

    Notes: Lactate dehydrogenase (LDH) and pyruvate kinase (PK), immobilized on solid glass beads by diazotization, were used in packed bed reactors to analyze for both pyruvate (PYR) and phosphoenolpyruvate (PEP) through the disappearance of β-nicotinamide adenine dinucleotide (NADH) monitored spectrophotometrically at 340 nm. Packed bed reactors containing PK and/or LDH were also capable of monitoring continuously varying concentrations of adenosine-5′-diphosphate (ADP), PEP, and PYR. The immobilized enzymes (∼40 μg/g glass) retained about 4% of the activity of the soluble enzymes. Preparations of immobilized LDH and PK exhibited enhanced stability when maintained in the presence of β-mercaptoethanol and NADH or EDTA, respectively, and were shown to regain 75% of the original activity after four months storage at 4°C.

    Notes: An investigation was made on the factors influencing the acetic acid concentration in the culture liquid of the aerobic fermentations where acetie acid was used as a carbon source. The acetic acid concentration in the culture liquid changed in proportion to the amount of unutilizable acid or base supplied. This was explained by the principle of conservation of electroneutrality.Another factor affecting the acetic acid concentration in the culture liquid was bicarbonate ions which were formed by the dissolution and dissociation of carbon dioxide in the gas phase of the fermentor. The increment in bicarbonate ion concentration was equal to the decrement in the acetie acid concentration in the culture liquid.
