    Notes: Abstract. Mcl-1, a member of the Bcl-2 family, has been identified as an inhibitor of apoptosis induced by anticancer agents and radiation in myeloblastic leukemia cells. The molecular mechanism underlying this phenomenon, however, is not yet understood. In the present study, we report that hyperpolarization of the membrane potential is required for prevention of mcl-1 mediated cell death in murine myeloblastic FDC-P1 cells. In cells transfected with mcl-1, the membrane potential, measured by the whole-cell patch clamp, was hyperpolarized more than −30 mV compared with control cells. The membrane potential was repolarized by increased extracellular K+ concentration (56 mV per 10-fold change in K+ concentration). Using the cell-attached patch-clamp technique, K+ channel activity was 1.7 times higher in mcl-1 transfected cells (NP o = 22.7 ± 3.3%) than control cells (NP o = 13.2 ± 1.9%). Viabilities of control and mcl-1 transfected cells after treatment with the cytotoxin etoposide (20 μg/ml), were 37.9 ± 3.9% and 78.2 ± 2.0%, respectively. Suppression of K+ channel activity by 4-aminopyridine (4-AP) before etoposide treatment significantly reduced the viability of mcl-1 transfected cells to 49.0 ± 4.6%. These results indicate that as part of the prevention of cell death, mcl-1 causes a hyperpolarization of membrane potential through activation of K+ channel activity.

    Description / Table of Contents: Zusammenfassung Die Nichtgleichgewichts-Austauschprozesse, die in einem Gas-Partikel-Gemisch hinter einer Stoßwelle ablaufen, werden in dieser Arbeit untersucht. Sie werden sowohl mit den Erhaltungsgleichungen für zwei interaktive Kontinua als auch mit den vereinfachten Gleichungen für Einzelpartikel in einem Gaskontinuum beschrieben. Der besondere Akzent liegt auf den Transportvorgängen sehr kleiner Partikel, die im Übergangsbereich zwischen Kontinuumsbedingungen und freier Molekülströmung stattfinden. Sie werden durch einfache Knudsenzahlansätze berücksichtigt. Beispiele zeigen die Geschwindigkeits-, Temperatur- und Partikelgrößenrelaxation hinter Stoß-wellen in Argon mit DOP-Tröpfchen und mit festen NaCl-Salzpartikeln.
    Notes: Abstract The nonequilibrium transfer processes, which take place in a gas-particle mixture behind shock waves, are investigated in this study. They will be described both by the conservation equations of two interactiv continua and by the simplified equations of a single particle in a gas continuum. The special accent is on exchange processes of very small particles, which take place in the transition regime between continuum and free molecular conditions. They are taken into account by simple Knudsen number corrections. Examples show the velocity, the temperature and the particle size relaxation behind shock waves in argon containing DOP droplets or solid NaCl particles.

    Notes: Abstract We study the influence of a variable neuronal threshold on fixed points and convergence rates of an associative neural network in the presence of noise. We allow a random distribution in the activity levels of the patterns stored, and a modification to the standard Hebbian learning rule is proposed for this purpose. There is a threshold at which the retrieval ability, including the average final overlap and the convergence rate, is optimized for patterns with a particular activity level at a given noise level. This type of selective attention to one class of patterns with a certain activity level may be obtained at the cost of reducing the retrieval ability of the network for patterns with different activity levels. The effects of a constant threshold independent of noise, time, and pattern are discussed. For high-(low-) activity patterns, the average final overlap is shown to be increased at high noise levels and decreased at low noise levels by a negative (positive) constant threshold, whereas a positive (negative) threshold always reduces the final average overlap. When the magnitude of the constant threshold exceeds a critical value, there is no retrieval. Rates of convergence towards the stored pattern with negative (positive) thresholds are greater than those with positive (negative) thresholds. These results are related to (de)sensitization and anesthesia. For certain threshold values and patterns with certain activity levels, hysteresis appears in the plot of the average final overlap versus the noise level, even for first order interactions. We make the analogy between the pattern-dependent neuronal threshold proposed in the present paper and the “task-related” modulation in neuronal excitability determined by cognitive factors, such as the attentional state of a higher animal. A constant threshold is associated with overall changes in neuronal excitability caused, e.g., by various drugs and physical injuries. Neurophysiological evidence of a dynamically variable neuronal threshold, such as accommodation and potentiation, is presented.

    Notes: Abstract 6Li+ (150 keV) was implanted into thin polypropylene foils at fluences of 1 x 1013 to 1 x 1014 cm−2. Subsequent neutron depth profiling measurements of the Li distributions revealed considerable deviations from the expected ballistic range profiles. This Li redistribution was simulated by a numerical computer calculation. The best fit between measurements and simulations was obtained by assuming that (i) Li redistributes immediately after its ballistic slowing-down, (ii) the Li mobility is enhanced in the radiation-damaged polymer region, the local diffusion enhancement being controlled by the target's electronic damage, (iii) mobile Li is readily trapped at radiation-induced defects, their density being proportional to the target's electronic damage, (iv) these traps are saturable ones, and (v) Li migration is not restricted to the ion track region, but proceeds also through the neighboring unirradiated bulk, though with slower speed.

    Notes: Abstract.  Examinations were performed on GeV-ion irradiated plexiglas. The radiochemical reaction products are studied by FTIR and UV spectrometry as a function of their depth of origin, and some of the possible radiochemical reaction mechanisms are examined in detail. The radiochemistry of commercial plexiglas is found to differ somewhat from the one of pure PMMA. No hint for ion-induced buckminsterfullerene formation could be found.

    Notes: Abstract Laser ablation of solid GaAs samples has been studied using one tunable pulsed dye laser. At relatively low laser power, enhancements of up to several hundred times have been observed in the yield of resonantly ionised Ga using laser wavelengths corresponding to the atomic transition 42 P 1/2-42 D 3/2. The influences of laser power and target geometry, on the ion yield and spectral profile, are discussed. It is argued that the resonant excitation and ionisation processes occur in the gas phase of the atoms ablated from the sample surface, and the observed asymmetric spectral profile results from laser-induced collisional processes, e.g., optical collisions, under conditions of relatively high atomic density in the interaction region. Potential applications are foreseen for resonant laser ablation in trace analysis.

    Notes: Abstract  The robust behavior, the degree of response linearity, and the aspect of contrast gain control in visual cortical simple cells are (amongst others) the result of the interplay between excitatory and inhibitory afferent and intracortical connections. The goal of this study was to suggest a simple intracortical connection pattern, which could also play a role in other cortical substructures, in order to generically obtain these desired effects within large physiological parameter ranges. To this end we explored the degree of linearity of spatial summation in visual simple cells experimentally and in different models based on half-wave rectifying cells (’’push-pull models’’). Visual cortical push-pull connection schemes originated from antagonistic motor-control models. Thus, this model class is widely applicable but normally requires a rather specific design. On the other hand we showed that a more generic version of a push-pull model, the so-called cascaded inhibitory intracortical connection scheme, which we implemented in a biologically realistic simulation, naturally explains much of the experimental data. We investigated the influence of the afferent and intracortical connection structure on the measured linearity of spatial summation in simple cells. The analysis made use of the relative modulation measure, which is easy to apply but is limited to moving sinusoidal grating stimuli. We introduced two basic push-pull models, where the order of threshold nonlinearity and linear summation is reversed. Very little difference is observed with the relative modulation measure for these models. Alterative models, like half-wave squaring models, were also briefly discussed. Of all model parameters, the ratio of excitation to inhibition in the simple cell exerts the most crucial influence on the relative modulation. Linearity deteriorates as soon as excitatory and inhibitory inputs are imbalanced and the relative modulation drops. This prediction was tested experimentally by extracellular recordings from cat area 17 simple cells and we found that about 62% showed a significant deviation from linear behavior. The problem that individual basic push-pull models are hard to distinguish experimentally led us to suggest a different solution. In order to generically account for the observed behavior (e.g., imbalance of excitation versus inhibition), we suggested a rather generic version of a push-pull model where it no longer mattered about (the hard-to-distinguish) fine differences in connectivity. Thus, we introduced a new class of biophysically realistic models (’’cascaded inhibition’’). This model class requires very little connection specificity and is therefore highly robust against parameter variations. Up to 25 cells are connected to each target cell. Thereby a highly interconnected network is generated, which also leads to disinhibition at some parts of an individual receptive field. We showed that the performance of these models simulates the degree of linearity and its variability in recal simple cells with comparatively high accuracy. This behavior can be explained by the self-regulating properties of a cascaded inhibitory connection scheme by which the balance between excitation and inhibition at a given cell is improved by the joint network effects. The virtues and the generic design of this connection pattern, therefore, allow to speculate that it is used also in other parts of the cortex.
